Objective To produce a new type of medical bed sheet suitable for hemodialysis .Methods Application effect of traditional methods compared hemostatic clamp fixed cycle trail tube with the use of dialysis sheets fixed cycle trail tube method in hemodialysis .Results Dialysis sheets of reasonable structure setting has the advantages of convenience and practicality than using sterilized forceps traditional fixed mode .The adaptability is strong, and preparation cost is low .The fixing device and the sheets are fixed as a whole , without also prepare disin-fection clamp fixed cycle trail tube , not circulating blood road pipe clamp bad , and put an end to the sheets caused by repeated clamping forceps to damage , and can be used as a piece of goods one -time cleaning and disinfection .Con-clusion The standard of energy saving and consumption reduction is achieved by dialysis bed sheets , which can a-chieve the goal of controlling and improving the quality of medical care .
2.Biocompatibility of freeze-dried antigen-extracted sheep cancellous bone scaffolds

BACKGROUND:Xenogeneic bone has a natural porous structure that is similar to human bone. In the treatment of bone defects, the porous structure is helpful to guide bone regeneration, but different degrees of immune responses wil be caused during the implantation process. OBJECTIVE:To prepare a freeze-dried antigen-extracted sheep cancelous bone scaffold and to evaluate its biocompatibility. METHODS: The sheep vertebral cancelous bone was colected to prepare two kinds of antigen-extracted heterologous bone scaffolds that were treated with chemical methods as chemical group and treated with chemical methods+cryopreservation at a-80℃ refrigerator for 4 weeks+drying in vacuum apparatus+60 RESULTS AND CONCLUSION:Freeze-dried bone had no cytotoxicity, no acute toxicity and heat reaction, and was negative for the intracutaneous stimulation test. The scaffold in the chemical group had cytotoxicity and mild irradiation as freeze-dried bone group. (1)Cytotoxicity test: Bone marrow mesenchymal stem cels isolated from sheep were cultured in extracts of the chemical group, free-dried bone group and Dulbecco’s modified Eagle’s medium/Ham’s nutrient mixture F-12. (2) Heat reaction and acute toxicity tests: Extracts from the chemical group, freeze-dried bone group and normal saline were respectively injected into the ear vein of rabbits. (3) Intracutaneous stimulation test: Extracts from the chemical group, freeze-dried bone group and normal saline were respectively injected subcutaneously into the back of rabbits. Co acute toxicity reaction, sent heat source and had mild irritation. Results show that after freeze drying processing, the sheep vertebral cancelous bone has good biocompatibility, can meet the requirements of bone tissue engineering, but the bone that through chemical processing exhibits a relatively poor biocompatibility that cannot achieve the safety standard of biological scaffold materials.
3.Xenogeneic bone properties prepared by calcination and freeze-dried methods

BACKGROUND:Xenogeneic bone structure and biological characteristics are similar to human bone tissue, and the xenogeneic bone has a decreased antigenicity after physicochemical treatment, with a natural porous structure and rich source, and can be kept for a long time, which is considered to be an effective way to solve the shortage of the autogenous bone and al ograft bone.
OBJECTIVE:To compare the physical and chemical properties of xenogenic bone materials prepared by two different methods.
METHODS:Sheep cancellous bone treated with chemical method was placed into the muffle furnace at 1 000 ℃for 2 hours to prepare calcined bone. Another cancellous bone was placed into an 80 ℃ refrigerator for 4 weeks and then placed into a vacuum instrument to prepare freeze-dried bone. Cancellous bone rinsed with ultra-pure water served as controls.
RESULTS AND CONCLUSION: Three groups of samples retained three-dimensional porous structure which similar with human bone tissue under microscopic observation. The framework was stil intact, with a smal pore of 55-650μm and high porosity of 65%-80%. For the calcined bone, the toughness was decreased and the brittleness increased significantly, but the freeze-dried bone had a little changes in the mechanical properties. Through diffraction analysis, hydroxyapatite was the main composition of the three groups. However, there was a smal amount ofβ-tricalcium phosphate in the calcined bone. Spectrum analysis confirmed that calcium and phosphorus content in these three groups were al close to the human body. The results suggest the cancellous bone treated with these two methods is similar to human bone structure, and the major elements are close to the body. In addition, the cancellous bone after processing has enough smal pore and higher porosity. However, calcination process has a more influence on the mechanical property of scaffold materials, and the freeze-dried bone has a little change but the antigen cannot be completely removed that can reach the basic requirements.
4.Clinical significance of combined detection of P53 and Ki67 in patients with esophageal carcinoma

