1.Experimental study on regeneration of articular cartilage defects with bone marrow stromal cells

[Objective]To retrieval a best cell factor which can induce bone marrow stromal cells (bMSCs) into chondrocyte in vitro and to explore an effective plan to repair rabbit's chondrocyte defect.[Method]Isolated bMSCs were cultured in vitro. rh Fibroblast Growth Factor 1(rhFGF-1)、rh Transforming Growth Factor-?1(rhTGF-?1)、rh Insulinlike Growth Factor-Ⅰ(rhTGF-Ⅰ) were utilizated. The proliferation of cells was detected by MTT assay, and the macroscopic histology , HE staining and immunohistochemical examinations were performed to seek the best cell factor. In vivo , to investigate the repair of the articular cartilage, bMSCs combined with fibrin glue and rhTGF-?1、rhIGF-I was compared with control group.[Result]The cells induced by rhTGF-?1 and rhIGF-I were similar to chondrocytes in morphology, and immunohistochemical examinations showed the cells possessed phenotype of chondrocytes. RhTGF-?1 and rhIGF-I could differentiate bone marrow stromal cells into cartilage cells in vivo, and repair the articular cartilage defect. The control group could not repair the articular cartilage defect efficiently.[Conclusion]rhTGF-?1 and rhIGF-I are best group which stimulate bMSCs to differenate into cartilage cells. BMSCs combined with fibrin glue and rhTGF-?1、rhIGF-I can repair the articular cartilage defect.

4.Effect of Acanthopanax trifo-liatus Alcohol Extract on the Contents of Inflammatory Factors in Serum of Adjuvant-induced Arthritis Rat

OBJECTIVE:To study the effect of Yi medicine Acanthopanax trifo-liatus alcohol extract on the contents of IL-1β, IL-6,IL-10,NO,TNF-αand PGE2 in serum of adjuvant-induced arthritis(AA)rats. METHODS:60 SD rats were randomly divid-ed into normal control group (distilled water),model group (distilled water),positive control group (nimesulide,0.03 g/kg), A. trifo-liatus high-dose,medium-dose and low-dose groups [7,3.5,1.75 g(medicinal material)/kg]. Except for normal control group,other groups were given 0.1 ml Freund’s complete adjuvant on posterior right toe to induce AA model. After modeling, those groups were given relevant medicine intragastrically,once a day. The contents of IL-1β,IL-6,IL-10,TNF-α,NO,PGE2 in rat serum were measured 30 days later. The pathological morphology of the rat ankle was also observed. RESULTS:Compared with normal control group,the contents of IL-1β,IL-6,TNF-α,NO and PGE2 in serum increased in model group while the content of IL-10 decreased,with statistical significance (P<0.01);compared with model group,the contents of IL-1β,IL-6,TNF-α,NO and PGE2 in serum decreased in A. trifo-liatus high-dose groups while the content of IL-10 decreased,with statistical significance (P<0.05). The pathology results showed that part of the slice synoviocytes mild thickening,fibrous tissue mild hyperplasia,in-flammatory cells infiltration at varying degrees,macrophages mild hyperplasia in A. trifo-liatus groups. Those changes were not found in normal control group. CONCLUSIONS:A. trifo-liatus alcohol extract has an obvious anti-inflammatory action to AA mod-el rat,by a mechanism of decreasing the contents of IL-1β,IL-6,TNF-α,NO and PGE2,and increasing the content of IL-10.

6.Transcranial approach for resecting spheno-orbital meningiomas

Objective To explore the effect and safety of transcranial approach for spheno-orhital meningioma. Design Retro- spective case series. Participants Thirty-two patients being operated with transcranial approach. Twenty-four cases were meningothelial meningiomas, 3 cases were fibrous meningiomas, 1 case was psammomatous meningioma, 2 cases were atypital meningiomas, 2 case were malignant meningiomas. Methods All patients underwent frontal-temporal craniotomy, the involved sphenoid wing bone and peri- orbit were removed to prevent recurrence. The superior orbital fissure and optic canal were decompressed, the dural and periorbital de- feet were repaired by autogenous temporal fascia or artificial dura. Main Outcome Measures Preoperative and postoperative exoph- thalmus and eyeball movement, the extent of tumor resection, the ratio of recurrence. Results The extent of tumor resection: 8 cases were Simpson gradeⅡ, 20 cases Simpson gradeⅢ, 4 cases Simpson grade IV. After surgery, proptosis were improved in all patients, ophthalmoplegia was found in 6 eases. There was no operation-related death or other significant complication. Tumor recurred in 6 cas- es. Conclusions Adequate exposure of the tumor and bony decompression of the cranial nerves can result from transcranial approach, all the involved bone should be removed in order to prevent recurrence. This approach is relatively safe and the ptoptosis are improved significantly. Complete surgical resection is difficult because of the involvement of the orbital apex, superior orbital fissure and cav- ernous sinus.
7.The clinical significance of combined measurement of anti-saccharomyces cerevisia antibody and antiperinuclear neutrophil cytoplasmic antibody in inflammatory bowel disease

