2.Pharmaceutical Care for a Postoperative Patient with Intracranial Glioma Treated by Chemotherapy

Objective:To investigate the role of clinical pharmacists in the process of chemotherapy for a patient with intracranial glioma. Methods:Usage and dosage of antitumor drugs, drug interactions, adverse drug reactions and medication education after dis-charged as the breakthrough points, clinical pharmacists and clinicians explored options for the patient treatment and adjusted the medi-cation timely according to the condition. Results:There were no serious adverse drug reactions in the adjusted therapeutic regimen and the treatment results of the patient were satisfactory. Conclusion:Clinical pharmacists provide pharmaceutical care can play an impor-tant role in improving the safety and effectiveness of drug use.

Solute Carriers (SLC) superfamily is one of the most important membrane transporter families in the cell membrane (including intracellular membrane) of human,it is involved in some essential physiological functions such as intercellular substance transporting,energy transfer,nutrition and metabolism,signal transduction and so on.Solute Carriers (SLC) superfamily contains 52 subfamilies,with more than 400 members.Studies have shown that abnormal protein expression or functional defects of SLC caused by human genetic mutations are closely related with a variety of serious diseases such as diabetes,hypertension and depression,so the function research of SLC had attracted much attention in recent years.The known three-dimensional structures of SLC transporters help to explain the precise molecular mechanism of substrate binding and transporting,it provides a fine structure basis to study the molecular mechanism of related diseases and structure-based drug discovery.In this review,we summarized the progress on the structure and function of SLC superfamily transporters of late years,try to provide some common rules of SLC superfamily transporters.
4.Establishment of malignant pleural effusion model of Lewis lung cancer based on Micro Echo Technology

Objective To establish a malignant pleural effusion model of Lewis lung cancer in C57BL/6 mice based on Micro Echo technology. Methods Single tumor cell suspension of Lewis lung cancer was injected into thoracic cavity.The pleural effusion was detected by Micro Echo technology.The volumes of effusion were quantified and the tumor cells were counted.Results After implanted of tumor cell, malignant pleural effusion can be detected by Micro Echo technology and observed after autopsy.Chemotherapy drugs such as Cyclophosphamide and Cisplatin can decrease the effusion volumes.Conclusion Pleural effusion model of Lewis lung cancer based on Micro Echo technology can be used to evaluate the efficacy of antitumor drugs.
5.The effect of endoplasmic reticulum stress in hyperlipidemia rats with acute pancreatitis

Objective To investigate the effect of endoplasmic reticulum stress in hyperlipidemia rats with acute pancreatitis(AP).Methods Forty-eight SD rats were divided into AP group(n=18,in- duced by injection of 40?tg/kg caerulein in normal rats twice with 2 hrs interval),normal control(n=6), byperlipidemia group(n=6,fed with a high-fat diet for 8 weeks)and hyperlipidemia AP group(n=18,in- duced by injection of 40?g/kg caerulein in hyperlipidemia rats twice with 2 hrs interval).The rats were sacrificed at 9,12,24 hrs(6 of each),respectively.Serum level of amylase was tested and the pathologi- cal changes of the pancreatic tissues were observed.The index of pancreatic apoptosis was assessed by TUNEL method.The expression of GRP78/Bip(glucose regulated protein)protein was determined by immunochemistry,the endoplasmic reticulum stress related molecules of XBP-1 splicing(X box binding protein),CHOP/GADD153(C/EBP-homologous protein or growth arrest and DNA-damage-inducible gene 153),caspase-12 were analyzed by RT-PCR.The dynamic expressions of GRP78/Bip and caspase- 12 were determine,d by Western blot.Results Artier 8 weeks of fat diet in hyperlipidemia rats,the ser- um levels of triglyceride[(0.99?0.38)mmol/L]and cholesterol[(3.17?0.18)mmol/L] were signifi- cantly increased(P
6.Posterior pedicle screw fixation for treatment of fracture and dislocation of atlantoaxial spine

