2.Effects of matriptase down regulation on invasion of human pancreatic cancer cells SW1990

Objective To investigate the effect of the down regulation of matriptase expression on invasion of human pancreatic cancers cells SW1990.Methods Small interfering RNA targeting at matriptase (Ma-siRNA) was transfected into human pancreatic cancers SW1990 cells,and nonsense siRNA (NC-siRNA) group was used as control.Real time PCR assay and Western blot were used to detect the expression of matriptase mRNA and protein.Transwell assay was used to examine the invasion ability of cancer cells.The enzymatic activity of matriptase and MMP-9 was determined by gelatin zymography assay.Results The expression level of matriptase mRNA in NC-siRNA group,12.5,25,50 nmol/L Ma-siRNA group were 1.000,0.417 ± 0.006,0.233 ± 0.068,0.221 ± 0.092;and the protein expression of matriptase were 0.736 ± 0.066,0.498 ± 0.036,0.341 ± 0.118,0.239 ± 0.050,respectively.The matriptase mRNA and protein expression in Ma-siRNA groups was significantly lower than those in NC-siRNA group,and the difference between the two groups was statistically significant (P < 0.05).The enzymatic activity of matriptase were 1.501 ±0.165,1.211 ±0.265,0.645 ±0.165,0.620 ±0.003,and the enzymatic activity of MMP-9 were 0.929 ± 0.260,0.484 ± 0.364,0.352 ± 0.113,0.346 ± 0.121,and the enzymatic activity of matriptase and MMP-9 in 25,50 nmol/L Ma-siRNA groups was significantly lower than that in NC-siRNA group,and the difference was statistically significant (P < 0.05 or < 0.01).The number of transmembrane cell was (256 ± 1)/per 200 power field,and it was (109 ± 3)/per 200 power field in 25 nmol/L Ma-siRNA group,and the invasion ability of the cells in 25 nmol/L Ma-siRNA group was decreased by (57.4 ± 5.4) % when compared with that of control group.Conclusions Down-regulation of matriptase inhibits invasion ability of pancreatic cancer SW1990 cells,and this result may be due to the down regulated enzymatic activity of matriptase and MMP-9.

4.Advances in novel carrier systems of chemical constituents from spice volatile oils.

Recent years, chemical constituents from spice volatile oils have gained worldwide concern owing to its multiple pharmacological effects and safety for using as the natural antibacterial agents. However, their poor dissolution, strong volatility, serious irritation, weak stability, easy oxidation and low bioavailability characteristics are the major obstacle in the preparation of effective oral formulation and practical application. Therefore, there is an urgent need to select a novel carrier system that can delivery the chemical constituents from spice volatile oils more efficiently with improving their stability as well as alleviating the irritation, and develop the functional food, health products and even medicine for exerting their pharmacological effects, which also is the focus and nodus of the research on their application. This review presents recent systematic studies on their novel carrier systems, including cyclodextrin inclusion complex, liposomes, nanoemulsions, nanoparticles, solid dispersion and so on, and summarizes the characteristics, application range and problems of each novel carrier systems, in order to provide some beneficial thoughts in further developing new products of chemical constituents from spice volatile oils.

CONCLUSIONMaillard reaction is involved in the process of rhizome C. barometz, which may be contributed to the activity between raw and processed C. barometz.

7.Risk factors for post-operative delirium and post-operative cognitive dysfunction in patients undergoing spine surgery

Objective To determine the risk factors for post-operative delirium(POD)and post-operative cognitive dysfunction(POCD)in patients undergoing spine surgery.Methods One hundred and twenty ASA Ⅰ-Ⅲ of both sexes aged 50-76 yr undergoing elective spine surgery under general anesthesia were studied.POD was assessed by Delirium Rating Scale revised 98 at 2 days after operation and the patients were assigned into POD and nonPOD group.Cognitive function was assessed by Mini-Mental State Examination(MMSE)at 1 day before and 3 days after operation.The patients were diagnosed as having POCD if MMSEpre-MMSEpost ≥ 3.The palients were assigned into POCD and nonPOCD group.Executive function and depression were assessed by stroop interference test and Beck Depression Inventory(BDI)at 1 day before operation.Age,sex,education,alcohol consumption per week,a history of psychiatric disease,ASA physical status,Charlson comorbidity score,type of anesthesia,anticholinergic drug administration and VAS score at 1 day after operation were recorded.If there was signifirant difference between the 2 groups,the factor was analyzed using multi-factor logistic regression to select risk factor for incidence of POD and POC).Results Eleven patients developed POD(9.2%)and 30 patients developed POCD(25.0%).Logistic regression model showed that lower Stroop-CW,higher BDI score,higher Charlson comorbidity score and a history of psychiatric disease were risk factors for POD,while lower Stroop-CW,higher BDI score,higher Charlson comorbidity score and higher alcohol consumption per week were risk factors for POCD.Conclusion Preoperative executive dysfunction,depression and greater preoperative comorbidity are risk factors for both POD and POCD.A history of psychiatric disease is a risk factor for POD and higher alcohol consumption is a risk factor for POCD in patients undergoing spine surgery.
8.Effects of α-crystallin on bioactivity of iNOS in the activated retinal microglial cells

