Objective To analyze the clinical feature of X-linked chronic granulomatous disease (X-CGD) and gene mutation of CYBB. Method The clinical data of X-CGD in one child and the results of CYBB gene detection in his family were reviewed. Results This boy had onset in the neonatal period and presented with recurrent severe pulmonary infection as his main manifestation. Results of nitroblue tetrazolium test (NBT) in both non-stimulation group and LPS stimulation group in the child were 0, and neutrophil oxidation index (NOI) was 1.15. Gene analysis showed a deletion mutation in exon 6 of CYBB gene in the child (579-582delATTA), which resulted in frameshift mutation started from coding sequence of 189—isoleucine (I) and stop codon occured in advance in the 212th amino acid (I189fsX212). Both the child's mother and grandmother were carriers of the mutated gene. The same deletion mutation was not found in the CYBB gene in the amniocyte from the mother's next child. Conclusion One case of X-CGD patient with CYBB gene mutation and his families were diagnosed by gene detection. Prenatal diagnosis can avoid the birth of children with X-CGD.

ObjectiveTo detect the mechanism of the growth inhibition and apoptosis of human acute leukemia cell line U937 cells induced by honokiol.MethedsThe proliferation inhibition was detected by MTT method.Cell apoptosis was tested by Hoechst 33342 staining and flow cytometry with Annexin V/PI staining.RT-PCR was used to detect the mRNA expression of the apoptosis gene Bcl-2,Bax,Caspase 3,Caspase 8 and Caspase 9.ResultsThe inhibition effect of honokiol(5 μg/ml,48 h) on U937 cells proliferation could he observed,and the inhibition rate of 10 μg/ml honokiol on cell proliferation reached above 50％ (48 h).U937 cells proliferation could be completely inhibited for 120 h. U937 cells apoptosis rate reached 26.8％ (P ＜0.01)after being treated with 10 μg/ml honokiol.After being treated with 10 μg/ml honokiol for 48 h,the Bcl-2 gene expression in U937 cells was reduced (control group:0.33 ± 0.02,experimental group:0.14 ±0.01,P ＜ 0.01 ),and the Bax gene expression was elevated ( control group:0.1 ± 0.01,experimental group:0.87 ± 0.08,P ＜ 0.01 ).The gene expressions of Caspase 3 ( control group:0.48 ± 0.01,experimental group:0.87±0.06,P ＜0.01),Caspase 8(control group:0.23±0.02,experimental group:0.41 ±0.07,P ＜ 0.01 ) and Caspase 9 ( control group:0.44 ± 0.05,experimental group:0.76 ± 0.06,P ＜ 0.01 ) were all increased.The activity of Caspase-3 was 0.325 ±0.089,which was significantly higher than that of the control group ( P ＜0.01 ).ConclusionHonokiol can significantly inhibit the proliferation and induce cell apoptosis of human acute leukemia cell line U937 cells.The mechanism is related to the up-regulation of Bax and down-regulation of Bcl-2,and the endogenous and exogenous pathways are both inolved in the apoptosis process.

Objective: To isolate and purify the polysaccharide from Cordyceps sinensis, and analyze its structure. Methods:Cordyceps sinensis was cultured by a liquid fermentation method. A water-extraction and alcohol-precipitation method was applied to ex-tract polysaccharide from Cordyceps sinensis fermentation liquor (EPS) and polysaccharide from Cordyceps sinensis mycelium (IPS). Sephadex gel chromatography was applied to isolate and purify the polysaccharide. The purity and relative molecular weight of the poly-saccharide were determined by a gel filtration method. The monosaccharide composition of the polysaccharide was identified by GC. The content of uronic acid was analyzed by sulfuric acid carbazole method. Results: The analysis results showed that the molecular weight of EPS was 78kDa. The content of polysaccharide and uronic acid was 94. 8% and 6. 0%, respectively. EPS was composed of mannose,glucose and galactose with the molar ratio of 4. 5∶8. 0∶1. 0. The molecular weight of IPS was 42kDa. The content of polysac-charide and uronic acid was 92. 5% and 4. 5%, respectively. IPS was composed of mannose,glucose and galactose with the molar ratio of 2. 8∶3. 0∶1. 0. Conclusion:Both polysaccharide EPS and IPS in Cordyceps sinensis are heteropolysaccharide.

Objective To discuss the psychological behavior intervention methods on reducing the violence behavior of schizophrenia patients. Methods 120 schizophrenia patients were admitted from January to December, 2007 and randomly assigned into the behavior intervention group (group A), the psy-chological support group (group B), the health education group (group C) and the control group (group D) with 30 cases in each group. Group A received routine nursing plus behavior intervention, group B was giv-en routine nursing plus psychological support, group C adopted routine nursing plus health education, group D only received routine nursing. The intervention effect was evaluated with MOAS and the relapse rate of violence behavior went through statistics. Results After intervention, the scores of MOAS decreased sig-nificantly in group A, B, C and D compared with those before intervention. The reduction of MOAS scores in group A, B and C was significantly higher than that of group D, with the highest reduction score in group A and group B and C followed subsequently. The relapse rates of the violence behavior in group A, B and C were significantly lower than those of group D. Conclusions Routine nursing, health education, psycho-logical support and behavior intervention can all reduce and decrease the violence behavior and the inci-dence of the violence behavior, among which behavior intervention proves to be the most effective, and psy-chological support and health education go subsequently.

