1.Hemangioblastic characteristics of human adipose tissue-derived adult stem cells in vivo

AIM: To investigate whether Flk1~+ CD31~- CD34~- cells isolated from human adult adipose tissue have characteristics of hemangioblasts in vivo. METHODS: After sublethally irradiated (300cGy) with a caesium source, the female non-obese diabetic/severe combined immunodeficient (NOD/SCID) mice were injected with human adipose tissue-derived Flk1~+ CD31~- CD34~- cells (10~5 cells per mouse) via tail vain with 0.4 mL Roswell Park Memorial Institute medium (RPMI-1640). The control mice received the same volume of RPMI-1640 medium. All mice were killed 2 months after transplantation for further study. The differentiation potential of Flk1~+ CD31~- CD34~- cells was assessed in bone marrow and gastrointestinal tract by the methods of flow cytometry, RT-PCR, FISH, and triple-color immunofluorescence. RESULTS: Flk1~+ CD31~- CD34~- human adipose tissue-derived adult stem cells differentiated into endothelial cells and hematopoietic cells at the single-cell level in vivo. CONCLUSION: Human adult adipose tissue-derived Flk1~+ CD31~- CD34~- cells bear characteristics of hemangioblast in vivo and may have potential application for the treatment of hematopoietic and vascular diseases.
2.Dimethyloxaloylglycine promotes spermatogenesis activity of spermatogonial stem cells in Bama minipigs

Background:
The testis has been reported to be a naturally O 2 -deprived organ, dimethyloxaloylglycine (DMOG) can inhibit hypoxia inducible factor-1alpha (HIF-1α) subject to degradation under normal oxygen condition in cells.
Objectives:
The objective of this study is to detect the effects of DMOG on the proliferation and differentiation of spermatogonial stem cells (SSCs) in Bama minipigs.
Methods:
Gradient concentrations of DMOG were added into the culture medium, HIF-1αprotein in SSCs was detected by western blot analysis, the relative transcription levels of the SSC-specific genes were analyzed using quantitative reverse transcription polymerase chain reaction (qRT-PCR). Six days post-induction, the genes related to spermatogenesis were detected by qRT-PCR, and the DNA content was determined by flow cytometry.
Results:
Results revealed that the levels of HIF-1α protein increased in SSCs with the DMOG treatment in a dose-dependent manner. The relative transcription levels of SSCspecific genes were significantly upregulated (p < 0.05) by activating HIF-1α expression. The induction results showed that DMOG significantly increased (p < 0.05) the spermatogenesis capability of SSCs, and the populations of haploid cells significantly increased (p < 0.05) in DMOG-treated SSCs when compared to those in DMOG-untreated SSCs.
Conclusion
We demonstrate that DMOG can promote the spermatogenesis activity of SSCs.
3.Current status of stress level among patients in observation room of emergency department and its influencing factors

Objective To explore the current status of stress level and its influencing factors among patients in observation room of emergency department .Methods Totally 300 patients in the observation room of emergency department from September 2014 to September 2015 were recruited in this study by convenience sample method.General situation questionnaire and stress-reacton questionnaire ( SRQ) were used to conduct this cross-sectional survey .Results The scores of SRQ and body reaction factor were ( 56.25 ±20.20) and (19.41±7.87), and they were all significantly higher than the national norm (t=2.231, 6.034;P<0.05).The score of behavior reaction was (10.65±4.82), and it was lower than the national norm (t=-9.143, P<0.01). There was no significant difference between the score of emotional reaction and national norm ( P>0.05) .Single-factor analysis showed that , the differences in the score of emotional reaction among patients with different working situation and monthly income were significant (F=3.684,4.109,P<0.05).The differences in the score of body reaction among patients with different working situation , occupation , monthly income and religion were significant (F=4.851,3.624,2.678,5.033;P<0.05).The differences in the score of behavior reaction among patients with different working situation and occupation were significant (F=4.181,2.489;P<0.05).Multiple stepwise regression analysis showed that religious faith and monthly income were identified as significant influencing factors of stress level (P<0.05).Conclusions Medical personnel should guide patients to correctly face the stress based on their religious faith and monthly income , so as to promote the recovery of health .
4.Correlation analysis of stress and coping styles among patients in Emergency Observation Room

Objective To explore the stress among patients in Emergency Observation Room and its correlation with coping modes.Methods A total of 300 patients in Emergency Observation Room of a Class Ⅲ grade A hospital were recruited by convenient sampling method for a cross-sectional survey and its correlation, from September 2014 to September 2015, using the general situation questionnaire, stress-reaction questionnaire (SRQ) and medical coping modes questionnaire (MCMQ). The data of survey was compared with norm, and Pearson correlation analysis was used to analyze the correlation between stress reaction and medical response level.Results The score of stress reaction was (56.25±20.20), in which somatic reaction was (19.41±7.87), higher than norm of hospitalized patients and the behavior reaction was (10.65±4.82),lower than the norm of hospitalized patients (t=6.034, -9.143;P<0.01). The total score of coping style of patients in emergency observation room was (39.26±6.97), in which the face dimension was (17.63±3.90), avoidance (13.96±3.28), yield (7.66±2.66), all lower than the norm of hospitalized patients (t=-8.200,-2.514,-7.485;P<0.05). The correlation was positive between stress reaction and coping styles (r=0.125-0.510,P<0.05). Conclusions The stress has close correlation with coping style for patients in Emergency Observation Room. Medical personnel should guide emergency patients to correctly face stress, and improve their coping styles.
5.Regulative function of telomerase and extracellular regulated protein kinases to leukemic cell apoptosis.