Objective To evaluate the clinical significance of combined measurement of antiperinuclear neutrophil cytoplasmic antibody(ANCA)and anti-saccharomyees cerevisia antibody(ASCA)for the diagnosis of inflammatory bowel disease(IBD)patients and difierentiation of Crohn's disease(CD)with ulcerative colitis(UC).Methods A total of 159 patients with IBD(97 UC,62 CD),167 patients with other non-IBD gastrointestinal conditions(NIBDC)and 25 healthy controls(HC)were recruited in our research.ASCA and ANCA were detected by enzyme-linked immunosorbent assay(ELISA)and indirect immunofluorescence assay.respectively.Results The prevalence of ASCA-IsA or IgG in CD group,UC group,NIBDC and HC were43.5%,14.4%,29.3%and 0,respectively.The prevalence of ASCA-IgA or IgG in CD group were higher than those in other groups(X2=16.76 or 4.12,P<0.01 or<0.05).The prevalence of ANCA in CD group.UC group,NIBDC and HC were 8.1%,56.7%,4.8%and 0,respectively.The prevalence of AMA in UC group were much higher than those in other groups(X2=38.08 or 90.47,P<0.01).The sensitivity specificity and positive predictive value(PPV)in ASCA+/ANCA-were 40.3%.93.8% and 80.6%,respectively,and in ANCA+/ASCA-were 48.5%,98.4% and 97.9%,respectively.Condusions ASCA or ANCA testing alone are not sensitive enoulgh for diagnosing CD and UC,but their combination asses are specific for differential diagnosis between CD and UC.Combined testing of ASCA-IgA with IgG can improve the sensitivity in screening CD patients.The ASCA positive pattern in Chinese CD group are correlated with surgery.
8.Impact of GSTM1 and GSTP1 polymorphism on lung cancer patients treated with platinum-based chemotherapy

Objective To investigate the relationship between glutathione S-transferases P1(GSTP1)Ile105Val and glutathione S-transferases M1(GSTM1)single nucleotide polymorphisms(SNP) and the sensitivity to chemotherapy among patients with ad-vanced non-small cell lung cancer(NSCLC) .Methods We used gene sequencing analysis to determine the SNP of GSTP1 Ile105Val and PCR analysis to GSTM1 in DNA from peripheral lymphocytes of NSCLC patients .Totally 89 patients with NSCLC were trea-ted with platinum-based chemotherapy ,and clinical response was evaluated after 2 cycles .The association between GSTP1 Ile105Val and GSTM1 SNP and chemosensitivity were analyzed .Results The overall response rate was 29 .2% .Chemotherapy re-sponse did not show statistically significant differences between the wild genotypes and the variant genotypes for the GSTP1 Ile105Val and GSTM1 gene(P>0 .05) .Conclusion The polymorphisms of GSTP1 Ile105Val and GSTM1 may be not associated with sensitivity to chemotherapy in NSCLC patients .
9.Altered Activity and Functional Connectivity of Superior Temporal Gyri in Anxiety Disorders: A Functional Magnetic Resonance Imaging Study.

OBJECTIVE: The prior functional MRI studies have demonstrated significantly abnormal activity in the bilateral superior temporal gyrus (STG) of anxiety patients. The purpose of the current investigation was to determine whether the abnormal activity in these regions was related to a loss of functional connectivity between these regions. MATERIALS AND METHODS: Ten healthy controls and 10 anxiety patients underwent noninvasive fMRI while actively listening to emotionally neutral words alternated by silence (Task 1) or threat-related words (Task 2). The participants were instructed to silently make a judgment of each word's valence (i.e., unpleasant, pleasant, or neutral). A coherence analysis was applied to the functional MRI data to examine the functional connectivity between the left and the right STG, which was selected as the primary region of interest on the basis of our prior results. RESULTS: The data demonstrated that the anxiety patients exhibited significantly increased activation in the bilateral STG than the normal controls. The functional connectivity analysis indicated that the patient group showed significantly decreased degree of connectivity between the bilateral STG during processing Task 2 compared to Task 1 (t = 2.588, p = 0.029). In addition, a significantly decreased connectivity was also observed in the patient group compared to the control group during processing Task 2 (t = 2.810, p = 0.012). CONCLUSION: Anxiety patients may exhibit increased activity of the STG but decreased functional connectivity between the left and right STG, which may reflect the underlying neural abnormality of anxiety disorder, and this will provide new insights into this disease.
