2.Caspase-3 gene silencing attenuates apoptosis in alveolar epithelial cells after Streptococcus pneumoniae infection

AIM: To investigate the influence of Streptococcus pneumoniae on the apoptosis of alveolar epithelial cell and the modulation of caspases-3 on apoptosis. METHODS: A study was undertaken to determine whether Streptococcus pneumoniae infection induced the apoptosis of A549 and facilitated the expression of pro-inflammative cytokine. After inhibited by siRNA, the expression of caspase-3 and apoptosis grade in A549 were determined. RESULTS: Streptococcus pneumoniae contributed to alveolar epithelial cell apoptosis in a time-dependent manner. Streptococcus pneumoniae facilitated the expression of caspase-3 and IL-6 in A549, while depressed IL-10 concentration in culture medium. SiRNA directly decreased the expression of caspase-3 to rescue A549 from apoptosis but not affected IL-6 and IL-10 concentration. CONCLUSION: caspase-3 gene silencing rescues epithelial cells from apoptosis initiated by Streptococcus pneumoniae infection.
3.Clinical effect of low molecular weight heparin calcium combined with vertebroplasty in osteoporotic vertebral compression fractures in elderly patients

Objective To investigate the clinical efficacy and value of low molecular weight heparin calcium combined with vertebroplasty in the treatment of osteoporotic vertebral compression fractures in elderly patients .Methods 67 elderly patients with osteoporotic vertebral compression fractures in our hospital from January 2013 to January 2016 were selected and randomly divided into control group 30 cases and treatment group 37 cases. All patients were treated with percutaneous vertebroplasty, the control group was given calcium carbonate D3 conventional treatment, the treatment group was treated with low molecular weight heparin calcium anti osteoporosis treatment, the course of treatment was one month.Measurement of bone mineral density before and after the operation of different follow-up time points ( BMD ) and the changes of the oswestry disability index ( ODI ) evaluation activities and the visual analogue scale (VAS) to assess the degree of pain, and the changes of vertebral height before and after operation were analyzed. Results All patients were followed up for an average of 16.4 months (12 to 21 months).Compared with before vertebroplasty, the BMD of the patients in the treatment group was higher than that in the control group at one month, three months and six months, the difference was statistically significant (P<0.05), the low molecular weight heparin treatment after January, treatment group VAS and ODI scores were significantly lower than the control group, and the subsequent different follow-up time points ODI and VAS score can effectively maintain, the difference was statistically significant ( P<0.05) .Compared with before vertebroplasty, postoperative anterior vertebral height, middle height better than preoperative vertebral height, the difference was statistically significant (P<0.05), and after one months of treatment, the treatment group of anterior vertebral height, middle height were better than the control group, the difference was statistically significant (P <0.05).Conclusion After treatment with low molecular weight heparin calcium, the healing process of vertebral body after vertebroplasty, to a certain extent, it can increase the bone mass and prevent bone destruction, increase bone strength, and further achieve the treatment of osteoporosis and prevention of osteoporotic fracture in elderly patients with osteoporotic vertebral compression fractures.
4.Laparoscopic Transcystic Duct Exploration for Cholecystolithiasis Complicated with Stones in the Non-Dilated Common Bile Duct

Objective To investigate the efficacy of laparoscopic transcystic duct exploration for cholecystolithiasis complicated with stones in the non-dilated common bile duct.Methods Before the resection of the cyst,the cystic duct was cut under a laparoscope,and then a C tube was inserted for cholangiography.Stones in the common bile duct were removed under the guidance of C-arm X-ray.Then,C-tube drainage was carried out after removal of the calculi.Results The stones were completely removed in all of the 36 cases with a mean operation time of 125.4 minutes(90-150 minutes).The C-tube was withdrawn in 3 or 4 days after the operation.The mean postoperative time of this series was 4 to 7 days.The patients were followed up for 3 months,during which no patients had biliary leakage,residual stones,or stenosis or dilation of the common bile duct.Conclusions Laparoscopic transcystic duct exploration is minimally invasive,safe,and effective for cholecystolithiasis complicated with stones in the non-dilated common bile duct.
5.Clinical significance of XAF1 expression in pancreatic cancer and its relationship with prognosis

Objective To investigate the expression ot XAF1 protein in pancreauc cancer,and its relationship with clinicopathological parameters and prognosis.Methods The tissue microarray was composed of 89 pancreatic cancer and 21 normal pancreatic tissues,and immunohistochemistry was used to examine the expression of XAF1 protein.The correlation between XAF1 expression and clinicopathological parameters was analyzed.The 89 patients were followed,and the survival was shown in Kaplan Meier curve.Cox proportional hazards regression was used to identify risk factors for the survival of pancreatic cancer patients.Results The negative expression rate of XAF1 was 44.9% (40/89),and it was 9.5% (2/21) in normal tissue,and the difference between the two groups was statistically significant (P <0.05).The expression of XAF1 was negatively associated with tumor staging (P < 0.05),but it was not associated with age,gender,lymph node metastasis,distant metastasis,pathological grading,surgical margins,vascular and neural invasion.Patients with negative expression of XAF1 had much shorter survival than patients with positive expression.Univariate analysis showed that negative expression of XAF1,pathological grading,lymph node metastasis,distant metastasis was associated with the survival (P < 0.05),and the multivariate analysis indicated that negative expression of XAF1 was an independent prognostic factor for survival of pancreatic cancer.Conclusions XAF1 may be involved in the development and growth of pancreatic cancer,and it is related with patient's prognosis.
7.Lentivirus-mediated shRNA interference of ghrelin receptor blocks growth of colorectal cancer cells

