3.Effects of hemoperfusion treatment on serum IL-23 and IL-17 levels in children with Henoch-Schönlein purpure.

OBJECTIVETo study the effects of hemoperfusion treatment on serum interleukin-17 (IL-17) and IL-23 levels in children with Henoch-Schönlein purpura (HSP).
METHODSEighty-seven children who were diagnosed with HSP and who had received hemoperfusion treatment between January 2011 and December 2012 were enrolled. Twenty-seven sex- and age-matched healthy children were recruited as normal controls. Enzyme-linked immunosorbent assay (ELISA) was used to measure serum concentrations of IL-17 and IL-23.
RESULTSThe serum IL-23 and IL-17 levels in the HSP group were significantly higher than in the control group (P<0.05). After hemoperfusion treatment, the serum IL-23 and IL-17 levels in the HSP group were significantly reduced to the levels of the control group. Serum serum IL-17 level was positively correlated with serum IL-23 level (P<0.05) in children with HSP.
CONCLUSIONSHemoperfusion treatment can reduce serum IL-23 and IL-17 levels in children with HSP, suggesting that the treatment may be effective for HSP.

4.The expression of interleukin-25 in blood and nasal tissue of patients with chronic rhinosinusitis with nasal polyps.

OBJECTIVE:
To explore the expression of interleukin-25 (IL-25) in chronic rhinosinusitis with nasal polyps (CRSwNP) and its potential significance in pathogenesis.
METHOD:
IL-25 expression in blood was detected by enzyme-linked immunosorbent assay (ELISA). IL-25 expression in tissue was detected by immunohistochemistry (LSAB method) from polyps (68 CRSwNP patients) and 55 inferior turbinate mucosa from patients with deviation of nasal septum served as control. Complete blood count and HE staining of blood and tissue eosinophil infiltration degree.
RESULT:
IL-25 expression in CRSwNP group were significantly higher than the control group, the difference was statistically significant (P < 0.01). IL-25 expression in local organizations was positively correlated with the number of eosinophil infiltration in CRSwNP group (r = 0.679, P < 0.01).
CONCLUSION
The expression of IL-25 in CRSwNP patients mutually reinforcing and might increase eosinophil infiltration and play an important role in the development of CRSwNP.

5.Expression of regulatory T cells and Th17 cells in idiopathic thrombocytopenic purpura and its significance.

OBJECTIVETo investigate the expression of regulatory T cells and Th17 cells in idiopathic thrombocytopenic purpura (ITP) and its significance.
METHODSThe quantity of CD4(+)CD25(+)T cells, CD4(+)CD25(high) T cells and CD4(+) IL-17(+)T cells in peripheral blood were measured with flow cytometry (FCM), the plasma level of IL-10 and IL-17 with ELISA and the expression of Foxp3 mRNA and RORc mRNA with RT-PCR in 29 newly diagnosed ITP patients and 28 healthy controls.
RESULTSThe ratio of CD4(+)CD25(+)T cells/CD4(+) T cells in the peripheral blood of ITP patients was significantly higher than that of the normal controls (P < 0.05). And the ratio of CD4(+)CD25(high) T/CD4(+) T cells in the patients was remarkably lower (P < 0.05). There was no difference in the percentage of CD4(+) IL-17(+)T/CD4(+) T cells between ITP patients and controls (P > 0.05). The ratio of Treg/Th17 was lower in ITP patients (P < 0.05). The plasma level of IL-10 in ITP patients was significantly decreased (P < 0.05), whereas no statistical difference was observed in the level of IL-17 (P > 0.05). The Foxp3 mRNA expression levels were largely reduced (P < 0.05), but the RORc mRNA expression was increased compared with that in controls (P < 0.05).
CONCLUSIONThe imbalance of Treg/Th17 plays an important role in the pathogenesis of ITP.

6.Detection of Th17/treg cell-associated cytokines in peripheral blood of patients with graft-versus-host disease and its clinical significance.

