2.Cryopreservation of Day 3 Mouse Embryos by Vitrification.

The use of hormonal stimulation in human in vitro fertilization and embryo transfer (IVF-ET) leads to increased production of embryos for ET. So to avoid high pregnancies and to allow conception in future, unstimulated cycles, cryopreservation of spare embryos is desirable. One of the improvement of cryopreservation methods is vitrification. We cryopreserved mouse day 3 embryos by vitrification using the three different vitrification solution (EFS40, VSll and VS3a). EFS40 solution is consisted of 40% (v/v) ethylene glycol, Ficol170 30% (w/v) and 0.5M sucrose and VSll is 6.0M ethylene glycol and 1.8M glycerol. And VS3a is 6.5M glycerol and 6% (w/v) BSA (bovine serum albumin). First we tested the toxicity of three vitrification solution by exposure to these solution during 3 min. After washing by thawing solution, the survival rates of each groups are 95.5%, 90.9% and 84.4% (EFS40, VS11 and VS3a). High percentages of them developed to expanded blastocyst and hatching embryos in culture 48hrs 94.2%, 97.7%, 100% and 97.4% (no treatment group, EFS40, VS11 and VS3a). So there is no significant differences among the each group. Second, after thawing of vitirfied embryos, the survival rates of each groups are 96.8% (slow freeze), 94.1% (EFS40), 85.5% (VS11) and 80.0% (VS3a, P vs. no freeze or EFS40 is 0.01). Vitrified embryos exhibited a high rate of development in vitro after 48hrs culture. The percentages of each group to blastocyst and hatching embryos are 88.7% (no freeze), 91.8% (slow freeze), 93.4% (EFS40), 87.7% (VS11) and 73.0% (VS3a, P vs. other group is 0.01). The results suggest that there is no significant differences in exposure of various vitrification solution and day 3 mouse embryos can be vitrified in solution EFS40 and VS11 by simple procedure.

3.Cryogenic Treatment of Herpes Simplex Keratits.

The results of 16 cases of herpes simplex keratitis treated by cryogenic method are presented. The cryoapplicators(2 mm and 4 mm in diameter of tip) were applied on the lesions 5-7 seconds and antibiotics and atropine solutions were instilled. No complication by cryogenic treatment was happened. The new vessels in the cornea were faded after cryocautery for the treatment of herpes simplex keratitis.

4.The study of cardiovascular changes by intravascular injection of contrast media

This investigation was aimed to study the effect of contrast media on the cardiovascular system. So in thisstudy, pithed rats were used whether alteration in cardiovascular system by contrast media were controlledcentrally. Furthermore, several hypertonic solutions were also used to clarify the effect of contrast media. Theresults are as follows: 1. Intravenous injection of contrast media in rats(2.5m/kg) caused hypotension andbradycardia. The effects were neither blocked by pretreatment of atropine nor pyribenzamine+atropine. 2. NaCl4.7%, dextrose 24.8%, urea 9.0% and glycerol 10.1%(v/v) which were equiosmolar with contrast media, causedhypotension, but did not affect the heart rate. 3. In pithed rats, intravenous injection of Angiografin increasedblood pressure in a dose-dependant manner, and caused decrease in heart rate compared with those of control rats. 4. In pithed rats, bradycardia by intravascular injection with Angiografin was partialy blocked by atropine. 5.Metrizamide of which iodine content was adjusted to 280 mg/ml caused increased in blood pressure when was injectedintravenously in pithed rats with little effect on heart rate. 6. When perfused with contrast media in rathindlimb at 15ml/kg speed both perfusion pressure and flow effluent incereased, simultaneously. These resultssuggest that hypotension might be caused by the central effect due to hyperosmolarity of contrast media andbrachycardia caused by both parasympathetic stimulation and direct inhibitory action on the cardiac conductivesystem.

5.Hemangiopercytoma of the Meninges: The immunohistochemical study for the relationship between hemangiopericytic meningioma and peripheral hemangiopericytoma.

Hemangiopericycic maningioma is clinically and pathologically similar to peripheral Hemangiopericytoma and now tends to be terned as hemangiopericytoma of central nervous system. The authors studied 3 cases of hemangiopericytic meningioma obtained from 3 patients, 1 case of meningotheliomatous meningioma, angioblastic meningioma and transitional meningioma, and 2 cases of peripheral hemangiopericytoma, which had operated from November 1988 to May 1989 at the department of neurosurgery, Pusan Inje University Hospital. The authors analysed and compared the immunohistochemical finding and light microscopic apearance. The results obtained were summarized as follows; 1) Classic maningioma (meningotheliomatous meningioma, transitional meningioma and angioblastic meningioma) shows reactivity to both EMA and vimentin. 2) Hemangiopercicytic meningioma and peripheral hemangiopericytoma are reactive only to vimentin, so the two tumors are suggested as same type of tumor. 3) One of 3 cases of hemangiopericytic meningioma shows whorling and interlacing bundles of spindle cells, the peculiar light microscopic features of transitional meningioma, suggesting transitional or mixed form of hemangiopericytic meningioma and transitional meningioma.

