1.Pingyangmycin-regulated expressions of adhesion molecules in human venous malformation endothelial cells.

Pingyangmycin (bleomycin A5 hydrochloride, PYM) is one of the anti-neoplastic agents which have been commonly used to treat venous malformations. However, the underlying mechanism by which PYM treats venous malformations remains poorly understood. It was reported that venous endothelial cells could recruit neutrophils via adhesion molecules (E-selectin, ICAM-1, ICAM-3, VCAM-1) during the acute/chronic inflammation and subsequent histological fibrosis after sclerotherapy with PYM. This study explored if the expression of E-selectin, ICAM-1, ICAM-3 and VCAM-1 in human venous malformation endothelial cells could be affected by PYM. HVMECs were cultured from human venous malformation tissue. Expressions of E-selectin, ICAM-1, ICAM-3 and VCAM-1 on HVMECs in response to PYM were analyzed by cell ELISA. The relative levels of mRNA expression in the cells were semi-quantified. The results showed that PYM up-regulated the expressions of E-selectin, ICAM-3, VCAM-1 and ICAM-1 in both time- and concentration-dependent manner. Our findings suggested that PYM could induce the expression of adhesion molecules in HVMECs, which might be a possible mechanism by which sclerotherapy by intralesional injection of PYM treats venous malformations.
2.Therapeutic Observation of Fire-needle plus Round-sharp-needle Acupuncture for Primary Trigeminal Neuralgia

Objective To observe the clinical efficacy of fire-needle plus round-sharp-needle acupuncture in treating primary trigeminal neuralgia.Method Sixty-two patients with primary trigeminal neuralgia were randomized into a treatment group and a control group, 31 cases in each group. The treatment group was intervened by fire-needle therapy at the trigger points followed by deep needling with round-sharp needle at Xiaguan (ST7), and the distal points Hegu (LI4) and Neiting (ST44) were also treated. The same acupoints were selected in the control group but treated with ordinary needling method. The clinical efficacies were evaluated after 2 treatment courses.Result The total effective rate was 93.5%(29/31) in the treatment group versus 77.4% (24/31) in the control group, and the difference was statistically significant (P<0.05).Conclusion Fire-needle plus round-sharp-needle acupuncture is an effective approach in treating primary trigeminal neuralgia.
3.The correlationship of plasma lipoprotein-associated phospholipase A2, hypersensitive C-reactive protein and D-dimer in coronary artery disease patients

Objective To study the correlationship among plasma lipoprotein associated phospholipase A2 (Lp-PLA2),hypersensitive C reactive protein (hs-CRP) and D-dimer levels of coronary heart disease (CHD) patients.To evaluate their relationship and prognostic values.Methods Case control study.This study enrolled 150 CHD patients,130 age and gender compared health control.The clinical data were recorded and plasma Lp-PLA2,hs-CRP and D-dimer were test.Lp-PLA2 was test by enzyme kinetics method,hs-CRP and D-dimer were test by immunonephelometry method.The blood were also test low density lipoprotein cholesterol and myocardial injury markers such as troponin I,myoglobin,creatin kinase MB.The numbers of lesion coronary arteries were confirmed by percutaneous coronary intervention operation.Student t-test and mann-whitney U test were used to compare two groups datas,analysis of variance was used to compare more than three groups datas,spearman test was used for correlationship analysis between groups.Results The Lp-PLA2,hs-CRP and Ddimer of CHD group were (43.62 ±13.23) g/L,5.09 mg/L(95% CI:0.56-74.15),173.0 mg/L(95% CI:103.0-966.5),respectively.They were higher than healthy control group whose values were (21.49 ± 7.63)g/L,2.61 mg/L(95 % CI:0.37-45.12),68.0 mg/L (95% CI:48.0-540.6),respectively.The difference were statistical significant (t =2.69,U =67.52,U =37.84,P < 0.001).After correlation with clinical data,the Lp-PLA2 was correlated to LDL(r =0.487,P < 0.001) and the numbers of coronary artery lesion (r =0.260,P =0.018).D-dimer was correlated to hs-CRP(r =0.263,P =0.011),it was also correlated to NYHA heart function level (F =23.78,P =0.001).Conclusions Lp-PLA2 is important in lipid metabolize,especially in LDL metabolize.Lp-PLAz can be used to predict severity of coronary artery lesion.Hs-CRP correlated to D-dimer level and cardiac injury markers.It stated link between inflammation and thrombosis.D-dimer is increasing with NYHA level.Lp-PLA2,hs-CRP and D-dimer are correlated to coronary artery lesions,they are good prognostic indices for coronary heart disease patients.
4.Impacts of electroacupuncture on constipation of intestine and lung syndromes and its correlative study.