Objective To investigate the clinical effect of pedicle screw fixation in the treatment of fracture and dislocation of atlantoaxial spine via posterior approach .Methods 19 patients with fracture and dislocation of atlantoaxial spine in this hospital from June 2011 to December 2013 were selected and treated with open reduction and pedicle screw fixation via posterior approach . The X‐radiographs were postoperatively re‐examined at regular time for understanding the correction of fracture and dislocation and implant fusion results ,the neurological functions were evaluated according to the Japanese Orthopaedic Association(JOA) scores . Results All cases got bony fusion without the occurrence of internal fixation loosening ,broken screw or broken rod .The JOA score was improved from preoperative (7 .35 ± 2 .39) points to postoperative (13 .21 ± 2 .53) points (P<0 .05) .Conclusion The posteri‐or atlantoaxial pedicle screw fixation and fusion for treating upper cervical spine injury has satisfactory effect .
7.Icariin induces apoptosis of esophageal cancer cell through enhancing Fas-FasL expression activity

Objective To investigate the inhibitory effect of icariin(ICA) on the xenograft tumors growth of esophageal car‐cinoma and to preliminarily investigate its mechanism .Methods The MTT assay and Giemsa staining were applied to detect and observe the in vitro inhibitory effect of ICA on esophagus cancer cell lines Eca‐109 and TE‐13 .The xenograft tumor model of nude mouse esophagus cancer cell was constructed and divided into 3 groups ,6 cases in each group .Each mouse in the experimental group was intraperitoneally injected by ICA 50 mg/kg ;while the control group was injected by the same volume of normal saline and the positive control group was injected by cis‐platinum 2 mg/kg ,once every 2 days ,a total of 14 days .The tumor volume was measured once per 3 d .After experiment ,the tumor weight was measured;the TUNEL staining was used to observe the morphological chan‐ges and cell apoptosis of tumor tissue in each group .The changes of Fas and FasL protein expression in tumor tissues were analyzed by immunohistochemistry .The FasL and IFN‐γlevels in peripheral blood were tested by the ELISA assay .Results ICA exerted no obvious inhibitory effect on the proliferation of Eca‐109 and TE‐13 cell in vitro .The average volume and weight of xenografts tumor had statistical difference between the experimental group and the positive control group (P<0 .05) .The TUNEL staining results showed that the tumor tissues had obvious apoptosis ,the number of apoptosis cells was significantly increased compared with the control group(P<0 .05) .The immunohistochemistry experimental results showed that the expression of Fas and FasL was signifi‐cantly increased(P<0 .05) .The ELISA experimental results demonstrated that the FasL and IFN‐γlevels of peripheral blood in the experimental group were significantly increased(P<0 .05) .Conclusion ICA had no obvious inhibitory effect on esophageal cancer cell proliferation in vitro ,but could induce in vivo apoptosis through the Fas expression and secretion of FasL and IFN‐γ,thus plays the role of anti‐esophageal cancer .
8.Effects of Sijunzi decoction and Danggui Buxue decoction on immunologic function in tumor-bearing mice treated by chemotherapy

