1.Investigation of correlative factors of non-operative treatment in the senile cataract and countermeasure of health education

ObjectiveTo investigate correlative factors of non-operative treatment in the senile cataract, and provide gist for the health education measures.Methods1513 retiring old people had a whole medical examination, and the data was analyzed.ResultsThe proportion of binoculus cataract was 45.9%. Predilection age was 70~79 years old. Cognition information and iatrogenic effects were major risk factors in the non-operative therapeutic agents. Cataract was the third in the elder chronic disease.ConclusionMany senile patients with cataract have not enough knowledge about cataract operation although having better medical treatment and economy conditions. So the closed attention to the health education must be paid in the future.
2.Combination of four medicines in treating postoperative complications of Ring mixed hemorrhoids

Objective To eveluate the clinical curative effects of combination of Ma Yinglong She Xiang Zhi Chuang Gao,Jin Xuan Zhi Ke Xun Xi Xan,Diosmin,Macrogol 4000 powder in treating postoperative complications of ring mixed hemorrhoids.Methods Ninty cases of postoperative ring mixed hemorrhoids patients were divided into two groups randomly from January 2008 to June 2009.Experimental group:From the first day on Jin Xuan Zhi Ke Xun Xi San 55 g and the 1000 mL boiling water were added flushing,and Maying Long She Xiang Zhi Chuang Gao 2.5 g,2 times daily.The Macrogol 4000 powder 10 g and water 200 mL were admistrated orally,Diosmin 1.0 g orally,2 times daily.Oral administration of two kinds of medications was done each two hours.Control group:Using 1:100 Sterile warm salt water hip bath,and Ma Yinglong she xiang zhi chuang gao 2.5 g,2 times daily;Phenolphthalein tablets 100 mg orally,2 times daily.Results The experimental group surpassed the control group in the anus ache,the hemorrhage,edema (P<0.05).The heal time reduced obviously(P<0.01).Conclusion To combination of Ma Yinglong She Xiang Zhi Chuang Gao,Jin Xuan Zhi Ke Xun Xi San,Diosmin,Macrogol 4000 powder has the distinct improvement in the anus ache,the hemorrhage,dropsy of ring mixed hemorrhoids and reduces the injured area heal time obviously.
3.Network pharmacology-based analysis of effective components and mechanism of Rhizoma coptidis in treating diabetes

Objective: To identify the active ingredients, potential targets, and mechanism of Rhizoma coptidis by bioinformatics method, and to explore the hypoglycemic effect of Rhizoma coptidis by in vitro experiments. Methods: The chemical components of Rhizoma coptidis were collected through database search, and oral bioavailability and drug-likeness were used for preliminary screening. The targets of Rhizoma coptidis and diabetes-related targets were collected by database retrieval and reverse docking techniques, and the biological process of cross-set proteins was analyzed. The inhibitory effects of Rhizoma coptidis on α-glucosidase, α-amylase activity, and advanced glycation end products (AGEs) were determined via in vitro experiments. In addition, the effects of Rhizoma coptidis on pre-adipocyte differentiation, absorption of glucose by adipocytes, and the level of intracellular triglyceride were investigated using the adipocyte differentiation model. Results: There were 11 potentially active ingredients in Rhizoma coptidis. IL-6, caspase-3, epidermal growth factor receptor (EGFR), MYC, and estrogen receptor 1 were considered as the key genes. The bioinformatics analysis showed that Rhizoma coptidis played an anti-diabetic role mainly via biological processes and signaling pathways including hormone receptor activity, glutathione binding, steroid binding, etc. In vitro experiments showed that the extract of Rhizoma coptidis inhibited the activities of α-glucosidase and α-amylase, and the generation of AGEs; meanwhile, the extract promoted the absorption of glucose by adipocytes. In addition, the extract of Rhizoma coptidis decreased triglyceride level. Conclusions: Our network pharmacology and in vitro experiments demonstrate the anti-diabetic effects and possible underlying mechanisms of Rhizoma coptidis extract.
4.RhoA/ROCK pathway mediated DHT regulates function of early endothelial progenitor cells

