OBJECTIVETo investigate the effect of baicalein on the gap junction intercellular communication (GJIC) in the TM4 Sertoli cells of the mouse testis and its related mechanism.
METHODSWe measured the cytotoxicity of different concentrations of baicalein on the TM4 Sertoli cells in the mouse testis by MTT, detected the fluorescence transfer of the TM4 Sertoli cells by parachute assay, and determined the expression of the protein connexin 43 ( Cx43) in the baicalein-treated cells by Western blot and immunofluorescence assay.
RESULTSBaicalein produced no obvious cytotoxicity on the TM4 Sertoli cells at the concentration below 60 µmol/L but significantly increased their GJIC at 0-20 µmol/L (P < 0.01). Western blot and immunofluorescence assay showed that 0-20 µmol/L baicalein remarkably elevated the expression of Cx43 in the TM4 cells (P < 0.01) and on the membrane of the TM4 cells.
CONCLUSIONBaicalein at the concentration of 0-20 µmol/L can significantly enhance GJIC in mouse TM4 Sertoli cells by increasing the expression of the Cx43 protein.

4.Effect of Dexmedetomidine Combined Electrical Stimulation on Coanitive Function of Patients Receiving Extracerebral Intervention.

OBJECTIVETo explore the effect of dexmedetomidine combined electrical stimulation on cognitive function of neurosurgical diseases patients treated by extracerebral intervention.
METHODSTotally 122 patients with neurosurgical diseases who underwent selective intervention were randomly assigned to the observation group and the control group, 61 cases in each group. Patients in the control group recieved anesthesia by dexmedetomidine. Those in the observation group received electrical stimulation at Baihui (DU20), Yintang ( EX-HN3), and Neiguan (PC6) before dexmedetomidine anesthesia. The cognitive function of patients at preoperative day 1 and postoperative day 1 was respectively evaluated by Mini-Mental State Examinations (MMSE). Serum NSE, S-100β, IL-1β, IL-6, and TNF-α levels were detected in the two groups before intervention and immediately after intervention using ELISA.
RESULTSMMSE scores of two groups were significantly reduced at post-intervention day 1, as compared with one day before intervention. MMSE score of the observation group at post-intervention day 1 was (23.15 ± 1.87) points, significantly higher than that of the control group [ (19.34 ± 1.64) points , (P < 0.05)]. The postoperative cognitive dysfunction (POCD) incidence rate of the observation group was 16.4% (10/61), significantly lower than that of the control group [39.3% (24/61); P < 0.05]. Compared with before intervention, NSE and S-100β protein levels, IL-1β, IL-6 and α-TNF levels of the two groups increased (P < 0.05). Post-intervention NSE and S-100β protein levels, IL-1β, IL-6 and α-TNF levels were significantly lower in the observation group than in the control group (P < 0.05).
CONCLUSIONDexmedetomidine combied electrical stimulation could effectively prevent the occurrence of postoperative cognition, and reduce levels of NSA, S-100β, IL-1β, IL-6 and TNF-α.

5.Application of vacuum sealing drainage and cryopreservation technology in hand and foot skin replantation.

OBJECTIVETo summarize the clinical effect of avulsed skin replantation of hand and foot via vacuum sealing drainage (VSD) combing low temperature technique.
METHODSFrom March 2012 to October 2013,13 cases with avulsed skin replantation of hand foot using combined technique included 8 males and 5 females with an average age of 32 years old ranging from 18 to 62 years. The time from injury to hospital was 1 to 4 hours (2.4 hour in average). The reasons of injury included machine injury in 7 cases and rolling over by cars in 6 cases. The parts of injuried involved finger in 2 cases,back of the hand in 5 cases and dorsum of foot in 6 cases. The area of avulsed skin was 5 cm x 6 cm to 12 cm x 16 cm,tendon and bone exposure was found in 4 cases. VSD was operated in all patients and the avulsed skin was refrigerated in the temperature of -4 °C or -80 °C. After 4 days, the skin stored in the -4 °C was replanted to the wounded place in 5 cases and in 3 cases the skin was planted to the donor site of flap. The skin stored in the -80 °C was replanted in 4 cases after 7 or 8 days, 1 case after 45 days.
RESULTSOf the 13 cases, 1 case of degloved injury from lower leg to dorsal foot,the replanted skin was necrosis completely; 1 case of degloving injury with fourth finger,the skin which replanted after 45 days survived approximately 30%,cured after skin-graft many times. In the other cases, the survival area of replanted skin was more than 85%, all cured after dressing. According to the standard of skin survival area evaluation by Jia et al, 11 cases showed excellent, 1 showed medium and 1 showed inferior. There were no complication about grafted skin rupture after the skin survived in 11 patients,after 4 to 22 months follow-up, the resiliency of grafted skin showed good. Sensation recovery was measured by BMRC standard: 3 cases of S3, 5 cases of S3, 3 cases of S2.
CONCLUSIONVSD combining lower temperature technique in skin replantation provides time and space for wound preparation and treatment plan for the patients who need second surgery, especially for the large area skin degloving,this method could utilize the degloved skin efficiently, decrease the donor site area, alleviate the pain and financial burden,reduce the scar formation of donor site and impediment.

