1.Baicalein induced Nrf2 activation and its protection against hepatotoxicity

Aim To study the activation of Nrf2 in-duced by baicalein ( BAI ) , and its protection against carbon tetrachloride ( CCl4 ) , ethanol and acetamino-phen ( APAP )-induced hepatotoxicity. Methods A reporter gene assay was conducted in human normal liver L-02 cells to detect the activation of transcription factor Nrf2 induced by baicalein. APAP ( 10 mmol · L-1 ) , CCl4 (10 mmol·L-1 ) and Ethanol (100 mmol · L-1 ) were used to induce hepatotoxicity in L-02 cells. After the pre-incubation with Baicalein (0, 1, 10, 25, 50, 100 μmol·L-1 ) for 15 min, cells were administrated with or without those above hepatotoxins. 48 h later, cell viability was detected by 3-(4, 5-dim-ethylthiazol-2-yl ) 2 , 5-diphenyltetrazolium bromide (MTT) method. Results Baicalein (25, 50 μmol· L-1 ) induced the activation of Nrf2 ( P <0. 01 , P <0. 05) in the reporter gene assay. As compared with control, three hepatotoxins ( APAP, CCl4 , Ethanol ) all decreased cell viability ( P<0. 01 ) , and baicalein significantly reversed such decreases in a concentra-tion-dependent manner ( P<0. 01 ) . Conclusion Ba-icalein can induce the activation of transcription factor Nrf2 , which is probably one of the mechanisms con-tributing to the protection of baicalein against hepato-toxins (APAP, CCl4, Ethanol)-induced hepatotoxici-ty.
2.Aminoglycoside-modifying Enzyme and 16S rRNA Methylase Gene Expressions in Acinetobacter baumannii

OBJECTIVE To understand aminoglycoside-modifying enzyme and 16S rRNA methylase gene expressions in Acinetobacter baumannii in Xinjiang region.METHODS 20 A.baumannii strains were isolated and test the anti-bacterial drug sensitivity.PCR methods were used to test aminoglycoside-modifying enzyme and 16S rRNA methylase gene.RESULTS From thirteen A.baumannii strains detected the aminoglycoside-modifying enzyme genes,the detection rate was 65%;in which aac(3)-Ⅰ gene was positive in 4 strains(20%),aac(3)-Ⅱ gene in 8 strains(40%),aac(6′)-Ⅰad gene in 4 strain(20%),ant(3″)-Ⅰ gene in 4 strain(20%),and ant(2″)-Ⅰ gene was positive in 1 strain(5%).The aac(6′)-Ⅰ b and aac(6′)-Ⅱ gens and were not detected;the 16S rRNA gene methylation was negative.CONCLUSIONS There are aminoglycoside-modifying enzyme genes existing in A.baumannii,no 16S rRNA methylase gene was detected in Xinjiang region.
3.Character of cardiac valvular calcification in uremia patients and its influence on cardiac structure and function

Objective:To evaluate the character of cardiac valvular calcification in uremia patients and its effect on cardiac structure and function.Methods:Thirty five cases with mitral and aortic valve calcification were selected from 326 uremia patients.Control group consisted of 33 uremia patients without valvular calcification.Their age,LVEDD,LAD,AOD,LVWT,LVEF and blood pressure matched with calcification group.The observing indexs included distribution of calcification,degree of regurgitation,LVEDD,LAD,AOD,LVWT and LVEF.The observing duration was one year.Results:The results showed that characters of valvular calcification with uremia were similar to elder valvular calcification.The degree of regurgitation was more greater in calcification group than in control group after one year.The LAD was greater in calcification group after one year.Conclusion:Cardiac valvular calcification of uremia may cause the increase of regurgitation and enlarge LAD. 4.Expert consensus on 3D-printing template assisted CT-guided radioactive 125I seed implantation brachytherapy

As an interstitial brachytherapy,radioactive seed implantation could provide high doses in the local site and minimal doses at surrounding normal tissues.It has become one of the best choice for early stage prostate carcinoma.Radioactive seeds were implanted under the guidance by ultrasound,CT and MRI,featured with surgical and interventional treatment.Based on dosimetry for target and organs at risk,radioactive seed implantation is multiple disciplinary.In order to acquire the accurate and high quality seed implantation,it is necessary to set up a team including surgeons,radiation oncologists,interventional doctors and nuclear medicine doctors.The content of the consensus is as follows:radiation physics and dosimetry,indications,side-effects and 3 D-printing template work-follow.Despite the benefit of radioactive seed implantation for solid carcinoma,there still a compelling need for prospective randomized and stage Ⅲ clinical trials from multiple centers,so as to upgrade the evidencebased level,above all confirm the role of radioactive seed implantation in the comprehensive treatment of tumors.
5.Dosimetry evaluation of navigation system-assisted and CT-guided seed implantation in the treatment of recurrent malignant pelvic tumors

