1.Effect of Antisense Oligodeoxynudeotides Targeting Nuclear Factor ?B on Expression of Caspase-3 in Glomerulosclerosis

Objective To investigate the effect of antisense oligodeoxynudeotides(ODN)targeting nuclear factor ?B(NF-?B)on the expressions of Caspase-3 and NF-?B in glomerulosclerosis.Methods Male SD rats were divided into 3 groups:sham operation(group A,n=6),glomerulosclerosis(group B,n=6),intervention(group C,n=9).Group C was divided into 3 groups:sense ODN(group C1,n=3),non-sense ODN(group C2,n=3),antisense ODN(group C3,n=3).Glomerulosclerosis models were made for SD rats by unilateral nephrectomy and being injected with adriamycin into caudal vein.After 8 weeks,various ODN applied to corresponding group for intervention.At the end of the 9th week,kidneys were taken out from all rats for the measurement of expressions of NF-?B p65 and Caspase-3 by immunohistochemistry staining.Results After intervention,on the d7,the rats urine protein,glomerular sclerosis index(GI)and expressions of Caspase-3,NF-?B p65 in group C3 were significantly lower than those in group B,C1,C2(Pa0.05).Conclusion Antisense ODN targeting NF-?B can inhibit the activities of NF-?B,Caspase-3 in rats kidneys and retard glomerulosclerosis and glomerular intrinsic cell apoptosis.
2.Endothelial nitric oxide synthase gene polymorphism and acute myocardial infarction.

Objective To investigate the association of the Glu298-Asp(894G→T)mutation at exon 7 of the endothelial nitric oxide synthase gene and acute myocardial infarction.Methods By using the designed primers based on flanking sequences of the Glu298-Asp mutation at exon7 of eNOS, 894G→T fragments were amplified by nested PCR from genomic DNA of healthy control and AMI subjects, digested by restriction enzyme after amplification and detected by agarose gel electrophoresis for 894G→T genotyping. To count the genotype and allele frequencies, the significance of difference in the genotype and allele between two groups were assessed by statistical analysis.Results Three genotypes containing GG, GT, TT were identified on the 894 site of the eNOS gene exon 7 in both AMI and controls. In AMI group, there were 25 patients having 9 homozygotes, the other 16 having heterozygotes. In controls, there were 13 cases with G894T mutation,all having heterozygotes. 5.Standardization of Clinical Application of Fat-soluble Vitamin for Injection(Ⅱ)in Our Hospital by Using PDCA Cycle

OBJECTIVE:To evaluate the effects of PDCA cycle management method on clinical application of Fat-soluble vita-min for injection (Ⅱ). METHODS:By retrospective analysis,500 discharge medical records of Fat-soluble vitamin for injection (Ⅱ)were collected from our hospital during Jul.-Aug. in 2015(before intervention),Sep.-Oct. in 2015(after first intervention cy-cle),Nov.-Dec. in 2015(after second intervention cycle),respectively. RESULTS:After 2 PDCA cycles,the proportion Fat-solu-ble vitamin injection (Ⅱ) use without indication decreased from 65.4% to 39.2%;the reasonable rate of drug dosage increased from 53.2% to 97.2%,and that of usage increased from 96.4% to 99.8%;the reasonable rate of medication course increased from 69.6% to 96.6%;the reasonable rate of the clinical application of Fat-soluble vitamin injection (Ⅱ) increased from 18.8% to 56.4%,with statistical significance (P<0.05). CONCLUSIONS:PDCA cycle can effectively promote the clinical reasonable of Fat-soluble vitamin for injection(Ⅱ).
8.Expression and purification of four single-stranded DNA-binding proteins and their binding on HCV RNA.

OBJECTIVEExpress and purify four single-stranded DNA-binding (SSB) proteins, and evaluate the binding of SSB proteins on HCV RNA.
METHODSThe expression plasmids of four SSB proteins were conducted, termed TTH, SSOB, KOD and BL21, respectively. The BL21 (DE3) was transformed by the expression plasmid of TTH, Transetta (DE3) were transformed by the expression plasmid of SSOB, KOD and BL21, then protein expression was induced with IPTG, the expression products were analysised by SDS-PAGE. To evaluate the binding of SSB on HCV RNA, RNA-SSB protein complexes were applied to a 1.2% TAE agarose gel.
RESULTSSuitable competent cells were transformed with the expression plasmids, induced by IPTG. SSB proteins were purified by affinity chromatography, to visualize their purity all SSB proteins were applied to SDS-PAGE analysis. All four proteins showed single clear bands. We have successfully obtained the SSB protein expression plasmid, expressed and purified SSB protein. TAE agarose gel electrophoresis was used to confirm SSB protein-RNA binding activity. The each of SSB-RNA complex migrated more slowly than the sole RNA, which suggested SSB protein could specifically bind to RNA.
CONCLUSIONSWe have expressed and purified four SSB proteins, and for the first time found that SSB protein can bind HCV RNA. Our results may provide a basis for future studies of the novel functions of SSB proteins on RNA.
