2.Expression Difference and Its Mechanism Study of miR-27a-3p and HOXB8 Protein in Cervical Cancer Tis-sues of HPV16-positive and HPV16-negative Patients

OBJECTIVE:To investigate the expression difference and its mechanism of miR-27a-3p and HOXB8 protein in cer-vical cancer tissues of HPV16-positive and HPV16-negative patients. METHODS:A total of 120 patients with cervical cancer in our hospital during Jan. 2012-Jan. 2016 were divided into HPV16-positive group(60 cases)and HPV16-negative group(60 cases) according to HPV16 infection situation. The expression of miR-27a-3p mRNA and HOXB8 mRNA in cervical cancer tissue were de-tected by RT-qPCR. The expression of HOXB8 protein was detected by Western blotting assay. DNA methylation level of miR-27a-3p promoter region was detected by nested-down methylation specific PCR (nMS-PCR). Histone methylation level of miR-27a-3p promoter region was detected by chromatin immunoprecipitation PCR (CHIP-PCR). The expression of miR-27a-3p mRNA and HOXB8 mRNA were detected after transfecting miR-27a-3p mimic and inhibitor into Human cervical cancer cell line SiHa,respectively. RESULTS:The expression of miR-27a-3p in HPV16-positive group was significantly lower than HPV16-nega-tive group,while HOXB8 mRNA and protein expression,DNA and histone methylation levels of miR-27a-3p promoter region were significantly higher than HPV16-negative group,with statistical significance (P<0.05 or P<0.01). After transfecting miR-27a-3p mimic into SiHa cells,the expression of miR-27a-3p was increased significantly,while that of HOXB8 mRNA was de-creased significantly;after miR-27a-3p inhibitor transfection,the expression of miR-27a-3p was decreased significantly,while that of HOXB8 mRNA was increased significantly,with statistical significance(P<0.01). CONCLUSIONS:HPV16 may down-regu-late the expression of miR-27a-3p through DNA methylation and histone methylation of promoter region,so as to influence the gen-eration and development of cervical cancer. HOXB8 may be the target protein of miR-27a-3p.
3.Mining inter-transaction association rules from time series microarray data

Objective To deduce the interactions between genes from time series microarray data.Methods We used inter-transaction association rules mining technique and GO (Gene Ontology) annotation to analyze the microarray data. Results Using 2-fold-change method, 119 differential expression genes were identified from total 10 080 genes or ESTs, whose expression levels varied significantly on 6 periods of fetus cerebellar development. As a result, about 1 300 inter-transaction association rules were extracted and 10 top rules were kept for their maximum J-measure values. A genes association network graph was deduced based on the 10 top rules. Conclusion Inter-transaction association rules are able to deduce the interactions between genes from time series microarray data and the gene expression status can be predicted based on the association rules.
4.Progress of ventilator related events monitoring and influencing factors

Ventilator related events is a new concept proposed by the America Center for Disease Control and Prevention of the United States. Compared with ventilator-associated pneumonia, ventilator related events has objectivity in diagnosis. In 2013, the National Health Safety Network replaced ventilator-associated pneumonia monitoring in adult inpatients with ventilator-associated event monitoring. This review mainly introduces the concept of ventilator-related events, the evaluation of the relationship between ventilator-related events and respiratory pneumonia, the monitoring situation at home and abroad, and the related factors of ventilator-related events, so as to lay a foundation for the further study in the future.
5.Effect of enalapril combined with folate acid on endothelial function and urine microalbumin in patients with hypertension

Objective To explore the effect of enalapril combined with folate acid on endothelial function and urine microalbumin(UMA) in patients with hypertension.Methods 120 patients with hypertension were randomly divided into two groups:control group (n =60) was given enalapril 10.0mg/d,observation group (n =60) received enalapril 10.0mg + folic acid 0.4mg/d.The total treatment period was 8 weeks.Blood pressure,plasma homocysteine (Hcy),flow mediated dilation (FMD) and UMA were examined.Results The efficacy of pressure releasinghad no significant difference between two groups.Hcy[(10.2 ± 5.8) μmol/L vs (16.6 ±-8.1) μmol/L,t =3.641],FMD[(14.8 ±5.4)% vs (8.2±3.5)%,t =7.325] and UMA[(14.8 ±5.4)mg/L vs (31.6 ±9.5)mg/L,t =8.221] of two groups were significantly different after treatment.Conclusion Combination therapy of enalapril and folate acid can decrease plasma Hey and UMA,restore vascular endothelium function in patients with hypertension.
6.Analysis of the Amount of Antibacterial Ophthalmic Preparation and Drug Resistance of Ocular Infection Bacteria in a Hospital during 2013-2016

