1.Analysis of protoscoleces-specific antigens from Echinococcus granulosus with proteomics combined with Western blot.

OBJECTIVETo establish and optimize the proteomic analysis of protoscoleces-specific antigens from Echinococcus granulosus. To provide a foundation for identifying specific antigens in the soluble proteins of E. granulosus protoscoleces for further research.
METHODSBrood capsules were collected aseptically from fertile E. granulosus cysts from the livers of an infected patient. The fertile E. granulosus cysts were fractured, and protoscoleces were collected by centrifugation. The soluble proteins of protoscoleces were acquired using the 2D Quant kit according to the manufacturer's instructions. We employed two-dimensional electrophoresis (2-DE) combined with immunoblot assay (Western blot) to analyze the soluble components of E. granulosus protoscoleces antigens. The 2-DE and immunoblot maps obtained were analyzed with PDQuest 8.0 image analysis software.
RESULTSAbout 233 soluble protein spots were identified with Coomassie-stained gels. Most of the proteins had a molecular weight of 16,000 Da to 117,000 Da, and an isoelectric point value of 3.0 to 10.0. 2-DE immunoblot was conducted and 57 specific antigen spots were observed, among which 23 spots were identified.
CONCLUSION2-DE combined with Western blot is the key to successful proteomic analysis and presents a new possibility for searching the specific E. granulosus protoscoleces antigens.

2.The Effect of Preemptive Analgesia of Morphine Injected to Subarachnoid Space on Postoperative Epidural Analgesia with Morphine

Objective To study the effect of preemptive analgesina of morphine injected to subarachnoid space on postoperative epidural analgesia with morphine. Methods Sixty ASA Ⅰ-Ⅱ patients were randomly divided into two groups. All patinents received combination spinal-epidural anesthesia(CSEA). In experimenta group (n=30), 0 5% bupivacaine 2ml and morphine 0 5mg were injected into the subarachnoid space of the patients for CSEA, and morphine 2 8mg was used for postoperative epidural analgesia. In control group (n=30),0 5% bupivacaine 2ml was injected into the subarachnoid space of the patients for CSEA, and morphine 3 3mg was used for postoperative epidural analgesia. The numerical rating score (NRS) and Ramsay sedation scores were performed after operation. The postoperative analgesic duration and frequency of side effect were recorded. Rusult NRS was significantly less and postoperative analgesic duration was obviously longer in experimental group compared with control group. Ramsay sedation scores and the frequency of side effect significantly increased in the experimental group compared with the control group. But the shaking frequency in experimental group was significantly less than that in the control group. Conclusions Preemtive analgesia of morphine injected to subarachnoid space could improve postoperative epidural analgesia with morphine and increase analgesia time. But it had more side effects.
3.A role of p38 mitogen-activated protein kinase in regulating expression of Peroxisome proliferator-activated receptors-? in rat mesangial cells

Objective To investigate the relationship between p38MAPK and PPAR-?,and to study the role of p38MAPK and PPAR-? for diabetic nephropathy. Methods The expression of p38MAPK protein and PPAR-? mRNA was investigated in rat mesangial cells line HBZY-1 that was incubated respectively with 25 mmol/L glucose,100 nmol/L insulin,100 ?mol/L H_2O_2 and 100 mg/L AGEs. The relationship between p38MAPK and PPAR-? expression was studied by using SB203580,a specific inhibitor of p38MAPK. Results p38MAPK had significantly higher expression,while the expression of PPAR-? was significantly decreased in rat mesangial cells line HBZY-1 incubated with 25 mmol/L glucose,100 nmol/L insulin,100 ?mol/L H_ 2 O_ 2 and 100 mg/L AGEs respectively. PPAR-? activity was significantly reduced when p38MAPK was inhibited by SB203580. Conclusion p38MAPK is involved in development of diabetic nephropathy and may antagonize the expression of PPAR-?. PPAR-? activity might protect the function of kidney.
4.Processing of Gait Data of Hindlimbs of Spinal Cord Injured Rhesus Monkeys

