1.Local recurrence after radical resection with total mesentery excision (TME) for the treatment of rectal cancer

ObjectiveTo evaluate the effect of radical proctectomy with TME for the treatment of rectal carcinoma in the prevention of postoperative local recurrence. Method78 patients undergoing this procedure were followed up for 5 years.ResultLocal recurrence developed in 10 cases during 5 year′s follow up, with recurrence related mortality of 7/78, local recurrence rate of 10/78 (13%). Tumor malignancy and Dukes′ grade were in direct proportion with the recurrence. The 5 year′s survival rate was 88%(69/78). The postoperative 5 year′s survival rate in Dukes′ A, B and C patients was respectively 100%(9/9), 92%(51/54), and 64%(9/14). ConclusionRadical proctectomy with TME is superior to that without TME in terms of low postoperative local recurrence rate for the treatment of rectal carcinoma.

3.Preparation of a toxicity fusion protein targeting to CD123 on leukemia stem cell

Objective To construct a fusion protein that used for treatment of resistance and palindromia in leukemia and studied its biological activity. Methods IL-3 and LP gene fragments were amplified by PCR. After enzymatic digestion and T4 ligation, the fusion gene was cloned into expression vector pAYZ. The product was purified by exchange chromatography and anti-Etag affinity chromatography. IL3-G4SLP fusion protein was analyzed by SDS-PAGE and Western blot. Protein biological activity was detected by FACS. Results The fusion protein was expressed as soluble protein by E.Coli 16C9. The protein expression level was about 1 mg/L, its purity was over 95 %, and the expression level was about 1 mg/L. The fusion protein can combined specificely with CD123 on leukemia stem cells. Conclusion Fusion protein IL-3-G4S-LP can target on leukemia stem cells and maybe as a potential drug used for treatment of resistance and palindromia in leukemia.
4.Prevention effects of AduoLa Fuzhenglin on brain injury Induced by microwave radiation in rats

Objective To study the prevention effects of AduoLa Fuzhenglin(ADL)Oll the brain injury induced by microwave radiation in rats.Methods A total of 140 male Wismr rats were divided randomly into 5 groups,including control group,microwave exposed group,low dosage(0.75 g·kg-1·d-1)group.middle dosage(1.5 g·kg-1·d-1)group and high dosage(3 g·kg-1·d-1)group.Rats in three ADL groups were lavaged with ADL per day for 2 weeks before radiation.After administration,rats were exposed to microwave at 30 mW/cm2 for 15 min.The abilities of learning and memory were detected by Morris water maze,and the contents of amino acids neurotransmitter of hippocampus were detected by HPLC, then the histology and uhrastrncture of hippocampus were observed with light and electron microscope at 6 h,7 and 14 d after exposure.Results The abilities of learning and memory were declined(F=0.000-0.043,P<0.05)from 6 h to 7 d after exposure,and the contents of four kinds of amino acid neurotransmitter in hippocampus were decreased,of which GLU,GLY and GABA were decreased significantly(F=0.000-0.007,P<0.01)at 6h after exposure,then tissue edema,neuronal degeneration,neuron mitoehondria swelling and cavitation,endocytoplasmie rotieulum broaden,synaptic cleft blurred,and perivascular space widen were found in the hippocampus at 6 h and 7 d after exposure.The changes in low dosage group were similar to those of the radiation group.However,in middle and high dosage groups,the abilities of learning and memory were normal to some extent with the significant differences compared to the radiation group from 6 h to 7 d after exposure(F=0.015-0.028.P<0.05).The contents of four kinds of amino acid neurotransmitter were not decreased,especially GLU contents close tO normal level.There were significant differences between middle and high dosage groups and radiation group at 6 h after exposure(F=0.000-0.042,P<0.05).Moreover,no obvious injury in the hippocampus was observed in middle and high dosage groups at 6 h and 7 d after exposure.Conclusions Exposure to 30 mW/cm2 microwave radiation could decrease the abilities of learning and memory,induce amino acid neurotransmitter turbulence,and injure the histology and uhrastructure of hippocampus.ADL at the dosages of 1.5 and 3 g·kg-1·d-1 would have preventive effects on the injury induced by microwave exposure.The concentration of 1.5 g·kg-1 ·d-1 of ADL might be the effective dosage to prevent the brain damage after microwave exposure.

