1.The 1 H-Magnetic Resonance Spectroscopy Study of Auditory Cortex Metabol ism in Patients with Metabol ic Syndrome

Objective To investigate the characteristic changes of the metabolism products in the auditory cortex (transverse temporal gyrus)in patients with metabolic syndrome combined with sensorineural deafness using 1 H magnetic resonance spectroscopy (1 H-MRS),and to find out the early warning indicators of sensorineural deafness in patients with metabolic syndrome.Methods The pure tone audiometry (PTA)was performed in 142 pa-tients with metabolic syndrome (diagnosed by endocrinology department),and 15 healthy subjects as the control group.The patients were divided into four groups:the metabolic syndrome group;the metabolic syndrome group with unilateral,the bilateral deafness group,and the control group.Cerebral metabolism was studied by assessing the ratios of nitro-acetyl aspartate contrast to choline (NAA/Cho)as well as to creatine (NAA/Cr),myo-inosi-tol to creatine (mI/Cr)and choline to creatine (Cho/Cr)ratios in the auditory cortical separately in these groups. ROC curves were made for those metabolism markers to find the best diagnostic threshold.Results Significantly lower values of NAA/Cho ratio and higher values of Cho/Cr were observed in metabolic syndrome in the control group (P<0.05),Cho/Cr higher than those (P<0.05).NAA/Cho ratios in the injured and uninjured auditory cortex of MS with unilateral deafness were significantly lower than those in the control group (P<0.05).A self-comparison was made between the inj ured and uninj ured auditory cortex,the result showed that NAA/Cho ratio had significant differences.All of the metabolisms were tested by the curve of ROC.The area of NAA/Cho under the ROC curve was 81%,which had a higher accuracy.NAA/Cho equating to 1 .82 could be used as the boundary indi-cators between metabolic syndrome without deafness and with deafness groups.The areas of the other indicators un-der the ROC curve were <50%.Conclusion NAA/Cho may be the early warning marker of sensorineural deafness in patients with metabolic syndrome.
2.Application of Planar Microelectrode Array Modified by Nano-structure Titanium Nitride on Dual Mode Neural Information Recording

The nano-structure TiN was modified on the laboratory self-made planar microelectrode array pMEA by magnetron sputtering method. The performance of modified pMEA was investigated. Research on neuroelectrical and neurochemical recording was studied in vitro. The impedance of the modified pMEA was decreased almost one order of magnitude, and the background noise level was reduced to ±6 μV. In the same testing environment, the signal-to-noise ratio (SNR) of modified electrodes was 1. 7 times of bare electrodes. The SNR of neuroelectrical recording on the brain slice of SD rats reached 10:1 , and the weak signal such as ±12 μV was separated easily. For neuroelectrical recordings, the detection limit of dopamine ( DA) solution reached 50 nmol/L with the 2:1 (S/N). During the concentration range of 0. 05-100 μmol/L, the linearly correlation coefficient of the DA oxidation currents was 0 . 998 . The modification of nano-structure TiN on pMEA reduced pMEA impedance and background noise level, meanwhile the SNR was increased. The weak signals of neuroelectrical and neurochemical recording were successfully recorded.
3.Influence on vestibular function caused by acute low-tone sensorineural hearing loss.

OBJECTIVE:
To study the vestibular function of patients with acute low-tone sensorineural hearing loss (ALHL). So we can know whether their vestibular function was influence.
METHOD:
Vestibular function tests were performed on fifty-two ALHL patients by recording electronystagmography (ENG) and static posturography (SPG) before and after their therapy. Then made a comparison with normal.
RESULT:
Length of locus and velocity of postural sway of the ALHL subjects were significantly increased in comparison with normal (P < 0.05) when eyes closed. After therapy, two (3.8%) ALHL subjects were still recorded abnormal ENG results.
CONCLUSION
Most ALHL subjects without vertigo symptom, but their vestibular function was abnormal, so vestibular function tests should be performed on patients with ALHL.

4.Investigation of SLC26A4 mutations associated with inner ear malformations.

