1.Research Progress of Taking Key Enzyme in Metabolism as Selectable Marker in Transgenic Plant

In plant genetic engineering, selectable marker is needed to distinguish transformant. As the commercialization of transgenic plants, people are more and more paying close attention to their safety, among which mainly refers to the safety of selectable marker. In order to increase the safety of transgenic plants, biologists began to search for biosafe selectable marker.Current research progress of taking key enzyme in metabolic pathways were overviewed,which include glycometabolism, amino acid metabolism, hormone metabolism ,nucleotide metabolism and protein metabolism et cetera, as selectable marker in transgenic plants.
2.Study on Mutated DHDPS Gene as Selectable Marker of Transgenic Plant

The rodipicolinate synthase gene was cloned from Nicotiana tabacum L.cv,which encodes the key enzyme in the synthesis of lysine,and then a vitro molecular reform reliefing feedback suppression was made. Some resistant seedlings were acquired using the mutated gene as selectable marker and analog of lysine as selectable reagent,and the detection results were positive by means of PCR and Real-Time PCR,however the phenotype is somewhat abnormal.
3.Inhibitory effects of ?-lipoic acid on activation of NF-?B induced by high glucose in rat mesenteric cells

Objective To investigate the effects of ?-lipoic acid on the proliferation and activation of NF-?B induced by high glucose (HG) in rat mesenteric cells (MCs). Methods The rat mesenteric cells were cultured in the medium with normal glucose (5.6mmol/L, NG), high glucose (25mmol/L, HG), HG+100 ?mol/L ?-lipoic acid, or HG+200?mol/L ?-lipoic acid and HG+PDTC (a NF-?B inhibitor). Activation of nuclear factor-?B (NF-?B) of rat mesenteric cells were measured by electrophoretic mobility shift assay (EMSA). The cell proliferation was assessed by MTT. 4.Application of prism alternate cover test and perimeter strabismometry in strabismic operation

AlM:To discuss the application of prism alternate cover test ( PACT) and perimeter strabismometry in strabismic operation.METHODS: A total of 145 patients with concomitant esotropia or exotropia, aging 2 ~ 50 years old, were included. They were examined by PACT and perimeter strabismometry before operation. The strabismic operations were performed, the orthotropia rate of postoperation was assessed.RESULTS:Using PACT and perimeter strabismometry, there was a significantly difference of deviating angle ( P<0. 05) in mild and severe strabismic group. But there was no significantly difference of deviating angle (P>0. 05) in moderate strabismic group. The functional cure rate of postoperation was respectively 70. 83%, 67. 01%; the clinical cure rate was 20. 84%, 24. 74%; invalid rate was 8. 33%, 8. 25% .CONCLUSlON: The application of PACT and perimeter strabismometry in strabismic operation design, referring to the eye axial length, this can be helpful of improving the cure rate postoperation and decreasing the danger of reopreation.
5.The value of blood lipid and CA125 in identification diagnosis between lung cancer and pulmonary tuberculosis

Objective To explore the value of blood lipid and CA125 in identification diagnosis between lung cancer and pulmo‐nary tuberculosis .Methods The blood lipid and serum CA125 levels in 131 patients with lung cancer( Ⅰ - Ⅳ stage) ,53 patients with pulmonary tuberculosis and 60 normal controls were determined by biochemstry ,immunoturbidimetry and electrochemilumi‐nescence immunoassay and was done compared study .Results The serum TC and HDL‐C level in 131 patients with lung cancer were decreased with severity and were lower than in those of in 53 patients with pulmonary tuberculosis(TC :P< 0 .05 ,< 0 .05 ,<0 .01 and < 0 .001 respectively ;HDL‐C :P< 0 .05 ,< 0 .05 ,< 0 .01 and < 0 .01 respectively) .The serum LDL‐C level was no differ‐ences between lung cancer( Ⅰ - Ⅲ stage) and in 53 patients with pulmonary tuberculosis(P> 0 .05) only the lung cancer( Ⅳ stage) was a little increased(P< 0 .05) .The serum TG and ApoB/ApoA1 ratio levels in 131 patients with lung cancer( Ⅰ - Ⅳ stage) were increased with severity and were significantly higher than in those of 53 patients with pulmonary tuberculosis (TG :P < 0 .05 ,< 0 .05 ,< 0 .01 and < 0 .01 respectively ;ApoB/ApoA1 :P< 0 .05 ,< 0 .05 ,< 0 .01 and < 0 .01 respectively) .The level of CA125 in 131 patients with lung cancer was significantly higher than in those 53 patients with pulmonary tuberculosis(P< 0 .001) .Conclusion The serum TC and HDL‐C ,TG ,ApoB/ApoA1 ratio and CA125 levels may be the indexs of identification diagnosis between lung cancer and pulmonary tuberculosis .
6.Influence of Total Rhizoma Panacis Japonica Saponins on Hemorheology in Rats with Occlusion of the Middle Cerebral Artery

