"It is not clear yet how the laminin is made in cytoplasm and is secreted by cells, though the significance of the interaction between laminin and cells during the development, cell differentiation, tumor growth, and metastasis is well known. Furthermore, it has been reported that aberrant expression. and secretion of laminin in malignant tumor cells, In this study I found a major cytoplasmic laminin binding protein (LBP), which was revealed as the heat shock cognate protein 70 (hsc70), in human colon carcinoma cells by cell surface labeling with '""I, laminin and gelatin affinity chromatography, peptide microsequencing, and immunoblot experiments. This suggests another function of hsc70 as the cytoplasmic LBP, which may play an important role in biosynthesis, assembly, and secretion of laminin in tumor cells."

6.The Basic Studies on the Application of Galectin - 3 as a Target Protein for Delivering Anticancer Agents.

Gastric cancer cells express large amounts of galectin-3 on the cell surface. This fact may provide the possibility to use galectin-3 protein as a surface target for delivering cytotoxic anticancer agents. To investigate the possibility of application of galectin-3 protein as a target protein in delivering cytotoxic anticancer agents, we synthesized doxorubicin immunoconjugate by using maleimidocaproic acid and conjugated doxorubicin immunoconjugate to anti-galectin-3 mAb. The anticancer effect of immunotoxin was assayed on NIH3T3, AGS and KATO III cell lines. The anticancer effect of immunotoxin on AGS cell line is highest and that of KATO III is higher than that of NIH3T3. This results relate to that of flow cytometry analysis previously shown and indicate that galectin-3 protein can be used as a target protein on the surface of gastric cancer cell for delivering cytotoxic anticancer agents.

7.Diagnostic Significance of Serum IGF-I Level in Growth Hormone Deficiency.

PURPOSE:Insuline-like growth factor I(IGF-I) is polypeptide mitogen and mediate growth effect of growth hormone(GH). It's serum level is regulated by GH. The aim of this study is to evaluate whether -2 standard deviation of IGF-I level in normal short stature after insuline and L-dopa stimulation test has any diagnostic significance in GH deficiency. METHOD:We included 64 children with GH deficiency(complete GH deiciency 18 cases, partial GH deficiency 46 cases). Their height was below 10 percentile of korean children's standard growth chart. Control group was 175 children whose test results were normal after insuline and L-dopa stimulation test. Serum growth hormone level was measured by IRMA(immunoradiometric assay) with "Daiichi" kit(Japan) and serum IGF-I level was measured with 125I RIA kit (U.S.A). RESULTS: 1)Serum IGF-I level in normal stimulation test result group was increased with the age and the level was higher in female than that of male. 2)Using the cut-off value of -2SD of IGF-I level in control group, sensitivity was 17.2%, specificity was 98.86%, positive predictive value was 84.62%, negative predictive value was 76.55%, and test accuracy was 76.99%. Sensitivity and test accuracy was 44.44% and 93.26% in th complete GH deficiency, respectively. 3)Serum IGF-I level was significantly correlated with peak GH level with insuline stimulation test in control and GH deficiency group(Y=0.018889X+11.32 r= 0.23930 P=0.0014, Y=0.008592X+4.189 r=0.28141 P=0.0267). But serum IGF-I level was was not correlated with peak GH level with L-dopa stimulation test(Y= 0.005609X+13.88 r=0.06625 P=0.3823, Y=0.008293X+2.98 r=0.20895 P=0.1031). CONCLUSION: Serum IGF-I level in GH deficiency group was lower than that of control group and had wide variation of normal range. Based upon above results IGF-I level has limited clinical value in the diagnosis of GH deficiency.

9.The keratolytic effect of several enzymes and irritants.

BACKGROUND: Several keratolytic agents have been used in many dirmatologic conditions such as callus, keratosis palmaris of plantaris, however the evaluation method of the effects of keratolytiic agents has not been good enough in clinical use. OBJECT: We have conducted an investigation to measure the effect of several keratolytic agents using an in vitro model. METHOD: We measured the fegraded protein of keratin by the bui iet method after adding enzymes such as trypsin, pepsin and papain, and irritants, salicylic acid and sodii m lauryl sulfate to the collected callus. RESULT: The order of the keratolytic effect of the enzymes was trypsir >pepsin>papain. It was difficult to detect the keratolytic effect of salicylic acid becaues of color hiidrance and there was an increasing tendency of keratolyti effect of sodium lauryl sulfate, however it was prominent mainly at a high concentration. CONCLUSION: These results suggested that the potency of similar types of keratolytic enzymes such as pepsin, trypsin and papain can be possible. However it was not such a sitable model to check the potency of the keratolytic effect of salicylic acid and the eoncentration tevel may be an important factor for certain kinds of chemicals such as sodium lauryl sulfate.