7.Effects of Thera-Band(R) Resistance Training Including Isometric Exercise in Total Knee Replacement Patients.

BACKGROUND: The purpose of this study was to assess the effects of an exercise program using isometric and isotonic thera band exercises. METHODS: Data were collected from 30 total knee replacement patients hospitalized in the orthopedics department at 'B' hospital (control=15, experimental=15). The control group performed isometric exercises for one week while the experimental group performed three or more rounds of both isometric and isotonic exercises, including Thera-Band(R) exercises, everyday for four weeks. RESULTS: The experimental group improved significantly in muscular strength (Z=-1.99, P=0.047) and self-efficacy (Z=19.48, P=0.000), and showed lower levels of depression than the control group (Z=8.60, P=0.003). However, no difference was found in pain experience between the two groups. CONCLUSIONS: Exercise programs using the isometric and Thera-Band(R) resistance training improve muscular strength and self-efficacy and relieve depression in total knee replacement patients.

8.Detection of Human Papillomavirus DNA 16/18 in Cervical Adenocarcinomas by Polymerase Chain Reaction.

Twenty-five paraffin-embedded tumor tissues were analyzed for detection of HPV 16 and 18 in cervical adenocarcinoma by polymerase chain reaction with type specific primers and by non-radioactive Southern blot hybridization for confirmation . The suitability of paraffin-embedded tissue as PCR material was confirmed by successful amplification of 100% of cervical specimens with human -globin specific primer. Eighty four percent of the cervical adenocarcinoma tissues were positive for HPV 16 and/or 18. HPV 16 positive rate was 68%, HPV 18 was 60%. The double infection with HPV 16 and 18 was found in 44%. Three cases of the negative specimen in PCR for each type of HPV DNA 16 and 18 were positive in Southern blot hybridization. The total positive rate was 92% for HPV 16 and/or HPV 18, HPV 16 positive rate was 80%. HPV 18 was 72%. The double infection with HPV 16 and 18 was 60%. These results suggest that the pattern of HPV types 16 and 18 is closely associated with carcinogenesis of cervical cancers. HPV type 18 appears to be preferentially related to cervical adenocarcinoma and the poor prognosis of these patients. Therefore, determination of HPV DNA type in cervical carcinoma patients is important in treatment and prognosis.

9.Acute Myocarditis Complicated by Permanent Complete AV Block Associated with Salmonella Group B Gastroenteritis.

Acute infectious myocarditis in children can be caused by many pathogens, including bacteria, viruses, fungi and protozoa. Patients may be asymptomatic or may have 'flu-like symptoms' to life-theratening arrhythmias or sudden death. We herein report a case of acute myocarditis complicated by complets and permanent AV block, associated with Salmonella group B gastroenteritis in a previously healthy 5-year-old boy. Presenting symptoms were dyspnea, cyanosis, vomiting, and diarrhea. Electrocardiography on admission showed very slow ventricular escape rhythm at 37 beats/minute. He was treated with antibiotics, inotropic agents, ventilatory support, and transvenous temporary pacemaker. Ten days later, permanent dual chamber pacemaker was implanted. All three consecutive stool cultures done after admission grew Salmonella group B. His ventricular function recovered rapidly and completely and he became fully active 2 months after the onset. However, his conduction system was totally destroyed and at last follw up evaluation 2 years after the onset, he is still totally dependent on the pacemaker without any escape rhythm.

10.A Study on Nurses' Perception for Nutrition Education Working in Kyungnam area.

This study was performed to investigate nurses' perception for nutrition education. The subjects of this study were 197 nurses and 94 nursing assistants working at hospitals in the Kyung-nam area. The survey was conducted by using a self-administered questionnaire in November, 2001. The results were as follows : Sixty-seven point seven percent of the subjects were nurse, 32.3% were nursing assistant. Average age of subjects was 26.9 years old, average nursing experience was 5.7 years, and 70.7% of the subjects graduated from a junior college. The average nutrition knowledge score was 14.3+/-2.5 out of possible 20 points. Seventy-four point two percent of the subject responded that nutrition education is very necessary for patient, and positive responses in the nurse were higher than that nursing assistant(p<0.001). Only 8.6% of the subjects had nutrition education training. The perceptions about a suitable person for nutrition counseling and nutrition education indicated dietitian(69.3%) and nurse(21.3%). Fifty-two point eight percent of the subjects responded that they would not participate in nutrition education themselves and the main reason for this was that they believed lack of expert knowledge(43.4%), and that such courses should be taught by specialist(40.5%). Forty-eight point eight percent of subjects had nutrition counseling experience for patient, and nurses, married nurses and those over 2 years of nursing careers had significantly higher nutrition counseling experience for patient than nursing assistant, unmarried nurses and those under 2 years of nursing careers(p<0.05~p<0.001)
