1.Breeding Aspergillus oryzae Strain with High Protease Activity by Ultraviolet Induced Mutation

Based on the assessment of protease (acid,neutral and alkaline protease) activity and amylase activity (?- amylase and ?- amylase),one Aspergillus oryzae K61 was selected from five wild Aspergillus oryzae which are original from spontaneous fermented soybean paste.One Aspergillus oryzae Y29 was screened with ultraviolet radiation.The Aspergillus oryzae Y29 can produce higher protease activity than original Aspergillus oryzae K61,and its hereditary stability is higher.Compared with Aspergillus oryzae 3.042,the Aspergillus oryzae Y29 showed higher protease activity,however,the both strains did not have significant difference on the amylase activity,growing speed of the strain and number of spore.Both strains were applied to producing soybean paste.The soybean that was produced by Aspergillus oryzae Y29 has full-bodied soybean paste flavor,and the soluble nitrogen (0.77g/100ml) is higher than the Aspergillus oryzae 3.042.

3.Antimicrobial resistance monitoring of bacteria isolated from blood culture in Xinjiang area during 2013

Objective To understand the distribution characteristics and drug resistance of bacteria isolated from blood cul‐ture in Xinjiang area during 2013 .Methods The identification of isolated bacteria were performed by adopting the France VITEK‐Compact and the ABI series bacterial identification instruments .The antimicrobial susceptibility test was carried out by using the minimum inhibitory concentration (MIC) and Kirby‐Bauer (K‐B) methods .Results A total of 3 962 strains of bacteria were isola‐ted from clinical blood culture ,in which Gram‐positive bacteria and Gram‐negative bacteria accounted for 50 .8% and 49 .2% respec‐tively .The most frequent strains were coagulase‐negative staphylococci (31 .7% ) ,Escherichia coli (23 .2% ) ,Staphylococcus aureus (9 .5% ) ,Klebsiella pneumoniae (8 .7% ) ,Acinetobacter baumannii (3 .6% ) ,Enterobacter cloacae (2 .5% ) ,Enterococcus faecium (2 .5% ) ,Pseudomonas aeruginosa (2 .2% ) ,Enterococcus faecalis (2 .0% ) and Streptococcus pneumoniae (1 .1% ) .The detection rate of extended‐spectrum beta‐lactamase (ESBLs) producing Escherichia coli ,Klebsiella pneumoniae and Proteus mirabilis were 69 .8% ,62 .6% and 66 .7% respectively .The detection rates of methicillin‐resistant Staphylococcus aureus(MRSA) and methicillin‐resistant coagulase‐negative Staphylococcus (MRCNS) was 36 .2% and 86 .3% respectively .The pan‐drug resistant (XDR) strains of Acinetobacter baumannii ,Pseudomonas aeruginosa ,Klebsiella pneumonia were 14 strains(9 .9% ) ,1(1 .2% ) ,2 strains(0 .1% ) ,16 strains(0 .6% ) .No strains resistant to vancomycin or linezolid were found in Staphylococcus and Enterococcus faecalis .Conclusion Among blood culture isolated bacteria in Xinjiang area ,the proportion of Gram‐positive bacteria and Gram‐negative bacteria have little difference .The diversity of bacterial species exist .The resistance to commonly used antibiotics is serious .The distribution situ‐ation of blood culture isolated bacteria should be timely understood .The bacterial drug resistance monitoring should be strengthened to control the nosocomial infections ,guide rational drug use in clinic and control the generation and spread of drug resistant bacterial strains .

5.Problems and solutions on transformation of scientific research achievements of acupuncture.

With more and more attention and investment on acupuncture scientific researches, considerable outcomes and achievements has been acquired, but the shortcoming of low transformation rate of acupuncture research achievements is gradually exposed. Nowadays there is no related report on this problem, so based on achievement translational research in other areas and practical situation of acupuncture, the existing problems and solutions are analyzed. As a result, the existing problems include (1) the research content is mainly basic research and clinical research but less acupuncture device research, leading to limited transformation efficiency; (2) the evaluation system and transformation pattern are still needed to be perfect. The solutions are (1) to properly evaluate the research achievements of acupuncture, (2) to advocate the concept and method of translational medicine, (3) to reform the policy and system, and (4) to establish valid platforms covering research, outcomes and transformation.

6.Percutaneous screw fixation for the treatment of pelvic fractures under C-arm fluoroscopy.