OBJECTIVE:
To study the molecular pathogenesis of SLC26A4 mutations associated with inner ear malformations including large vestibular aqueduct syndrome (LVAS), Mondini dysplasia and inner ear malformations but not accompanied with LVAS.
METHOD:
DNA sample and clinical material were obtained from 14 sporadic LVAS probands, six Mondini dysplasia probands and seven inner ear malformations excluding IVAS probands. SLC26A4 gene mutation was analyzed by direct sequencing for its 20 coding exons. GJB2 gene and also mt12SrRNA were analyzed by direct sequencing.
RESULT:
In 14 cases of LVAS, two mutations were detected in 12 patients (85.7%, either homozygous or compound heterozygous mutations), and one mutation was found in two patients (14.3%). In six cases of Mondini dysplasia, two mutations were detected in all of patients (100%). No mutation could be found in the seven cases of other inner ear abnormalities not accompanied with LVAS. No pathogenic mutation was detected in all of these 27 probands in GJB2 gene and mt12SrRNA 1555/1494T.
CONCLUSION
We have shown that LVAS and Mondini dysplasia closely correlate with SLC26A4 gene. No mutation was detected in seven probands of inner ear malformations not accompanied with LVAS. We should study the molecular pathogenesis of this disease in depth.

5.A case of liver cirrhosis due to hypopituitarism in adolescence after nasopharyngeal carcinoma radiotherapy

Nonalcoholic fatty liver disease is a metabolic stress liver injury, including simple hepatic steatosis, nonalcoholic steatohepatitis (NASH), cirrhosis, and hepatocellular carcinoma (HCC). Liver fibrosis in NASH patients progressed by an average of 7 to 10 years, and the incidence of cirrhosis in NAH patients was only 0.6%to 3.0%after 10 to 20 years of follow-up. This case reports a 21-year-old man with pituitary dysfunction and cirrhosis, whose condition develops rapidly. The mechanism may be that growth hormone deficiency affects liver signaling transduction pathways to make the liver metabolism disordered, causing nonalcoholic fatty liver disease. In this report, the pathogenesis, diagnosis and treatment of this case of cirrhosis duo to hypopituitarism in adolescence are reviewed retrospectively to improve the understanding of the diagnosis and therapy of this disease.
6.Effect of topical corticosteroids on expression of PKC, Bcl-2 and Bax in nasal polyps.

OBJECTIVE:
To study the effect of intranasal glucosteroid on the expression of PKC, Bcl-2 and Bax in nasal polyps.
METHOD:
The expression of PKC, Bcl-2 protein and Bax were detected with immunohistochemistry in nasal polyps from the patients (n=16) pre and post treated for 4 weeks with intranasal glucosteroid. And which compared with normal mucous membrane of inferior turbinate concha.
RESULT:
The PKC and Bcl-2 protein expressed significantly higher in the pretreated patients (P < 0.01), which were significant reduced in the post treated patients compared with pretreated ones. The expression of Bax protein significantly higher in the post treated patients (P < 0.05).
CONCLUSION
Our findings indicated that abnormal expression of apoptosis related genes in nasal polyps tissues might play an important role in the occurrence and progression of nasal polyps. The treatment of intranasal glucosteroid may regulate the expression of apoptosis related genes.

7.Evaluation on the application effects of quality control circle on improv-ing the accuracy of pupil observation for nurses in the department of neurosurgery

Objective To explore the application effects of quality control circle (QCC) on improving accuracy of pupil observation for nurses in the department of neurosurgery. Methods Quality control circle was established. The theme of the activity was determined according to urgency and circle capacity, and activity proposal was decided. Investigation on current situation was carried out, and objective was set according to the focus on improvement and circle capacity. Reasons were analyzed, and feasible strategies were proposed and implemented. Accuracy of pupil observation before and after the QCC activity was compared, and the effects after the implementation of QCC was confirmed. Results As for tangible results, the scores of examination on specialized theoretical knowledge for nursing practitioners was (83.11±4.55) before the QCC activity, and the scores were (90.59±3.57) after the activity, the specialized theoretical knowl-edge for nursing practitioners significantly improved, and the difference was statistically significant (P<0.05). Accuracy of pupil observation was 47.50% before the QCC activity, and the accuracy was 90.83% after the activity, the accuracy of pupil observation after the activity significantly improved compared to that before the activity, and the difference was statistically significant (P<0.05). As for intangible results, self scores from the circle members in 8 aspects including a-bility of solving problems, responsibility, ability of coordination and communication, team coherence, confidence, enthu-siasm, ability of applying quality control circle and happiness were all higher than those before the activity, and the differences were statistically significant (P<0.05). Conclusion The activity of QCC is able to improve the level of spe-cialized nursing knowledge for nurses in the department of neurosurgery, enhance the ability of scientific research, un-derstanding of QCC, application ability and execution for nursing practitioners, and improve the accuracy of pupil ob-servation.
8.Significance of expression and promoter methylation of LITAF gene in B-cell lymphoma.

