1.Effects of siRNA on VEGF expression and cell proliferation of HeLa cells

Objective To investigate the effects of small interference RNA (siRNA) targeting vascular endothelial growth factor (VEGF) on VEGF expression and cell proliferation of cervical carcinoma HeLa cells,in order to provide a theoretical basis for the treatment of human cervical carcinoma.Methods siRNAs plasmids targeting VEGF165 were constructed and stably transfected into HeLa cells.HeLa group,Mock group and VEGF siRNA group were set up.The expressions of VEGF mRNA and protein in stable transfectants were confirmed by RT-PCR,Western blotting and ELISA assay,respectively.The cell proliferation was analyzed by MTT and flow cytometry.Apoptosis was measured by Hoechst33342 staining.Results VEGF siRNA stably transfected cell lines were successfully established.The siRNA targeting human VEGF gene effectively decreased the expression of VEGF at RNA and protein levels,and inhibited the proliferation of HeLa cells (P
2.Status quo and research of medical students′professional perception and humanistic education

Objective To better understand students' professional perception,humanistic education status quo and their preference to different humanistic education methods.Methods Questionnaire was conducted for 990 undergraduate students (2006 grade- 2010 grade) at medical school of Shanghai Jiao Tong university.SPSS 17.0 statistical software was used to analyze results of the questionnaire.Results Nearly 45% students chose to major in clinical medicine out of personal interest.The identification of students for humanistic quality was lower than that for clinical knowledge and skills.More than 50% students believed doctors were with high humanistic quality and there was a gap between students′ action and their understanding of the medical career identity and medical humanistic cultivation.More than 50% students preferred activities that train medical and auxiliary skills.Nearly 67% students also showed a shallow reading habit from regular internet surfing.Conclusion The professional perception of medical students are influenced by both social and historical factors influence.The gap between understanding and action is a consequence of current humanistic educational system.The humanistic educational methodology should conform to the psychological and s~ial characteristics of contemporary students.
3.Oxidative Stress and Expression of Bax and Bcl-2 in Rat Corpus Striatum Induced by 6-Hydroxydopamine

Objective To study the oxidative stress and apoptosis relative protein expression in rat striatum during the pathogenesis of Parkinson's disease (PD) induced by 6-hydroxydopamine (6-OHDA). Methods 6-OHDA was stereotacticly injected into the right striatum of the rats at two sites to produce PD models. Activities of superoxide dismutase (SOD), glutathione peroxidase (GSH-Px), contents of reduced glutathione (GSH) and malondialdehyde (MDA) in the injured and normal striatum were measured using assay kits; and levels of Bax and Bcl-2 were detected by Western blotting in injured striatum. Results In 10 successful PD rats, compared with either the sham group or the normal group, activities of SOD and GSH-Px and contents of GSH in the right striatum significantly decreased while contents of MDA increased obviously (P<0.05); And levels of Bax significantly increased while expression of Bcl-2 obviously decreased. Conclusion Oxidative stress plays a key role in the pathogenesis of PD. Furthermore, Bax and Bcl-2 were involved in the regulation of apoptosis under oxidative stress induced by 6-OHDA.
4.Behavior and Neuronal Apoptosis in Substantia Nigra of Rats with Parkinson's Disease

ObjectiveTo assess rat behavior, observe apoptotic morphology of neurons and measure expression of caspase-3 in substantia nigra(SN) of Parkinson's disease(PD) animal model. Methods6-OHDA was stereotacticly injected into the right striatum of the rats at two sites to produce PD models. After 5 weeks, the behavior tests including modified Morris Water Maze and narrow beam test were measured. Then tyrosine hydroxylase (TH) histochemistry, Hoechst 33258 staining, Western blotting for caspase-3 in right substantia nigra was separately conducted.ResultsEscape velocity significantly decreased and its latency was obviously enlarged in modified Morris Water Maze, and the latency and total time in narrow beam test were also markedly increased (P<0-05) in 15 successful PD rats compared with either the sham group or the normal group. Furthermore, there were obvious less TH-positive neurons in lesioned SN while more apoptotic cells appeared there. In addition, the expression of caspase-3 was highly upregulated in lesioned SN.Conclusion6-OHDA induced neuronal apoptosis in SN is associated with high levels of caspase-3, and the results of rat behavior tests are correspondent with the morphological changes including TH immunohistochemistry and Hoechst 33258 staining. Thus modified Morris Water Maze and narrow beam test are beneficial for assessments of effects of new drugs in PD animal model.
5.Morroniside Inhibiting Ca2+ after Platelet Aggregation Induced by Adenosine Diphosphate in Rabbits

Objective To explore the effects of morroniside on Ca2++ in the condition of platelet aggregation in rabbits induced by adenosine diphosphate (ADP). Methods The mobilization of cytosolic-free calcium after platelet aggregation induced by ADP was detected by Ca2++-sensitive fluorescent indicator, Fura-2 AM and time scan measurement. Results Compared with the controls, morroniside significantly inhibited the increase of Ca2++ induced by ADP (P<0.001). Conclusion Morroniside acts as an effective platelet aggregative antagonist by inhibiting the increase of platelet Ca2++.
6.Effects of Morroniside on Inhibiting Thromboxane B2 after Platelet Aggregation Induced by Adenosine Diphosphate in Rabbits

Objective To study the effects of morroniside on thromboxane B2 in the condition of platelet aggregation in rabbits inducedby adenosine diphosphate (ADP). Methods The level of TXB2 was determined by enzyme-linked immunosorbent assay (ELISA). ResultsCompared with the controls, morroniside significantly inhibited the increase of TXB2 induced by ADP (P<0.001). Conclusion Morronisidecan inhibit the level of TXB2 by inhibiting the platelet aggregation in rabbits induced by ADP.