In order to investigate the regulative function of telomerase and phosphorylated (activated) extracellular regulated protein kinase (ERK) 1 and 2 in the leukemic cell lines HL-60 and K562 proliferation inhibition and apoptosis, three chemotherapeutic drugs Harringtonine (HRT), Vincristine (VCR) and Etoposide (Vp16) were selected as inducers. The proliferation inhibition rate was detected by MTT method, the cell cycle and cell apoptosis was analyzed by flow cytometry and the telomerase activity was detected by the telomeric repeat amplification protocol (TRAP) assay and bioluminescence analysis method. The phosphorylated ERK1/2 protein expression was detected by western blot method. The results showed that HRT, VCR and Vp16 could inhibit cell proliferation, induce apoptosis, inhibit telomerase activity and down-regulate the protein expression of phosphorylated ERK. It was suggested that ERK signal transduction pathway was involved in the down-regulation of telomerase activity and the onset of apoptosis in the leukemic cells treated by HRT, VCR and Vp16.

6.Study of celluar toxicity and antibacterial property of nano-silver coating modified denture base

Objective To investigate celluar toxicity and antibacterial property of nano-silver coating modified denture base and to provide a theortical basis for clinical application.Method The samples were divided into coating group and denture base group.Samples in coating groups were treated with silver ion sputtering and were further divided into the 10,20,40,60,80,100 s subgroups according to their sputtering time.Surface microstructure of samples in coating groups and denture base group were observed by scanning electron microscopy (SEM),and composition of material surface were determined with energy dispersive spectrum (EDS) analysis.Methyl thiazolyl terazolium(MTT) method was used to analysis L929 cells proliferation rate and cell toxicity grade while the coating groups(n =6) and group test specimens (n =6),negative control (polyethylene,PE) (n =6) and positive control group (PVC-org.Sn) (n =6) were co-cultured with L929 cells.The antibacterial properties of coating groups were investigated by using the bacterial membrane adhering method with Streptococcus mutans and Candida albicans.Results SEM observation showed that the surface of control group specimens were smooth,whereas dense and uniform distributed nano silver particles were visible on the surface of coating group specimens.The silver particles were round,and the diameter size was 10 nm.MTT assay showed that after being co-cultured with specimens in control group,L929 cell had higher proliferation rate than 100% at 24,48,72 h,and cytotoxicity were graded 0.The membrane adhering method results showed that all of bacterial colonies were found in control group,but without anything at the coating groups.Conclusions Deposition of nano silver on the denture base surface by ion sputtering method is a viable method.There's no cytotoxicity of series Ag-coating material.Nano-silver modified denture base material benefits L929 cell surface proliferation and has antibacterial action.
7.Progress in the diagnosis and treatment of reactive arthritis in children

Reactive arthritis(ReA) is a kind of sterile, non-purulent arthritis that occurs after microbial infections far from the joints.The disease has a broad spectrum, and it can be classified into three categories based on clinical characteristics: human leukocyte antigen B27(HLA-B27)-associated ReA, acute rheumatic fever(ARF), and post-streptococcal reactive arthritis(PSRA).In addition to joints, it may also involve the gastrointestinal tract, skin, eyes, and heart.Unlike adults, the pathogenesis of ReA in children is more complex.HLA-B27-associated ReA is more common after gastrointestinal and respiratory infections, with less involvement of the central axis and sacroiliac joints and more involvement of the hip and peripheral joints and attachment point inflammation.ARF is most common in children aged 5 to 15 years, characterized by migratory and multiple arthritis.The duration of onset of PSRA in children is shorter than that in adults.This article reviews the epidemiology, clinical manifestations, diagnosis and treatment of ReA in children to improve clinicians′ understanding of ReA in children.
8.Interpretation of clinical practice guidelines for juvenile idiopathic arthritis associated uveitis

Juvenile idiopathic arthritis associated uveitis (JIA-U) is a primary extra-articular complication of juvenile idiopathic arthritis, which can cause symptoms such as red eyes, eye pain, photophobia, floating shadows in front of the eyes, or decreased vision.In severe cases, it can induce blindness and cause significant harm to children and their families.The American College of Rheumatology, the Multinational Interdisciplinary Working Group for Uveitis in Childhood, the Pediatric Rheumatology European Society and the Canadian Rheumatology Association have released relevant clinical guidelines and expert consensus on this disease.This article mainly interprets the above-mentioned clinical guidelines and expert consensus regarding the diagnosis, treatment, and long-term management of JIA-U, to improve the efficiency and management quality in clinically diagnosing and treating JIA-U.
9.Single hydrogen-methane breath test for the diagnosis of small intestinal bacterial growth