OBJECTIVETo investigate promoter methylation status of LITAF gene in B-cell lymphoma and to explore transcription regulation of 5-Aza-2'-deoxycytidine (5-Aza-CdR) on LITAF gene.
METHODSOne hundred and five paraffin specimens including 54 cases of diffuse large B cell lymphoma (DLBCL), 15 small lymphocytic lymphoma (SLL), 8 mucosa-associated lymphoid tissue lymphoma (MALT) and 6 follicular lymphoma (FL) were included. Five reactive lymphoid hyperplasia samples were collected as control. Methylation status of CpG island in LITAF gene in the specimens and in Raji, Pfeiffer and Daudi cell lines were detected by methylation-specific PCR (MSP). LITAF expression in Raji, Pfeiffer and Daudi cell lines with or without 5-Aza-CdR treatment was detected by Western blot and immunohistochemistry. The inhibitory ratio in the three cell lines was measured by MTT assay.
RESULTSThe frequency of LITAF gene methylation in B-cell lymphoma was 89.5% (94/105) . Among them, 3.8% (4/105) showed complete hypermethylation. In control group, however, there was no methylation in CpG island of LITAF gene promoter. The expression of LITAF was recovered or increased along with the cell growth inhibition when the cells exposed to demethylating reagent.
CONCLUSIONSLITAF gene silencing with aberrant CpG methylation is probably one of the critical events to the oncogenesis of B-cell lymphoma, which may have important implications as a candidate marker for diagnosis and target gene therapy.

9.Screening and identification of long non-coding RNAs in metastatic laryngeal squamous cell carcinoma

[Abstract] Objective: To search valuable candidate molecular markers for LSCC by screening and identifying differentially expressed long non-coding RNAs (lncRNAs) in metastatic laryngeal squamous cell carcinoma (LSCC) with high throughput gene microarray technique. Methods: Four pairs of LSCC tissues and corresponding para-cancer tissues that pathologically confirmed with lymph node metastasis were collected from Bio-sample lab of Otorhinolaryngology Department, the Second Hospital of Hebei Medical University. After total RNA extraction, the SBC-lncRNA (human 4x180k) chip assay was then applied to detect the differentially expressed lncRNAs and mRNAs, and Fold-change and Student's t-test methods were used to identify differentially expressed genes; the Fold Change (linear)≤0.5 or≥2.0, P<0.05 was used to identify the differentially expressed genes. The reliability of the chip results was confirmed by quantitative reverse transcription polymerase chain reaction (qRT-PCR). Results: The gene expression profiles of metastatic LSCC tissues and corresponding para-cancer tissues were significantly different. There were 3 073 differentially expressed lncRNAs, including 1 967 up-regulated and 1 106 down-regulated lncRNAs in cancer tissues. There were 2 809 differentially expressed mRNAs, including 1 791 up-regulated and 1 018 down-regulated mRNAs in cancer tissues. The differentially expressed lncRNAs were mainly involved in cell proliferation and apoptosis, immune response biological process, and were associated with cytokine and cytokine receptor interaction, chemokine signaling pathway, cell cycle regulation, P53 signaling pathway, etc. In addition, 10 significantly differentially expressed lncRNAs were chosen and validated by qRT-PCR in 25 cases of LSCC tissues, and the results were in agree with microarray detection. Conclusions: There were obvious differences in lncRNAs expression between metastasis LSCC and corresponding para-cancer tissues; in-depth analysis of these differences may has important significance on clarifying the mechanisms of invasion and metastasis of LSCC, which can provide the theoretical basis for biomarker screening and effective targeted therapy for LSCC.·