Investigations on the mechanism of Wnt signaling in neurogenesis and angiogenesis are under way, and these investigations contribute to the recognition of the restoration and regeneration after the brain injury.
8.The applied anatomy of iliac tissue flap pedicled on the iliolumbar artery

Objective To provide anatomical basis for elevating iliac tissue flap pedicled on the iliolumbar artery.Methods The course,number,outer diameter and distribution of iliolumbar artery were observed on 13 human cadavers.Results All the iliolumbar artery originated from the internal iliac artery.The iliolumbar artery gave off 2 branches (iliacus branch and lumbar branch) when passing between the obturator nerve and the lumbosacral trunk,posteriorly to the psoas major.The mean distance between origin of the iliolumbar artery and bifurcation point to iliacus and lumbar branches was 7.1 (7.1 ± 0.5) cm.The iliacus branch divided into two branches:one artery curved forward and anastomosed with the iliacus branch of deep circumflex iliac artery; the other artery supplied the tissue around the posterior superior iliac spine.The lumbar branch supplied the psoas major and the quadratus lumborum.Conclusion The iliolumbar artery and the iliacus branch is one of the most constant and reliable vessels supplying the iliac bone,and can be used as the pedicle of free or pedicled iliac tissue flaps.
9.Treatment timing for kyphoplasty with bone cement injection in patients with thoracolumbar vertebral compression fractures

BACKGROUND:Related studies have shown that after kyphoplasty with bone cement injection, the vertebral height restoration is closely related to the injury time. Surgical timing also has an important influence on the incidence of postoperative complications. OBJECTIVE:To compare the clinical efficacy of kyphoplasty with bone cement injection at 2 and 2-4 weeks after thoracolumbar vertebral compression fractures, and to investigate the best timing for kyphoplasty. METHODS:Eighty-two thoracolumbar fracture patients, aged 55-85 years old, were included. Thirty-nine cases were subjected to kyphoplasty with bone cement injection within 2 weeks after injury. Another 43 cases were subjected to kyphoplasty with bone cement injection within 2-4 weeks after injury. The visual analog scale score, restoration of anterior and central vertebral height, volume and leakage of bone cement after treatment were compared between two groups. At 6 months after treatment, the daily activities of patients in the two groups were evaluated using Oswestry disability index. RESULTS AND CONCLUSION:Immediately and at the 6th month after treatment, the scores on the visual analog scale and the Oswestry disability index were lower than those before treatment (P < 0.05). The visual analog scale score immediately after treatment in the treatment group within 2 weeks was higher than that in the treatment group within 2-4 weeks (P< 0.05). After 6 months of treatment, there was no significant difference in the restoration rate of anterior and central vertebral height between these two groups, but the loss rate of the anterior and central vertebral height in the treatment group within 2 weeks was lower than that in the treatment group within 2-4 weeks (P< 0.05). Bone cement injection volume and leakage rate had no significant differences between two groups. These results demonstrate that patients appeared to have obvious pain after percutaneous kyphoplasty with bone cement injection within 2 weeks, but the percutaneous kyphoplasty with bone cement injection had smal influence on the short-term loss rate of vertebral height. Therefore, percutaneous kyphoplasty with bone cement injection with 2 weeks after injury is the optimal treatment timing for patients with thoracolumbar compression fractures.
10.Effect of Sox2 on invasion and migration of cervical cancer via Wnt signaling pathway

ABSTARCT:Objective To investigate the effects of the transcription factor SRY-related high-mobility-group box 2 (Sox2)related to stem cells on the invasion and migration of cervical squamous carcinoma cell line SiHa and its mechanism.Methods The expression of Sox2 was detected in Sox2 stably over-expressed cell line SiHa-Sox2 and negative control SiHa-EGFP cells by RT-PCR and Western blotting,respectively.The effects of Sox2 on the invasion and migration capacities of SiHa cells were detected by wound-healing assay and transwell assay.The expression of β-catenin was detected by Western blot.Results Compared with that of SiHa-EGFP cells,the expression of Sox2 at both mRNA and protein levels was obviously upregulated in SiHa-Sox2 cells.The migration and invasion capacities of SiHa-Sox2 cells were increased significantly (P <0.01 ),and the expression of β-catenin increased dramatically compared with that of the control cells.Conclusion Sox2 promotes the invasion and migration capacities of SiHa cells by increasing the expression of β-catenin and activating Wnt signaling pathway, which contributes to the development of cervical cancer.
