2.Construction and Identification of CD147 siRNA Expression Vector

Objective To construct CD147 siRNA expression vector in o rder to analyze the role of CD147 in the invasion and metastasis of tumors deriv ed from skin.Methods According to CD147 cDNA sequence in the Genebank,a pair of 64-nt oligonucleotides,each containing the sites of restriction endonucleas e at both ends,were designed and synthesized.Oligonucleotides were annealed an d ligated with linearized pSUPER by T4DNA ligase.The recombinants (named pSUPER /CD147 siRNA ) were finally sequenced and identified by PCR and restriction end onuclease digestion.Results CD147 siRNA expression vector was successfully co nstructed and identified by double endonuclease digestion.Sequence analysis of inserted fragment revealed the same sequence as synthesized siRNA oligonucleotid es.Conclusions CD147 siRNA expression vector has been successfully constructed,which paves the way for studying the role of CD147 in the invasion and metasta sis of tumor cells derived from skin as well as in tumor therapy.

5.Evaluation the quality of life of different age group of CRS patients and analysis of influential factors before and after the surgery

Objective To evaluate how functional endoscopic sinus surgery(FESS)modifies patients symptom profiles and the quality of life and analyze the influential factors.Methods During 2010 to 2012,90 cases were investigated to evaluate the quality of life of different age groups to use SNOT-20.Results The quality of life after the surgery of different age groups were significantly higher than it before the surgery(P<0.05).The juvenile group:first dimension(nasal congestion,70%),the second dimension (memory,30%),the third dimension(worried about the condition,30%)and the fourth dimension(social influence,40%).The young aged group:first dimension(nasal congestion,73.3%),the fourth dimension(social influence,43.3%),second dimension (quality of sleep,20.0%)and the third dimension(irritability,16.7%).The middle and old aged group:first dimension(nasal con-gestion,66.7%),the third dimension(trepidation,20%),the second dimension(quality of sleep,20.0%)and the fourth dimension (financial burden,40.0%).Preoperative QOL related factors:complications,age,gender,disease duration and urban-rural;Postop-erative QOL related factors:complications,disease duration,age,perioperative management,gender and urban-rural.Conclusion The quality of life of different ages after FESS is improved in different aspects.To increase the level of perioperative compliance can improve the quality of life.
6.Ultrasonic condition for extraction of flavone from Radix Astragali by orthogonal design

Objective:To ascertain the optimized ultrasonic condition for extraction of flavone from Radix Astragali by orthogonal design. Methods: The contents of calycosin-7-O-?-D -glucoside and formononetin were taken as the indices and were determined. The ultrasonic time (10 min, 20 min, and 30 min), concentrations of methanol (50%, 75% and 100%) and times of extraction (1, 2, and 3) were analyzed by orthogonal design; the best ultrasonic condition was ascertained and compared with those of soak extraction and Soxhlet extraction. Results: Ultrasonic with 100% methanol twice (20 minutes each time) was the optimized condition for extraction of flavone from Radix Astragali. The efficiency of ultrasonic extraction was better than those of soak extraction and Soxhlet extraction. Conclusion: Compared with other methods, the ultrasonic extraction of flavone from Radix Astragali is efficient, quick and simple.
7.CD147 Expression in Keratinocytes and Its Role in Keratinocyte Differentiation

Objective To investigate the expression and role of CD147in the differentiation of nor-mal keratinocytes and squamous cell carcinoma(SCC)cells.Methods CD147expression was analyzed immunohistochemically in20cases of verruca vulgaris(VV),various benign,premalignant and malignant epidermal tumors including21cases of seborrheic keratosis(SK),20actinic keratosis(AK),20Bowen' s disease(BD)and57squamous cell carcinoma.SCCs were classified using Broder's system of grading.Im-munoblot analysis was used to observe CD147expression in normal keratinocyte(HaCaT)and SCC cell(HSC-5)in vitro during their differentiation process induced by calcium.The effects of high-calcium medi-um culture and CD147antibody on the differentiation-related morphology of HaCaT and HSC-5cells were also observed.Results The same staining pattern of CD147was shown in20VV and21SK specimens as in normal epidermis.Positive CD147staining throughout the lesion was shown in4of20AK and7of20BD specimens.Positive CD147staining at the periphery of tumor nests was shown in8of20gradeⅠSCC specimens.CD147expressed throughout tumor nests in16of20gradeⅡSCC and all of the17gradeⅢSCC specimens.Immunoblot analysis revealed decreased CD147expression in both HaCaT and HSC-5cells during differentiation process induced by calcium.Not only high-calcium medium but also CD147antibody could induce differentiation-related morphology of both HaCaT and HSC-5cells in vitro.Conclusion These results suggest that CD147is a novel low-differentiation marker of keratinocyte,which might inhibit the dif-ferentiation of both normal keratinocyte and SCC cell.
8.Distribution of bevacizumab in ocular tissue and its toxic effect after injection of anterior chamber