To investigate the peripheral levels and clinical significance of Th17/Treg cell-associated cytokines in patients with acute graft versus host disease (aGVHD) or chronic GVHD (cGVHD), blood samples were collected from 39 hematopoietic stem-cell transplantation patients and 20 healthy donors. The patients included 10 patients with aGVHD, 13 patients with cGVHD and 16 patients without evidence of GVHD. Th17/Treg cell-associated cytokines such as IFNγ, IL-4, IL-6, IL-10, TGF-β(1), IL-17 and IL-23 were detected by ELISA. The results showed that the plasma levels of IFN-γ, IL-4, IL-6, IL-17 and IL-23 significantly increased in patients with aGVHD or cGVHD, compared with the patients without clinical signs of GVHD and the healthy donors (p < 0.05), while IL-10 and TGF-β(1) were obviously lower than that of them (p < 0.05). After aGVHD and cGVHD patients were treated effectively, the plasma levels of IL-6, IL-17 and IL-23 were significantly decreased, and IL-10, TGF-β(1) were significantly increased, while the levels of IFN-γ and IL-4 did not markedly change. The TGF-β(1) level were negatively correlated with IL-6 (r = -0.36, p < 0.05), IL-17 (r = -0.51, p < 0.05) and IL-23 (r = -0.44, p < 0.05) respectively, while there were positive correlations between IL-6 and IL-17 (r = 0.62, p < 0.05), IL-6 and IL-23 (r = 0.71, p < 0.05), IL-17 and IL-23 (r = 0.93, p < 0.05). It is concluded that Th17/Treg cell-associated cytokines may play an important role in the development of a/cGVHD, which helps to find novel targets for developing new strategies of GVHD treatment.

7.Effects of Cytokines on Early Death in Patients with Newly Diagnosed Acute Promyelocytic Leukemia.

OBJECTIVE:
To explore the effect of cytokine levels on early death and coagulation function of patients with newly diagnosed acute promyelocytic leukemia (APL).
METHODS:
Routine examination was performed on 69 newly diagnosed APL patients at admission. Meanwhile, 4 ml fasting venous blood was extracted from the patients. And then the supernatant was taken after centrifugation. The concentrations of cytokines, lactate dehydrogenase (LDH) and ferritin were detected by using the corresponding kits.
RESULTS:
It was confirmed that cerebral hemorrhage was a major cause of early death in APL patients. Elevated LDH, decreased platelets (PLT) count and prolonged prothrombin time (PT) were high risk factors for early death (P <0.05). The increases of IL-5, IL-6, IL-10, IL-12p70 and IL-17A were closely related to the early death of newly diagnosed APL patients, and the increases of IL-5 and IL-17A also induced coagulation disorder in APL patients by prolonging PT (P <0.05). In newly diagnosed APL patients, ferritin and LDH showed a positive effect on the expression of IL-5, IL-10 and IL-17A, especially ferritin had a highly positive correlation with IL-5 (r =0.867) and IL-17A (r =0.841). Moreover, there was a certain correlation between these five high-risk cytokines, among which IL-5 and IL-17A (r =0.827), IL-6 and IL-10 (r =0.823) were highly positively correlated.
CONCLUSION
Elevated cytokine levels in newly diagnosed APL patients increase the risk of early bleeding and death. In addition to the interaction between cytokines themselves, ferritin and LDH positively affect the expression of cytokines, thus affecting the prognosis of APL patients.

8.Function of peripheral blood Th17 cells in patients with multiple myeloma.