OBJECTIVETo investigate the applications of percutaneous screw fixation for the treatment of pelvic fractures and its related surgical considerations.
METHODSFrom June 2010 to June 2012,19 patients with pelvic fractures were treated with percutaneous hollow screws. There were 13 males and 6 females, with an average age of 41 years (ranged from 22 to 58 years). Fractures were caused by traffic accidents in 11 cases, by falling down from high place in 8 cases. Based on the Tile classification, there were 15 cases of Tile C type and 4 case of Tile B type. The indexes such as screw inserting time, intraoperative blood loss, complications, functional recovery and reduction conditions were observed. Fixation methods included sacroiliac screws, cannulated screw fixation of the pubic ramus and cannulated screw fixation of the pubic symphysis separation.
RESULTSAnatomical reduction achieved in 7 cases, satisfactory reduction 11 cases, and unsatisfactory reduction 1 case. Union time of fracture union ranged from 8 to 12 weeks (mean 10 weeks). Wound infection,ununion of fracture and nerve injuries were not found. According to the Majeed standards, 12 patients obtained an excellent results, 6 good and 1 fair.
CONCLUSIONPercutaneous screw fixation for the treatment of pelvic fractures under fluoroscopy has several advantages such as less trauma, less blood loss, fewer rates of complications, reliable fixation and no blood transfusion, which can reconstruct the stability of the pelvic ring, but it needs adequate preoperative preparation and high requirements for the surgeon.

7.Role of Epidermal Growth Factor in Invasion and Metastasis of Pancreatic Cancer Cells

Objective Overexpressions of epidermal growth factor(EGF)and EGF receptor have been associ- ated with progression and invasive phenotype of pancreatic cancer.However,the underlying molecular mechanism by which EGF worked in pancreatic cancer cells has not been completely understood.In this study,effect of EGF on the invasion and metastasis of pancreatic cancer cells and its regulatory mechanism were investigated.Methods The effects of EGF on the proliferation,adhesion and invasion of pancreatic cancer cells were detected by WST-1 prolif- eration assay,adhesion assay and invasive assay,respectively.The activity and expression of MMP-2 and MMP-9 were examined by zymography,Western blot and RT-PCR,respectively.The activity of NF-?B was examined by EMSA.Results EGF could significantly promote the invasiveness of pancreatic cancer cells but did not affect cell proliferation or adhesion.The expressions of NF-?B and MMP-9 were significantly increased by EGF,but EGF did not affect the activity and expression of MMP-2.Furthermore,EGF stimulated the NF-?B binding activity.Pre- treatment with NF-?B inhibitors,pyrrolidine dithiocarbamate(PDTC),could significantly inhibit the activity of NF-?B induced by EGF.Meanwhile,the EGF-induced expression and activity of MMP-9,as well as cell invasiveness were also inhibited by NF-?B inhibitor.Conclusion EGF could increase the expression and promote the invasiveness of MMP-9 via the activation of NF-?B in pancreatic cancer cells,which implies that NF-?B inhibitant,such as PDTC,may diminish the invasiveness of pancreatic cancer cells.

Aim The effects of midazolam on the whole-cell sodium currents in rat sympathetic neurons were studied to explore the mechanisms where by midazolam mediates hypotension. Methods Whole-cell patch-clamp recordings were performed on enzymatically isolated rat superior cervical sympathetic neurons. Objective:To explore the accuracy of CT-guided 125I seed implantation assisted by a navigation system and 3D-printing template in the treatment of recurrent malignant pelvic tumors by comparing pre-plan and intraoperative physical dosimetric parameters. Methods:This study involved 15 patients with recurrent malignant pelvic tumors who received CT-guided radioactive 125I seed implantation assisted by a navigation system and 3D-printing template in the Peking University Third Hospital from Dec 2018 to Feb 2020.Seven of the patients had cervical cancers, seven had rectal cancers, and one had prostate cancer.The median age was 55 years (34-84 years old). The prescription dose was 100-150 Gy.The pre-plan and post-implant data were compared, including the number of implanted seeds, implantation needle number, and some dosimetric parameters such as the minimum prescription doses delivered to 90% and 100% of target volume( D90, D100), mean percentages of volume receiving 100%, 150%, and 200% of the prescription doses( V100, V150, and V200), conformity index(CI), external index(EI), and homogeneity index(HI)of the target volume. Results:The median lesion volume was 29.20 cm 3, the median seed number was 54, and the median D90 was 150 Gy.The post-implant V150 was lower than pre-plan V150 (64.1% vs. 67.1%, t=2.937, P=0.011), and the post-implant mean HI was higher than pre-plan HI (32.01% vs. 26.68%, t=-2.950, P=0.011). There were no significant differences in other dosimetric parameters before and after seed implantation. Conclusions:With CT-guided radioactive seed implantation assisted by a navigation system and 3D-printing template in the treatment of recurrent malignant pelvic tumors, the actual postoperative dose could meet the preoperative plan requirement, ensuring the accuracy and consistency of the dose delivered.
