1.Determination of nucleosines in Ningxinbao Capsules by HPLC

AIM:To establish HPLC method for making a assay of nucleosines in Ningxinbao Capsules(Cepha-(lospovium) sinensis Chen.sp.nov). METHODS:The assay was performed on a C_(18) column(4.6 mm?250 mm,5 ?m).The mobile phase was composed of phosphate buffer(pH 7.8-8.0)-acetonitrile(83∶17).The flow rate was at 1.0 mL/min.The detection wavelength was at 260 nm. RESULTS:The linear ranges of uridine,adenosine and inosine lay in the concentration range of 4.168-83.36,4.480-89.60,5.230-104.6 ?g/mL,respectively(r=0.999 9). The average recoveries were 0.4%,1.8%,2.5%,respectively. CONCLUSION:The method is simple,rapid.It is suitable for the assay of nucleosines in Ningxinbao Capsules.
2.Development and validation of real-time fluorescence quantitative PCR method for murine virus

Objective To develop and validate the real-time fluorescent quantitative PCR(Q-PCR)method for the detection of 8 murine viruses. Methods The specificity,sensitivity and precision of the Q-PCR method were verified by four laboratories,and the virus simulated contamination test and blind sample detection were carried out simultaneously,of which the detection results were compared. The Q-PCR method was used to detect 26 batches of monoclonal antibody cell lines for SARS-CoV-2 vaccine production and 15 batches of other products of murine origin. Results The Q-PCR method used for detecting 8 kinds of murine viruses had no cross reaction with the same family and genus or other murine viruses. Except the sensitivity of laboratory 2 to ectromelia virus(EctV/Mouse Pox,MPV)was 2 × 10~2copies/μL,the sensitivity of laboratory 2 to other 7 viruses and 3 other laboratories to 8 murine viruses was 2 × 10~1copies/μL. Except the inter-assay CV of the copy number of mouse adenovirus(MAdV)detected by laboratory 3 was 37. 58%,the intra-assay and inter-assay CVs of the Ct and copy number of other 7 viruses detected by laboratory 3 and those of 8 viruses detected by other 3 laboratories were all less than 25%.The sensitivity of virus simulated contamination test met the parameter requirements. The coincidence rate of blind sample detection results by 4 laboratories was 100%. All the 26 batches of monoclonal antibody cell lines for SARS-CoV-2 vaccine production and 15 batches of other murine derived products were negative for 8 murine viruses. Conclusion Q-PCR method for murine virus has good specificity,sensitivity and precision,and can be used for the detection of murine derived biological products.
3.Preparation of Monoclonal Antibody Against "a" Determinant With Mutation on Site 145 and Rapid Diagnosis Assay for HBV Mutant Strain

Hepatitis B virus surface antigen with mutation on site 145 may cause omission in detection of hepatitis B virus with HBsAg diagnostic reagent. Monoclonal antibody against this mutation was obtained through hybridoma technique. The sub-class of the monoclonal antibody was determined to be IgG1(? type, the titer is 1∶9?104), and the monoclonal antibody has no cross-reaction to other hepatitis virus. The method of AC-ELISA which was established with monoclonal antibody and polyclonal antibody showed high analysing susceptibility for HBV mutant samples.
4.Impairment of left ventricular function and mechanism analysis after intermittent hypoxia

Intermittent hypoxia (IH) is a direct consequence of obstructive sleep apnea syndrome (OSAS), which results in left ventricular dysfunction , remodeling and myocardial cell injury .Understanding the mechanisms of OSAS is important for us to prevent and treat the cardiovascular complications in the clinical practices .We summarize the effects of IH on left ventricular function , and analyze the mechanisms at the cellular and molecular levels , including oxidative stress, ion exchangers and inflammation .In addition, the effects of endothelin-1 and hypercapnia on IH-induced cardiac injury are discussed .
5.Advances in the treatment of parotid gland adnoid cystic carcinoma

Adenoid cystic carcinoma is regarded as one of the most common malignant tumors in parotid gland. The local control rate and long term survival rate of salivary gland adenoid cystic carcinoma, which tends to spread along the perinerve and metastasis distantly, are not good. 6.EFFECTS OF DIETARY COPPER DEFICIENCY ON THEENZYMATIC COMPONENTS OF THE PRIMARYFREE RADICAL DEFENSE SYSTEMAND SERUM HEMOLYSININ RATS

Weanling Wistar rats were fed a modified AIN-76 diet ad libitum for 25 to 57 days, either copper-deficient (0.49mg/kg; CuD) or copper-adequate (6.44mg/kg;CuA). The effects of dietary copper deficiency on the enzymatic components of the primary free radical defense system-super-oxide dismutase (SOD), catalasc (CAT) and glutathione peroxidase (GSH-Px), lipoperoxides (LPO) and serum hemolysin were examined. The amount of LPO exhibited no significant change even though SOD activity decreased significantly in the tissues of CuD rats, which indicated that Cu-ZnSOD activity responded directly to copper status of the body but the compromises in the overall protection against free radicals were not evident. The profound reduction in scrum hemolysin in CuD rats suggests that humoral immune function is copper dependent. The results also suggest that as an index of evaluating copper nutriture CuZnSOD is more sensitive than hemoglobin to copper deficiency.
7.Research progress of the relationship between HGF/Met and invasion of thyroid carcinoma

Activation of HGF/Met has been complicated in tumorigenesis, invasion and metastasis of various tumors. We summarize current knowledge on their association with invasion of thyroid carcinoma. By discussing the expression of Met and its possible mechanism and analyzing the activation of Met with tumor motility, angiogenesis and inflammatory reaction, we conclude that the research of the possible role of HGF/Met in thyroid carcinoma will advance knowledge on the diagnosis and management of this disease,as well as tumor invasion.

10.Selection and clinical significance of the extent of hepatectomy for gallbladder cancer

Gallbladder cancer is the most common malignant neoplasm in biliary system,with a high degrade of malignancy,invasion and metastasis.The prognosis is heartbreaking.Radical surgery is the only possible way for curing.The current understanding of China's doctors at all levels on radical surgery of gallbladder cancer is chaotic.Based on the latest version eighth TNM staging of the American Joint Committee on Cancer(AJCC)of gallbladder cancer,making a review of the literature,combined with the author's personal experience,we have interpreted the reasonable selection of hepatectomy of gallbladder cancer,in order to improve the doctors at all levels to regulate the understanding of hepatectomy and the prognosis of gallbladder cancer.
