1.Expressions of five matrix metalloproteinase genes in papillary thyroid carcinoma

Objective To study the expressions of matrix metalloproteinase (MMP) gene family in papillary thyroid carcinoma (PTC) and its relationship to the pathogenesis of PTC. Methods The thyroid tissues of patients were collected in surgical operation, including 8 cases with papillary thyroid carcinoma, 7 cases with Hashimoto′s thyroiditis, 8 cases with adenoma and 8 normal tissues. The total RNA of these samples were extracted and purified by the method of acidic phenol. The gene expressions of MMP-2, MMP-9, MMP-14, MMP-25 and MMP-26 were assayed by RT-PCR and 1% agarose gel electrophoresis. The fluorescent degree of electrophoretic bands was detected by automatic gel scanner and analyzed statistically in order to compare the levels of gene expressions among 4 groups. 2.THE DIGESTIBILITY AND THE EFFECT ON DRY STOOLWEIGHT OF DIETARY FIBER FROM APPLE, BRAN, CABBAGE, CARROT AND PEAR IN RATS

The 5 foods, apple, bran, cabbage, carrot and pear, were fed to rats as part of 5 diets each containing 2.5% neutral detergent fiber (NDF) and the effect on increasing dry stool weight (DSW) and the digestibility of dietary fiber (DF) components of the foods were studied. The results showed that DSW increased with increased NDF intake, cabbage being the most effective. The mean DSW of the test food groups was 2.27 times compared with non-fiberous controls. The SDW was correlated with intake of NDF and cellulose from analysis of individual variation. The regression equation was y =- 1.9040 + 3.5644X1 (NDF) + 3.5644X2 (cellulose).The digestion of DF components from carrot was the most complete, while hemice-llulose was 1.3 times as digestible as cellulose. Lignin was found to have an influence on DF breakdown. Increasing hemicellulose level might decrease the digestibility of cellulose. The variation of DF digestibility of di- fferent food may also be ascribed to the physical and biological properties of the food.
3.THE RELATIONSHIP OF ERYTHROCYTE MEMBRANE FLUIDITY WITH LIPID PEROXIDATION IN RIBOFLAVIN DEFICIENT RAT

The effect of riboflavin deficiency on the fluidity of erythrocyte (RBC) membrane, the level of membrane MDA, the activity of RBC SOD and lipid peroxidation were studied. Growing male rats were fed with an experimental (riboflavin-deficient, RD) or a control (riboflavin-supplemented, RS, 22mg/ kg) diet for 7 weeks.The RBC membrane of RD rats contained higher level of MDA (0.6868?0.1732 nmol/mg protein) compared with the control (0.5548?0.0980 nmol/mg protein), while the activity of RBC SOD (7.745210.6101 nU/mg protein) of RD rats were significantly decreared compared with the control (8.2685?0.3010 nU/mg protein), which indicated the lipid peroxidation was elevated.Membrane fluidity was studied with fluorescence polarization (P) and mean microviscosity (?) using DPH as probe. The value of P and? of RD rats (0.2976 ? 0.0198, 3.9483? 0.3680, respectively) were significantly higher than the RS rats (0.2760 ? 0.0207, 2.875310.4634, respectively), which showed the membrane fluidity in RD rats was decreased according with the increase in lipid peroxidation. This study demonstrated that the RBC membrane fluidity was dicrcased in riboflovin deficient rats. It was relative to the increasing lipid peroxidation and decreasing RBC SOD activity.

5.Preparation of a new surface-enhanced Raman scattering substrate and its applica-tion to the detection of E.coli

Objective To develop a new type of Raman-enhanced substrate for rapid detection of E.coli based on label-free surface-enhanced Raman scattering( SERS) technology.Methods Stober’ s improved method was used to prepare 360 nm silica ( SiO2 ) nanospheres.Prepared gold core-silver shell nanoparticles( Au@Ag) of different size were attached to 360 nm SiO2 to fabricate the nanocomposites ( SiO2-Au@Ag ) that were characterized by transmission electron microscopy (TEM) and UV-visucl light adsorption spectra (UV-Vis).PATP was detected to select SiO2-Au@Ag with optimal SERS effect.This optimal SiO2-Au@Ag was used to obtain the sensitivity of PATP and E.coli detection after a simple mixed culti-vation.Results TEM images showed that Au@Ag aggregated with the size of Au@Ag attached to 360 nm SiO2 .UV-Vis spectra indicated that the maximum absorption of Au@Ag and SiO2-Au@Ag had a red shift with the invrease of Au@Ag size.The experiment results suggested that detection sensitivity of PATP by SiO2-100 nm Au@Ag 10 -10 mol/L, while the lowest detectable E.coli concentration was 105 CFU/ml.Conclusion The 360 nm SiO2 binding with 100nm Au@Ag exhibits great potential for SERS applications.

