This study was aimed to explain the relationship between chemical component and therapy objective and to identify effective component group (ECG). The objective of this study is to build a new method to identify the ECG. Traditional Chinese medicine (TCM) formula and one of its treated diseases were selected as the re-search object. The entity grammar systems (EGS) were used as the theoretical framework. A grammatical method for ECG identification was constructed. The component-disease relationship was inferred and the ECG was identi-fied with these inference results. In this paper, 16 compounds which act to 12 proteins in Type 1 diabetes melli-tus (T1D) disease network were identified from Bai-Hu Tang plus Xiao-Ke Fang and 9 chemical compounds were determined as the candidate effective components group. The results indicated that this method can be used to identify ECG and provide a new way to elucidate the molecular mechanism of TCM formula.

OBJECTIVETo construct microRNA-223 overexpression and suppression lentivirus vectors and determine their effects after infecting oral squamous cell carcinoma (OSCC) cell line.
METHODSLentivirus vectors GV229 and GV232 were cut by the restriction sites of Age I and EcoR I and connected to the target gene, which contained mature microRNA-223 and microRNA-223 oligonucleotide. Real-time polymerase chain reaction (PCR) method was used to detect the microRNA-223 expression level after infecting the recombinant lentivirus vector into the OSCC cell line.
RESULTSThe successful construction of microRNA-223 recombinant lentivirus vectors was confirmed by the PCR method and DNA sequencing. HN-30 cell infected with microRNA-223 overexpression vector showed a significant increased in microRNA-223 expression, whereas HN-30 cell infected with microRNA-223 inhibitor vector suppressed microRNA-223 expression.
CONCLUSIONThe microRNA-223 overexpression and suppression lentivirus vectors are successfully constructed. These vectors could alter the expression level of microRNA-223 in OSCC cell line significantly, and provide a stable cell line for functional studies in the future.

OBJECTIVE To explore the resistance of Pseudomonas aeruginosa isolated in clinic against five antiseptics involving in povidone iodine(Iodophor),glutaraldehyde, chlorhexidine, symclosene(trichloroisocyanurate) and benzalkonium bromide.METHODS The susceptibility test of P.aeruginosa was determined by PhoenixTM-100 system.Minimun inhibitory concentration(MIC) of povidone iodine, glutaraldehyde, chlorhexidine, symclosene and benzalkonium bromide was detected by liquid dilution method.RESULTS The resistant rates of ampicillin/sulbactam,chloramphenicol,tetracycline and trimethoprim-sulfamethoxazole in 190 isolates of P.aeruginosa were all more than 98.0%.However,P.aeruginosa was to imipenem and meropenem were 15.3% and 6.8%.It was found that P.aeruginosa possessed the most resistant to glutaraldehyde and symclosene with its MIC50 being 32 ?g/ml and 64 ?g/ml.But the MIC50 of chlorhexidine and benzalkonium bromide were only 1 ?g/ml and 2.4 ?g/ml.Meanwhile,time-kill assays indicated that chlorhexidine could still produce rapid and powerful bactericidal effects at a concentration of 1MIC after 10 min treatment.CONCLUSIONS There are distinct differences in P.aeruginosa against povidone iodine, glutaraldehyde, chlorhexidine, symclosene and benzalkonium bromide. It is very important that antiseptics should be used rationally.Measurements should be taken to decrease dissemination of resistant bacteria and prevent nosocomial infection.

Functional targets are the objects that Chinese herbal medicines act directly upon. If the relationships between the properties of Chinese herbs and their functional targets were analyzed clearly, it would benefit the overall understanding of the holistic mechanisms of Chinese herbal treatments. In this paper, data regarding the properties of Chinese herbs and their functional targets were collected from the 2005 edition of The People's Republic of China Pharmacopoeia. After analyzing and assessing the data, the relationships were defined between the four qi, meridian entry and medicinal functional targets and between the four qi, five flavors and mode of function. Then the relationships between a single herbal medicine and a prescription were analyzed, and the results conformed with the traditional knowledge of Chinese herbal nature and efficacy. This demonstrated that the holistic mechanisms of the properties of Chinese herbs adhere to the findings, which may be beneficial for the development and compatibility of Chinese herbal medicines.

Chinese Journal of Clinical Infectious Diseases 2011;4(3):144-148
Objective To investigate the molecular mechanism of Klebsiella pneumoniae resistant to carbapenem. Methods The minimal inhibitory concentrations ( MICs) of the antimicrobial agents were determined by E-test. The 23 β-lactamase genes and 2 porin genes were amplified by polymerase chain reaction (PCR) , then the products were purified and their sequences were analyzed. Results The MICs of piperacillin, piperacillin/sulbactam, amoxicillin/clavulanic acid, cefoperazone/sulbactam, cefotaxime, cefepime and aztreonam to 5 strains of Klebsiella pneumoniae were all higher than 128 μg/mL, and those of imipenem or meropenem were higher than 32 μg/mL. All isolates carried blaTEM-1 and blaDHA-1 genes. Deletion of ompK35 and ompK36 were observed in Kp01 and Kp03, and the deletion of ompK35 was also observed in Kp02 and Kp05. Base insertion of ompK36 occurred in Kp02, Kp04 and Kp05. Compared with GenBank (GU945384) , ompK35 gene mutations of G→C at base 465 and T → C at base 466 in Kp04 lead to Gln to His substitution at position 155 and Tyr to its substitution at position 156, and it might be a new subtype. Conclusion The production of DHA-1 β-lactamase combined with the loss of OmpK36 or OmpK35 in porin genes may contribute to high-level carbapenem resistance in Klebsiella pneumoniae.

Objective To use bioinformatics methods to analyze large amounts of data generated by gene chips and to screen common key genes in hepatocellular carcinoma in human and rat. Methods For search of the medical literature,3 sets of gene chip with data which met our predetermined criteria were downloaded from the GEO database. The data were standardized by using the bioconductor and R version of the 2.10.1 version. The original data of the affymetrix platform were normalized with background correction, standardized and transformed into log2 by using the algorithm of the affy packages RMA.The TTEST function of the excel was then used to calculate the significance of each gene. The DAVID was used for gene ID conversion and a table was established for samples and the corresponding gene expression data.A meta analysis was performed to calculate the common genes of human and rat.An enrichment regulation pathway was gained with the KEGG in the DAVID library. Results There were 26 common expression genes in the development process of hepatocellular carcinoma in human and rat.Five of these genes were up-regulation genes, while twenty-one were down-regulation genes. An enrichment pathway, which is a focal adhesion pathway, was found and this pathway has been reported to be associated with development of hepatocellular carcinoma. Conclusion With bioinformatics,we were able to screen common key genes and a pathway which were closely related to development of hepatocellular carcinoma in human and rat.