Objective:To analyze the effects of DHT on the proliferation and migration of endothelial progenitor cells (EPCs) and the role of RhoA/ROCK pathway in this process.Methods:Early EPCs were isolated from peripheral blood of healthy adults, and cultured in serum-free EBM-2 medium for 24 h before incubation with various concentrations of DHT (1, 10, and 100 nmol/L). EPCs proliferative and migrative capacities were measured. The adherent cells were collected and randomLy divided into: control group, DHT group, C3 exoenzyme+DHT, Y-27632+DHT group. EPCs proliferation and migration were assayed by MTT assay and modified Boyden chamber assay respectively.Results:DHT significantly increased the proliferation and migration ability of EPCs in a dose- and time-dependent manner, maximum at 10 nmol/L, 24 h ( P<0.05). C3 exoenzyme [(0.22±0.02) vs (0.26±0.05), P>0.05] and Y-27632 [(0.21±0.04) vs (0.26±0.05), P>0.05] can attenuate the proliferative capacities of EPCs induced by DHT compared with the DHT group, but there was no statistical significance. The influence of DHT on EPCs migrative capacities can be abolished by C3 exoenzyme [(35.26±4.27) vs (46.92±5.46), P<0.05] and Y-27632 [(33.61±5.33) vs (46.92±5.46), P<0.01]. C3 exoenzyme [(116.75±7.42) vs (156.80± 21.74), P<0.05] and Y-27632 [(121.73±5.33) vs (156.80 ±21.74), P<0.01] could noticeably attenuate DHT-induced EPCs secretion of VEGF respectively. Conclusions:DHT can modulate EPCs proliferation, migration and the RhoA/ROCK pathway plays an important role in this process.
5.Mesenchymal stem cells homing to rat lung emphysema followed by differentiation into pulmonary vascular endothelial cells

BACKGROUND:Mesenchymal stem cells have unique homing,immunoregulation and anti-inflammatory properties.After intravenous injection,mesenchymal stem cells can home to the damaged target organs and tissues,and function to repair damaged tissues.OBJECTIVE:To observe the pathological changes of lung tissues in rats with emphysema after mesenchymal stem cells transplantation.METHODS:Twenty-four female Wistar rats were randomly divided into experimental group,control group and healthy control group.In the first two groups,the model of pulmonary emphysema was established by the method of dropping porcine pancreatic elastase.BrdU-labeled mesenchymal stem cells were injected into the tail vein of the rats in the experimental group,and PBS was injected in the control group.After 14 days,the pathological changes of lung tissues were observed.Tumor necrosis factor-α level,alveolar wall apoptotic index,anti-CD34 and anti-BrdU immunohistochemical changes in the bronchoalveolar lavage fluid were detected.RESULTS AND CONCLUSION:Compared with the healthy control group,the tumor necrosis factor-α level and apoptotic index of alveolar wall cells increased (P < 0.01),and the relative areas of anti-Brdu and anti-CD34 decreased (P < 0.01) in the control group.Compared with the control group,the level of tumor necrosis factor-α and apoptotic index of alveolar wall cells decreased (P < 0.01),and the relative area of anti-Brdu and anti-CD34 increased (P < 0.01) in the experimental group.The histopathological findings showed that both the control group and the experimental group showed emphysema-like changes,but these changes were milder in the experimental group than the control group.To conclude,mesenchymal stem cells can inhibit inflammatory response and apoptosis in experimental emphysema,improve the pathological changes of the lung,and moreover,bone marrow mesenchymal stem cells can differentiate into lung vascular endothelial cells.
6.Huaiqihuang granule in adjuvant treatment of childhood asthma:a randomized controlled,multicenter study

Objective To observe the clinical efficacy and safety of GINA regimen and GINA regimen combined with oral Huaiqihuang granule in the treatment of children with bronchial asthma.Methods A ran-domized,single blind,multicenter,parallel controlled clinical trial wascarried out.A total of 1 128 patients with bronchial asthma in children were randomized into two groups.The observation group were treated with GINA regimen combined with oral Huaiqihuang granule.The GINA regimen treatment group was treated by GINA reg-imen.Clinical assessment and C-ACT scores was observed in first month,third month,sixth month after treat-ment.Clinical assessment included the times of upper respiratory tract infection occurrence,bronchitis and pneu-monia,asthmatic attacks,application of emergency medicine,hospitalizations due to asthmatic.Drug adverse effect in the two groups was compared.Results The times of upper respiratory tract infection,bronchitis and pneumonia,asthmatic was significantly decreased(P <0.05 ),and C-ACTscores were significantly higher(P <0.05)in the first month,the third month,and the sixth month after treatment.There were 16 cases of drug relat-ed adverse reactions.Seven cases were in observation group(n ﹦7)(1.15%)and 9 cases in the GINA regimen treatment group(n ﹦9)(1.73%).There was no significant difference of adverse effect between the two groups (P >0.05).Conclusion The treatment of bronchial asthma in children with GINA regimen combined with oral Huaiqihuang granule can significantly reduce the incidence of respiratory infections and the number of asthmatic attacks dramatically and safely improve clinical curative effect,asthma control.
7.Relationship between driver gene mutation and clinicopathological features in 300 cases of non-small cell lung cancer based on next generation sequencing

