1.Research Advances in Baculovirus Occlusion-derived Virions.

Baculoviruses are a family of arthropod-specific viruses that produce two morphologically distinct types of virions (budded and occlusion-derived) in their lifecycle. Baculoviruses establish infection in the midgut of their host via the oral route: occlusion-derived virions have pivotal roles in these processes. This review summarizes the basic characteristics of baculoviruses, and discusses the composition and classification of baculovirus occlusion-derived virions. The latter focuses mainly on the evolution and role of multiple occlusion-derived virions in the lifecycle of baculoviruses. These achievements should aid understanding the evolution and infection mechanisms of baculoviruses.

2.The community succession of sarcosaphagous insects on pig carcasses in summer indoor and outdoor environment in Shenzhen area.

OBJECTIVE:
To explore the growing development and community succession of main sarcosaphagous insects on pig carcasses in summer indoor and outdoor environment in Shenzhen area and to estimate the postmortem interval (PMI).
METHODS:
From early May to August in 2013, in Forensic Medical Examination Center of Shenzhen Public Security Bureau, the main insect species and the decomposition process were observed in two adult pig carcasses of simulative indoor and outdoor environment. The different decomposition stages and the community succession of insects were recorded.
RESULTS:
The indoor and outdoor pig carcasses showed skeleton 412.5 and 325 hours after death, respectively. The main species of flies on pig carcasses were Chrysomya megacephala, Chrysomya rufifacies and Chrysomya chani. The main species of beetles were Crecphilus maxillosus, Necrobia ruficollis, Saprinus splendens and Dermestes maculatu. The dominant species of flies in the outdoor pig carcasses obviously produced the second generations due to the effect of mass rainfall, nor in the indoor pig carcasses.
CONCLUSION
There are regular patterns on the community succession of insects on pig carcasses in summer indoor and outdoor environment in Shenzhen area. The activity patterns of seven typical insects and their larva show important value for estimating PMI.

3.Breeding system of Paris polyphylla var. yunnanensis.

OBJECTIVETo provide the basic guidance for seed breeding and cross-breeding of Paris polyphylla var. yunnanensis.
METHODThe floral behavior and pollinators were observed; 0.5% TTC solution was used for the pollen viability test and benzidine and -H2O2 was used for estimation of the stigma receptivity. The mating systems were tested by out crossing index (OC1), and pollination experiment was carried out by bagged and emasculated test in the field.
RESULTCommonly, stigma lobes spread slightly, and anthers started presenting the pollen from the outer ring while the flower was just beginning to open. Consequently, the distance between the stigma and its own pollen was relatively far, this "floral behavior" may be conducive to outcrossing. Then the flower entered the later period, while the stigma lobes spread widely, anthers all split, and this "floral behavior" shortened the stigma and its own pollen's distance, which may be conducive to selfing. P. polyphylla was partly protogynous. Stigma life-span was about 10-12 d. After anther dehiscence, the pollen viability maintained about 10% within 2 days, and 20% within 10 days. The value of out crossing index (OC1) was 4. By pollination experiment and pollinators observed, P. polyphylla was self-compatible, but no capacity for autonomous self-fertilization; In natural circumstances, outcrossing fructification rate was low, and mainly anemophilous. Assisted selfing-fertilization fructification rate was higher, spider was the main pollinators.
CONCLUSIONP. polyphylla has a mixed mating system with self-pollination and cross-pollination characteristics. Floral behavior has important adaptive significance in avoiding female and male interference, outcrossing, and delayed selfing. P. polyphylla is ambophily (a combination of both wind and insect pollination), pollinators changes due to environment. Pollen limitation is the main cause of low fructification rate under natural conditions.

4.Genetically engineered corn rootworm resistance: potential for reduction of human health effects from pesticides.

OBJECTIVE AND METHODSInsecticide use, grower preferences regarding genetically engineered (GE) corn resistant to corn rootworm (CRW), and the health effects of using various CRW insecticides (organophosphates, pyrethroids, fipronil and carbamates) are reviewed for current and future farm practices.
RESULTSPest damage to corn has been reduced only one-third by insecticide applications. Health costs from insecticide use appear significant, but costs attributable to CRW control are not quantifiable from available data. Methods reducing health-related costs of insecticide-based CRW control should be evaluated. As a first step, organophosphate insecticide use has been reduced as they have high acute toxicity and risk of long-term neurological consequences. A second step is to use agents which more specifically target the CRW.
CONCLUSIONWhereas current insecticides may be poisonous to many species of insects, birds, mammals and humans, a protein derived from Bacillus thurigiensis and produced in plants via genetic modification can target the specific insect of CRW (Coleoptra), sparing other insect and non-insect species from injury.

5.Study of antibacterial effect of Galla Chinensis on the bacteria biofilm in chemostat.

OBJECTIVETo compare the antibacterial effect of Galla Chinensis with that of chlorhexidine by means of chemostat and provide experimental foundation for caries prevention with Chinese medicine instead of chemicals in future.
METHODA multispecies consortium biofilm (BF) model was construted in the chemostat. The growth-inhibition on bacterial biofilm coated on HA was investigated by means of the colony-forming units (CFU). Acid-inhibition was inspected by continual pH-recording in the flow cells. And the profile on the biofilm treated by both experimental medicines was observed with the scanning electromicroscopy (SEM).
RESULTBoth of the experimental medicines could inhibit the growth of the BF bacteria. And Galla Chinensis had no obvious influence on the ecological composing of the BF bacteria. Both of the experimental medicines could inhibit the acid producion of experimental bacteria. But the final pH of Galla Chinensis was lower than that of chlorhexidine. A typical biofilm formed on HA was observed by SEM. Both of Galla chinensis and chlorhexidine could inhibit the production of extracellular matrix and make the BF profile on the surface of the HA clearer and simpler.
CONCLUSIONGalla Chinensis had definite antibacterial effect. No obvious difference is found between Galla Chinensis (4 g x L(-1)) and chlorhexidine (0.5 g x L(-1)). Therefore, it is possible for Chinese medicine to gradually replace the chemical medicine in clinic and it will give a new choice for caries prevention.

6.Enhancing hGH expression level in insect cells by shortening the 5'-UTR of hGH cDNA.

The regulation of foreign gene expression in Insect-Baculovirus Expression System is very complex. In this report, the effect of 5'-UTR in the expression of hGH gene in cultured Sf9 cells was examined. A 18 bp length in the end of 5'-UTR of hGH (human Growth Hormone, hGH) cDNA including a stem-loop structure was deleted by PCR. The truncated hGH cDNA, delta 1hGH was cloned in pFastBac1, named pFast-Bac-delta 1hGH. After transforming into E. coli. DH10Bac, which have a shuttle vetor-Bacmid, the delta 1hGH was integrated into Bacmid by site-specific transposition, and an expression vector, rBacmid-delta 1hGH DNA was acquired. By transfecting the cultured Sf9 cells with the recombinant expression vector DNA, pure recombinant virus, rAcV-Bac-delta 1hGH was obtained, and hGH gene was expressed. Immuno-blot and Chemiluminescent assay revealed that the expressed hGH had normal immunological activity, the amount of hGH expression level in Sf9 cell supernatant infected with rAcV-Bac-delta 1hGH containing the truncated 5'UTR was four to five times higher than that infected with rAcV-Bac-hGH.
