1.EFFECT OF GINSENOSIDE Rg1 ON LEARNING AND MEMORY IMPAIRMENT INDUCED BY β-AMYLOID PEPTIDE(25-35) AND ITS MECHANISM OF ACTION

AIM To study the effect of ginsenoside Rg1 on the learning and memory impairment in mice induced by aggregated β-AP(25-35). METHODS Mice were administered Rg1 (5, 10 mg*kg-1, ip) for 10 d and control mice received daily ip injections of saline after the intracerebroventricular injection of aggregated β-AP(25-35). After the final treatment, passive avoidance and performance in the Morris water maze (MWM) were assessed. and the activity of cortical and hippocampal ChAT and AchE were detected after the final behavior test. RESULTS Ginsenoside Rg1 (5, 10 mg*kg-1, ip) significantly ameliorated the learning and memory impairment induced by β-AP(25-35). Rg1 (5, 10 mg*kg-1) decreased the latencies and swim distances of mice to reach a hidden platform and improved the corresponding changes in search strategies occurred in the Morris water maze, and Rg1 (10 mg*kg-1, ip), increased step-through latencies also. Biochemical analysis showed that Rg1 (5, 10 mg*kg-1, ip) prevented the cortical and hippocampal ChAT activity decline induced by β- AP(25-35), and showed inhibition of the activity of AchE, although β-AP(25-35) showed no effect on the cortical and hippocampal AchE activity. CONCLUSION These data showed that ginsenoside Rg1 significantly improved the learning and memory impairment induced by β-AP(25-35), and this effect could be attibuted to its inhibition of AchE and increase of ChAT activity.
2.Feasibility of stem cells from umbilical cord blood as seed cells for tooth regeneration

BACKGROUND: With the development of tissue engineering, more attention has been paid to tooth regeneration. However, the resource of best seed cells is still uncertain. Therefore, alternative sources should be attached to intensive investigation. OBJECTIVE: To review the feasibility of stem cells from umbilical cord blood as seed cells for tooth regeneration. METHODS: A PubMed search was performed for articles published between January 1998 and January 2009. Key words were "tooth regeneration, seed cells, tissue engineering, umbilical cord blood, mesenchymal stem cells". Only studies written in English were included. Simultaneously, databases of China National Knowledge Infrastructure, Wanfang, and VIP published between January 1998 and January 2009 were also retrieved, using the key words of "tooth regeneration, seed cells, tissu engineering, umbilical cord blood, mesenchymal stem cells". Only studies written in Chinese were included. Totally, 40 literatures were included. RESULTS AND CONCLUSION: Seed cells, such as odontoblasts from dental germ, stem cells from dental pulp and deciduous teeth, and ectomesenchymal cells from the firstbranchial arch showed odontogenic differentiation potential in basic research. However, these cells are not practical to harvest in clinic. Although bone marrow mesenchymal stem cells have odontogenic capacity, their differentiation abilities significantly decrease with the increasing age of the donors. Stem cells from umbilical cord blood have many clinical advantages over bone marrow mesenchymal stem cells, and exhibited typical mesenchymal stem cells characteristics. Thus, we propose the hypotheses that stem cells from umbilical cord blood could be induced into odontogenic lineage and might be used as suitable seed cells for tooth regeneration to replace the lost tooth.
3.Effect of Gongxuening capsule on inflammation and bleeding of instant placement IUD in cesarean section

Objective To explore the effect of Gongxuening capsule on inflammation,postpartum hemorrhage and the amount of lochia duration of instant placement intrauterine device(IUD)in patients with cesarean section.Methods 210 cases of cesarean section immediately placed IUD patients as the research object,were divided into the observation group and the control group by simple random number table method,control group of 105 patients with cesarean section immediate postplacental IUD, observation group were given Gongxuening gapsule on the basis of the patients in the control group.The inflammatory cytokines(IL-6,IL-8,IL-10,TNF-α)levels,the amount of postpartum hemorrhage,puerperal pathology and duration of lochia related indicators were compared between two groups of patients.Results After treatment,the IL-6,IL-8,IL-10,TNF-αlevels in observation group were significantly lower than that of control group(P<0.05),postpartum hemorrhage 24h were significantly less than the control group(P<0.05).There was no significant difference in average body temperature,white blood cell count,neutrophil ratio and puerperal disease rate between observation group and control group.The maternal postpartum <28 days wa 33 cases,which was higher than 26 cases in control were,the difference was statistically significant (P<0.05).There was no significant difference in maternal postpartum from 28 d to 42 d,or >42 d between two groups.Conclusion Gongxuening capsule helps reduce postpartum hemorrhage,the degree of inhibition of inflammatory reaction in patients of cesarean section immediately placed IUD.
4.A study on non-T-cell depleted haploidentical bone marrow transplantation using Basiliximab for GVHD prophylaxis

Objective To explore the effects of Basiliximab (Simulect) on reducing the incidence of severe acute GVHD in haploidentical bone marrow transplantation (BMT). Methods Nine patients with leukemia received haplotype Allo-BMT from HLA two or three loci mismatched related donor. Most patients were classified as high risk category. The donors of patients were administrated with G-CSF 250 ?g/day for 7 doses prior to marrow harvest. In addition to combination of CsA, MTX, ATG and Mycophenolate mofetil for GVHD prophylaxis, Simulect was administered to prevent severe GVHD. A total 40 mg Simulect was given in two doses of 20 mg each by 30 min intravenous infusion on 2 h before transplantation and day 4 after transplantation.Results All patients were engrafted. 100 % donors hematopoietic cells after transplantation was determined by cytogenetic evidence analysis. None developed the Ⅱ-Ⅳ acute GVHD. Eight patients could be evaluated for chronic GVHD. All experienced chronic GVHD confined to the skin. The median follow-up duration was 14 months (range 12~20 months). One patient died from CMV infection on 3 months and one patient died from disease relapse on 14 months. The remaining 7 patients were survived in disease free situation.Conclusion The use of Simulect in haploidentical bone marrow transplantation is effective on preventing acute severe GVHD and improving disease-free survival.
5.Basiliximab for prophylaxis of graft-versus-host disease in mismatched bone marrow transplantation

