1.Study on the potential mechanisms of leukemia cell resistance to TRAIL-induced apoptosis.

OBJECTIVETo explore the potential mechanisms of leukemia cell resistance to tumor necrosis factor-related apoptosis-inducing ligand (TRAIL) -induced apoptosis.
METHODSCells apoptosis, changes of mitochondrial membrane potential, activity of NF-kappaB, activity of caspase-8 and expressions of apoptosis-related proteins in TRAIL treated K562 and CEM cells, were detected by flowcytometry, ELISA and Western blotting methods, respectively.
RESULTSAfter treated with TRAIL, the apoptosis indexes were 29.98% and 14.1%, and mitochondrial membrane potential were decreased to 73.25% and 25.4% in K562 and CEM cells respectively. Constitutive level of caspase-8 expression in CEM was lower than that in K562 cells. Both cells became over-expressed Bcl-xL and down-regulated Bax. The ratio of Bcl-xL/Bax in CEM cells was higher than that in K562 cells. Compared with that in K562, the NF-kappaB activity increased significantly in CEM after treatment with TRAIL in early stage.
CONCLUSIONCEM cells were more resistant to TRAIL-induced apoptosis than K562 cells did. The potential mechanisms associated with CEM drug resistance might be the lower expression of the constitutive level of caspase-8, lower sensitivity of mitochondrial inner membrane, early increase in NF-KB activity and altered expression of Bcl-2 proteins family.

2.Study on inhibitory effect of antisense VEGF RNA on the growth of hepatocellular in vitro in vivo.

OBJECTIVETo determine the effects of PCMV-FGEV transfection on the profile of SMMC-7721 hepatocellular in vitro in vivo.
METHODSSMMC-7721 hepatocellular was transfected with PCMV-FGEV antisense, PCMV-VEGF sense and empty vector plasmid encapsulated by lipofectamine. The positive cell clones were selected with G418. The stable transfection and expression of VEGF in the SMMC-7721 hepatocellular were determined by in situ hybridization and immunochemical analysis. The effect of PCMV-FGEV transfection on SMMC-7721 hepatocellular proliferation was observed by MTT colorimetric assay. Flow cytometry was used to determine the effects of PCMV-FGEV transfection on cell apoptosis of SMMC-7721. The growth of transfected cells was also observed in nude mice.
RESULTSThere was reduced VEGF expression in SMMC-7721 transfected with PCMV-FGEV confirmed by in situ hybridization and immunohistochemical analysis. There was no effect of PCMV-FGEV transfection on cell proliferation and cell apoptosis of SMMC-7721 in vitro. The growth of cell with PCMV-FGEV transfected was slow in nude mice (vivo) and accompanied with obvious apoptosis. The latent time of tumor in the antisense mice group was 25.0+/-1.8 days, which was longer than that in sense and control group significantly (F=19.455, P<0.01). On the other hand, the average tumor weight in antisense group (0.96 g+/-0.28 g) was the smallest among the three groups (F=21.501, P<0.01).
CONCLUSIONSThe expression of VEGF was inhibited by PCMV-FGEV. There was no effect on cell proliferation and cell apoptosis of SMMC-7721 by transferring PCMV-FGEV gene into SMMC-7721 cells in vitro. But in vivo it can inhibit tumor growth and induce cell apoptosis.

OBJECTIVETo explore the effects of mitochondrial pathways on apoptosis in colon carcinoma cells induced by Tumor necrosis factor related apoptosis inducing ligand and offer evidences for TRAIL application in clinic.
METHODSApoptosis, integration of mitochondria (including DeltaPsim, cardiolipin), activity of Caspase-9 and release of cytochrome c in colon carcinoma cells SW1116 treated with TRAIL, were detected by means of flowcytometry, fluorometer method and western-blot at the different time point.
RESULTSAfter treated with TRAIL for 4 hours, the apoptosis index was 32.98%, and the damage of mitochondria occurred with DeltaPsim, cardiolipin decreased, and the activity of Caspase-9 and cytochrome c increased. The Caspase-9 activity at 24 hour was (48.12+/-2.21)micromol.L(-1).h(-1).mg(-1) protein. Mitochondrial damage induced by TRAIL could be inhibited by Caspase inhibitor Z-VAD. fmk.
CONCLUSIONMitochondrial pathways involved in the apoptosis of colon carcinoma cell induced by TRAIL. Cytochrome c was released and Caspase-9 was activated in the Caspase-dependent manner after the damage of mitochondrial.

