The study of peptide drugs has been an important direction in research and development of new drugs. However, lots of natural macromolecular peptides are limited in clinical use by their metabolic instability and low bioavailability. In recent years, the active small peptidomimetics open up a new hotspot of peptide drug development with the characteristics of low molecular weight, high bioactivity and structural modification. Many peptidomimetics are on the market or on the clinical study. This paper elaborated the small peptidomimetics approved by American Food and Drug Administration (FDA) from 2005 to 2014, and reviewed their researching status with source, synthetic method, chemical structure, marketing time, indication, clinical efficacy and safety. Research prospects in this field were discussed.

Purpose To detect and isolate cancer stem cells ( CSC) in fresh specimens of primary and metastatic carcinoma tissues from 10 cases of colorectal cancer to study the biological functions and mechanisms of migrating cancer stem cells. Methods Suspen-sions of primary and metastatic colorectal cancer cells were labeled with antibodies CD133-PE and CD44-FITC, and then flow cytometry was used to detect and isolate the CD133-PE-positive and CD44-FITC-positive cells. Results There were certain percentages of CD133-positive and CD44-positive cells present in primary and metastatic colorectal cancer and the proportion of CSC in metastatic colorectal cancer was higher than that in primary cancer tissue (P<0.05). Conclusions There are CSC in both primary and meta-static colorectal cancer and the proportion of CSC in metastatic colorectal cancer is significantly higher than that in primary cancer tis-sue, suggesting that CSC play an important role in the metastasis of colorectal cancer.

Objective To investigate the correlation between Presenilin 1 gene methylation (PSEN1) and sporadic Alzheimer's disease (SAD).Methods Massarray method was used to perform an analysis of DNA methylation in the promoters of PSEN1 gene in lymphocytes from 35 SAD patients,33 mild cognitive impaired (MCI)patients and 22 age-and gender-matched controls.Reporter gene plasmid of PSEN1 was built by genetic recombination methods,and then transiently transfected into the SH-SY5Y and HeLa cells.After that,cells were treated with several agents including serum deprivation,Aβ25-35,5-aza-2'-deoxycytidine (5-aza Cdr) and S-adenosyl-Lmethionine (SAM).A dual-luciferase reporter assay system was used to calculate the relative luciferase activity (RLA).Results Some CpG sites of PSEN1 (Cytosines at-173,95 and +76) showed hypomethylation patterns in SAD cases as compared with MCI and control group (P＜0.01).A dual-luciferase reporter assay showing significant differences in PSEN1 transcription activities were found in both cell lines under SAM treatment [SY5Y,SAM:(6.40±0.81),control:(9.13±1.67),P=0.000; Hela,SAM:(2.06±0.34),control:(2.99±0.59),P=0.000].Conclusions DNA hypomethylation of PSEN1 gene promoter is significantly associated with SAD susceptibility.PSEN1 may change the gene transcription level by methylation,further affecting the activity of amyloid protein-like γ-secretase,leading to the onset of AD.

Objective To anlayze the imaging diagnosis of spinal primary tumor.Methods Fifty and nine cases of spinal primary tumors confirmed by pathology were analyzed retrospectively.Results Of 59 cases,there were chordoma in 23,hemangioma in 7,giant cell tumor of bone in 11,aneurysmal bone cyst in 5,osteoblastoma in 7,osteogenous sarcoma in 2,myeloma in 2,spinal eosinophilic granuloma in 1 and primary bone lymphoma in 1.These tumors had its own imaging characteristics.Conclusion Imaging examinations are of significant value for the clinical treatment of spinal primary bone tumor.

Objective To explore the application and effectiveness of simulated patient role play in the inquiry and the medical record writing teaching.Methods Carrying out the teaching model of simulated patient role play in the Second Clinical Medical College of Southern Medical University to the undergraduate students of grade 2009 snd 2010 in diagnose inquiry and medical record writing training class.A self-designed student questionnaire mainly about atmosphere of class,study enthusiasm,Self-evaluation of learning efficiency,the effect of the medical record writing in-class training and so on was used to do the survey.On the other hand,questionnaire for teachers included students' performance,students' writing ability,etc.100 students and 50 teachers were investigated with the survey.The percentage of the options of each question on the questionnaire was calculated in order to analyze the new teaching mode.Results The random questionnaire survey showed that 85％ (85/100) of the students believe that the new teaching mode can activate classroom atmosphere and improve learning motivation.About 62％(62/100) of the students said medical record writing errors can be obtained and corrected timely in-class training.Most students in grade 2009 have better capacity in medical record writing than those students who did not contact with new teaching mode.Conclusion The teaching model of simulated patient role play in the analogue inquiry and medical record writing is efficient and feasible in medical education,which can improve students' practical ability and is expected to be applied in undergraduate medical education.

Necroptosis is a newly discovered form of programmed cell death, which is activated by the binding of death receptors and their ligands and executed through specific path ways. At present, this form of cell death has been proved to be involved in a variety of diseases,such as cancer, autoimmune diseases, traumatic brain injury and brain ischemia-reperfusion injury.

Objective To compare the effectiveness and safety of two different protocol of chlopidogrel therapy on intra-stent thrombus prevention in patients undergone coronary interventions. Methods Sixty-three consecutive patients underwent elective coronary interventions in Beijing Mining General Hospital were enrolled in this retrospective study. The patients were divided into two groups according to the protocol of chlopidogrel therapy. For the testing group (n=29), 300 mg and 225 mg of chlopidogrel were given 48 hours and 24 hours prior to the intervention procedure respectively followed by 75 mg daily. And for the control group (n=34), 300 mg cholpidogrel were given 6 hours prior to the procedure followed by 75 mg daily. Acute, subacute and late in-stent thrombosis and bleeding were recorded during a 2-40 months follow-up. Infants cholestasis is a common disorder of hepatobiliary system which is caused by abnormal cholestasis of hepatocytes or substances accumulation in the liver, blood and extrahepatic tissue due to the disorder of the intrahepatic or extrahepatic bile ducts.Serum Gamma-glutamyl transpeptidase(GGT) was widely distributed in one side of the capillary bile duct and the whole bile duct system of hepatocytes.When hypersynthesis in the liver or disorder of cholorrhagia, serum GGT increased.This review summarized the changes of GGT cholestasis of different causes in infants and its significance in differential diagnosis, for better understand the disease and provide references for clinical practice.

With the characteristics of complicated etiologic factors,multiple complications and high mortality,severe acute pancreatitis (SAP) has been treated by all kinds of clinical management.Management of SAP is a hot topic in recent decades.Researches on the pathophysiology of SAP have advanced dramatically during the past 20 years.As the number of randomized controlled studies of SAP has steadily increased,mainly in the United States and Europe,evidence based management was demanded for the management of SAP.The main improvement of the treatment results was achieved following internationally accepted recommendations; however,some aspects of the management are different and need to be discussed.There has been no consensus regarding the treatment of SAP.Failure to provide definitive treatment may potentially put a patient at risk of a fatal episode of SAP.The aims of this study,therefore,were to survey the currently adopted practice of the management of SAP,to evaluate the impact of various subspecialty interests on the practice adopted,and to suggest pathways towards a better implementation of the guidelines and other evidence-based management approaches.