6.The analysis of sexuality situation and the influencing factors in 1 732 migrants of Chongqing

Objective To analyze the sexuality situation and the influencing factors of 1 732 cases of migrants in Chongqing , and provide the scientific basis for improving the sexual health of migrants and the service level of the relevant departments .Meth‐ods The multi stage random cluster sampling method was used to select the subjects .A total of 1 732 married and unmarried mi‐grants who had sexuality were included into the structured questionnaire survey .The SAS9 .3 software was use to statistical analy‐sis .The χ2 test and Ordinal regression analysis and Logistic regression analysis were use to univariate and multivariate analysis .Re‐sults The communication frequency of sexuality of male migrants was higher than the females ,OR=1 .53(95% CI:1 .24-1 .89);young people had the high communication frequency of sexuality ;the frequency of low education groups lower than higher .The fre‐quency of subjects worked in Hong Kong ,Taiwan ,overseas‐funded enterprises are higher than other workplace .The sexual life fre‐quency of male migrants is higher than the female ,OR=1 .84(95% CI:1 .40-2 .42);the sexual life frequency of low age group was higher than high age group ,the frequency of sexual life in factory workers was lower than the Hong Kong ,Taiwan ,overseas‐funded enterprises workers .Compared with the subjects who communicate the sexuality issues often ,the frequency of sexual depression and suppression was higher in the migrants who communicate general and rarely ,the OR was 2 .75 (95% CI:1 .71 -4 .41) and 2 .60 (95% CI:1 .45-4 .66) .Conclusion Male ,younger ,highly educated migrants have a high sexual issues communication frequency . The age and the workplace was the most important factor of the frequency of sexual life .Ease the work pressure and increase the communication of sexual issues will help to relax the sexual depression and depression .

8.Transurethral Resection of the Prostate with a Bipolar Tissue Management System Compared to Conventional Monopolar Resectoscope: One-Year Outcome.

The purpose of the present study was to evaluate the efficacy and safety of bipolar transurethral prostatectomy (TURP) using the GyrusTM PlasmaKinetic System compared with conventional monopolar TURP. This study included 102 patients with benign prostatic hyperplasia (BPH) who underwent TURP from January 2003 to March 2005. In all, 49 consecutive patients had bipolar and 53 had monopolar TURP. All patients were assessed by preoperative and postoperative International Prostate Symptom Score (IPSS), uroflowmetry, transrectal ultrasonography, operative time, weight of resected tissue, change in serum sodium and hemoglobin, duration of catheter use, length of hospital stay, and complication rates. Significant improvement was seen postoperatively in both groups, and no difference was observed in the resection time, weight of resected tissue, change in serum sodium and hemoglobin, improvement of IPSS and peak flow rate (Qmax), or complication rates over the 12-month follow-up in both groups. There was, however, a significant difference in duration of catheter use and hospital stay. Duration of catheter use (2.28 days vs. 3.12 days) and hospital stay (3.52 days vs. 4.27 days) were shorter in the bipolar group (p = 0.012 vs. p = 0.034, respectively). Our results demonstrate that bipolar TURP using the Gyrus(TM) Plasma Kinetic System is as effective as conventional monopolar TURP with the additional advantage of reduced length of catheter use and hospital stay. Bipolar TURP is a promising new technique that may prove to be a good alternative to conventional TURP in the future.

9.Alphavirus replicon-vectored plasmid DNA-based vaccine elicits protective immunity against classical swine fever virus.

We have shown previously that a Semliki Forest virus (SFV) replicon vectored DNA vaccine (pSFV1CS-E2) expressing the E2 glycoprotein of classical swine fever virus (CSFV) conferred full protection for pigs immunized three times with 600 microg of the vaccine. This study aims to evaluate the efficacy of the DNA vaccine with lower dosage and fewer inoculations. Pigs were immunized twice with 100 microg pSFV1CS-E2 (n = 5) or control plasmid pSFV1CS (n = 3), respectively. Pigs immunized with pSFV1CS-E2 developed high titers of specific neutralizing antibodies against CSFV after the booster, and the antibody titers increased rapidly upon challenge. The immunized animals showed no clinical symptoms except short-term fever and low-level viremia, whereas the control pigs immunized with the control plasmid produced no detectable antibody before challenge and showed obvious clinical signs following challenge, and 2 pigs died on 10 or 11 days post-challenge. All control animals developed extended viremia as detected by nested RT-PCR and real-time RT-PCR. Severe pathologic lesions typical of CSFV infection were observed at necropsy. We conclude that the alphavirus replicon-vectored DNA-based vaccine can be potential marker vaccine against CSFV.

10.Bipolar Transurethral Resection of the Prostate: A Comparative Study with Monopolar Transurethral Resection.

PURPOSE: Transurethral resection of the prostate (TURP) using bipolar electrocautery and 0.9% saline is a new technology in the field of surgery for benign prostatic hyperplasia (BPH). This randomized prospective study was conducted to compare the efficacy and safety of the Gyrus(TM) Plasmasect loop bipolar TURP and the conventional monopolar TURP for the treatment of BPH. MATERIALS AND METHODS: This study included 50 patients who were randomized 1:1 to undergo bipolar or monopolar TURP from August 2003 to October 2004. Preoperatively, the patients were assessed by the symptom score, uroflow and transrectal ultrasonography, and the two groups were comparable with regards to these measures and the mean group age. The perioperative and postoperative parameters we studied included the operative time, the resected prostate volume and the change in serum Na and Hb. Postoperatively, the patients were assessed for the symptom score and uroflow at both 1 and 6 months. RESULTS: Postoperative improvements in the symptom score and the Qmax were significant for both groups and the improvements were similar for the two groups. There was no difference in the operative time and resected prostate volume. The serum Na dropped by 4.2mEq/l in the monopolar group, whereas it fell only 1.1mEq/l in the bipolar group (p<0.001). Significant smaller reduction in serum Hb, a shorter postoperative catheterization time and a shorter hospital stay were noted for the bipolar group. CONCLUSIONS: Bipolar TURP using the Gyrus(TM) system is as effective as conventional monopolar TURP and it has additional advantages too. Thus, bipolar TURP may be a good alternative to conventional TURP. However, more follow-up is necessary to assess its long-term efficacy.