Osteoarthritis (Osteoarthritis, OA) is a common clinical degenerative joint disease with increased incidence rate in recent years. Animal experiment is one of the important ways to explore pathogenesis and treatment of OA, while induced animal model is the most important part in animal experiment. Intra-articular injection of drugs is a classical method for establishing animal model of OA. Choose of animal should follows the principle of correlation, appropriateness and practicability, injections should perform in accordance with experimental purposes and subject, detections means and evaluation methods also should corresponding to experimental reality. The gold standard of OA animal model and intra-articular injections has not build, need further study.

5.Investigation of the Relationship between Cognitive Function and MRI Manifestation in Patients with SHPT

Objective To investigate the relationship between cognitive function and magnetic resonance imaging (MRI) manifestations in patients with secondary hyperparathyroidism (SHPT) by analyzing brain MRI diffusion-weighted imaging,diffusion tensor imaging,serological markers,and cognitive function.Methods We evaluated 49 random patients with SHPT.We adopted two functional scales to evaluate cognitive function and performed MRI fractional anisotropy (FA) and apparent diffusion coefficient (ADC) to evaluate white matter function of brain functional areas.We recorded serological markers,and conducted statistical analysis to analyze correlation among cognitive function scores,brain MRI in white matter functional areas,and biochemical parameters.Results We found that at the frontal area,the FA and ADC values were correlated with the MMSE language and memory function scores;at the parietal area,the FA and ADC values were correlated to the MoCA visuospatial executive function scores;the other areas had no significant correlation with cognitive function scale score.Cognitive function was negatively correlated with immunoreactive parathyroid hormone (iPTH) levels but positively correlated with education level,while it was not correlated with sex,age,blood pressure,and serum calcium levels in patients with SHPT.Conclusion In patients with SHPT,cognitive function is correlated with iPTH level and education level.The FA and ADC values of brain MRI in the frontal and parietal lobes might play a diagnostic role in evaluating cognitive function and locating injury for patients with SHPT,which requires clinical attention.
6.Study of the effect of hepatitis C virus core protein on interferon-induced antiviral genes expression and its mechanisms.

To study the effect of HCV core protein on the interferon-induced antiviral genes expression and its mechanisms. Methods HepG2 cells were transiently transfected with HCV core protein expression plasmid and the blank plasmid respectively. RT-PCR was used to analyze the effect of HCV core protein on PKR and 2'-5'OAS expression. The effect of HCV core protein on ISRE-medicated gene expression was detected by luciferase activity assay. Western-blot assay was performed to observe the change of mRNA and protein levels of SOCS3, STAT1 and p-STAT1 following HCV core expression. In the presence of HCV core protein, the transcription of PKR and 2'-5' OAS are down-regulated. ISRE-medicated reporter gene expression and STAT1 phosphorylation were inhibited. The transcription and expression of SOCS3 were induced compared with blank plasmid-transfected group. In HepG2 cells, HCV core protein can down-regulate the expression of some interferon-induced antiviral genes, which involves the induction of SOCS3 and the inhibition of STAT1 phosphorylation.

7.Soft tissue changes observed through musculoskeletal ultrasound in the treatment of knee osteoarthritis with needle-knife.

OBJECTIVETo study soft tissue changes observed through musculoskeletal ultrasound (MSUS) in the treatment of knee osteoarthritis with needle-knife, so as to provide MSUS basis for needle-knife in the treatment of knee osteoarthritis.
METHODSForty patients with knee osteoarthritis treated in the Third Affiliated Hospital of Beijing University of Chinese Medicine from December 2011 to December 2012 were selected according to inclusion and exclusion criteria. All the patients were treated with needle-knife release method. The VAS scores and knee joint circumference were recorded before treatment and 2 weeks after treatment. The changes of knee joint hydrops articuli and joint synovial thickness were measured through MSUS.
RESULTSThe knee pain index was 6.850 +/- 1.417 before treatment and 2.790 +/- 1.299 after treatment;the index after treatment was lower than that of before treatment. The knee joint circumference was 407.320 +/- 45.151 mm before treatment and 391.240 +/- 41.129 mm after treatment; the knee joint circumference decreased after treatment. The amount of hydrops articuli observed by musculoskeletal ultrasound showed that 47 knees were cured, 19 knees improved and 2 knees failed. The synovial membrane thickness: 43 knees cured, 17 knees improved and 8 knees failed.
CONCLUSIONThe hydrops articuli and synovial thickness of knee joint of patients with knee osteoarthritis observed under the MSUS is consistent with the main symptoms and signs, which suggests that MSUS observation on soft tissue changes before and after needle knife in the treatment of knee osteoarthritis with high reliability.