Objective:To investigate the diagnostic value of a single hydrogen-methane breath test (SHMBT) for small intestinal bacterial overgrowth (SIBO).Method:The current investigation was a cross-sectional study. Questionnaires and SHMBTs were administered to 162 patients with gastrointestinal symptoms (case group) and 69 healthy volunteers (control group). Differences in SHMBT results between the two groups were assessed,and cut-off values of CH 4 (methane) and H 2 (hydrogen) were analyzed via receiver operating characteristic (ROC) curves. Lastly,archived SHMBT data from 2 655 patients with gastrointestinal symptoms (validation set) were used to evaluate the diagnostic value of the SHMBT with respect to SIBO. The Chi-square test,the Mann-Whitney U test,Spearman′s Rank correlation analysis,and the Z test were used for statistical analysis. Results:Based on the international recommended diagnostic criteria for SIBO,which are fasting CH 4 ≥10 ppm (parts per million) or H 2 ≥20 ppm,the SHMBT-positive rate in the case group was significantly higher than that of control group (35.2% vs. 21.7%, χ2=4.08, P=0.043). Levels of CH 4 and H 2 were higher in the case group than in the control group [CH 4: 3(2,7) vs. 3(1,3) ppm, H 2: 11(4,22) vs. 10(5,15) ppm],and the difference in CH 4 levels was statistically significant ( Z=6.22, P=0.001). ROC curves were generated based on whether the subjects had gastrointestinal symptoms. The areas under the ROC curves were 0.633 for CH 4 alone,0.531 for H 2 alone, and 0.620 for CH 4 combined with H 2. The cut-off values were fasting CH 4≥4 ppm,fasting H 2≥13 ppm,and fasting CH 4 ≥5 ppm (or CH 4≥4 ppm and H 2≥24 ppm),respectively. Measuring CH 4 alone and CH 4 combined with H 2 was effective for determining the presence of gastrointestinal symptoms ( P<0.05). When CH 4 alone or CH 4 combined with H 2 were used as diagnostic indicators of SIBO, the respective SHMBT-positive rates in the validation set were 34.2% and 30.4%. These rates did not significantly differ from the SIBO-positive rate of 32.0% obtained via the international recommended diagnostic criteria ( P>0.05). The specificity of CH 4 alone was 79.9%,and the accuracy of CH 4 alone was 68.8%. The specificity of CH 4 combined with H 2 was 85.0%,and the accuracy of CH 4 combined with H 2 was 71.7%. Conclusion:Rapid one-time determination of CH 4 and H 2 in exhaled breath may a viable diagnostic method for SIBO, and using CH 4 combined with H 2 ( i.e.,fasting CH 4≥5 ppm, or CH 4 ≥4 ppm and H 2 ≥24 ppm) as cutoff values may be feasible.
10.Effect of hyperbaric oxygen therapy on postoperative cognitive dysfunction in elderly patients undergoing general anesthesia

Objective To evaluate the effect of hyperbaric oxygen (HBO) therapy on postoperative cognitive dysfunction (POCD) in elderly patients undergoing general anesthesia.Methods A total of 112 patients,aged 65-75 yr,of American Society of Anesthesiologists physical status Ⅱ or Ⅲ,undergoing elective non-cardiac surgery with general anesthesia,were randomly divided into control group (C group,n =54) and HBO group (n =58).Patients were exposed to hyperbaric oxygen in a hyperbaric oxygen chamber once a day from day 3 to day 12 after surgery in both groups.Pressure was slowly increased to 2 atmosphere absolute within 20 min,pure oxygen was inhaled for 35 min by mask,5 min later pure oxygen was inhaled for another 35 min,oxygen inhalation was then stopped and pressure was slowly increased to 1 atmosphere absolute in HBO group.Patients inhaled air at 1 atmosphere absolute for 70 min in C group.Cognitive function score was assessed using Mini-Mental State Examination,language ability test,visual identification function test,digit span backwards task and Hasegawa's Dementia Scale (HDS) at 2 days before surgery and 7 and 13 days after surgery.The development of POCD was recorded.Results Compared with the baseline at 2 days before surgery,language ability test,digit span backwards task and HDS scores were significantly decreased at 7 and 13 days after surgery in C group,and digit span backwards task scores were significantly decreased at 7 days after surgery in HBO group (P<0.05 or 0.01).The language ability test and HDS scores were significantly higher,and the incidence of POCD was lower at 7 and 13 days after surgery in HBO group than in C group (P<0.05).Conclusion HBO therapy can reduce POCD in elderly patients undergoing general anesthesia.