OBJECTIVETo observe the improvements in constipation differentiated as intestine system syndrome and lung system syndrome treated with electroacupuncture (EA) and explore their correlation.
METHODSSeventy cases of severe functional constipation were randomized into an EA group and a pseudo-EA group, 35 cases in each one. In the EA group, the needles were inserted deeply at Tianshu (ST 25) and Fujie (SP 14) on bilateral sides, and directly went to parietal peritoneum. EA was attached to the needles, dense-disperse wave, 2 Hz/15 Hz, and 0.1 to 1.0 mA. The perpendicular insertion was done at Shangjuxu (ST 37), 25 mm in depth. After qi arrival, the needle was lifted, thrusted and rotated once every 10 min, for 3 times totally. The needles were retained for 30 min. In the pseudo-EA group, the pseudo-points lateral to Tianshu (ST 25), Fujie (SP 14) and Shangjuxu (ST 37) on bilateral sides were punctured shallowly. The electric stimulation was pretended to connect but with the electric wire cutting off. The needles were retained for 30 min. The treatment was given 5 times weekly in the first two weeks and 3 times weekly in the later 6 weeks. Totally, 28 treatments were required. TCM intestine and lung syndrome scale was used for evaluation. The changes in TCM syndromes were observed before and after treatment in the patients of the two groups.
RESULTSThe total score of intestine and lung syndrome and the score of individual syndrome were all reduced after treatment as compared with those before treatment in the two groups (both P<0.01). The improvements of the EA group in the total score of intestine system syndrome, the scores of large. intestine syndrome and stomach syndrome, the total score of lung system syndrome and the score of lung dysfunction in dispersing and descending syndrome were superior to those of the pseudo-EA group (P<0.01, P<0.05). The differences in the scores of lung qi deficiency syndrome and throat syndrome were not significant between the two groups (all P>0.05). Simultaneously, the very strong positive correlation (P<0.01) and positive linear correlation (P<0.01) were presented in the total score of intestine and lung syndrome of the two groups.
CONCLUSIONEA obviously improves in intestine system syndrome (including large intestine syndrome and stomach syndrome), as well as lung system syndrome (lung dysfunction in dispersing and descending) in the treatment of constipation. But the improvements are not apparent in the treatment of lung qi deficiency and throat disorder. Additionally, the obvious correlation is displayed in the improvements in intestine and lung syndrome.

5.Expression and clinical significance of serine-threonine kinase/mammalian target of rapamycin/p70 S6K signal path- way in oral squamous cell carcinoma.

OBJECTIVETo investigate the expressions of serine-threonine kinase (Akt)/mammalian target of rapamycin (mTOR)/p70 S6K in oral squamous cell carcinoma (OSCC) and provide references for early diagnosis and prognosis evalua- tion of OSCC.
METHODSA total of 51 cases of OSCC, 10 cases of paracancerous mucosa, and 10 cases of normal oral mucosa were collected. The expressions of Akt/mTOR/p70 S6K in these cases were detected using the SP method of immunohisto- chemistry. The correlation between their expressions in OSCC was also analyzed.
RESULTSThe positive expressions ofp-Akt, p-mTOR, and p70 S6K in OSCC were significantly higher than those in normal oral mucosa and paracancerous mucosa. The expressions of p-Akt, p-mTOR, and p70 S6K in OSCC were not correlated with age, gender, and clinical stage; by comparison, these expressions were correlated with lymph node metastasis and pathological grade. Strong positive correlations were also observed between the expressions ofp-Akt, p-mTOR, and p70 S6K in OSCC.
CONCLUSIONAkt/mTOR/p70 S6K signaling molecules exhibit active expressions in OSCC and may be implicated in the occurrence and development of OSCC.

