1.P2 X7 receptor and renal diseases
Journal of China Pharmaceutical University 2015;(5):629-634
P2 X7 receptor is a member of ATP-gated non-selective cation channels;and is one subtype family of the P2X purinergic receptor.Signaling pathways of P2X7 receptor couple with several key inflammatory molecules including IL-1β;IL-6 and COX-2;which suggests that P2 X7 receptor plays a key role in the development of dis-eases via increased inflammation;indicating its potential as a novel therapeutic target.Recent studies show that P2 X7 receptor has a relationship with renal diseases. This article reviews the effect of P2 X7 receptor on renal diseases and its mechanism.P2 X7 receptor may be a new drug target;and its selectivity antagonist may be a new therapeutic agent for the treatment of renal diseases.

Methods:The varieties, sales amount and DDDs of insulin and its analogs in 43 hospitals of Nanjing during 2011 to 2013 were analyzed. Results:The use of insulin and its analogs was increased year by year from 2011 to 2013 with the average annual growth rate of 16%. Glargine, pre-mixed insulin 30R, aspart 30, human insulin and aspart ranked the top five in the sum of money with the proportion above 80%. Pre-mixed insulin 30R, aspart 30, glargine, human insulin and common insulin ranked the top 5 in DDDs. Conclusion:The application of insulin and its analogs is wide in clinics, and the usage and sum of money both show rapid growth trend.

Aim To investigate the protection effect of icariin on isoproterenol induced neonatal rat cardiomyocytes injury. Methods Isolation of cardiomyocytes from Sprague-Dawley rats aging 1~3 d were performed.Cardiomyocytes were grown in DMEM culture medium containing 15% fetal bovine serum. The cell density was adjusted to 1?105~5?105 cells?L-1,and the suspension was pipetted into each well of culture plates.Cells were incubated at 37 ℃ in 5% CO_2 atmosphere for 72 h.Cultured rat cardiomyocytes were divided into different groups:control,0.5 mmol?L-1 isoproterenol(model),and therapeutic groups with 0.1,1 and 10 ?mol?L-1 icariin.Cell viability was analyzed using MTT assay. The potential of mitochondrial membrane was determined by laser scanning confocal microscope.Apoptotic cells were detected by using flow cytometric analysis. Results Compared with the control group, the cell activity was lower in model group(P

AlM: To evaluate the effectiveness of Spot Vision Screener on vision screening of children without cycloplegia.METHODS:A total of 87 children (174 eyes) aged from 2~9 years old were examined with Spot Vision Screener and optometrist before cycloplegia.RESULTS: Statistical analysis demonstrated that the cylinder diopter and axis, the equivalent spherical diopter in both eyes, and the spherical diaopter in left eye had no significant change ( P>0. 05 ). However, the spherical diaopter in right eye had statistical significance. CONCLUSlON: Spot Vision Screener is a suitable instrument in vision screening of children without cycloplegia.

Uncoupling protein 4 (UCP4) is a member of the multigenic uncoupling proteins (UCPs), specific expressing in the cerebral cortex and hippocampus. UCP4 plays an important role in Parkinson's disease, multiple sclerosis, epilepsy, stroke, brain trauma and other central nervous system diseases by uncoupling, decreasing mitochondrial membrane potential, regulating Ca2+ homeostasis and oxidative stress. This article reviews UCP4 and its roles in the central nervous system diseases in order to provide certain basis for the development of UCP4targeted medication.

AIM: To investigate the effect of lipopolysaccharide (LPS) priming on macrophage(M?). METHODS: Macrophage cell line RAW264.7 were pretreated with or without LPS for 1 h, then challenged with PMA, or LPS, muramyl dipeptide(MDP), Zymosan, formyl-methionyl-leucyl-phenylalanine(FMLP) for 1 h . O 2 production in supernatants and intracellular free calcium([Ca 2+ ]i ) were measured, and changes in [Ca 2+ ]i and LPS induced O 2 production were compared. RESULTS: LPS pretreatment significantly increased O 2 production in RAW264.7 cells challenged with the stimuli, and in a certain extent, both O 2 production and increase of resting intracellular [Ca 2+ ]i were dose- and time-dependent on LPS pretreatment.Furthermore, the peak [Ca 2+ ]i was significantly higher in LPS pretreated groups than that of LPS unpretreated groups when challenged with PMA. Pretreatment with Ca 2+ inophore A23187 mimicked the LPS priming effects on O 2 production, but pretreatment with Ca 2+ chelator BAPTA and EGTA blocked this priming effect. CONCLUSION: LPS induced M? priming effect on O 2 production is dependent on elevation of resting intracellular [Ca 2+ ]i .

Chinese Pharmacological Bulletin 1986;0(05):-
The dried powder of cultural mycelium of ordyceps sinensis was extractedwith 1.25 mol ? L-1sodium hydroxide and precipitated with ethanol to afford the CsOH, in 2.58% yield. CsOH was consisted of 5 monosaccharide measured by paper chromatogra-phy (PPC) and gas- liquid chromatography (GLC). CsOH given intragastrically at the dose of 500 mg?kg-1and 250 mg?kg-1,ip at a dose of 100 mg?kg-1and 50 mg?kg-1had significant hypoglycemic activities in normal mice, alloxan-induced diabetes mice and streptozotocin-induced diabetes mice, and showed a dose-effect relation.

Acta Pharmaceutica Sinica 2014;49(11):1578-1581
Two new sesquiterpenoids, named as tyromols A and B (1 and 2), were isolated from cultures of basidiomycete Tyromyces chioneus, along with two previously reported 15-hydroxy-6 α, 12-epoxy-7β, 10αH, 11βH-spiroax-4-ene (3) and agripilol C (4). Compounds 1-4 were separated and purified by silica gel, RP-18, Sephadex LH-20 column chromatography. Their structures were elucidated on the basis of extensive spectroscopic analysis including IR, MS, 1D and 2D NMR experiments.
