1.Laminin B1 mRNA expression in squamous cell carcinoma of the skin.

BACKGROUND: Laminin, which is present in all basement membrane, is a multifunctional glycoprotein with diverse biological activities. Like fibronectin, it can influence cell adhesion, growth, morphology, differentiation, and migration as well as the assembly of the extracellular matrix. Laminin primarily affects cells of epithelial origin, and response varies depending on the cells. OBJECTIVE: Studies about the relationship of laminin and the pathogenesis or metastasis of squamous cell carcinoma(SCC). MEHTODS: In this study, we have examined the laminin B1 mRNA expression and localizing pattern of laminin in SCC, using Northern and slot-blot hybridization, in situ hybridization and immunoperoxidase stain. RESULTS: In Northern blot hybridization, 5.6Kb mRNA transcript was detected in control and SCC specimens. Slot-blot hybridization indicated theat amounts of laminin B1 mRNA expression in SCC was 677.0+/-268.5 densitometer absorbance unit, in control was 228.0+/-16.37 unit respectively. There is a 3-fold increase in SCC compared to control. In in situ hybridization, 35S-labeled laminin B1 cDNA-mRNA hybrid granules were detected along the basement membrane and adjacent cells of SCC. In immunoperoxidase stain, laminin has stained strongly and continuously along the basement membrane of the tumor nest in well differentiated SCC, and weakly and interrupted in poorly differentiated SCC. CONCLUSION: These results suggested that laminin might be related to the pathogenesisand metastasis of squamous cell carcinomas.

2.Quality of Life in Survivors of Patients after Hematopoietic Stem Cell Transplantation and Received Chemotherapy.

PURPOSE: This study is to assess the quality of life(QOL) of hematic cancer survivors after hematopoietic stem cell transplantation(HSCT) and received chemotherapy(RC) to prepare basic information for nursing interventions in order to improve the patients' QOL. METHOD: The data were collected by self-reporting questionnaire from January to March, 2003 intended for outpatients at the Cancer center of D university hospital in Busan. All 44 of them were diagnosed as hematic cancer and had spent 100 days after getting HSCT and complete remission(CR) throughout RC. The collected data were analyzed with descriptive statistics, t-test. ANOVA using SPSS/WIN 10.0 program. RESULTS: The total mean score of the QOL was moderate. In case of survivors in HSCT, the total mean score of the QOL was 5.81+/-.08, and that of survivors in RC was 5.94+/-.13. The facts above has not been considered statistically as the result of analysis of differences in each domain of the QOL depending on the general characteristics of the objects of this study. CONCLUSION: The total mean score of the QOL was at moderate levels, indicating that the survivors after HSCT and RC were perceiving their QOL as moderate. In the nursing business aspect, the most important thing is to understand the QOL which the 2 groups of the survivors perceive, and the plans of nursing intervention that can be helpful to more qualitative life should be studied constantly.

3.Alterations of COX-1, 2 mRNA and Protein Expression in Pregnant Rat Kidney.

The pregnancy causes the marked changes in maternal renal hemodynamic and volume homeostasis. During pregnancy, renal sodium and water retention result in an expansion of extracellular fluid and plamsma volume. This study was to examine the alteration of expression and localization of COX-1, 2, mRNAs and proteins in the kidneys of non-pregnant (NP) and pregnant rats using RT-PCR, Western blot analysis and immunohistochemistry. Pregnant Sprague-Dawley rats were evaluated on various time sets : days 10.5 (P10.5), 12.5 (P12.5), 17.5 (P17.5), and 19.5 (P19.5). In RT-PCR, COX-1 expression was gradually increased from P10.5 to P19.5 compared with NP rat. COX-2 expression was gradually decreased from P10.5 to P17.5 compared with NP rat, but restored NP level at P19.5. In Western blot analysis, COX-1, 2 proteins were detected in ~70, ~72 kDa, respectively. COX-1 expression was gradually increased from P10.5 to P17.5 and peaked at P19.5 compared with NP rat. COX-2 expression of pregnant rats was slightly decreased compared with NP rat. In NP rat, immunoreactivity of COX-1 was detected in entire collecting duct, glomerular epithelium, and medullary interstitial cells. In pregnant rats, the pattern of cellular labeling and signal intensity of COX-1 protein was identical to NP rat, but signal intensity was markedly increased in the inner stripe of outer medulla and inner medulla at P19.5. COX-2 immunoreactivity of NP rat was detected in the cortical thick ascending limb and macula densa. In pregnant rats, the pattern of cellular labeling of COX-2 protein was identical to NP rat, but signal intensity was slightly decreased. These results suggest that the expansion of extracellular fluid volume and water retention may be partly regulated by COX-1 rather than COX-2 during the pregnancy, especially at late stage.