6.Antibiotic Resistance of Staphylococcus aureus Isolates

OBJECTIVE To investigate the antibiotic resistance of Staphylococcus aureus isolates from the clinical samples from 2005 to 2007.METHODS The susceptibility tests and D-test were performed by the disk diffusion method and their results were analyzed by the software WHONET 5.4.RESULTS The rate of MRSA isolates accounted for 64.1%.The resistance rate to 12 kinds of the antimicrobial agents,such as penicillins,cephalosporins and carbapenems was 100%.The MRSA and MSSA were susceptible to linezolid and vancomycin.No isolates were resistant to vancomycin in our hospital;the positive rate of D-test was 78.7%.CONCLUSIONS MSSA is still sensible to most of the antimicrobial agents,but MRSA shows multidrug resistance.The D-test is of great clinical importance.
7.Analysis of association between PTEN gene single nucleotide polymorphism and stomach neoplasms

8.Impact of nano-structured porous polycaprolactone combined with bone marrow stromal cells on CTGF expression in rabbit impaired articular cartilage

Objective To study nano-structured porous polycaprolactone combined with bone marrow stromal cells on CTGF expression in rabbit impaired cartilage. Methods 50 rabbits were randomly divided into normal group, model group, NSP-PCL group, BMSCs group, or NSP-PCL + BMSCs group. A model of rabbit impaired cartilage was surgically established. Then NSP-PCL, BMSCs, and NSP-PCL + BMSCs were separately administered to the latter three groups once a week from the 2nd week to the 5th week after the procedure. The impaired cartilage tissues were collected 24 h after the last administration. The cartilage tissues were pathologically examined by H & E staining. Number of surviving BMSCs was detected by Hoeehst33342 Markers 1 week and 3 weeks after transplantation. Levels of CTGF protein in cartilage were determined by immunohistochemistry and Western blotting. Results In the model group, cartilage layer became thinner, with proliferation of fibroblast cells and a obvious cartilage surface hollow; New cartilage cells appeared in the surface hollow of the impaired cartilage in each treatemnt group, with a thicker layer. The number of transplanted BMSCs after 3 weeks was significantly increased in BMSCs group and NSP-PCL + BMSCs group. As compared with the model group, levels of CTGF protein were increased in each treatment group, with significant differences (P < 0.05). Conclusions Nano-porous polycaprolactone combined with bone marrow mesenchymal stem cells can repair cartilage damage by enhancing the expression of CTGF protein.
9.An approach of diabetic foot ulcers nature and its predilection site

Objective To investigate the feature of diabetic foot ulcer and the relative mechanism, and to analyze its predilection site. Methods The precipitating factors and the areas of ulcer were analyzed in 121 diabetic patients with foot ulcer. The peripheral neuropathy was diagnosed by measuring the skin sensation, tendon reflex, electromyogram or vibration perception threshold (VPT). The diabetic peripheral neuropathy (DPN) was evaluated by positive symptoms, ultrasound or magnetic resonance angiography. According to morbid changes, the patients were divided into neuropathic ulcer group (n = 33), ischemic ulcer group (n = 27), and mixed type ulcer group (n = 61). Results The ulcers of 97 cases (80. 2%) were distributed in digital pedis and plantar pedis. In neurogenic ulcer group, there were 48.5% lesion at planta pedis, 30.3% at dorsum pedis, and 21.2% at digiti pedi. In ischemie ulcer group, there were 59.3% at digiti pedis, 29.6% at dorsum pedis, and 11.1% at planta pedis. In mixed type ulcer group, there were 49. 2% at digiti pedis, 41% at planta pedis, and 9.8% at dorsum pedis. The difference of ulcer distribution was apparent in these groups. Among these three groups, patients in mixed type ulcer group was the oldest, and had the longest duration of diabetes (P<0.05) and the largest area of ulcer at diagnosis. However, the BMI and blood pressure in isehemic ulcer group were higher than those of the other groups (P>0.05). Conclusions The difference of lesion basis affects the position of diabetic foot ulcer, and the thumbs and plantar pedis are the predilection area of diabetic foot.
