2.Application of TOPSIS Method in the Assessment of TCM Appropriate Technology ;Packages

Objective To evaluate the promotional effectiveness of different TCM appropriate technology packages; To provide a scientific basis for improving the technology package. Methods 8 assessment indexes, including input costs, the number of experts, the number of people with senior professional post, the number of grassroots people under the guidance of experts, the number of training hours, the number of people receiving training, evaluating rate and the number of users were selected. TOPSIS method was used to evaluate the promotional effectiveness of five kinds of TCM packages in grassroots. Results The Ci values for appropriate technology packages were 0.76 for pains in neck, shoulder, waist and lower extremities, 0.49 for gynecological diseases, 0.44 for infantile diarrhea, 0.66 for chronic gastropathy, and 0.00 for tumors. Conclusion The effectiveness of promotion of TCM appropriate technology packages from high to low is pains in neck, shoulder, waist and lower extremities package, chronic gastropathy package, gynecological diseases package, infantile diarrhea package, tumors package.
3. Phenotypic and functional characteristics of natural killer cells in HBV cirrhotic patients

Objective To analyze the phenotypic and functional characteristics of hepatic and peripheral natural killer (NK) cells in HBV cirrhotic patients, and further confirm the key role of NK cells in the immunopathogenesis of liver fibrosis. Methods Thirty HBV cirrhotic patients were recruited for this study in the First People's Hospital of Zhengzhou and Fifth Medical Center of Chinese PLA General Hospital from January 2015 to January 2017, meanwhile thirty age-and sex-matched healthy individuals were enrolled as health controls. Liver biopsies were collected from 10 HBV cirrhotic patients, and 8 healthy liver tissue samples were obtained from the healthy donors whose livers were used for liver transplantation. The frequency, phenotypic and functional characteristics of hepatic and peripheral NK cells from the two groups were analyzed by using multicolor flow cytometry. The killing activities of NK cells against HSCs were explored using LX-2 cell model (a cell line of HSCs) in vitro. Results Compared with health controls, the percentages of hepatic and peripheral NK cells were reduced significantly in cirrhotic patients (U=8.5, P=0.006; U=184.0, P<0.001, respectively); the expression levels of activation markers CD69 (U=102.0, P=0.009), HLA-DR (U=82.5, P<0.001) and CD38 (U=0.0, P=0.029) were increased; the expressions of functional molecule granzyme B (U=0.0,P=0.004) was decreased in peripheral NK cells, and perforin and Granzymes were also decreased in hepatic NK cells except TRAIL that was up-regulated (U=4.0, P=0.026), especially the change of perforin (U=4.0, P=0.034); the functional decrease in IFN-γ production (U=2.0, P=0.032) was observed in vitro in hepatic NK cells and the reduction of CD107a degranulation and IFN-γ production were both observed in hepatic and peripheral NK cells (U=88.0, P=0.018; U=13.0, P<0.001, respectively); purified NK cells from peripheral blood of HBV cirrhotic patients could induce less 7AAD– Annexin V+ early apoptotic LX2 cells and 7AAD+ Annexin V+ late apoptotic LX2 cells compared with those from health controls (U=6.5, P=0.025; U=2.0, P=0.002, respectively). Conclusions NK cells from HBV cirrhotic patients displayed a decreased frequency, activation increase, functional decrease in CD107a degranulation and IFN-γ production, and cytolytic activities decrease against HSCs in response to various stimulators in vitro compared with those from health controls. These findings demonstrate an impaired anti-fibrotic function of NK cells' in HBV cirrhotic patients and further clarify the immunopathogenesis of liver fibrosis.
4.Proliferation, collagen production and related gene expression in keloids and normal skin fibroblasts

Objective To study the proliferation,collagen production and related gene expression in keloids and normal skin fibroblast.Methods Isolated primary cells of keloid fibroblasts (KFb,n=12) and normal human dermal fibroblasts (NFb,n=12) were identified,the cell viability and proliferating potential and the cell cycle were detected,and the difference on the collagen synthesis between KFb and NFb were compared.The expression of cell cycle-associated genes such as p21,p16,and p27 was dectected by real-time fluorescent quantitative PCR.Results The phase contrast optical microscopy imaging showed that both KFb isolated from keloid tissues and NFb from normal skin tissues possessed classic and similar fibroblast morphology.But there was a significant difference between cell proliferation,Hyp [(2.30±0.10) μg/ml vs.(1.66±0.13) μg/ml,P<0.05] and collagen levels [(17.19±0.75) μg/ml vs.(12.37±0.94) μg/ml,P<0.05].Compared with NFb,KFb exhibited more percentage of G2/M phase cells [(5.90±0.62)% vs.(16.94 %±1.93)%,P<0.05]and less percentage of G0/G1 phase cells [(90.24 ±2.27)% vs.(75.65±1.92)%,P<0.05].Cell cycle related genes p16,p21 and p27 were low expressed.Collagen type Ⅰ was highly expressed at mRNA levels in KFb than that in NFb [0.84±0.11,1.32±0.2,1.69±0.12,4.33±0.27 in KFb vs.1.43±0.13,2.56±0.26,2.89±0.37,1.40±0.12 in NFb,P<0.05].Conclusions There are cell dysfunction and abnormal cellular dynamics in keloid fibroblasts.The formation of keloid likely involves aberrant interactions of some genes that affected its development at different extents.
5.Dynamic Changes of Type Th22 Cell Immunological Response During Atherosclerosis Process in Experimental Mice

