1. Biological significance of the expression of B-myb in hepatocellular carcinoma and the correlation with cyclin D1

Objective: To study the expression of the B-myb and cyclin D1 in human hepatocellular carcinoma (HCC) tissue and explore their correlation with initiation and progression of HCC. Methods: The expressions of B-myb and cyclin D1 in 60 cases of HCC tissue and the corresponding pericancerous liver tissue were detected by immunohistochemical method. Results: The positive rates of B-myb and cyclin D1 in the HCC tissue were higher than those in the pericancerous liver tissue (56.67% vs 36.67% and 50% vs 31.67%, P < 0.05) respectively. The expression of B-myb in the HCC tissue significantly correlated with the clinical stage, the extrahepatic metastasis, post-operative recurrence, and the number of tumor nodes but not correlated with the tumor thrombus in portal vein, the tumor diameter, the serum level of alpha-fetoprotein (AFP), and tumor differentiation. The expression of cyclin D1 in the HCC tissue was significantly associated with the clinical stage, the tumor thrombus in portal vein, extrahepatic metastasis, post-operative recurrence, the number of tumor nodes, and the differentiation of tumor but not associated with the tumor diameter and the serum level of AFP. The expression of B-myb had positive correlation with expression of cyclin D1 in HCC tissues. Conclusion: The overexpressions of B-myb and cyclin D1 in HCC tissue may contribute to the proliferation of hepatoma cells, and have close correlation with the initiation and progression of HCC.
2.Oncogene DEK shows synergestic effect with transcriptional factor AP-2α to promote HER2 overexpression in breast cancer

Objective To investigate the pathological significance of DEK-AP-2α interaction in HER2 overexpres-sion and breast tumorigenesis. Methods The protein level of DEK,AP-2α and HER-2 in breast tumor tissues was detected by Western blot. The interaction of DEK and AP-2α in MDA-MB-453 mammary cancer cells was detected by immuno-coprecipitation. Furthermore the impact of DEK and AP-2α on HER2 expression was investigated by siRNA in MDA-MB-453 mammary cancer cells followed by semi-quantitive RT-PCR and Western blot. Results A correlation between DEK, AP-2α and HER2 levels in breast cancer tissues were found. The interaction between DEK and AP-2α in MDA-MB-453 cells was verified by co-immunoprecipitation assay. Depletion of DEK and AP-2α in MDA-MB-453 cell by siRNA cooperatively repressed HER2 expression at both the mRNA and protein levels. Conclusion Oncogene DEK has synergestic effect with AP-2α transcriptional factor to promote HER2 overexpres-sion in breast cancer.
3.Expression of Oct4 gene in cervical cancer

Objective To study the expression of Oct4 gene, an embryonic stem cell marker, in cervical carcinoma cell lines and tissues for the purpose of finding the relationship between this gene and cervical cancer. Methods The expression of Oct4 in normal cervical tissues and cervical carcinoma tissue and cell lines was detected by the immunohistochemistry and RT-PCR. Results ① The expression of Oct4 mRNA in cervical carcinomas (73.3%, 22/30) was significantly higher (P<0.01) than that in normal cervical tissues (25%, 3/12); ② The rate of Oct4 protein expression in cervical cancer (53.7%, 22/41) was higher (P<0.01) than that in normal cervical tissues (7.7%, 1/13); ③ Oct4 protein was expressed in cervical carcinoma cell lines and primary tumorspheres. ④ The expression of Oct4 in tumorspheres was down-regulated during cell differentiation. Conclusion Our results imply that Oct4 may be one of the cervical cancer stem cell markers, but this still needs further research data to support.
5.Effect and treatment of Mason type-Ⅲ radial head fractures with type-T plate

[Objective]To analyze the result of mini-type T plate for treatment of Mason type-III radial head fracture.[Method]Seven cases of Mason type-Ⅲ radial head fractures,6 male and 1 female,including 2 cases with radial neck fractures,were treated by open reduction and mini-type T plate fixation associated with Kirscher wires.If necessary,bone graft was placed between radial head and neck.[Result]The patients were followed up for an average of 20.3 months(ranged from 14 to 39 months).All fractured radial heads were heal and no case appeared ischemic necrosis.According to the Brobrg and Morrey functional elbow index,the averaged score was 87 points and the outcome was excellent in 2 patients,good in 4,the excellent and good rate was 85.7%.[Conclusion]Treatment of Mason type-Ⅲ radial head fractures by open reduction,mini type T plate fixation assocated with Kerscher's wire and autogenous bone graft showed a high union rate of fracture and satisfactory joint function but no ischemic necrosis with the advantages of reservation of radial head and decreasing the rate of re-section and prosthesis replacement.Further observation should be taken to get more clinical cases number.
7.Application Value of Doctor-patient Communication Courses to Medical Staff