Objective To explore the protective effect of Sijunzi decoction and Danggui Buxue decoction on the immunological function in tumor-bearing mice treated by cyclophosphamide.Methods Transplantable carcinomas animal model were constructed by inoculating tumor cell to mice for 24 hours,and randomly divided into five groups.Tumor-bearing control group were reveived normal saline 0.2 mL per mice one day by intraperitoneal injection,and normal saline 0.4 mL per mice each day by intragastric administration.Cyclophosphamide group were reveived CTX 20 mg per kg weight each day,and normal saline 0.4 mL per mice each day.Sijunzi decoction and Danggui Buxue Decoction low dose group were reveived CTX 20 mg per kg weight each day and traditional Chinese medicine 0.1 g per mice each day.Sijunzi decoction and Danggui Buxue Decoction mediate dose group were reveived CTX 20 mg per kg weight each day and traditional Chinese medicine 0.2 g per mice each day.Sijunzi decoction and Danggui Buxue Decoction high dose group were reveived CTX 20 mg per kg weight each day and traditional Chinese medicine 0.4 g per mice each day.The weight indices of thymus gland and spleen,NK-cell activity and the proliferation of T-lymphocyte in five groups were measured after ten 10 days treatment.Results The weight indices of thymus gland and spleen,NK-cell activity and the proliferation of T- lymphocyte were increased in all three Sijunzi decoction and Danggui Buxue decoction groups,especially in Sijunzi decoction and Danggui Buxue decoction mediate and high dose group,the differences between them and CTX group were statistically significan(P<0.05,P<0.01).Conclusion Sijunzi decoction and Danggui Buxue decoction can antagonize the decrease of immunological function in tumor-bearing mice treated by chemotherapy.
9.Evaluation of the proficiency of laboratories for detection of rabbit hemorrhagic disease virus antibody

Objective Through the detection of rabbit hemorrhagic disease virus( RHDV) antibody, to investigate the capacity of experimental animal quality control laboratories, so as to improve their detection proficiency.Methods According to the program approved by CNAS, the screened samples were numbered randomly and tested for their stability and homogeneity.The random samples were issued to the participant laboratories with the Standard Operation Procedure ( SOP) .The participant laboratories must submit the test reports and original records in time.The feedback results were judged by the rate of concordance with the anticipated results.Results Twenty laboratories from 14 provinces were en-rolled in the evaluation, and all of them submitted detection results on time.ELISA methods were used in 14 laboratories, and hemagglutination inhibition ( HAI) assay was used in 6 laboratories.The results of 17 laboratories were marked as pass or excellent, with a rate of pass of 85%.Conclusions The ability for detection of RHDV antibody in animal test labora-tories in China is high.The implementation of capacity testing can reflect the level of quality control laboratories.
10.Development and application of duplex PCR for detection of H-1 and KRV strains

Objective To develop a duplex PCR assay for detection of rat parvovirus H-1 and KRV and its application.Methods To design specific primers on the basis of H-1 ( NC_001358 ) and KRV ( U790330 ) genome sequences published in NCBI and establish a duplex PCR assay using H-1 and KRV DNA as templates.To verify the sensitivity and specificity of the method after optimizing PCR.The rats were infected by oral inoculation.The rats were divided into three groups:H-1 infection, KRV infection and mixed infection groups.To collect feces at the 4th, 6th, 8th and 10th days postinfection.Rats were euthanized on the 10th day and samples from heart, liver, spleen, lung, kidney and cecal contents were collected from each rat, then all the samples were screened with the duplex PCR.Results The 183 bp and 302 bp bands were amplified using H-1 and KRV as templates.The sensitivity test showed that the PCR method can detect as low as 3.8 pg/mL H-1 and 0.73 pg/mL KRV.There were no bands amplified when mouse minus virus, canine parvovirus and feline parvovirus were used as templates, showing that the specificity of the duplex PCR assay is very good. The nucleic acids of H-1 or KRV were detected in all rat feces on the 2th day postinfection and there was no obvious clinical symptoms in all the infected rats.The positive rates of H-1 were as follows:50%(4/8) heart tissues, 50%(4/8) liver tissues, 62.5%(5/8) spleen tissues, 50%(4/8) lung tissues, 37.5%(3/8) kidney tissues and 62.5%(5/8) cecum contents, and the positive rate of single infection group was higher than that of mixed infection group.The positive rates of KRV were as follows:0 (0/8) heart tissues, 25% (2/8) liver tissues, 87.5% (7/8) spleen tissues, 12.5% (1/8) lung tissues, 25%(2/8) kidney tissues and 62.5%(5/8) cecum contents, and the positive rate of mixed infection group was higher than that of single infection group.Conclusions The duplex PCR assay for H-1 and KRV established in this study can effectively detect H-1 or KRV infection in rat feces and other tissues, and can be used as an effective supplement to the national standard of lab animals.