Objective:To explore the accuracy of CT-guided 125I seed implantation assisted by a navigation system and 3D-printing template in the treatment of recurrent malignant pelvic tumors by comparing pre-plan and intraoperative physical dosimetric parameters. Methods:This study involved 15 patients with recurrent malignant pelvic tumors who received CT-guided radioactive 125I seed implantation assisted by a navigation system and 3D-printing template in the Peking University Third Hospital from Dec 2018 to Feb 2020.Seven of the patients had cervical cancers, seven had rectal cancers, and one had prostate cancer.The median age was 55 years (34-84 years old). The prescription dose was 100-150 Gy.The pre-plan and post-implant data were compared, including the number of implanted seeds, implantation needle number, and some dosimetric parameters such as the minimum prescription doses delivered to 90% and 100% of target volume( D90, D100), mean percentages of volume receiving 100%, 150%, and 200% of the prescription doses( V100, V150, and V200), conformity index(CI), external index(EI), and homogeneity index(HI)of the target volume. Results:The median lesion volume was 29.20 cm 3, the median seed number was 54, and the median D90 was 150 Gy.The post-implant V150 was lower than pre-plan V150 (64.1% vs. 67.1%, t=2.937, P=0.011), and the post-implant mean HI was higher than pre-plan HI (32.01% vs. 26.68%, t=-2.950, P=0.011). There were no significant differences in other dosimetric parameters before and after seed implantation. Conclusions:With CT-guided radioactive seed implantation assisted by a navigation system and 3D-printing template in the treatment of recurrent malignant pelvic tumors, the actual postoperative dose could meet the preoperative plan requirement, ensuring the accuracy and consistency of the dose delivered.
6.Interaction of butylphthalide with rat and human liver CYP450 isoenzymes.

The work aims to study the drug metabolizing enzymes involved in the metabolism of butylphthalide and evaluate the induction and inhibition activities of butylphthalide on CYP450 isoenzymes by using in vitro (liver microsome incubation system of rats and human) and in vivo (CYP induced model of rats) method. Butylphthalide was incubated with selective inhibitors of CYP450, and its metabolic rate was determined to identify the metabolizing isoenzymes of NBP in rat (normal and induced rats) and human liver microsomes. The in vitro inhibition effect of butylphthalide on 6 main liver microsomal CYP450 isoenzymes was evaluated by using probe drugs; the induction and inhibition activities in vivo of butylphthalide on CYP450 isoenzymes were evaluated by NBP ig dosing (160 mg x kg(-1)) and iv dosing (20 mg x kg(-1)) in rats. After adding the specific inhibitors of CYP2C11, 2E1 and 3A 1/2 for rat, CYP2C19, 2E1 and 3A4/5 for human, the metabolism of NBP in rat and human liver microsomes were reduced 38.8%, 86.2%, 78.4% and 51.0%, 92.0%, 58.9% of control, respectively. The metabolic rates of NBP in CYP2E1 and 3A 1/2 induced rat liver microsomes were increased 25.5% and 68.9%. High concentration of NBP (≥ 200 μmol x L(-1), in vitro) could inhibit the activities of CYP1A2, 2C6, 2C11 and 2D2 in rats, and high concentration of NBP ( ≥ 15 μmol x L(-1), in vitro) could inhibit the activity of CYP2C19 in human. All the results indicated that NBP should be mainly metabolized by CYP2E1, 2C11 and 3A 1/2 in rats and CYP2E1, 2C19 and 3A4/5 in human. High concentration of NBP could inhibit human CYP2C19 in vitro. No significant induction/inhibition effects of NBP were observed on rat liver CYP450 isoforms after ig 160 mg x kg(-1) NBP or iv 20 mg x kg(-1) NBP.

7.Influence of Excessive Na Stress on Growth and Element Accumulation of Three Species of Ectomycorrhizal Fungi

This study deals with colony expansion characteristics, biomass increment and Na accumulation in mycelia of three ectomycorrhizal fungi ( Boletus edulis, Xerocomus chrysenteron and Gomphidius viscidus ) under treatment of Na2SO4 and NaCl. The experimental results showed that no impact was observed from Na2SO4 and NaCl treatments on the growth mode of the mycelia, but the biomass increment of X. chrysenteron and G. viscidus was significantly restrained under 0.1 mol/L Nacl. Under the treatment of Na2SO4, biomass increment of X. chrysenteron is significantly higher than the control, but the biomass of G. viscidus is lower than that of the control, and no significant impact was measured on growth of B. edulis under both treatments of Na2 SO4 and NaCl. The experiment also indicated that Na accumulation in mycelia varied significantly among the three tested strains, highest Na accumulation was measured in X. chrysenteron under treatment of NaCl, while under the treatment of Na2SO4 , accumulation of Na in B. edulis is much higher than the other two strains.
8.Purification of Recombinant Fusion Protein Staphylokinase-Hirudin Expressed by Escherichia coli and Analysis of its Dimer

The recombinant fusion protein staphylokinase-hirudin(rSFH) was purified from the high density-fermented engineered E.coli by means of ion-exchange chromatography (IEC) and gel filtration (GF). The purity of rSFH reached to more than 98% determined by RP-HPLC and SDS-PAGE, and the yield was up to 0.7g per liter of fermentation broth. The analysis of homologous dimmer of rSFH appeared during the purification and calculation of the surface hydrophobic area had been carried out by means of hydrophobic chromatography and MALD-TOF. The influence of sodium chloride and temperature on the behavior of rSFH reversible dimerization was analyzed by high performance sized- exclusive chromatography(HPSEC). It is concluded that the hydrophobic interaction played an important role in the reversible dimerization of rSFH.

Organic solvent tolerant microorganism(OSTM) is a novel extremophile and it hasn't been systematically studied until 1980s.Relying on certain mechanisms,the OSTM is able to effectively de-fend and decrease the toxicity from organic solvents,which enable the OSTM to be potentially applied in the industrial fields such as whole-cell catalysis and environmental treatment,etc.The comprehen-sively understanding of the mechanisms involved in organic solvent tolerance of OSTM could be com-bined with genetic engineering in order to modify and optimize the various specifications of OSTM,and further broaden its application in other industrial areas.Latest studies on the tolerant mechanisms of OSTM,in this paper,will be reviewed from four aspects such as vesicle exocytosis and changes of phos-pholipid composition in membrane,etc.Besides,the application of OSTM in whole-cell catalysis and other fields will be introduced.