Objective: To investigate the significance of doctor -patient communication courses to medical staff.Methods:Using self-made questionnaire and self -evaluation of anxiety scale (SAS), selection in our u-niversity study of 200 cases of medical personnel , in accordance with the doctor -patient communication before and after the course is divided into classes before and after teaching group , respectively on two groups before and after learning the medical staff on the SAS scores and questionnaire survey .Results:Compared with group before teach-ing, teaching group after medical staff thought through the study of doctor -patient communication course , is con-ducive to cultivating communication consciousness , improve doctor -patient communication skills as well as pre-venting the doctor-patient contradiction (P<0.05).After teaching group SAS score (34.50 ±6.88), signifi-cantly lower than the teaching group SAS score (45.51 ±6.97), difference was statistically significant (t =15 .622 ,P<0 .01 ) .Conclusion:Doctor-patient communication courses are effective way to improve the ability to communicate , to alleviate the anxiety of the medical staff at the same time also has a certain value .
8.Investigation of relevant genes to the pathogenesis and metastasis of osteosarcoma with SBC gene microarray

[Objective]To investigation the relavant genes to the pathogenesis and metastasis of osteosarcoma,and to discuss the significance of gene expression in the pathogenesis and metastasis of osteosarcoma. [Method]Two well-known ATCC osteosarcoma cell lines and an osteoblastic cell line were collected.After the total RNA was extracted,the finally synthesized biotinylated cRNAs were hybridized to SBC arrays.After the design and synthethesis of the primers,four of the differentially expressed genes were then chosen at random to further confirm the array results using the SYBR Green real-time RT-PCR method in freshly collected osteosarcoma specimens and the cell lines.The data analysis depended on the ABI Prism 7300 system.[Result]By an entrance limit of ≥2.0 or ≤0.5,189 up-regulated and 84 dowm-regulated genes which were considerable differentially expressed in the two osteosarcoma cell lines compared with the osteoblastic cell line hfob1.19 were finally detected.Many of the genes were first reported to be related to the pathogenesis of osteosarcoma.The array results were further confirmed by the real-time RT-PCR.[Conclusion]Many genes are involved in the pathogenesis and metastasis of osteosarcoma.It is helpful to apply gene array to find the laws of gene expression and to study the gene-gene interactional relationships in this tumor.
9.Effects of Integripetal Rhodiola Herb on Ethology and Anti-oxydation Potential of Brain Tissue of Alzheimer’s Disease Model Rats

Objective To study the intervention effect and mechanism of integripetal rhodiola herb (IRH) on Alzheimer’s disease model rats. Methods Alzheimer’s disease model was made by injecting D-galactose (10 mg/mL) in abdominal cavity and taking AlCl3 (20 mg/mL) by intragastric administration, and injecting scopolamine. Morris water maze testing system was undertaken to observe the change of learning and memory ability in rats, biochemical indicator tested was to contrast the state of oxidative stress, common HE staining and immunohistchemistry staining were to observe the morphology change of the hippcampus and cortex cell. The model rats were administrated IRH by means of intragastric administration contrasting with hydergine to compare the effect. Results The memory ability of model group was obviously decreased, cellula nervosa were damaged badly. A? and senile plaques appeared on hippcampus and cortex. All of the test indexes in Morris water maze showed that the abilities of space search, learning and memory in different groups remarkably improved compared with that of the model group (P
10.Percutaneous lumbar discectomy for contained herniation of lumbar disc.

Objective To evaluate the influence of lumbar disc herniation type on the curative effect of percutaneous lumbar discectomy (PLD). Methods 158 patients were classified into two groups including contained and discontained herniation according to clinical symptoms and diagnostic images. They were treated by meanes of the same technique in nonvascular intervention. Their curative effects were compared with each other in this study. Results The curative and total effective rates were 81.1% and 98.5% respectively in contained group, 7.7% and 38.5% in discontained group without serious complications. The duration of follow up ranged from 3 months to 73 months. Conclusions PLD is a effective procedure for lumbar disc herniation, which is relatively simple and safe with rapid recovery and without serious complications. It should be confirmed in simple contained herniation only.