Objective To study the effect of total rhizoma panacis japonica saponins (tRPJS) on hemorheology in rats with occlusion of the middle cerebral artery. Methods Ischemia rat models were made by using the method of thread inserting right middle cerebral artery occlusion. The effects of tRPJS on whole blood viscidity, erythrocyte deformability and erythrocyte congregate in model rats were observed. Results tRPJS 200, 100, 50 mg/kg could significantly improve the erythrocyte deformability, reduce whole blood viscidity and erythrocyte congregate. Conclusion tRPJS can improve the hemorheology after cerebral ischemia. It may be one of the mechanisms for tRPJS in treating ischemic stroke.

Objective To investigate the role of rosiglitazone in high glucose(HG)-induced intercellular adhesion molecule-1(ICAM-1) expression of rat mesangial cells.Methods The rat mesangial cells(MCs) were cultured in the medium with normal glucose(5.6 mmol/L,NG),high glucose(25 mmol/L,HG),HG+5 ?mol/L rosiglitazone,HG+20 ?mol/L rosiglitazone and HG+PDTC(?NF-?B inhibitor).ICAM-1 mRNA expression was measured by semi-quantitative RT-PCR assay.Activation of nuclear factor-?B(NF-?B) of rat mesangial cells was measured by electrophoretic mobility shift assay(EMSA).The levels of ICAM-1 in the supernatants were determined by enzyme-linked immunosorbant assay(ELISA.) Hemodynamic monitoring is an essential part in the care of children with congenital heart disease during perioperative period to guide clinical management.Currently,there are several methods available for hemodynamic monitoring.The invasive methods include the Fick method,thermodilution method,using the Swan-Ganz catheter and the pulse contour method.The noninvasive methods include partial carbon dioxide resorption,impedance method.In this paper,the principle,advantages and disadvantages of these monitoring methods in children undergoing cardiopulmonary bypass surgery were reviewed.
10.Survey of 15 liver cirrhosis patients without upper gastrointestinal hemorrhage after transjugular intrahepatic portosystemic shunt

Objective To investigate the survival status of liver cirrhosis patients without upper gastrointestinal hemorrhage after transjugular intrahepatic portosystemic shunt (TIPS).Methods From 2004 to 2013,15 liver cirrhosis patients without upper gastrointestinal hemorrhage volunteered received TIPS treatment were followed up to find out the difficulty and the success rate of TIPS procedure,the incidence of hepatic encephalopathy,upper gastrointestinal hemorrhage and improving of hypersplenism.Results The success rate of operation was 100%.The average of operation time was 60 minutes.During follow-up,no stent angulation occurred,no gastrointestinal hemorrhage happened and no one died in all 15 patients after TIPS operation.There were four patients with hepatic encephalopathy in eight weeks after operation.The anemia of four patients improved compared with that before operation.Conclusions TIPS is a safe and effective threapy in the prevention of gastrointestinal hemorrhage in the patients with liver cirrhosis accompanied with severe gastroesophageal varices.It may become the primary prophylaxis for liver cirrhosis patients without upper gastrointestinal hemorrhage.