OBJECTIVE:To provide reference for suitable selection of antibacterial ophthalmic preparation in the clinic. METHODS:The application of antibacterial ophthalmic preparation in a hospital during Jan. 2013-Dec. 2016 were summarized and analyzed to calculate DDDs. The results of bacterial culture and identification,drug sensitivity test were collected. The correlation of DDDs of antibacterial ophthalmic preparation with resistance rates of bacteria was analyzed by using Pearson test. RESULTS:During 2013-2016,there were 7 categories of antibacterial ophthalmic preparations used in this hospital,involving 10 types. To-tal DDDs showed an upward trend with average annual growth rate of 26.16%. DDDs of Tobramycin (dexamethasone) eye drops were the highest for consecutive 3 years (2013-2015). Gatifloxacin eye drops/eye gel and Ofloxacin eye drops/eye oint-ment ranged 2nd-4th place in the list of DDDs. A total of 26143 ocular specimens were examined,and 21 kinds of bacteria were detected,involving 6221 strains with detection rate of 23.80%. Top 3 bacteria in the list of detection rate were Staphylo-coccus epidermidis (3067 strains),Streptococcus pneumoniae (470 strains) and Staphylococcus aureus (321 strains). Resistance rates of above 3 kinds of bacteria to erythromycin were all higher than 70%,while they were sensitive to rifampicin,chlorampheni-col,gentamicin and levofloxacin,etc. Compared to previous year,the average resistance rates of rifampicin(in 2015),gentamicin (in 2015,2016),tetracycline(in 2015),ofloxacin(in 2014,2015),levofloxacin(in 2016)and gatifloxacin(in 2016)had statisti-cally significant differences(P<0.05). DDDs of tetracycline was positively associated with its average resistance rate(r=1.000,P<0.05),and DDDs of other antibacterials had nothing to do with resistance rate(P>0.05). CONCLUSIONS:The main ocular infec-tions were S. epidermidis,S. pneumoniae and S. aureus. DDDs of tetracycline is certainly associated with resistant rates. Regulating clinical use of antibacterial ophthalmic preparation is important for delaying the development of bacterial resistance.
7.A study of case-mix method by clinical pathway

Objective: To investigate the case-mix method by clinical pathway. Methods: K-MEANS cluster analysis was applied to case-mix classification and artificial neural network was used for case-mix prediction. Results: Five hundred and twenty three inpatient records constructed a case-mix classification scheme of 4 groups.Statistical significant difference of costs existed in 4 groups.The training error of artificial neural network was low(0.0 029) and the predicting result was accurate(98.91%). Conclusion: Case-mix result was more reasonable using records under clinical pathway.The existing models of case-mix depend on dividing individual variables, but artificial neural network does not.
8.Normalization strategies for microarray data

Objective To design a set of software Gene_Norm for the data processing of genechips by normalization methods. Methods Hybridization spots were divided into two groups: "good" and "bad". Turn on off spots were extracted from the "bad" spots. Normalization processing of data was performed by using average ratio. Results Application of Gene_Norm in the processing of high density microarray data (10 080 spots, 42 records of each spot) resulted in satisfactory processing results. Conclusion The software of genechip can be used to process large amount of data. Some parameters can be chosen manually, so the software is of simple operation and wide application.
9.Expression and significance of MYC/BCL2 protein coexpression in diffuse large B-cell lymphoma

Objective To explore the condition of MYC/BCL2 protein coexpression in 245 patients with diffuse large B-cell lymphoma (DLBCL)and to find the correlations among MYC/BCL2 protein coexpres-sion,the germinal center B-cell-like (GCB)subtype and non-GCB subtype.Methods Paraffin-embedded lymphoma samples from 245 patients with DLBCL were studied using immunohistochemistry for MYC,BCL2, CD10,BCL6,MUM1 and KI67.And the protein expressions of them were analyzed.Results In the speci-mens of 245 patients with DLBCL,the patients with non-GCB were 163 (66.5%),and the patients with GCB were 82 (33.5%).The group of MYC/BCL2 protein coexpression comprised 35.9% (88/246)of the all patients.Non-GCB subtype group had a significantly higher frequency of MYC/BCL2 protein coexpression than the GCB subtype group (41.7%∶24.4%;χ2 =7.116,P=0.008).Conclusion The data shows that MYC/BCL2 protein coexpression occurs significantly more commonly in the non-GCB subtype.We can predict prog-nosis and choose therapeutic regimen base on the assessment for MYC and BCL2 protein expression.
10.Synthesis of A Novel Fluorescence Switch Conjugated Polymers and Its Discrimination Ability Toward S-Adenosylmethionine

A novel fluorescent switch was constructed based on poly 3-{[ 1-( 2-hydrazino-2-oxoethyl ) piperidin-4-ylidene] methyl}thiophene ( PMTH ) in the presence of Pb2﹢ or S-adenosylmethionine ( SAM ) . The switch turned off in the presence of Pb2﹢owing to complex effect, and the fluorescence intensity of PMTH solution was efficiently quenched by Pb2﹢ ions. Upon adding SAM to the Pb2﹢-PMTH solution, which was stronger Pb2﹢ chelators, it could form more stable SAM-Pb2﹢, the Pb2﹢ ion was displaced from PMTH and the fluorescence of PMTH was recovered. By triggering the turn-on signal of PMTH, a new discrimination ability toward SAM method for the determination of SAM was established. The effects of the interaction on the fluorescence spectral characteristics of these Pb2﹢-PMTH and Pb2﹢-SAM complexes in H2 O-CH3 CH2 OH(4:1, V/V) solution, suitable reaction conditions and influencing factors were investigated. This method offered good sensitivity and selectivity for detecting SAM in the presence of amino acids and metal ions. Under optimum conditions, the concentration of SAM in the range from 1. 0 × 10-8-2. 0 × 10-6 mol/L exhibited a linear relationship with the relative fluorescence intensity. The regression equation was △I = 68. 51﹢72. 32c (μmol/L), the correlation coefficient r=0. 9982. The limit of detection was 8. 72×10-9 mol/L. The system was successfully applied for detecting SAM in human serum, urine and injection samples.
