1.Changes of hydroxyprolin in the slake lime burn treated by maduxin

Maduxin ointment is totally extracts from the leaves of the Maduca pasquieri and has been applied in burn treatment in Vietnam since 1987. We studied the effects of Maduxin on the collagenization at burn wounds by following up the changes of OH-Pro content at slaked lime burn wounds - a popular type of burn in Vietnam in comparison with the effects of SSD in burn treatment. Our study was carried out in 21 slaked lime burn patients who were treated at the Vietnam Burn Institute from May to September 1997. The results indicated that OH-Pro concentration increases related to collagen synthesis during the burn wound healing. In the slaked lime burn wounds treated by Maduxin OH-Pro level in burn tissue increases more than in the wounds treated by Silvesufadiazin during the healing.

2.Advances on microbial synthesis of L-proline and trans-4-hydroxy-L-proline.

L-proline (L-Pro) is the only imino acid among the 20 amino acids that constitute biological proteins, and its main hydroxylated product is trans-4-hydroxy-L-proline (T-4-Hyp). Both of them have unique biological activities and play important roles in biomedicine, food and beauty industry. With the in-depth exploration of the functions of L-Pro and T-4-Hyp, the demand for them is gradually increasing. Traditional methods of biological extraction and chemical synthesis are unable to meet the demand of "green, environmental protection and high efficiency". In recent years, synthetic biology has developed rapidly. Through the intensive analysis of the synthetic pathways of L-Pro and T-4-Hyp, microbial cell factories were constructed for large-scale production, which opened a new chapter for the green and efficient production of L-Pro and T-4-Hyp. This paper reviews the application and production methods of L-Pro and T-4-Hyp, the metabolic pathways for microbial synthesis of L-Pro and T-4-Hyp, and the engineering strategies and advances on microbial production of L-Pro and T-4-Hyp, aiming to provide a theoretical basis for the "green bio-manufacturing" of L-Pro and T-4-Hyp and promote their industrial production.

3.Effects of Intake of Alcohol on the Concentration of Urinary 4-hydroxyproline.

BACKGROUND: We studied whether alcohol intake influences in urinary 4-Hyp concentration and whether 4-Hyp concentration is change by time in healthy office workers for 2 weeks. METHODS: Three healthy male workers, who has a same job, recruited for this study, aged from 29 to 33 years, who had been any diagnosed or treated to the alcohol or bone related diseases. Urine samples were picked about 30 ml in screw capped vials for 2 weeks, and analysing method were used to previous published method for determination of urinary 4-Hyp with dansyl chloride. RESULTS: From the ANOVA-test results about objects' Hyp concentration that classify by urine picking time, When subjects did not ingest alcohol, urinary 4-Hyp concentration was difference according to subjects for-work and post-work. But, did not show difference when subjects ingested alcohol. While, T-test result that do alcohol intake existence and nonexistence and urine picking time by variable, urinary 4-Hyp concentration did not show difference for-work and post-work, but there was difference in post-work that is next day that ingest alcohol. When did not ingest alcohol, mean concentration of urinary 4-Hyp increased go by post-work (3.80+/-6.47 nmol/mg creatinine) in pre-work (1.46+/-1.61 nmol/mg creatinine). But when ingested alcohol, mean concentration of urinary 4-Hyp decreased by the next day morning (3.32+/-6.36 nmol/mg creatinine), order of for-work (1.54+/-1.59 nmol/mg creatinine) in alcohol intake future generations post-work (5.09+/-6.41 nmol/mg creatinine). CONCLUSION: We confirmed that alcohol is influencing in urinary 4-Hyp concentration of three healthy male workers, who has a same job, for 2 weeks, and mean concentration of 4-Hyp increasing go by the post-work in the pre-work, could confirm that influence in the next day morning if ingest alcohol in the evening.

4.Experimental Studies on the Vitreous Substitutes.