Objective To analyze the status of human metapneumovirus in children with respiratory tract infection in Shenz-hen and Shantou and the clinical characteristics of the diseases with hMPV infection.Methods The viral nucleic acid of 1 217 samples were amplified by RT-PCR(1 137 from children with respiratory tract infection,80 from health children),and five positive PCR products were chosen randomly for sequencing.Nucleotide sequences alignment and phylogenetic analysis were performed with DNAstar software.The clinical data of the children with hMPV infection were analyzed.Results The positive rate of hMPV infection identified from 1 137 children wtith respiratory tract infection was 4.49% (51/1 137), hMPV was significantly prevalent in February,March and April.Most of the positive cases was under 3 years old.Five hMPV gene fragments of sequence were closely related to hMPV in the GenBank.Similarity of hMPV partial N gene with which published at nucleotide levels varied from 80.8%~98.4%.Phylogenetic tree of nucleotide revealed two different gen-otypes.All hMPV positive patients suffered from fever,cough and wheezing.The clinical diagnoses were wheezing pneumo-nia (17),bronchiolitis (9).None of the nasal swabs from 80 healty subjects were tested hMPV specific gene fragment.Con-clusion hMPV is an important viral pathogen in children with respiratory tract infection in Shenzhen and Shantou.Two genotypes may circurate in these areas.
8.Effects of light intensity on associated enzyme activity and gene expression during callus formation of Vitis vinifera.

We analyzed the best light intensity for callus induction and maintenance in Vitis vinifera and explored the mechanism of grape callus browning. Tender stem segments of grape cultivar "gold finger" were used to study the effects of different light intensities (0, 500, 1 000, 1 500, 2 000, 2 500, 3 000 and 4 000 Lx) on the induction rate, browning rate and associated enzyme activity and gene expression during Vitis vinifera callus formation. The callus induction rate under 0, 500, 1 000 and 1 500 Lx was more than 92%, significantly higher than in other treatments (P < 0.05). A lower browning rate and better callus growth were also observed during subculture under 1 000 and 1 500 Lx treatments. We found that chlorogenic acid, caffeic acid, p-hydroxybenzoic acid and coumaric acid contents were correlated with the browning rate of callus, among which chlorogenic acid content was positively correlated with the browning rate (P < 0.05). Peroxidase (POD) and polyphenol oxidase (PPO) activities were negatively correlated with the browning rate of callus (P < 0.01). The POD, PPO and phenylalanine ammonialyase (PAL) expression levels were positively correlated with the browning rate at P < 0.05 or P < 0.01. An appropriate light intensity for the tissue culture of Vitis vinifera was 1 000-1 500 Lx, higher or lower light intensities significantly impaired normal callus growth.

9.Correlation of cancer-related fatigue and life quality of lung cancer patients after chemotherapy

Objective To explore the correlation of fatigue and life quality of lung cancer patients after chemotherapy. Method A survey was conducted among 54 lung cancer patients after chemotherapy with revised piper fatigue scale ( PFS ) and medical outcomes study (SF-36). Result The total score of PFS of lung cancer patients was (6.44 ± 1.62). The score of SF-36 related area include physical dimension (58.47 ± 5.48), psychological dimension (55.04 ± 4.91) and social dimension (49.21 ± 4.77) environment dimension (52.86 ± 4.98). The total score of PFS was negatively correlated with physical, psychological and environmental dimensions (P<0.05), and not correlated with social dimension (P<0.05). Conclusions The cancer-related fatigue and life quality of lung cancer patient is at medium-severe level, and their life quality is at low level. Positive and effective psychological intervention and psychological nursing can reduce cancer-related fatigue and improve their life quality.

Objective To evaluate the effects of the improved monitoring system for pressure ulcers? Method The monitoring system for pressure ulcers was improved and the measures were as follows:completing the organizational system for managing ulcers, fulfilling the regulations,revising the report form and enforcing classified nursing training? Result The accuracy rate of risk assessment and the methods for using wet dressings and negative pressure drainage were significantly improved and the effects by using decompression and skin-prevention pads were significantly better than those before improving the monitoring system(P < 0?05)?Conclusion The improved monitoring system for pressure ulcers may help to improve the clinical practice of pressure ulcers?