OBJECTIVE: To analyze the impact of extraction and nonextraction in soft tissue profile repair. METHODS: Databases of CNKI, Wanfang, VIP Chinese Periodical Database, CBM, Medline (ovid) and Science direct had been searched for studies of soft tissue profile after orthodontic treatment with and without extractions published in recent decade, with orthodontics, corrective, tooth extraction, soft tissue profile as key words. Hand searching was also done to obtain any further information about the studies. Meta analysis was used to comprehensively quantitatively analyze the upper (lower) lip with esthetic plane and soft tissue plane before and after extraction and nonextraction treatment. According to homogeneity test, fixed effect model and random effect model were adopted to calculate weighted mean difference (WMD) and 95% confidence interval (95% Cl).Revman 5.0 was used for data management. RESULTS: Five studies with 356 patients were included. Homogeneity test showed that lower lip-esthetic line and lower lip-soft tissue line Ⅰ~2 < 50%, without heterogeneity, so fixed effect model was adopted. Upper lip-esthetic line and upper lip-soft tissue line were more than 50%, indicating heterogeneity, so random effect model was adopted. Result of meta-analysis showed that, the WMD value and 95% Cl of lower lip-esthetic line, lower lip-soft tissue line, and upper lip-soft tissue line has statistically significance. CONCLUSION: The extraction treatment significantly decreased the distance of lower lip-esthetic line, lower lip-soft tissue line, and upper lip-soft tissue line.

ObjectiveTo observe the effects of salmeterol fluticasone on the glucose metabolism and bone density of the elderly patients with chronic obstructive pulmonary disease (COPD) combined with type 2 diabetes mellitus (T2DM).MethodsThirty-one patients with COPD combined with T2DM were divided into 2 groups by random digits table,14 cases in control group were given conventional therapy,17 cases in experimental group were given conventional therapy and fluticasone propionate 50/500 μg,twice a day,for 3 months.The fasting blood glucose,postprandial 2 hours glucose,glycosylated hemoglobin,plasma cortisol and bone density respectively before and after treatment were detected.ResultsIn experimental group,the fasting blood glucose,postprandial 2 hours glucose,glycosylated hemoglobin and bone density before treatment was (5.25 ± 0.21 ) mmol/L,(7.14 ± 0.33 ) mmol/L,(5.58 ± 0.26 )％,( 1.96 ± 0.11 ) g/cm2,and after treatment was(5.31 ± 0.27 ) mmol/L,(7.22 ± 0.29 ) mmol/L,(5.67 ± 0.23 )％,(2.03 ± 0.15 ) g/cm2,there was no significant difference (P ＞ 0.05).In control groups,the fasting blood glucose,postprandial 2 hours glucose,glycosylated hemoglobin and bone density before treatment was (5.33 ± 0.35) mmol/L,( 7.26 ± 0.29 ) mmol/L,( 5.62 ± 0.19 )％,( 1.88 ± 0.20 ) g/cm2,and after treatment was ( 5.36 ± 0.31 ) mmol/L,(7.30 ± 0.35 ) mmol/L,( 5.69 ± 0.26 )％,( 1.98 ± 0.17 ) g/cm2,there was no significant difference (P ＞ 0.05 ).There was no significant difference in plasma cortisol before and after treatment in two groups (P ＞0.05).ConclusionInhaling salmeterol fluticasone for elderly patients with COPD combined with T2DM is safe.

Objective:To observe the effects of Astragalus polysaccharides on antigen presentation of dendritic cells(DC).Methods:DC and CIK cells were generated by culturing PBMC of healthy blood donor.The typical DC and DC pulsed by Astragalus polysaccharides were co-cultured with CIK cells,respectively.Then the changes in the phenotypes of the DCs pulsed with Astragalus polysaccharides were determined.Cell surface markers were analysed by FACS method.IFN-? and IL-12 secreted by co-cultured cells were detected by ELISA.The cytotoxicity of effector cells on A549 cells in vitro were measured by MTT assays.Results:Astragalus polysaccharides could increase the expression of CD40,CD80 and HLA-DR on DC surface.The Astragalus polysaccharides pulsed DC-CIK cells resulted in an enhanced killing activity to A549 cells than that of unpulsd DC-CIK cells(P

Objective To summarize the key points in surgical cooperation of laparoscopic left lateral segment liver resection. Methods The clinical data of 24 patients undergoing laparoscopic left lateral segment liver resection from March to September of 2013 were analyzed retrospectively to summarize the key points in surgical cooperation of preoperative preparation and intraoperative coordination.Result All the operations were successful with the operation mean time of(72.0±26.8)min,no intraoperative transfer or termination of the operations.Conclusion Sufficient preoperative preparation together skilled surgical nursing cooperation in laparoscopic left lateral segment liver resection is critical for the success of operation.

Objective To explore the transport properties of the radionuclide strontium 90Sr in the soil-plant system,and discuss the accumulation characteristics of strontiumes in different vegetable tissues.Methods Three kinds of vegetables,including Chinese cabbage,radish and celery,were planted in this experiments using stable nuclide 88Sr instead of 90Sr.Strontium concentrations in these kinds of plant and soil were measured with inductively coupled plasma atomic emission spectrometry (ICP-AES),and the transportation factor (TF) of strontium and the concentration ratio (CR) of these plants were calculated.Results The contents of strontium in different tissues of the vegetables increased significantly in response to the strontium concentrations in soil (F =3.75-139.39,P ＜ 0.05).The contents of strontium at 398.33,477.99 and 557.57 mg/kg in different tissues of these vegetables were significantly higher than that in control group (t =-10.49-7.41,P ＜0.05).Except celery stems,there was a positive correlation about contents of the strontium between other tissues of these vegetables and their experimental soils (r =0.88-0.99,t =7.41-15.96,P ＜0.05).Based on dry weight of examples,the TF in each tissue sample of cabbage and radish was higher than 1,and the concentration ratio,for cabbage and radish,was higher in stems than in roots (F =8.22,91.49,P ＜ 0.05).Conclusions Three kinds of vegetables have been shown to have strontium accumulation ability in soil environment.