6.Epidemiologic features and trends of leukemia in Shenzhen during 2001-2015

Objective To describe the incidence trend of leukemia in Shenzhen during 2001 to 2015, and to provide base data for designing prevention and treatment strategies on leukemia. Methods The leuke-mia incidence data and population data collected by Shenzhen Cancer Registry from 2001 to 2015 were used in our analysis. The crude incidence,age-standardized incidence rate by Chinese standard population (ASR China)and age-standardized incidence rate by world standard population (ASR world)were calculated. The annual percentage change (APC)of the incidence was analyzed by Joinpoint regression. Results Shenzhen Cancer Registry registered 2106 new cases of leukemia from 2001 to 2015. The crude incidence was 6. 31 per 100000,with 6. 75 per 100000 ASR China and 7. 15 per 100000 ASR world. Cumulative rate (0-74 years) was 0. 63%,and truncated rate (35-64 years)was 7. 03 per 100000. From the perspective of gender distribu-tion,the incidence of male was significantly higher than female,with a sex ratio of 1. 38 : 1. In terms of time trend,the incidence of leukemia was stable,and the Joinpoint regression showed that APC = - 0. 09%(95% CI:- 1. 60% -1. 41%,P = 0. 92). In terms of subtypes,acute myelocytic leukemia (AML)accounted for 17. 66% of the total cases,and the incidence of AML has increased during 2001 to 2015 (APC = 13. 34%, 95% CI:5. 71% -21. 51%,P < 0. 01). The median age of leukemia patients was 36 years old,and the mean age was 37. 29 years old. The two peaks of the incidence were 1-4 and 80-84 age groups,and the ASR inci-dences were 9. 13 per 100000 and 39. 40 per 100000 respectively. Conclusion The incidence of leukemia is very high in Shenzhen. Children and the elderly are at high risk of leukemia. Government needs to guide insti-tutions to carry out research to reduce the incidence of leukemia.

Vertebrobasilar dolichoectasia(VBD ) can increase the risk of stroke. Significant expansion, elongation and tortuosity of the vertebrobasilar arteries are the main morphology manifestations of VBD. However, there is no consensus on the quantitative imaging assessment of VBD. Many studies showed that some quantitative parameters, such as basilar artery length and bending length, basilar angulation, vertebral tortuosity index can be used for the quantitative imaging assessment of VBD.

OBJECTIVETo observe the protective effects of total glycosides Rubus parviflolius (TGRP) on local cerebral ischemic.
METHODThe local cerebral ischemia in rat was made by middle cerebral artery occlusion(MACO). The infraction weight was determined by TTC stain. SOD, MDA, GSH and apoptotis were determined with different method respectively.
RESULTTGRP 20, 10 mg x kg(-1) ig markedly improved the abnormal nervous symptoms, incredsed the SOD, GSH activity and reduced contentes of MDA in brain of MACO rat, TGRP 20 mg x kg(-1) ig significantly decreased the numbers of apoptotic cells in ischemic cortex.
CONCLUSIONTGRP has protective effects against cerebral infraction, and its mechanism may be related to anti-apoptotis and free radical.

9.The biological behavior of hepatocarcinoma stem cells in rats.

OBJECTIVESTo study the biological behavior of hepatocarcinoma stem cells in rats.
METHODSPrimary liver carcinomas were induced in rats using diethylnitrosamine. Tumor cells from 8 rats were separated according to rats oval cell (OVC) markers CD34, c-Kit, Thy-1, AFP, CK7, CK8, CK14, CK18, CK19 and GGT and then they were separately injected into the livers of nude mice. The tumors grown from the different subpopulation of OVC markers in the nude mice livers (10 OVC markers negative or positive cells) were weighted 1 month after the inoculations. The hepatocarcinoma cell subpopulations with higher ability in causing tumor growths were further studied in vitro. The cell cycles and DNA content of those subpopulation cells were investigated using flow cytometry.
RESULTS(1) Subpopulation cells with CK7(-), Thy-1(+) and AFP(+) markers had a higher ability in causing tumors in nude mice; (2) Subpopulation cells, exhibiting characters of TSC, had a low growth rate in vitro.
CONCLUSIONS(1) Different subpopulations of hepatocarcinoma cells had different abilities in causing tumors in rats. Some subpopulation cells, such as CK7(-), Thy-1(+) and AFP(+) cells, have characteristics of tumor stem cells. (2) The hepatocarcinoma stem cells may have a low growth rate in vitro.

OBJECTIVETo compare the tissue formation and the content of polysaccharide between the wild Dendrobium candidum and the cultured ones and to find any existed differences.
METHODBare-handed microtomy and photomicrography; The content of polysaccharide is determined by phenol-sulphuric acid method.
RESULT AND CONCLUSIONThere are no marked noticeable differences between the wild D. candidum and the cultured ones in terms of the tissure formation and the content of polysaccharide.