Background Bevacizumab has been widely used in the treatment of new blood vessel disease in ophthalmology.The investigation of the pharmacokinetics and safety after intracameral injection of bevacizumab can offer the basis for the management of iris neovascularization and neovascular glaucoma.Objective The present study was to observe the distribution of bevacizumab(avastin)in eye tissue and toxic effects following the injection of anterior chamber.Methods Twenty-four New Zealand albino rabbits were divided into two groups randomly.0.05 ml (1.25mg)of Bevacizumab was intracamerally injected into the left eyes in the experimental group,and a balanced salt solution of 0.05 ml was injected in the same way into the left eyes of the control group.The anterior segment of eyes and ocular fundus were examined by slit-lamp microscope and direct ophthalmoscope after injection.Intraocular pressure was measured and corneal endothelial microscopy was performed before and after the injections.Five rabbits of the two groups were sacrificed on the first day,the fourth day,the seventh day,the fourteenth day,and the thirtieth day after injection,and the eyeballs were enucleated for histopathological examination.The ultrastructure of eye tissue was observed under the transmission electron microscope on the fourth day and the thirtieth day,and then immunofluorescence staining were performed to assess the distribution of bevacizumab in the eye tissues.This experiment complied with the Regulations for the Administration of Affair Concerning Experimental Animals by State Science and Technology Commission(Version 1988).Results No abnormality in the cornea,lens,vitreous and retina was observed after the injection of bevacizumab under the slit lamp microscope and direct ophthalmoscope.No significant differences were found in intraocular pressure and corneal endothelial cell density in the bevacizumab group compared with the control group before injection and 2 hours,1 day,7 days,14 days,30 days after injection(P =0.760,P =0.956).No histopathological and ultrastructural changes of the cornea,lens,chamber angle,iris,ciliary body and retina were seen after the injection in the experimental group and control group under the light microscope and transmission electron microscope.Bevacizumab was distributed in the anterior chamber angle,iris,ciliary body,choroid and retina in injected eyes and fellow eyes after intracameral injection with red fluorescence and presented the dynamic changes with the lapse of time.The immunofluorescence response of eye tissue to bevacizumab was weaker in the fellow eyes compared with injected eyes.Bevacizumab was mainly distributed in the vessel wall and lumen.Conclusions Bevacizumab can quickly distribute in the vascular tissue of the anterior chamber angle,iris,ciliary body,choroid and retina in injected eyes after intracameral injection without obvious toxic effects to eye tissue.Bevacizumab administered intracamerally may be a new strategy or a joint strategy for iris neovascularisation.
9.Olfactory function in patients with idiopathic rapid eye movement sleep behavior disorder.

OBJECTIVETo analyze the correlation between olfactory bulb (OB) volume with depth of olfactory sulcus (OS) and olfactory function in patients with idiopathic rapid eye movement sleep behavior disorder (iRBD).
METHODSFifty patients with iRBD and fifty controls were assessed with polysomnography (PSG). The results of olfactory function T & T testing, OB volume and depth of OS assessed with magnetic resonance imaging (MRI) were compared. SPSS 11.0 software was used to analyze the data.
RESULTST & T olfactory testing revealed that iRBD patients had higher scores (3.1 ± 0.5) than those in controls (0.6 ± 0.1), and the difference was significant (t = 7.913, P < 0.05). Both men and women with iRBD were affected by the same extent of olfactory loss (t = 1.015, P > 0.05). OB volume of left side in iRBD patients was (33.75 ± 4.11) mm(3), right side was (34.57 ± 4.21) mm(3), average OB volume was (33.94 ± 4.15) mm(3); OB volume of left side in controls was (51.68 ± 7.71) mm(3), right side was (52.31 ± 7.77) mm(3), average OB volume was (51.94 ± 7.74) mm(3); OB volume were lower in iRBD patients as compared to controls (t value were 9.013, 8.889 and 8.923, all P < 0.01). OS depth study revealed no statistical difference between iRBD patients and controls (t value were 0.923, 0.897 and 0.904, all P > 0.05). Olfactory discriminate threshold was negatively correlated with OB volume in iRBD patients (r = -0.61, P < 0.05), but no correlated with depth of OS (r = -0.24, P > 0.05).
CONCLUSIONSThe OB volume was lower in iRBD patients as compared to controls. The depth of OS showed no significant changes in iRBD patients. The OB volume was correlated with olfactory function, while the depth of OS was no correlated with olfactory function.

10.Protection of Transcutaneous Acupoint Electrical Stimulation for Brain Injury Undergoing Intervention: a Clinical Observation.

OBJECTIVETo observe the effect of transcutaneous acupoint electrical stimulation (TAES) combined dexmedetomidine on hemodynamic of intracranial aneurysmal subarachnoid hemorrhage patients undergoing intervention, and their protection for brain Injury.
METHODSTotally 108 intracranial aneurysmal subarachnoid hemorrhage patients undergoing intervention were randomly assigned to the electroacupuncture (EA) group and the control group according to random digit table, 54 in each group. All patients were anesthetized with dexmedetomidine. Patients in the EA group were needled at bilateral Neiguan (PC6), Lieque (LU7), and Yunmen (LU2). Parameter setting was as follows: The dilatational wave at 1. 5 Hz, strength 2 - 4 mA, 30 min. Systolic blood pressure (SBP), diastolic blood pressure (DBP), mean arterial pressure (MAP), and heart rate (HR) were compared between the two groups immediately after entry into the room (T0), after administration (T1), intubating (T2), resuscitation (T3), extubation (T4), and leaving the operating room (T5). Levels of S100β protein (S100β) and neuron specific enolase (NSE) were compared between the two groups at T0, immediately after surgery (T6), 6 h after operation (T7), 12 h after operation (T8), and 24 h after operation (T9).
RESULTSCompared with the same group at T0, SBP, DBP, MAP, and HR were significantly reduced in the two groups at T1-T5(P <0. 05), serum levels of S100β and NSE in the two groups were significantly increased at T6-T9 (P<0. 05). Compared with the control group at T1 - T5, SBP, DBP, MAP, and HR decreased in the EA group (P <0. 05). Compared with the control group at T6-T9, serum levels of S100β and NSE decreased in the EA group (P <0. 05).
CONCLUSIONTAES combined dexmedetomidine could effectively maintain stable hemodynamics of intracranial aneurysmal subarachnoid hemorrhage patients undergoing intervention, and regulate their serum levels of S100β and NSE.