Purpose To explore the mutation characteristics of common driver genes in non-small cell lung cancer (NSCLC) and its relationship with clinicopathological features.Methods Next generation sequencing (NGS) was used to detect the mutations of common driving genes such as EGFR, KRAS, ALK, ROS1, BRAF, MET, RET and HER-2 in 300 paraffin-embedded NSCLC tissues. Results In 300 patients with NSCLC, the mutation rates of EGFR, KRAS, ALK, ROS1, BRAF, MET, RET, HER-2 were 52.00%, 10.33%, 6.67%, 1.67%, 3.67%, 3.33%, 1.00%, and 2.33%, respectively. A case of EGFR 21 exon L858 R mutation was combined with LINCO1446-EGFR gene fusion. EGFR 20 th exon C797 S and T790 M existed in cis or trans form and merged with EGFR sensitive mutations in 1 case each. 3 cases of EGFR gene point mutation was associated with MET gene copy number amplification. EGFR mutations were more commonly detected in non-smoking women with lung adenocarcinoma (P<0.05).KRAS mutations were more commonly found in smoking men (P<0.05). ALK mutations were associated with age (P<0.05), and more commonly noted in patients younger than 60 years.ROS1 fusion mutations were associated with gender (P<0.05), more commonly detected in women. BRAF, MET, RET, and HER-2 gene mutations were not associated with gender, age, smoking, histological type, and c TNM stage. Conclusion EGFR can coexist with other driver gene mutations. Gene mutations and clinicopathological features like gender, age, smoking, and histological types have corresponding links. The incidence of BRAF, MET, RET, and HER-2 mutations is low, and its clinical significance remains to be explored. Coexisting gene mutations and rare mutations discovered by NGS should be taken seriously.
8.Prevalence and Molecular Characterization of Echinococcus granulosus Sensu Stricto in Northern Xinjiang, China

Echinococcus granulosus is an important zoonotic parasite globally causing cystic echinococcosis (CE) in humans and animals. In this study, prevalence of CE and variation of cox1 gene sequence were analyzed with isolates E. granulosus collected from different areas in northern Xinjiang, China. The survey showed that 3.5% of sheep and 4.1% of cattle were infected with CE. Fragment of cox1 was amplified from all the positive sheep and cattle samples by PCR. In addition, 26 positive samples across the 4 areas were included. The isolates were all E. granulosus sensu stricto (s.s.) containing 15 haplotypes (Hap1-15), and clustered into 2 genotypes, G1 (90.1%, 91/101) and G3 (9.9%, 10/101). Hap1 was the most common haplotype (48.5%, 49/101). Hap9 were found in humans samples, indicating that sheep and cattle reservoir human CE. It is indicate that E. granulosus may impact on control of CE in livestock and humans in the region.

9.Clinical effect and action mechanism of Weicao Capsule in treating gout.

OBJECTIVETo study the therapeutic effect of Weicao Capsule (WCC) on gout.
METHODSTwo hundred gout patients were assigned to two groups. The treated group was treated with WCC and the control group was treated with Tongfengding Capsule. Both groups were given the respective treatments orally 3 times a day, 2 capsules each time with 2 weeks as one course and all patients received 2 successive courses of treatment. Changes of blood beta(2)-microglobulin (beta(2)-M), hemoglobin (Hb), 24 h urinary protein (24 h UP), pH value of urine and blood uric acid (BUA) as well as kidney function were observed.
RESULTSAfter treatment, level of beta(2)-M got lowered significantly, Hb and 24 h UP, blood urea nitrogen, serum creatinine and the clearance rate of creatinine, as well as blood lipids all improved obviously in the treated group (all P<0.01), while these parameters remained unchanged in the control group (P>0.05). The pH value of urine was improved in both groups showing an insignificant difference between them (P>0.05). BUA was decreased in both groups with a decrease to a larger extent in the treated group (P<0.01). The total effective rate was 87% in the treated group, which was superior to that in the control group (62%, P<0.05).
CONCLUSIONWCC has a favorable therapeutic effect on gout and its mechanism of action for improving renal function and reducing urinary protein could be related with the lowering of blood beta(2)-M, BUA and lipids.

10.Detection of common deletions and mutations causing α-thalassemia in Southeast Asians and Southern Chinese with denaturing high performance liquid chromatography.

OBJECTIVETo establish a comprehensive and simple assay using denaturing high performance liquid chromatography (DHPLC) for the diagnosis of most common mutations and deletions of α-thalassemia gene in Southeast Asians and Southern Chinese.
METHODSThis assay has included a duplex polymerase chain reaction (PCR) followed by DHPLC analysis. An improved PCR was also performed followed by DHPLC analysis. With this assay, a blinded study of 160 samples was screened for three common mutations and three common deletions.
RESULTSThe duplex PCR-DHPLC combined with the improved PCR-DHPLC analysis has detected all mutations and the wild-type allele. The results were consistent with those by the original methods.
CONCLUSIONThis molecular assay may be used for the diagnosis of α-thalassemia patients from this geographical region. The method is accurate, rapid, semi-automatic and cost-effective, which makes it suitable for large-scale screening.