[ ABSTRACT] AIM:To investigate the therapeutic effects of lentivirus-mediated shRNA targeting growth hormone secretagogue receptor 1a (GHSR1a) on colorectal cancer cell line SW480 both in vitro and in vivo.METHODS: Human GHSR1a sequence was used for the design of shRNA targeting GHSR1a, which was introduced to lentivirus, followed by transfection into SW480 cells.CCK-8 assay was performed to detect cell viability.The mRNA expression of GHSR1a and PTEN in colorectal cancer cells was detected by RT-PCR.The protein levels of GHSR1a, ghrelin, PTEN, p-AKT and p53 were determined by Western blot.Stable GHSR1a silencing in SW480 xenografts in nude mice was established.After the mice were sacrificed and weighted, immunohistochemistry was used to detect the positive expression of Ki-67 and PTEN in the tumors.RESULTS:GHSR1a was over-expressed in the malignant cells in comparison with the normal cells in vitro. The tumor specific lentivirus-mediated shRNA targeting GHSR1a gene and GHSR1a knockdown SW480 cells were success-fully constructed.After transfection with GHSR1a shRNA, the expression of GHSR1a at mRNA and protein levels was markedly inhibited in the SW480 cells.Furthermore, GHSR1a silencing by specific shRNA showed increased PTEN, inhi-bition of AKT phosphorylation and promotion of p53 release in the SW480 cells.In vivo results demonstrated that down-re-gulation of GHSR1a in the SW480 cells significantly decreased the expression of Ki-67 and increased the expression of PTEN in the tumor tissues, leading to a marked reduction in tumor weight in comparison to blank control or negative control
tumors.CONCLUSION:Down-regulation of GHSR1a by shRNA technique inhibits the growth of colorectal cancer cell line and xenograft tumor through activation of the PTEN/PI3K/AKT signaling pathways.
8.Clinical study of phacoemulsification in the treatment of angle-closure glaucoma

AlM: To study the application value of phacoemulsification in clinical treatment of angle-closure glaucoma.?METHODS:A total of 60 cases ( 82 eyes ) , the best corrected visual acuity, anterior chamber depth, intraocular pressure, width of chamber angle and adverse reaction of patients were researched and analysed before and after treatment.?RESULTS:ln this group, 60 cases (82 eyes) compared with that before treatment, the best corrected visual acuity in patients after treatment significantly improved, with significant difference between the two groups ( P<0. 05 ); anterior chamber depth of patients before treatment were 2. 0±0. 3mm, anterior chamber depth after treatment was 4. 4 ± 1. 0mm, treatment of patients with anterior chamber depth comparison significantly improved after treatment, with statistically significant difference between the two groups ( P<0. 05 ); lOP in patients with postoperative effective control over an extended period, the difference before treatment was statistically significant ( P<0. 05 ); the width of chamber angle after treatment compared with before treatment significantly increased, with statistically significant difference between the two groups ( P < 0. 05 ); after treatment by the emergence of 5 corneal edema, 3 mydriasis, an iris atrophy, no one cases of retinal detachment.?CONCLUSlON: Phacoemulsificationin the treatment of angle - closure glaucoma has a high value, can be effectively corrected visual acuity, improve patient anterior chamber angle width and depth, a small incidence of adverse reactions.
9. Research progress of PI3K/Akt pathway in diabetic cardiomyopathy

Diabetic cardiomyopathy(DCM) is a type of primary cardiomyopathy caused by diabetes, changing in the structure and function of the myocardium, which is distinguished from secondaiy cardiomyopathy resulted from coronary atherosclerosis, hypertension, and viruses. Phosphatidylinositol 3-kinase (PI3K)/Akt is an important signal transduction pathway in human body involved in the growth and reproduction of body. Regulatory factors of the PDK/Akt signaling pathway include silencing information regulator 1, homologous phosphatase-tensin, glycogen synthase kinase 3(3 and the rapamycin target protein. The role of PDK/Akt signaling pathway in the pathogenesis of DCM includes regulating blood sugar level, regulating lipid metabolism, protecting endothelial cells, reducing inflammatory response , improving cardiac function, resisting fibrosis, and regulating myocardial apoptosis. The development of PI3K-Akt signaling pathway inhibitors provides the possibility for PDK-Akt pathway targeted therapy. This article reviews the progression of the PDK/Akt pathway in DCM.
