1.Influence of fenofibrate on plasma BNP level and heart rate variability in aged patients with acute coro-nary syndrome

Objective:To explore influence of fenofibrate on plasma level of brain natriuretic peptide (BNP) and heart rate variability (HRV) in aged patients with acute coronary syndrome (ACS) .Methods :A total of 150 aged ACS patients undergoing treatment in our hospital were selected ,randomly and equally divided into routine treat‐ment group and fenofibrate group .Changes of plasma BNP level ,HRV parameters , blood lipid levels ,and chest pain were compared between two groups before and after treatment .Results:After treatment ,BNP level signifi‐cantly reduced ,and HRV , blood lipid levels significantly improved in both groups , P<0.01 all .Compared with routine treatment group ,plasma BNP level significantly reduced [ (132.5 ± 22.1) pg/ml vs .(91.2 ± 13.7) pg/ml] ( P<0.01);there were significant rise in standard deviation of normal to normal RR intervals calculated over the 24 h period (SDNN) ,adjacent normal RR interval difference > 50ms stroke accounted for a percentage of 24h total RR interval (PNN50) ,root‐mean square of differences between successive normal to normal intervals (rMSSD) and high frequency (HF) ,and significant reduction in low frequency (LF) ,P<0.05 all;significant reductions in levels of TC ,TG and LDL‐C ,and significant rise in HDL‐C level ( P<0.01 all);there were significant reductions in on‐set duration [ (9.53 ± 5.34) min vs .(6.32 ± 2.13) min] and frequency [ (3.87 ± 1.13) times/d vs .(2.99 ± 1.14) times/d] of chest pain after treatment in fenofibrate group , P<0.01 both .Conclusion:Routine therapy combined fenofi‐brate can significantly reduce BNP level ,regulate blood lipid and autonomic nervous function in aged ACS patients .
2.Screening of lentiviral vectors carrying effective siRNA targeting S1P2 gene

AIM: To screen the lentiviral vector carrying siRNA with higher efficiency of suppressing the sphingosine-1-phosphate receptor 2 (S1P2) gene expression in the primarily cultured corpus cavernosum smooth muscle cells of spontaneously hypertensive rats (SHR).METHODS: SHR and SD rats (n=5 each) were used for primarily culturing corpus cavernosum smooth muscle cells.The cells were randomly divided into 6 groups: SHR siRNA-1, SHR siRNA-2, SHR siRNA-3, SHR GFP, SHR control (SHR non-transfection group), and SD control (SD rat control group).Each group had 5 samples with 1.0×105 cells of each sample.At 72 h after transfection (MOI=60) with lentiviral vectors carrying S1P2 siRNA into the SHR corpus cavernosum smooth muscle cells, the expression of GFP was observed under fluorescence microscope.The protein expression of S1P2, ROCK1, ROCK2 and eNOS in the corpus cavernosum smooth muscle cells, and the mRNA expression of S1P2, ROCK1 and ROCK2 were determined by by Western blot and RT-PCR.RESULTS: The transfection efficiency of the corpus cavernosum smooth muscle cells in SHR siRNA-1, SHR siRNA-2, SHR siRNA-3 and SHR GFP groups were>80%.Compared with SHR control group, the mRNA levels and the protein expression of S1P2, ROCK1 and ROCK2 in SHR GFP group showed no remarkable changes, while those in SHR siRNA-1, SHR siRNA-2, SHR siRNA-3 and SD control groups were significantly lower than those in SHR control group (P<0.05).The protein expression of eNOS in SHR siRNA-1, SHR siRNA-2, SHR siRNA-3 and SHR GFP groups were not significantly changed as compared with SHR control group, but that in SD control group was significantly higher than that in SHR control group.CONCLUSION: Three groups of siRNA lentiviral vectors targeting S1P2 inhibit the expression of S1P2 in the corpus cavernosum smooth muscle cells of SHR, and by silencing the S1P2 expression, the expression of ROCK1 and ROCK2 is inhibited.Among them, siRNA-1 has the highest inhibitory efficiency.
3.Granulocyte-Colony Stimulating Factor Mobilizing Autologous Peripheral Blood Stem Cells Transplantation to Form Cardiac Myoid Cells and Angiogenesis

Objective To study autologous peripheral blood stem cells (PBSCs) transplantation for cardiac myoid cells formation and angionesis after recombinant human granulocyte-colony stimulating factor (rhG-CSF) mobilizing. Methods The 60 white Japanese big-ear rabbits were divided into 3 groups, i.e. transplantation (T) mobiliztor (M) and control (C) groups, each group with 20 rabbits. Myocardial infarction (MI) model was developed by ligating the anterior descending coronary artery. G-CSFs were given continually for 7 d in T and M groups since 1 h after MI model development. Cell suspension which derived from the peripheral blood and labeled with BrdU which prepared 1 week ago were injected into infarction regions and borders, while in C groups only the same doses saline was injected. The survival and differentiation of the implanted cells were detected with histological analyses and capillary densities. Results BrdU positive cells which were taken on immature cardiac myoid cells were observed at infarcted areas in T group, which were detected as the Actin positively. HE stains showed that the structures of infarction regions were deranged in C group, but in T and M groups cell arrangements were arranged regularly. Meanwhile, there were a large amount of neogenesis capillaries. The capillaries densities were respectively (58.2 ± 11.5) and (52.3±6.0) per high-power field in T and M groups, while in C group was (21.6±4.9) (P<0.01 ). In C group blue collagen fibers were much more than T and M group under Masson stains (P <0.01). In T and M groups the cardiac functions were much better than in the C group at the end of 4 weeks, especially ejection fraction were respectively (65.34±2.54)% and (63.40±2.84)% in M and T groups. In C group it was only (50.51 ±6.47)% (P<0.01). Conclusion After G-CSF mobilizing the implanted PBSCs may survive and differentiate into cardiac myoid cells in infarcted areas and vicinities, at the same time promote neogenisis and improve cardiac function. It is significant that cell transplantation will treat the cardiac infarction in future.
4.Study on the association between vitamin D receptor gene polymorphism of ApaI and BsmI and vitamin D deficiency rickets