Objective To establish a new processing method for gait data on Matlab to evaluate the hindlimbs behavior of non-human primates. Methods Gait analysis was tested on three rhesus monkeys 6 weeks after spinal cord injury, and kinematics data of hindlimbs were obtained using the VICON system. The raw data of kinematics were filtered and extracted, which were achieved through VICON 3D motion capture system with the Excel link combining Matlab with Microsoft Excel, and calculated in the Matlab environment. Results The kinematic parameters such as step length, step height, knee joint angle, and malleolus joint angle were gained by calculating. The mean values of step length (F=2.869, P=0.088) and step height (F=1.148, P=0.344) showed no significant difference at three speeds, which implied a higher repeatability of the data model. Angle-time curve reflected the joint function and movement. This system initially described the foot gait trajectory which could be used in gait repetitive analysis, and also generated the gait 2D/3D trajectories of hindlimbs. Conclusion The implement of these functions makes the post-processing of data more flexible and open whitout VICON system, and the calculated parameters and space tracing of gait trajectory basically meet the need of hindlimb behavior evaluation for nonhuman primate.
5.De-noising of Impedance Cardiography Differential Signal and Detection of the Feature Points Based on Wavelet Transformation.

Calculation of cardiac hemodynamic parameters is based on accurate detection of feature points in impedance cardiogram. According to these parameters, doctors can determine heart conditions, so it is very important to accurately detect the feature point of impedance differential signals. This article presents a process in which we used wavelet threshold method to de-noise signals, and then detected the feature points after six layers wavelet decomposition by using bior3. 7. The experimental data were collected from healthy persons in our laboratory and twenty two clinical patients in Chongqing Daping Hospital by using KF_ICG instrument. The results indicated that this method could precisely detect feature points whether it was from healthy people or clinical patients. This helps to achieve the application of noninvasive detection cardiac hemodynamic parameters in clinical treatments by using impedance method.

6.Evaluation of the efficacy of intensive pulse light on facial beauty

Objective To assess the efficacy and safety of intensive pulse light (IPL) device for facial rejuvenation and the treatment of hyperpigmented lesions, facial telangiectasias, acne vulgaris and hair removal. Methods One hundred females who claim to improve their skin texture, hair re-moval and patients with hyperpigmented lesions, facial telangiectasias and acne vulgaris were treated with IPL device. Patients received five treatments with the time interval of 3 weeks to 1 month. Pho-tographs were assessed 1 month after the last treatment. Results For facial skin texture, the total im-provement were scored 100 %. For hyperpigmented lesions and facial telangiectasias, the total im-provement reached to 90%. For ache vulgaris, the total improvement reached to 75 %. For hair re-moval, the total improvement was 95 %. Conclusion The IPL device is an effective and safe modality for the improvement of skin texture, hyperpigmented lesions, facial telangiectasias and hair removal, and a novel modality for the treatment of acne vulgaris.
7.Effects of intranasal dexmedetomidine as premedication on postoperative behavioral outcomes in children

Objective To observe the effects of intranasal dexmedetomidine (DM)as premedi-cation on postoperative behavioral outcomes in children.Methods Sixty ASA physical status Ⅰ or Ⅱchildren of both genders,2-5 years,weighing 10-30 kg,undergoing hernia surgery,were equally as-signed into three groups (n =20 each)using a random number table:control group (group C),mid-azolam group(group M)and dexmedetomidine group(group D).Thirty minutes before anesthesia in-duction,the children were respectively received intranasal normal saline 0.02 ml/kg (group C),in-tranasal midazolam 0.2 mg/kg (group M)and intranasal DM 2 μg/kg (group D).The sedation score of children apart from parents,the receipt score of face mask for sevoflurane anesthesia induction,the postoperative recovery time,adverse effects,and the percentage of patients requiring rescue analgesic were recorded.To observe the postoperative behavioral outcomes on 1th、7th、30th day using the PH-BQ.Results Compared with group C,the sedation score and the receipt score of face mask of groups M and D were significantly increased (P <0.05).Compared with groups C and M,the adverse effects and the percentage of patients requiring rescue analgesic of group D were decreased (P <0.05).The incidence of the postoperative behavioral outcomes of group C was higher than groups M and D on 1th,7th day (P <0.05).And on 30th day after operation,there was no significant difference among three groups.Conclusion Intranasal dexmedetomidine as premedication can significantly decrease the incidence of the postoperative behavioral outcomes in children.
8.Treatment of pediatric patients with refractory acute lymphoblastic leukemia with IHDA regimen

