1.Detection of Chromosome Aberration in Interphase Nuclei of Tumor Cells by Nonradioactive In Situ Hybridization Using Chromosome-specific Probes.

An improved protocol for in situ hybridization(ISH) to routinely processed, paraffin-embedded tissue sections from lung carcinoma is presented. For this study, DNA probes for alpha-satellite chromosome 7 and 17 were used. The protocol to detect numerical chromosome aberrations involved treatment of sections with 1 M sodium thiocyanate prior to pepsin digestion, resulting in reproducible ISH reactions. The effect of avidin-biotin detection system. Four layer avidin methods and triple biotin methods, using avidin-PO, goat antiavidin, biotinylated antigoat IgG, avidin-PO or anti-biotin, biotinylated antirabbit IgG, avidin-PO, markedly enhanced the intensity of positive signals. More than 80% of the tumor and stromal cells showed distinct chromosome hybridization signals in 6 micrometer-thick sections. Lung carcinoma cells showed multiple chromosome signals(2~5 spots), contrasted by one or two signals in the stromal cells in the same section. These results suggest that chromosome polysomy can be reliably detected in tissue sections using in situ hybridization. This capability will prove to be an important tool for determining the underlying genetic basis for tumor development, tissue phenotype heterogeneity and progression by allowing genetic determination to be made on paraffin-embedded tissue sections where tumor histologic architecture is preserved.

4.Morphologic Change of Rat Liver Induced by Repeated Administration of Carbon Tetrachloride and Dimethylnitrosamine.

Carbon tetrachloride and dimethylnitrosamine, both potent hepatotoxic agents, affect the hepatic lobules with fatty changes and central necrosis, and hemorrhagic necrosis. To study the effects on morphologic changes of the hepatic lobules in cases of single and repeated treatment of both hepatotoxins, sublethal doses of carbon tetrachloride, 0.4ml/kg, and dimethyl nitrosamine, 40 mg/kg of rats were given introperitoneally single, twice and triple. With interval of 3 days, and the results were as follows: 1. The fatty changes and central necrosis of the hepatic lobules were milder and more quickly disappeared in the rats with twice or triple treatment than single administration of carbon tetrachloride, and regenerative changes of hepatic and sinusoidal cells achieved fater in the rats with repeated administration of carbon tetrachloride than those with single treatment. 2. The hemorrhagic necrosis of the hepatic lobules was not significantly influenced by the times of DMN treatment, but the hyperplastic changes showed more active to animals, with multiple administration of DMN.

7.Perception on Nutrition Labeling of the Processed Food among Elementary School Teachers in Busan .

This study was carried out to investigate the perception on nutrition labeling of the processed foods among elementary school teachers in Busan. The survey was conducted from November 1 to November 30, 2008 by questionnaires and data which were analyzed by SPSS program. The results are summarized as follows. Ninety five point seven percent of the male teachers and 98.2% of the female teachers recognized the nutrition labeling (NL). The necessity of a NL system and comprehension of NL were higher in the female group than the male group. NL was trusted by 22.9% of male teachers and 19.7% of female teachers. The main reason for checking NL of the teachers was for food safety. For the most important NL items at purchasing the food, the teachers replied 'trans fatty acid' and 'cholesterol'. Five point seven percent of male teachers and 17.0% of female teachers were educated for NL. Seventy two point nine percent of males and 91.0% of females recognized the necessity of NL teaching. It is necessary to prepare education programs for both teachers and students about accurate recognition of NL.

9.Study on Fatigue, Stress and Burnout of Pregnant Nurses.

PURPOSE: This study is a correlative study to assess the relationships of fatigue, stress, and burnout among the pregnant nurses in hospital. METHODS: The subjects in this study were 103 nurses aged between 22 and 40 who were confirmed as pregnant by the OBGY (Obstetrics and Gynecology) hospital and working at medical facilities located in J-province of G city during the period of July 15 through September 15, 2010. RESULTS: The level of fatigue of pregnant nurses showed an average of 5.60 out of 10 points, and the level of stress showed an average of 25.91. In terms of burnout, the average was 2.72 out of 5 points. The results of the correlative study to assess relationships among fatigue, stress and burnout showed significant positive correlation between burnout and fatigue (r=.47, p<.001) and stress (r=.53, p<.001) and between stress and fatigue (r=.58, p<.001). Therefore, the results show that increased level of fatigue and stress leads to increased level of burnout. CONCLUSION: The results of the study showed that higher level of fatigue and stress led to higher level of burnout. Therefore, to minimize the burnout of pregnant nurses, many efforts are required in the hospital policy and for the strategies to reduce burnout.
