1.AN EXPERIMENTAL STUDY OF THE ANTICANCER EFFECT OF POLYSACHARIDES MIXTURE

We have studied the inhibitory effect of polysaccharides mixture on Ehrlich Ascites Carcinoma (EAC), Sarcoma 180 (S180), reticulocytic leukemia (L615) in mice. It is found that polysaccharides mixture could obviously prolong the survival duration of mice suffering from tumour and inhibit neoplastic proliferation. Thymus and adrenal glands atrophy in mice caused by vaccination of cancer cells was also remarkably resisted.The results of our experiments show that polysaccharides mixture is a safe,effective and promising pharmaceutic preparation of traditional Chinese medicine. It deserves to be further studied and developed.
2.Changes of protein kinase C ? in renal tissue of diabetic rats

Objective To study the expression and transcription of PKC? in renal tissue of streptozotocin-induced diabetic rats. Methods Hyperglycemia was induced with streptozotocin(55 mg/kg) in Sprague-Dawley rats. After 5 weeks, the expression of PKC? protein and mRNA was measured by immunohistochemistry, reverse transcription-polymerase chain reaction (RT-PCR) and Western blot analysis. Results In diabetic animals, the expression of PKC? was greatly enhanced especially in the proximal tubules and the glomeruler mesangial areas with upregulating of the membrane-associated PKC?. RT-PCR analysis showed that mRNA level of PKC? increased by 1.67 folds in diabetic rats as compared with the normal ones. Conclusion The expression of PKC? protein and mRNA is upregulated in early diabetic nephropathy, which suggests an interaction between PKC? and the pathogenesis of diabetic nephropathy.
3.Lengthening the right renal vein by a v-shape excision of vena cava for kidney graft

Purpose:To introduce a method of lengthening the right renal vein for kidney graft. Methods:AV-shaped excision of the neighbouring vena cava wall may lengthen a short rigth renal vein. Results: 6 cases ofpatients have been done, and all obtained satisfactory outcome. Follow up for 1.5 month to 1.5 year, all 6 pa-tients had their transplanted kidney function well. Conclusions:A V-shaped excision and sewing up the rest venacava is a simple, effective measure for a short right renal vein during kidney transplantation.
4.Comparative analysis of POCT and routine test in BNP testing

Objective To analyze the relevance between the point-of-care testing (POCT) and routine test in BNP testing .Meth-ods The whole blood samples or plasma samples from 40 inpatients were detected brain natriuretic peptide (BNP) by the Alere Triage? MeterPro fluorescence immunoassay analyzer (POCT ) or the Beckman Coulter Access ?2 chemiluminescence analyzer (routine test) ,respectively .The acquired data were subjected to comparative analysis according to the CLCS EP 9-A2 .Results The linear regression of the BNP content in the blood samples detected by POCT and the routine test was good ,the correlation coeffi-cient(r) was 0 .999 7 .Conclusion POCT and the routine test have good correlation in BNP testing .POCT for BNP testing has higher reliability and is applicable for clinical detection .
5.Model making of radioactive optic nerve injury