Objective To observe the efficacy and adverse effect of IHDA [Idarubicin(IDA) + high-dose Cytarabine(HD-Ara-C)] as a remedy regimen in the treatment of pediatric patients with refractory acute lymphoblastic leukemia(ALL).Methods Twelve children with refractory ALL were treated by IHDA regimen as follows:IDA,10 mg/(m2·d),d1-3;Ara-C,1.0g/m2,q12h,d1-3.The children who achieved complete remission(CR)could get into the following sequential regimens or allogeneic hematopoietic stem cell transplantation (allo-HSCT).The same regimen was given to the children who didn't achieve CR when WBC >2.0×109/L.The efficacy and hematology or non-hematology adverse effect were evaluated.Results CR/partial remission (PR)/non-remission (NR) were respectively 4/3/5 cases after giving the first regimen,and CR/PR/NR were 5/3/4 cases after giving the second regimen,respectively.The overall remission was 66.7% (8/12 cases),of which 5 cases(41.7%) achieved CR,3 cases (25.0%) reached PR and 4 cases(33.3%) reached NR.Grade Ⅳ myelosuppression occurred in all patients,but no severe infection and hemorrhage happened after the application of granulocyte colony stimulating factor (G-CSF),platelet transfusion and anti-infection treatment.Some reversible side effects like liver toxicity,myocardial damage and nerve injury were observed in some patients.There was no chemotherapy related mortality in all the patients.Two cases relapsed again followed up to October 2015.One achieved CR after applying chimeric antigen receptor T-cell immunotherapy and was receiving allo-HSCT now.Another was dead after applying FLAG (Prednisone+Fludarabine+Ara-C+G-CSF) save regimen.The time of the other 3 cases achieving CR was 26,10,4 months,respectively.Among the remaining 7 cases,3 cases were forced to receive hematopoietic stem cell transplantation,2 cases abandoned treatment and 2 cases failed to follow up.Conclusions The IHDA regimen is a well-effective and tolerated treatment for pediatric patients with refractory ALL,and could create an opportunity for the application of allo-HSCT.
9.Quality control of statistical analysis in data management of clinical trials.

Data is the basis and soul of clinical trials. To obtain accurate data, strict and standard data management is essential, which can be effectively supported by quality control in statistical analysis. In this paper, we briefly introduce the concept of the quality control in clinical trials, and describe its contents and methods. We hope that this work will be helpful to the application of statistical quality control in data management of clinical trials.

10.Bell’s palsy: the inducing factors survey

OBJECTIVE To investigate the induc-ing factors of Bell’s palsy. METHODS From Febru-ary to May 2005, 262 outpatients of Bell’s palsy were surveyed for 9 inducing factors. RESULTS The mean age of onset was 39?17 years old. The ratio of male to female and of left to right were 48:52. Forty-eight patients denied all inducing factors, while 214 pa-tients (81.7 %) had at least one factor. Cold was found in 53.5 % of the patients, fatigue in 22.5 %, viral infec-tion in 18.7 %, recurrence in 11.8 %, psychological stress in 11.5 %, family history in 7.6 %, puerperal period in 1.5 %, and molar infection of the affected side in 1.5 %. CONCLUSION Bell’s palsy is likely to be a set of disease. Most of patients with Bell’s palsy had inducing factors. Avoid these factors may reduce the incidence of Bell’s palsy. Further investigate will fractionize Bell’s palsy to several diseases includ-ing true idiopathic peripheral facial paralysis and some definite diseases.