Experimental replacements of rabbit vitreous by air, normal saline, dextran and Haemaccel were tried in albino rabbits. After aspiration of 0.7ml of liquid vitreous from the central portion of vitreous, 0.5ml of substitutes were injected. Clinically, these substitutes were well tolerated with slight uveal inflammation which subsided within one week. Intraocular pussure returned to normal value after 5 days. Hexosamine content of the whole vitreous samples showed moderate increase after Haemaccel injection which decreased slowly thereafter, whereas no significant changes were observed with other substitutes. Hydroxyproline content of the whole vitreous also showed marked increase after injection of Haemaccel, owing to the high hydroxyproline content of HaemacceJ. After 6 weeks 22.9% of the injected hydroxyproline remained in the vitreous. With other substitutes no change was observed.

5.The effects of ventilation with high density oxygen on the strength of gastrointestinal anastomosis.

PURPOSE: The aim of our study is to evaluate the effects of administration of perioperative supplemental oxygen on anastomoses. METHODS: Forty male Wistar albino rats were used in the study and randomized into 4 groups. Ischemia-reperfusion models were built in groups 3 and 4. Jejunojejunostomy was performed in all rats and assigned to an oxygen/nitrous oxide mixture with a fraction of inspired oxygen of 30% in groups 1 and 3 and 80% in groups 2 and 4. The measurements of perianastomotic tissue oxygen pressure, bursting pressure, level of hydroxyproline were evaluated and compared in all groups. RESULTS: The perianastomotic tissue oxygen pressures, bursting pressures and levels of hydroxyproline were identified as significantly high in groups 2 and 4, administered a fraction of inspired oxygen of 80%, compared to groups 1 and 3, administered a fraction of inspired oxygen of 30%. CONCLUSION: Perioperative supplemental oxygen contributes positively to the anastomotic healing.

6.An Analysis on the Priority of Selection of Supplier for School Food Service Materials.

The purpose of this study is to examine how nutritionists, principals and parents evaluated the importance and priorities of different selection factors for food materials suppliers in an attempt to suggest how to ensure the best supplier choice, since the selection of the best supplier was mandatory for the successful TQM of school food services. This study especially aims to develop priority alternatives based on relative rather than absolute assignments on selection of suppliers for school food service materials. To apply AHP, the selection factors of a food materials suppliers were grouped into five categories, which included quality, service, reliability, hygiene and price. And the five categories involved 28 elements. As the result, quality was considered most crucial on the whole, followed by service, reliability, hygiene and price; and food labeling in the category of quality evaluated as that of first priority in the totality evaluation elements.

7.Study on hydroxyproline tolerance of different cultivars of Chrysanthemum morifolium in vitro.

OBJECTIVETo investigate hydroxyproline tolerance of Chrysanthemum morifolium plantlets included " Boju", "Huaiju", "Chuju", "Gongju" and "Hangju",and provide references basis for excellent cultivar and breeding of Ch. morifolium.
METHODPlantlets in vitro from five kinds of Ch. morifolium were inoculated on medium added with different concentrations of hydroxyproline. Free proline in leaves from plantlets was determined, then the damage index and survival rate were compared.
RESULT AND CONCLUSIONThe results showed that hydroxyproline tolerance of " Boju" and "Huaiju" were superior, the survival rates and free proline of them were higher, but the damage index was inferior. The hydroxyproline tolerance of "Hangju" was the worst, and the survival rate was minimum. The survival rate of "Chuju" and "Gongju" was between "Boju" and " Hangju", and the hydroxyproline tolerance of them was also medium.

8.Coordination mechanism and antioxidant activity of L-hydroxyproline zinc(II).