ObjectiveTo study the distribution characters and linkage disequilibrium of vitamin D receptor(VDR) gene ApaI and BsmI polymorphism,and explore the genetic susceptibility of VDR and vitamin D deficiency rickets.MethodsVDR gene ApaI and BsmI polymorphisms were determined by PCRRFLP technology in 56 cases of rickets and 76 cases of normal children.The frequencies of the VDR genotype and allele were compared between the two groups.The software SHEsis was used to make linkage disequilibrium analysis.Results No significant difference was found in either the frequency distribution of VDR gene ApaI and BsmI polymorphism or allele of them between two groups; Two polymorphisms didn't show linkage disequilibrium and D' and r2 were 0.23 and 0.01,respectively.ConclusionsThe ApaI and BsmI polymorphism of VDR gene might be not associated with vitamin D deficiency rickets.Two polymorphisms didn't show linkage disequilibrium.
5.CT Scan Diagnosis in Gastrointestinal Tract Rupture after Blunt Abdominal Trauma

Objective To evaluate the role of CT in diagnosis of the gastrointestinal tract rupture after blunt abdominal trauma . Methods Twenty preoperative CT scans and clini cal data were obtained in 20 patients who subsequently had bowel ruptures verifi ed surgically. CT findings were analyzed retrospectively in these patients. Retr ospective interpretation was made by consensus of at least two radiologists. Results Twenty cases of CT scan showed intraper itoneal fluid (18 cases), pneumoperitoneum (18 cases), extravasations of gastro intestinal tract contents (2 cases), bowel wall findings (14 cases) and mesenter ic injury (15 cases). Conclusion CT is fast, sensitive and noninvasi ve in diagnosis of the gastrointestinal tract rupture after blunt abdominal tra uma.
6.Study of interleukin-18,interleukin-10 and matrix metalloproteinase-9 levels in restenosis after percutaneous coronary intervention

Objective To investigate the levels of IL-18,IL-10 and MMP-9 in coronary heart disease(CAD)patients with in-stent restenosis(ISR)after percutaneous coronary intervention (PCI)treatment,so as to discuss the influence of inflammatory fac-tors to ISR after PCI.Methods CAD patients with ISR after PCI were angiographically re-evaluated and formed the ISR group(n=68)and the non-ISR group(n=173)based on the presence or absence of ISR.109 subjects without angiographic evidence of CAD formed a reference control group(control group).The plasma IL-18,IL-10 and MMP-9 concentrations of subjects were measured. Results The concentrations of serum IL-18 and MMP-9 in ISR group and non-ISR group were significantly higher than control group,while IL-10 level was the opposite.Contrasted with non-ISR group,the concentrations of serum IL-18 and MMP-9 in ISR group were significantly higher,but IL-10 level was the opposite too.There were significantly statistical differences(P <0.05)Con-clusion There is significant correlation between ISR and serum levels of IL-18,IL-10 and MMP-9.The inflammation may have im-portant impact on the process of ISR.

AIM: To apply UASLG for Rana temporatia chensinensis David's identification. METHODS: Rana temporatia chensinensis David was made ultrasonic extraction. These solvents in turn were petroleum ether、chlorofo mn、ethyl acetate、n-butanol and 95%WTBZ# ethanol,then their absorbance in UV and one-order derivative spectrums were determined. RESULTS: Rana temporatia chensinensis David's UASLG was reproducible. CONCLUSION: UASLG's action spectrums for Rana temporatia chensinensis David can be identified for quality control.
9.Percutaneous stent implantation for treatment of inferior vena cave obstruction

Objective: To evaluate the effects of stent implantation for treatment of Budd-Chiari syndrome. Methods:A total of 16 patients with pasthepatic inferior vena cave (IVC) obstruction syndrome were treated by balloon catheters ( Polythene and Inoue ) in percutaneous transluminal angioplasty (PTA) , then stent were inserted into the obstruction. Results: The residual pressure gradient after PTA and stent implantation was decreased from (4.6?0. 3 ) kPa to (2. 1?0. 1) kPa. The diameter of IVC after PTA and stent implantation was increased from (3.8?1. 7)mm to (14. 9?2. 0)mm. Conclusion : PTA and stent implantation are effective treatment for pasthepatic inferior vena cave obstruction.