This study was aimed to investigate the effects of peripheral blood Th17 cells and their secreting IL-17, IL-21 in the occurrence and development of multiple myeloma (MM). A total of 55 patients with MM were divided into non-remission group (group A , n = 30), remission group (group B, n = 25); healthy volunteers were used as control group (group C , n = 30). The concentration of IL-17, IL-21 and IL-6 in the peripheral blood mononuclear cell (PBMNC) culture supernatant were determined with ELISA. The ratio of Th7 cells in PBMNC was determined by flow cytometry. The results showed that IL-17, IL-21 levels and ratio of Th17 cells in group A were much higher than those in group B and C (P < 0.05), IL-17, IL-21 and the ratio of Th17 cells between group B and group C were not significantly different (P > 0.05); IL-17 level in non-remission MM group positively correlated with IL-6 level (r = 0.782, P < 0.05), IL-21 level in non-remission MM group positively correlated with IL-6 level (r = 0.778, P < 0.05). It is concluded that Th17 cells as a initiating factor may be involved in the immune pathogenesis of MM patients, promoting the progress of the disease.

9.Expression level and significance of IL-17 and IL-23 in serum and nasal secretion of patients with allergic rhinitis and non-allergrie rhinitis.

OBJECTIVE:
To investigate the effect of IL-17 and IL-23 in the pathogenesis of allergic rhinitis(AR) and non. allergic rhinitis(NAR).
METHOD:
Selected 156 cases of patients with allergic rhinitis (AR group) and 59 cases of patients with non-allergic rhinitis (NAR group), 60 cases of healthy people (control group). All cases in AR group and NAR groups were evaluated by a visual analog scale (VAS) score of nasal symptoms. Collected peripheral blood and nasal secretions in all cases and then detected IL-17 and IL-23 expression levels.
RESULT:
There was no significant difference in VAS score of AR group and NAR group (P>0. 05). IL-17 and IL-23 levels of serum and nasal secretions in AR group and NAR group were both higher than control group, with a highly significant difference (P <0. 05). The research showed a clear correlation between expression of IL-17 and IL-23 both in serum and nasal secretions of AR group and NAR(P<0. 05).
CONCLUSION
IL-17 and IL-23 may be important cytokines and IL-23/IL-17 pathway may play a significant role in the pathogenesis of allergic rhinitis and non-allergic rhinitis.

10.Role of TGF-beta, IL-10 and IL-17 in allergic rhinitis patients with specific immunotherapy.

OBJECTIVE:
To detect the level of TGF-beta, IL-10 and IL-17 in the allergic rhinitis(AR) patients pre- and post-specific immunotherapy (SIT).
METHOD:
The level of TGF-beta, IL-10 and IL-17 in serum of 48 AR patients were detected by enzyme-linked immunosorbent assay before SIT. and the nasal symptom score (NSS) was evaluated in the first and second year after SIT. At the same time. 35 healthy persons were made as normal.
RESULT:
(1) The level of TGF-beta and IL-10 in serum of AR patients were lower than that of normal group (P < 0.05). The level of TGF-beta and IL-10 in AR patients taken SIT for two years was lower than that of patients with one year SIT (P < 0.05), but it was higher than that of patients pre-immunotherapy (P < 0.05). However, The level of TGF-beta and IL-10 in the group that taken SIT for two years was still lower than that of normal group (P < 0.05). (2) The level of IL-17 in serum of AR patients pre- immunotherapy was higher than that of normal group (P < 0.05). The level of IL-17 continuously declined after taking SIT, However it was higher than that of normal group two years later (P < 0.05). (3) In AR patients pre-immunotherapy and post- immunotherapy for 2 years, the level of IL-10 and TGF-beta showed negative correlation with NSS respectively, the level of IL-17 was positive related to NSS respectively (P < 0.05). But the level of IL-10, TGF-beta and IL-17 showed no correlation with NSS respectively (P > 0.05) when SIT was taken for one year.
CONCLUSION
IL-10 and TGF-beta were fluctuated in AR patients taking SIT, which raised first and then dropped. but the level of IL-10 and TGF-beta was higher than that of pre- immunotherapy. However, the level of IL-17 continuously declined in the whole process. It showed that the level change of IL-10, TGF-beta and IL-17 correlate with patients' symptom only two years after SIT,so the level change of IL-10, TGF-beta and IL-17 can be used as an index for evaluating the treatment effectiveness of SIT at that time.