4.Perinatal Prognosis of Single Umbilical Artery.

OBJECTIVE: To evaluate the perinatal prognosis of fetuses with a single umbilical artery. METHODS: From 1992 to 1998, nineteen cases with single umbilical artery(SUA) was observed in 8,704 deliveries at Chonnam University Hospital. RESULTS: Out of nineteen fetuses, thirteen fetuses with single umbilical artery were detected by antenatal ultrasonographic examination and six fetuses were detected after birth. The male to female ratio was 0.9: 1. Congenital malformations were observed in 8 babies(42.1%) and included leg deformity, esophageal atresia, imperforated anus, ventriculomegaly, meningocele, hydronephrosis, ventricular septal defect, joint contracture, cleft lip and palate, toe anomaly, imperforated anus, kyphosis, no urethra and testis, clubfoot, patent ductus arteriosus and rnild mitral regurgitation. Among 10 cases of karyotyping analysis three cases were diagnosed as trisomy 18. Fourteen fetuses(77.8%) showed growth restriction at delivery. Antenatal obstetric complications were hydramnios(n = 3), oligohydramnios(n =2), and severe preeclampsia(n = 3). CONCLUSION: Careful ultrasonographic evaluation for the identification of a SUA is necessary because of its frequent association with congenital anomaly, growth restriction and cytogenetic abnormality.

5.DNA Profiling via Short Tandem Repeat Analysis by Using Serum Samples.

Serum is free of cellular components. Because DNA is located in the nuclei or mitochondria of cells, serum could be assumed DNA free. Few previously published case reports to date have used serum for DNA typing. Here, we report on human genotyping via short tandem repeat (STR) analysis using serum as a sample, and discuss problems involved in the process.

6.The Results of Treatment in Femoral Neck Fractures: The Clinical Study

We analyzed ninety one fractures of femoral neck treated with surgery and followed for more than 2 years at Seoul National University from January 1977 to June 1983. Of these, 45(49%) were treated with internal fixation(compression hip screw or Knowles' pinning), 26(29%) with primary endoprosthesis, 20(22%) with total hip replacement arthroplasty. The criteria for internal fixation were fresh fracture that could be reduced satisfactorily and the age of the patient were under 65 years. The criteria for endoprosthesis were displaced subcapital or transcervical type or severe osteoporosis with intact acetabular cartilage. The criteria for total hip replacement arthroplasty were neglected fractured patients with significant medical problems, including cardiovascular, pulmonary, and renal disease, that posed significant risks for a second operation, and patients who had previous hip disease such as degenerative arthritis or rheumatoid arthritis. Following results were obtained. 1. In our series, the most frequent occurrence of femoral neck fractures was in the 7th decade(34 %), and the most common cause was fall down(72.6%). 2. In fresh fractures less than 3 weeks after injury, transcervical type (73%) according to anatomical classification and Garden's type III(44%) according to displacement were the most common type, respectively. 3. After internal fixation, 11% of avascular necrosis and 9% of non-union occurred. After endoprosthesis, 4% of acetabular protrusion, 8% of loosening and 12% of loosening and 12% of painful hip occurred. After total hip replacement arthroplasty, 10% of dislocation and 10% of loosening occurred. 4. Functional end results of excellent 56%, good 22%, fair 6% and poor 16% in internal fixation group, excellent 31%, good 38%, fair 12% and poor 19% in endoperosthesis group, and excellent 45%, good 30%, fair 10% and poor 15% in total hip replacement arthroplasty group were obtained.

10.The Effect of Job Demand, Leader Member Exchange, and Recovery Experience from Job Stress on Job Embeddedness of Workers in the Manufacturing Industry.

PURPOSE: This study was conducted to investigate factors affecting the job embeddedness of workers in the manufacturing industry. METHODS: The survey was conducted on 261 workers of the manufacturing industry in P city and Y city with the help of a structured self-report questionnaire, administered between June 10 and June 30, 2017. Data were analyzed using descriptive statistics, a t-test, ANOVA, a Scheffés test, Pearson's correlation coefficients, and stepwise multiple regression. RESULTS: There were significant differences in job embeddedness according to educational level, marital status, jobs and types of employment, satisfaction with salary, stress level, and the perceived health status of the subjects. There were significant positive correlations between role clarity of job demands (r=.45), leader member exchange (r=.48), recovery experience from job stress (r=.27), and job embeddedness. From the multiple regression analysis, the most significant factors affecting job embeddedness were found to be leader member exchange (β=.43), recovery experience from job stress (β=.22), and job demand (β=.15). These variables explained 35.0% of the total variance in job embeddedness. CONCLUSION: In order to increase job embeddedness of workers in the manufacturing industry, it is necessary to prepare measures to increase job demand, leader member exchange, and recovery experience from job stress.