Objective: To study the dynamic changes of type Th22 cell immunological response during atherosclerosis process in experimental mice in order to provide a new theoretical basis for atherosclerosis therapy. Methods: 8 weeks C57BL/6J mice were divided into 2 groups: Experiment group,n=24 ApoE-/- mice and Control group, n=24 normal mice. All animals received high fat diet and the following indexes were compared between 2 groups at 0, 4, 8, 12 weeks after treatment: aortic atherosclerotic lesions were deifned by Oil red O staining, dynamic changes of Th22 cells in spleen were measured by lfow cytometry, mRNA expressions of interleukin-22 (IL-22), IL-22R1, AhR and T-bet in aorta were examined by RT-PCR, blood levels of IL-22 was detected by ELISA. Results: Compared with Control group, Experiment group had the increased area of aortic atherosclerosis (the ratio of plaque area/lumen area) and Th22 cell (CD4+ IL-22+/CD4+T cell) amount, elevated mRNA expressions of IL-22, IL-22R1, AHR, T-bet in aorta and higher blood levels of IL-22 at all time points, the differences between each time point (except 0 week) had the statistic meaning,P<0.05. In Experiment group, the differences between 2 adjacent time points, for the area of aortic atherosclerosis and mRNA expressions of AHR, T-bet: 4 weeks vs 0 week, 8 weeks vs 4 weeks, 12 weeks vs 8 weeks all had statistic meaning; for Th22 cell amount: 4 weeks vs 0 week, 8 weeks vs 4 weeks had statistic meaning and 12 weeks vs 8 weeks had no real distinction; for mRNA expressions of IL-22, IL-22R1 and blood levels of IL-22: 4 weeks vs 0 week had statistic meaning and 8 weeks vs 4 weeks, 12 weeks vs 8 weeks had no real distinctions. Conclusion: Hyperactive immunological response of Th22 cells might be involved in atherosclerosis process, the relevant mechanism should be further studied.

7.Evaluation of Dental Implants Stability in Patients with Lipid Metabolism Disorder

Objective To study the expression of inflammatory factors in the saliva and the stability of implants after implant denture repair in pa-tients with dysplasia of the dentition defect ,and to analyze the prognosis of patients with lipid metabolic disorder. Methods The dentition defect patients with lipid metabolism disorder were selected as the experimental group. Patients with lipid defect were selected as the control group. Saliva was collected before implantation,1 month and 3 months after implantation. ELISA method was used to detect IL-6,IL-1βand TNF-α. Osstell ISQTM radio frequency analyzer was used to detect the stability of the implants and the periodontal status was detected by Florida probe. Re-sults There were no significant differences in the depth of implant probing(PD),bleeding index(BI)and ISQ between the two groups by radio frequency analyzer(all P>0.05). The levels of IL-6,IL-1βand TNF-αin the pre-implantation,1 month and 3 months after implantation were sig-nificantly higher than those in the control group(all P<0.05). The levels of IL-6,IL-1βand TNF-αin the experimental group were significantly higher than those in the control group before planting(all P<0.05) 3 months after planting,there were no significant differences of IL-6,IL-1β, and TNF-αbetween the experimental group and control group(all P>0.05). Conclusion The patients with dyslipidemia after implantation have good implant stability as those with normal blood lipid ,and the patients with dyslipidemia can also obtain good prognosis.
8.Distribution of the Activated Acupoints after Acute Gastric Mucosal Injury in the Rat

Objective To observe the dynamic distribution of the extravasated Evans Blue (EB) dye points (neurogenic inflammatory response) at the skin after acute gastric mucosal injury (AGMI) and its relation to the related regular acupoints in the locations in rats. Methods A total of 70 Wistar rats were randomized into normal control (n=10),normal saline (n=10),and AGMI (n=50) groups. The AGMI group was further divided into 5 h,2 d,3 d,4 d and 5 d subgroups with 10 rats in each. AGMI model was duplicated by intragastric perfusion of diluted hydrochloric acid (HCl,0.5 mol/L). Evans Blue Dye (50 mg/kg,50 mg/mL in 0.9% saline) was given to the rats before AGMI modeling. The plasma extravasated EB points at the skin of the whole body were observed after removal of the hair. Results The extravasated EB points presented a nerve-segmental distribution,with the proportion of the points in the location being 47.5% for "Geshu" (BL 17),58.82% for "Jizhong" (GV 6),88.23% for "Pishu" (BL 20),82.35% for "Weishu" (BL 21),17.64% for "Zhongwan" (CV 12),and 5.88% for "Shangwan" (CV 13),respectively. The plasma extravasation of EB seldom appeared in normal rats and only fewer points were found in rats accepted administration of 0.9% saline.