8.Urethroplasty with oblique preputial island flap for the treating of hypospadia.

OBJECTIVETo Explore the effect of oblique preputial island flap for the treating of hypospadias.
METHODSFifty-one patients were performed one-stage urethroplasty with oblique preputial island flap to repair hypospadias.
RESULTSAll cases resulted in a good contour of the penis without any redundancy and a normal anatomic position of slit-shaped urethral meatus. The urination was perfect. Six patients occurred complications (3 cases of urinary fistula, 3 cases of meatal stenosis).
CONCLUSIONWith extensive scope of materials, reliable blood supply of skin flap, satisfactory appearance of shaping penis and few complications, one-stage urethroplasty with oblique preputial island flap is an effective method to repair hypospadias of penile type and penile-scrotal type.

9.Expression of PH20 in primary and metastatic breast cancer and its pathological significance.

OBJECTIVETo evaluate the expression pattern of PH20 in primary and metastatic breast cancer and its relationship to tumor metastatic potential.
METHODSAnti-PH20 antibody was synthesized by injection of conjugated human PH20 peptides into rabbits. Immunohistochemical study was performed on 53 cases of human breast cancer. Western blot was used to detect PH20 expression in 5 cases of breast cancer with available fresh tissue. Two oligonucleotide probes were prepared for in-situ hybridization using breast tissue microarray.
RESULTSNormal breast tissue did not express PH20 (0/3), while 58.4% (31/53) of breast cancer cases did. The highest expression rate was found in metastatic foci in regional lymph nodes (83.3%), followed by primary breast cancer tissue in cases with lymph node secondaries (70.8%). The breast cancer cases with no any metastasis had an expression rate of 48.2%. The immunohistochemical staining results were further confirmed by Western blotting. In-situ hybridization showed PH20 RNA in 75% of the breast cancer tissue (21/28). Two of the 17 cases of normal breast tissue showed weak expression in some ductolobular units.
CONCLUSIONSThe expression of PH20 has a positive correlation with metastatic potential in breast cancer. It is possible that PH20 may play an important role in the invasive growth and metastasis of breast cancer cells, via mechanisms such as digestion of surrounding stromal tissue and release of FGF-2.

10. Mechanism of Tibetan medicine Lagotisbrachystachys Maxim in treatment of chronic alcoholic liver injury and acute gouty arthritis based on network pharmacological and molecular docking

Aim To explore the common mechanism of Lagotisbrachystachys Maxim in treating chronic alcoholic liver injury and acute gouty arthritis. Methods TCMSP, CTD database and the related literature were used to obtain the chemical composition and target of Lagotisbrachystachys Maxim, combined with the data of Gene Cards, OMIM to screen the common target; an active ingredient-disease-common target network diagram was constructed, GO, KEGG enrichment analysis were performed and molecular docking were carried out to verify the binding of components and common target. Results Seven compounds in Lagotisbrachystachys Maxim could act on 253 common targets to exert their therapeutic effects; 20 key targets, such as IL-6, ALB, TNF, TP53, VEGFA, STAT3, CXCL8 and IL- 10, and 44 signaling pathways, such as Toll-like receptor signaling pathway, nod-like receptor signaling pathway, JAK-STAT signaling pathway, FcϵRI signaling pathway and PPAR signaling pathway, were used to treat the two diseases. The results of molecular docking showed that bicuculline, chryseriol and luteolin might be effective compounds for treating the two diseases. Conclusions Through a preliminary analysis of the common molecular mechanism of Lagotisbrachystachys Maxim in the treatment of chronic alcoholic liver injury and acute gouty arthritis, this study provides i-deas for future research on the mechanism of action.