Objective To investigate effects of α-crystallin on proliferation of lipopolysaccharide (LPS)-activated retinal microglia and bioactivity of iNOS.Methods The retinal microglial cells cultured in vitro were analyzed and their purity was identified by cell immunofluorescence and flow cytometry.After microglia cells being intervened using LPS and α-crystallin at various concentrations,influence of α-crystallin on activity of LPS-activated retinal microglia was detected by MTT method and level of NO was measured by RT-PCR to observe changes of iNOS expression in microglia.Results Purity of primary cultured microglial cells was 94.15% by GSA-IB4 immunohistochemical identification and 93.34% by CD11b flow cytometry.α-crystallin of 10-4g/L awakened activity of microglia induced by 10-6g/L LPS (P < 0.01).Expressions of iNOS protein and mRNA showed significant decrease in combined treatment group (P < 0.05).Conclusion In clinical condition,α-crystallin decreases the harm of microglial cells on retinal ganglial cells (RGCs) after optical nerve injury by inhibiting the microglia cells to produce NO and iNOS.
9.The use of a multiplex real⁃time PCR assay for rapid diagnosing acute respiratory viral infections in children

Abstract:Objective The aim of the study was to evaluate a multiplex molecular real ⁃ time assay for the diagnosis of respiratory viruses of childhood acute respiratory virus infection,and analyze the symptoms associated with the pathogens which were detected from those samples. Methods From August 2018 to August 2019,a sample of 275 swallow swabs
from children with acute respiratory infections in Chengdu Children′s Specialist Hospital was collected and tested using the RespiFinder®SMART 22 test kit. The kit is based on multiplex PCR technology and can detect and distinguish 22 pathogens related to respiratory infections at one time. Results Of the 275 samples,245(89.1%)detected positive viral infections,
of which 160 were single virus infections,59 were multiple viral infections,17 were positive for Mycoplasma pneumoniae, and 9 were positive for Chlamydia pneumoniae. The most common pathogens were rhinovirus 77 cases(28.0%),adenovirus 45 cases(16.4%)and respiratory syncytial virus 34 cases(12.4%). Influenza virus was detected in 28(10.2%)samples, parainfluenza in 25 cases(9.1%),coronavirus in 15 cases(5.5%),Boca virus in 12 cases(4.4%),and human metapneumovirus in 9 cases (3.3%). Rhinovirus (28.0%) was the most common pathogen in upper respiratory tract infections,while respiratory syncytial virus(12.4%)was the most common pathogen in lower respiratory tract infections. It has been found that multiple viral infections in children were associated with severe respiratory symptoms in clinical manifestations. Conclusion Compared with ordinary PCR detection,the multiplex PCR could detect multiple pathogens at one time in experiment,which greatly shortens the detection time and had important clinical significance for rapid diagnosis of multiple respiratory pathogens. Thus,it could be useful for detecting several viruses causing respiratory tract disorders.
10.Study on Purification Technology of Total Alkaloid from the Flos of Aconitum kusnezoffii

OBJECTIVE:To optimize the purification technology of total alkaloid from the flos of Aconitum kusnezoffii. METH-ODS:The content of total alkaloid from the flos of A. kusnezoffii was determined by acid-base titration. The purification technology of total alkaloid from the flos of A. kusnezoffii was optimized by ion resin with resin type,mass concentration of loading liquid and exchange speed as factors,maximum adsorption quantity,desorption rate and mass fraction of total alkaloid as index,and verifica-tion test was conducted. RESULTS:The optimal purification technology was as follows as type 732 cation exchange resin,mass concentration of loading liquid 0.32 g/L,exchange speed of 7 column volume(BV)/h. In validation test,the content of total alka-loid was 86.88%(RSD=0.52%,n=3),and desorption rate was 92.81%(RSD=0.40%,n=3)averagely. The extraction trans-port rate of total alkaloid from 3 batches of the flos of A. kusnezoffii was 81.76% and purification transport rate was 89.47% in av-erage. CONCLUSIONS:The established method is stable and feasible,and shows high transport rate.