Background Radiation-induced optic neuropathy (RION) is a severe complication after radiotherapy for head and neck cancer,which threatens the visual acuity and quality of life of patients.Till now,there is no recognized treatment for RION.It is of great significance to study the natural progression of the RION,and to prevent and treat RION.Objective This study was to establish an ideal radioactive optic nerve injury animal model.Methods Healthy 8-week SD rats with hygiene grade were randomly divided into normal control group and model group,with 6 rats in each group.The total 30 Gy dose of radiation with 3 portions was used to irradiate the head model group rats;ELISA was performed to analysis the changes of endothelin-1 (ET-1) and Von Willebrand factor (vWF) concentrations in blood 2,4 and 8 weeks after irradiation.Hematoxylin-eosin staining and transmission electron microscope were performed to observe the changes of optic structure.The use and care of the experimental animals complied with the ARVO statement.Results The concentrations of ET-1 in the model group were (23.18± 0.11),(27.98 ±0.22),(33.90 ±0.1 1),(65.25 ±0.38) and (43.82 ± 0.09) pg/ml before irradiation,1 day,2,4,6 weeks after irradiation,those in the normal control group were (22.65 ± 0.14),(23.18 ± 0.19),(23.68 ± 0.15),(24.23±0.12) and (23.58±0.16)pg/ml.The concentrations of vWF in the model group were (63.16±2.21),(88.32± 2.06),(123.38 ± 1.36),(191.40 ± 0.61) and (141.69 ± 0.82) pg/ml before irradiation,1 day,2,4,6 weeks after irradiation,those in the normal control group were (62.82 ± 1.56),(63.35 ±2.06),(64.12 ± 1.76),(63.52±2.02) and (63.48 ± 1.55)pg/ml.There were significant differences of ET-1 and vWF concentrations among different groups and time points (ET-1:Fgroup =32.160,P =0.012;Ftime =21.180,P =0.023.vWF:Fgroup =73.110,P=0.001;Ftime =46.180,P =0.002).The nerve fiber bundles was swelled with disordered arrangement and vacuolization 8 weeks after irradiation.Axon swell and atrophy,axons with myelin sheath layer plate separation were obtained.The rates of axon demyelination in the normal control group and model group were (1.35 ±0.79) % and (14.44±2.32)%,respectively.There was a statistically significant difference between the two groups (t =14.07,P<0.01).Conclusions The total 30 Gy dose of radiation on the head of rats can make stable radioactive optic nerve injury model.This model making method is simple,cheap and practical,which is worth further study.

Objective To investigate the expression and translocation of protein kinase C (PKC) ?Ⅱ isoform in renal tissue of diabetic rats and the effects of blocking renin-angiotensin system (RAS) on them. 9.THE MEASUREMENT OF SERUM TNF LEVEL IN PATIENTS WITH ABDOMINAL INFECTION AND ITS CLINICAI SIGNIFICANCE

The blood level of tumor necrosis factor (TNF) was measured by the method of TNF cytotoxic activity in 20 patients with abdominal infection, 10 patients with gastrointestinal cancer and 30 normal subjects. The results showed that the blood level of TNF in patients with abdominal infection was significantly higher than that in normal subjects and in patients with gastrointestinal cancer. The blood level of TNF in serious abdominal infection was remarkably higher than that in moderate abdominal infection. The blood level of TNF in both group of patients was obviously declined after the infection had been controled. The blood level of TNF in the patients died from abdominal infection was constant during treatment. Therefore, the blood level of TNF is an important index of abdominal infection and has important clinical value in evaluating the seriousness of abdominal infection and predicting the development of infectious disease.
10.Confocal laser endomicroscopy for differential diagnosis of gastric ulcer

Objective To estimate the value of confocal laser endomicroscopy (CLE)for differential diagnosis of gastric ulcer. Methods Patients diagnosed as gastric ulcer by conventional endoscopic mode of CLE were enrolled. The eonfocal endoscopic mode of CLE was used to observe the borderline and/or the surface of the ulcers. Biopsy specimen was taken for histopathological examination and the result of CLE was compared with histopathological finding. Results A total of 12 769 CLE images were obtained from 150 sites of 43 patients. The sensitivity and specificity of CLE for phlegmonosis process were 83.54% and 85.92%, respectively, with positive predictive value of 86.84% and negative predictive value of 82.43%. Whereas the sensitivity and specificity of CLE for intraepithelial neoplasia were 53.57% and 88.52%, respectively, with positive predictive value of 51.72% and negative predictive value of 89.26%. As for diagnosis of gastric cancer, the sensitivity and specificity of CLE were 88.37% and 93.46%, respectively, with positive predictive value of 84.44 % and negative predictive value of 95.24%. The kappa values for agreement of histopathological findings and CLE for diagnosis of phlegmonosis process, intraepithelial neoplasia and gastric cancer were 0.69,0.42 and 0.81, respectively. Conclusions CLE can provide a real-time histopathological diagnosis of the gastrointestinal tract with high sensitivity, specificity, positive and negative predictive values. Targeted biopsy can be done under CLE and the detective rates of precancerous lesions and tumors will be increased.