Objective To investigate the effects of Basiliximab on alloreactive T lymphocytes precursors, hematopoietic progenitor cells by ex vivo assay and prophylaxis of graft-versus-host disease (GVHD) in mismatched bone marrow transplantation.Methods Two groups were set up: (1) Basiliximab group including 72 leukemia patients subject to haploidentical bone marrow transplantation (BMT) with Basiliximab for prophylaxis of GVHD without ex vivo T-cell depletion. (2) Control group having 15 patients receiving the same regimen before Nov. 2000, without Basiliximab for GVHD prophylaxis. Limiting-dilution method was used to determine the effect of Basiliximab on reactivity of cytotoxic T lymphocyte precursors (CTLP). In the semi-solid hematopoietic culture system, the effect of Basiliximab on the colony proliferation of CFU-GM, BFU-E and CFU-Meg was measured. Results In Basiliximab group, 72 patients established trilineage with full donor hematopoietic reconstitution. Engraftment rate showed no statistical difference between the two groups. 6.Research progress on biological function of PARP3

As a member of PARP superfamily, PARP3 shares a high homology with PARP1 and PARP2, which are all DNA-dependent enzymes that are catalytically activated by DNA strand breaks.Compared to PARP1 and PARP2, PARP3 exerted some special properties in tissue expression pattern and biological function.The evidence has shown that PARP3 could be activated by DNA double strand breaks and special DNA single strand breaks and synthesize mono(ADP-ribose) (MAR) covalently attached to target proteins including itself.PARP3 plays an important role in DNA double strand breaks, DNA single strand breaks, activation of PARP1 and development of nervous system.It has been reported that PARP3 is associated with glioma and breast cancers.In this review, PARP3 structure, activation mechanism, biological function and its relationship with diseases will be presented.

Diabetic cognitive dysfunction is one of the important complications of diabetes mellitus .Insulin plays an important protective role in cognitive function through MAPK and PI 3-K/Akt signaling pathway .Insulin resistance may give rise to excessive tau protein phosphorylation , formation of neurofibrillary tangles inhibits insulin degrading enzyme , reduces the degradation of amyloid β;enhances oxidative stress , further damages the integrity of the neu-ronal structure and function and eventually leads to cognitive dysfunction .
8.RAPD Analysis on different cultivars of Cornus officinalis

Objective To analyze the genetic relationship among the different cultivars of Cornus officinalis and provide some foundation for heredity breeding. Methods A modified method of extracting total DNA from the leaves of C. officinalis was selected by improving the traditional method-CTAB, the total DNA was analyzed by RAPD; the genetic similarity correlation was calculated by SPSS 10.0 DICE method, cluster analyses were carried out using Between-groups linkage method, and the genetic dendrogram was established. Results Random primers (22 10-bp) were selected to be used for the PCR, a total of 133 bands were amplified by 12 samples, among which 75 bands were polymorphic, accounting for 56.39%. The cluster analysis indicated that: spindleform itself was one group, short pear-shape and short cylindericform clustered together, the rest longer types clustered together, which reflected the result of artificial selection in the course of cultivating. Conclusion The result keeps accordance with the biologic character and territorial distribution of C. officinalis cultivars; RAPD analysis is an assistant mean for seed breeding of C. officinalis.
9.ALLOGENEIC BONE MARROW TRANSPLANTATION USING G-CSF MOBILIZED MARROW

20?10 9 /L was (18 1?3 0) days in the recipients.The incidences of grade I GVHD and II GVHD were 28 6% and 5 7%, respectively. Furthermore, the percentages of CD34 + and CD4 + cells in G CSF stimulated bone marrow increased, concomitant with a significant decline in CD8 + cell count. These data demonstrated that G CSF mobilization remodeled the density of hematopoietic progenitors and T lymphocyte subsets in the bone marrow, which in turn accelerated hematopoietic reconstitution and minimized the incidence of severe acute GVHD after allogeneic transplantation.Transplantation of marrow harvests from donors who are pretreated with G CSF might be a feasible measure for a successful allogeneic bone marrow transplantation.
10.Study on quality standard for Qingdai Powder

AIM: To establish the quality standard for Qingdai Powder (Indigo Naturalis, Radix Glycyrrhizae, Borax, Boreolum Syntheticum, Herba Menthae, Rhizoma Coptidis, Catechu, etc.) METHODS: Indigo Naturalis, Borneolum Syntheticum, Rhizoma Coptis, Radix Glycyrrhizae were indentified by TLC, and the content of indigo was determined by HPLC. RESULTS: Indigo Naturalis, Borneolum Syntheticum, Rhizoma Coptis, Radix Glycyrrhizae could be indentified by TLC. Indigo showed a good linear relationship at a rang of 27.35~ 410.25ng, r=0.9997. The average recovery of indigo was 99.03%( n =6) and RSD was 2.14%. CONCLUSION: The method is simple, accurate and high specificity with good repeatability. The method can be used for quality control of Qingdai Powder.
