Carvedilol is a non-selective beta-blocker with additional blockade of alpha_(1)-adrenoceptors.It has been applied in the treatment of hypertention、stable angina pectoris、chronic heart failure.In addition,Carvedilol exerts antioxidant and anti-apoptosis effects,it can inhibits vascular smooth muscle cell proliferation、migration and neointimal formation following vascular injury.The article makes a review on the molecular cytologic mechanism of Carvedilol in these aspects.
2.Adenovirus - mediated Herpes Simplex Virus Thymidine Kinase Gene Transference and Combined Drug Therapy Leads to Opoptosis of Human Epithelial Ovarian Cancer Cells

Objective To evaluate the treatment and apoptosis effect of adenovirus-mediated herpes simplex virus thymidine kinase (HSV 1-tk) gene transference followed by administration of ganciclovir (GCV) and acyclovir (ACV) on ovarian epithelial cancer cells. Methods Recombinant adenovirus was amplificated and purified by routine method. The expression of HSV 1-tk gene was assayed by polymerase chain reaction (PCR). The efficiency of recombinant Advtk transference was evaluated. The cytotoxicity efficacy of TYK cells that carry HSV 1-tk gene was evaluated followed by GCV、ACV administration after the transference of Advtk. The changes and apoptosis of TYK cells that carry HSV 1-tk gene were observed by means of analysis of DNA fragmentation and electronic microscopy. Results Adenovirus was amplificated and purified in large amount. PCR assay showed 404 bp special band. When the multiplicities of infection (MOI) was 100, the transduction rate was 98 9%. The inhibition rates of TYK cells that carry HSV 1-tk gene increased with the increase of MOI when the same concentration of GCV、 ACV were given. When the MOI was same, the inhibition rates were also increased with the increase concentration of GCV、ACV. Apoptosis of TYK cells that carry HSV 1-tk gene after administration of GCV was observed by means of analysis of DNA fragmentation and electronic microscopy. Conclusions The HSV 1-tk gene can effectively transfered into TYK cells by recombinant replicated-deficient adenovirus vector, GCV、ACV can effectively kill TYK cells that carry HSV 1-tk gene in vitro. Apoptosis may be the mechanism of the killing effect.
4.Effects of arsenic trioxide on different types of human ovarian cancer cell lines in vitro

Purpose:To study the effects of arsenic trioxide on the different types of human ovarian cancer cells growth in vitro.Methods:Methyl thiazolyl tetrazolium (MTT)was used to observe the growth inhibition rates of human ovarian cancer cell lines 3AO,SKOV 3 and TYK cells by various concentration arsenic trioxide at different times; Cell apoptosis percentage and cell cycles phase distribution of 3AO were measured by flow cytometry(FCM) assays; Apoptotic phenotype of SKOV 3 was observed by acridine dying. Results:Arsenic trioxide could inhibit the growth of 3AO?SKOV 3 and TYK effectively, depended on the action time and concentration of the medicine(P0.05). As a non-pungency inhaled agent with less respiratory irritation,sevoflurane has been widely used in various surgeries for induction and maintenance of general anesthesia.However,the incidence of pediatric emergence delirium has been significantly increased with sevoflurane general anesthesia which severely disturbs the surgical effect and increases the requirement of medical staff in post anesthesia care unit.This article systematically reviews the research advance in emergence delirium in children with sevoflurane general anesthesia and introduces the reasonable intervention for the improvement of anesthesia quality.
8.Study on regulatory effects of interleukin-6 on placental growth factor expression in trophoblast cells

Objective To investigate the expression pattern of Placental growth factor (PlGF) and regulatory effects of interleukin-6 (IL-6)on PlGF expression in the primary cultured trophoblast cells. Methods Cytotrophoblast cells werecollected by trypsin-collagenase digestion and Percoll gradient centrifugation for primary culture. Reverse transcriptase polymerase chain reaction(RT-PCR) were resorted to demonstrate the expression pattern of PlGF mRNA in trophoblast cells cultured in vitro. The effects of IL-6 with different concentration(100,10,1 and 0.1 ?g/L) and duration (6,12,24 and 48 h) on PlGF expression were observed. Results The 185 bp and 248 bp bands of PlGF were showed by RT-PCR. PlGF expression correlated with IL-6. PlGF began to increase at 6 h, and reached the climax at 12 h when recultured with 100 ?g/L IL-6. Conclusions PlGF expression has time and dose dependance on IL-6. It may play an important role in early pregnancy.
9.Repeated CT scan in improving the reproducibility of grass tumor volume for moving target

Objective To find a method to improve the range accuracy of moving target such as peripheral lung tumors, since a single CT snapshot may not be accurate during the treatment process.Methods A simple harmonic motion phantom, embedded with a cube and a circular ball, was used to simulate the tumor motion. Individualized moving targets were scanned 24 times with different amplitudes and frequencies. Then the images were fused from every 1, 2 or 3 sets of CT scans. The GTV volume variation of circular target and the length variation of the cube target along the z axis were contoured and analyzed. Results As motion amplitude increased, the maximum of both circular target volume and cube target length was increased, while the minimum of the factors was decreased. Motion frequency affected the target volume less than amplitude. For a cube target with the length of 3.3 cm at stationary phase, when motion frequencies was 20 and motion amplitude was 2 cm, the maximal length was 2. 4 times of the minimal length (5. 1 cm vs. 2. 1 cm). When it came to the cube target groups fused from every 1,2 and 3 sets of CT scans, the average length and standard deviation were (3.77 ± 1.20) cm, (4.18 ±0. 91)cm and (4.52 ±0. 59) cm, respectively. With the increase of fused scan number, targets became bigger, the standard deviation decreased, and the change of center positions was decreased. Conclusions The motion amplitude, frequency and the number of CT scans are the main factors affecting target definition, though, the optimized scanning phase is not certained. When 4DCT and respiration gating technique are not available,the efficient and practical method to solve this problem is to scan the target three or more times and fuse them in planning system, which will generate a larger, more reproducible GTV volume for moving targets.
10.Outcome and reproductive function after conservative surgery for borderline ovarian tumors

Purpose:To study reproductive function and disease outcome in women with borderline ovarian tumors (BOT) who were treated with primary conservative surgery. Methods:From January 1990 to May 2000, the clinical data eligible according to criterions in our hospital , which had been statistically managed by SAS software package, were retrospectively reviewed and questionnaires were accepted in all cases with BOT. Results:41 of the 43 patients with a median follow up time of 63 months, finished our questionnaires. Of the 43 selected patients, 26(60.5%) were classified as serous borderline ovarian tumors (SBOT), 17(39.5%) mucous borderline ovarian tumors(MBOT) . 29 patients were free after primary conservative surgery and 14 cases with recurrence including one patient who died of tumor recurrence and the other died of intercurrent disease .The median recurrence time after conservative surgery was 39.3 months and the recurrence was more frequent in patients treated with ovarian cystectomy than in those treated with oophorectomy alone (58% compared with 22%, Fisher exact probability =0.0351). The death rate due to conservative surgery was not higher than that seen in hysterectomy and bilateral adnexectomy (Fisher exact probability=0.64)?Moreover, twelve of the 24 patients attempting pregnancy(50%) had conceived of which the median age was 25 years, with a range of 23-34 years. Conclusions: Conservative surgery remains a therapeutic option in selected patients with BOT. Although the rate of recurrence is relatively high especially in those treated with ovarian cystectomy, mortality from cancer remains about the same in patients with conservative surgery or radical operations. Many patients who desire pregnancy are able to conceive and deliver healthy offspring.