The coordination compound of L-hydroxyproline (Hyp)-Zn (II) was synthesized with Hyp and zinc sulfate as raw materials in water medium, coordination Synthesizing Mechanism and Antioxidant Activity of Hyp-Zn(II) coordination compound has been researched. Compared with Hyp, the infrared spectrogram of Hyp-Zn (II) coordination compound emerge a new absorption peak at 1100 cm(-1). Conclusion could be obtained that there exists a coordination effect between Hyp and ZnSO4; TG and DSC curve of Hyp and Hyp-Zn(II) coordination compound were analysed. Compared with Hyp, the peak of Hyp-Zn(II) disappear at 290 degrees C and 375 degrees C. This phenomenon confirmed the front conclusion; At the NMR graph of Hyp-Zn(II) coordination compound, the disappearance of the alpha-carboxyl-hydrogen and alpha-hydroxyl-hydrogen's peak at 3.5-3.9 ppm could indicate that combination's position of Hyp is alpha-carboxyl and alpha-hydroxyl; Structure of Hyp-Zn(II) coordination compound were exosyndrome by the Atomic Force microscopy. It is showed that centr-atom Zn(II) was surrounded by several Hyp at Hyp-Zn(II) coordination compound's phase diagram; The proportion of Hyp-Zn(H) coordination compound was determined by dialysis experiment, the proportion is 4:1; Above-mentioned determination and exosyndrome indicated that the molecular formula of Hyp-Zn (II) coordination compound is Zn(Hyp)4.H2O. The results indicated that the Hyp-Zn(II) coordination compound can inhibit hydroxyl free radicals of Zn(II), and the Percentage of Inhibition is 75.5%; the total antioxidant activities of Hyp-Zn(II) coordination compound is 80.167 u/mL, the anti-superoxide activities of Hyp-Zn (II) coordination compound is 53.19 u/mL.

9.Passive Skeletal Muscle Excursion after Tendon Rupture Correlates with Increased Collagen Content in Muscle.

PURPOSE: This study was designed to measure time-dependent changes in muscle excursion and collagen content after tenotomy, and to analyze the correlation between muscle excursion and collagen content in a rabbit model. MATERIALS AND METHODS: Twenty-four rabbits underwent tenotomy of the second extensor digitorum longus (EDL) muscles on the right legs and were randomly assigned to three groups based on the period of time after tenotomy (2, 4, and 6 weeks). The second EDL muscles on left legs were used as controls. At each time after tenotomy, passive muscle excursion and collagen content, determined by hydroxyproline content, were measured bilaterally, and the ratio of each value to the normal one was used. RESULTS: The mean ratio of muscle excursion after tenotomy to the value of the control decreased in a time-dependent fashion: 92.5% at 2 weeks, 78.6% at 4 weeks, and 55.1% at 6 weeks. The mean ratio of hydroxyproline content in muscle to the value of the control increased in a time-dependent fashion: 119.5% at 2 weeks, 157.3% at 4 weeks, and 166.6% at 6 weeks. There was a significant negative correlation between the ratio of hydroxyproline content in muscle after tenotomy to the control values and the ratio of muscle excursion after tenotomy to the control values (r=-0.602, p=0.002). CONCLUSION: The decrease in muscle excursion seems to correlate with the increase in collagen content in the muscle in a time-dependent fashion following tenotomy.

10.The Effect of Hydroxyproline on the Improvement of Facial Wrinkles.

BACKGROUND: Many treatment modalities for wrinkle reduction have been developed in the field of dermatology. Hydroxyproline plays a critical role in stabilizing the triple helix of collagen. The triple-helical conformation gives collagen most of its unique properties and is essential for normal fibrillogenesis. OBJECTIVE: This study was performed to investigate the effect of hydroxyproline on the improvement of wrinkles in vitro and in vivo. METHODS: Cultured fibroblasts and 25 human volunteers were used for in vitro and in vivo studies respectively. Hydroxyproline was administered to a fibroblast culture system and was also applied to periorbital wrinkle lesions twice daily for 12 weeks. The effect of hydroxyproline was examined by using an image analysis system with skin replica, and subjective and objective assessment of wrinkles were evaluated. RESULTS: With the in vitro study, the number of cultured fibroblasts did not increase in the 10(-4), 10(-6) and 10(-8)microgram/ ml hydroxyproline groups compared to the control groups. Procollagen Type 1 C-peptide of cultured fibroblasts did increase in the 10(-8)microgram/ml hydroxyproline group compared to the control group (p<0.05). With the in vivo study, there was a significant decrease in wrinkles at 8 and 12 weeks after application in the 0.5% hydroxyproline applied group compared to the control group (p<0.001). In the subjective visual assessment by the investigator and the subjects at 8 and 12 weeks after application, the 0.5% hydroxyproline treated group showed a significant improvement of wrinkles and more personal satisfaction than the control group (p<0.05). CONCLUSION: 0.5% hydroxyproline had an effect in vivo and in vitro on wrinkle improvement. It is suggested that hydroxyproline might be a candidate compound for treatment or reduction of wrinkles.