Objective To investigate the clinical significance of expression of P53,Ki67 in proliferation,invasion and metastasis of cancer cells of patients with esophageal carcinoma.Methods Eighty-six patients with esophageal carcinoma were selected as tumor group from January 2006 to May 2008.And another 86 patients without tumor were as control group.ABC-immunohistochemistry was used to detect the expression of P53 and Ki67 in 172 cases of patients with esophageal carcinoma.Results The expression of P53 in patients with esophageal carcinoma was different from that in control subjects (x2 =4.045,P < 0.05),and the P53 expression rate in two group were 66.3% (57/86) and 51.1% (44/86) respectively.The rate of P53 expression reaching to + + in two groups was also significant (x2 =8.592,P < 0.01).Ki67 expression in patients with esophageal carcinoma was higher than that of control patients (76.7% (66/86) vs.60.5% (82/86),x2 =5.291,P < 0.05).Additionally,there was significant difference regarding of Ki6 expression reaching to + + positive(x2 =13.661,P < 0.01).Moreover,the expression of P53 and Ki67 were found to be a positively correlation(r =0.400,P =0.00).The expression of P53 and Ki67 were related with invasion and differentiation of cancer cells (x2 =3.945,5.794 respectively,all P < 0.05) and lymph node metastasis (x2 =5.570,4.354 respectively,all P < 0.05).Conclusion Over-expression of P53 and Ki67 were involved in the process of proliferation,invasion and metastasis of cancer cells in esophageal carcinoma,which coherently impacted the occurrence and development of esophageal cancer.The combined detection of P53 and Ki67 seems to help the early diagnosis of esophageal cancer and evaluation of the degree of malignancy,invasion,metastasis and prognosis.Over-expression of P53,Ki67 might be served as a reference marker in screening for chemotherapy for esophageal cancer.
5.The microstructure of antigen-extracted heterologous bone

BACKGROUND:Different methods to remove immunogenicity have different effects on the spatial microstructure of antigen-extracted heterologous bone.
OBJECTIVE:To compare the microstructure of the antigen-extracted heterologous bone via different methods to provide experimental data for tissue engineering industrialization.
METHODS:Fresh cancellous bones extracted from adult sheep vertebrae were prepared into cylinders. Their long axis direction was the same with orientation of the trabeculae. After vibration washing and different-frequency ultrasound rinsing, the cylinder samples were randomly divided into three groups:in physical calcined group, the samples were defatted, decellularized and deproteinized sequential y using methanol/chloroform and hydrogen peroxide, then bathed in sodium pyrophosphate and directly calcined at 1 000 ℃ for 3 hours;in chemical group, the samples were defatted, decellularized and deproteinized sequential y using methanol/chloroform and hydrogen peroxide;in control group, the samples were dried natural y at room temperature. Microstructure of the samples in each group was analyzed and compared through determination of porosity, scanning electron microscopy observation, X-ray diffraction analysis, X-ray atomic spectroscopy elemental analysis microscopic spatial structure.
RESULTS AND CONCLUSION:The physical calcined and chemical groups maintained natural network pore system to different extents. The size of the large pore was 50-600μm and that of the smal one was about 2μm. The porosity was 55%to 70%. Hydroxyapatite was the main component of the physical calcined group which was determined by X-ray diffraction, and a smal amount of theβ-Tricalcium phosphate was also determined. In the chemical group, the main component was only hydroxyapatite. The three-dimensional spatial structures of the deproteinized cancellous bones were not damaged greatly, and they had a natural pore network system. Antigen component of xenogeneic cancellous bone can be more thoroughly removed by physical calcination step. The scaffold material made by antigen-extracted heterologous bone may satisfy the demands for bone tissue-engineering scaffolds.
6.Vasodilatory effect of midazolam on pre-contractions of in-vitro porcine coronary artery and its mechanisms

Aim To investigate the effects of midazolam on porcine isolated coronary artery rings pre-contracted by potassium chloride(KCl)and the possible mechanism.Methods The vessel tension recorder system was used.Isotonic tension of porcine isolated coronary artery rings precontracted by KCl(30 mmol·L-1)was recorded.The vasorelaxing action of midazolam and effects of various drugs were observed in the rings.Results Midazolam(3×10-6~1×10-4 mol·L-1)respectively concentration-dependently reduced the contraction induced by KCl,and there was significant difference between the rings with intact and denude endothelium(P<0.05).On KCl-induced precontraction,midazolam′s relaxation was depressed by L-NAME and the blend of L-NAME and L-Arg(P<0.05),but was not affected by Indo,L-Arg and 1400W.The contraction was not prevented by pretreatment with the inhibitor of Na+/Ca2+ exchanger(KB-R7943).The inhibitor of KATP(Gli)restrained the diastolic function of midazolam(P<0.05),while the inhibitor of BKCa(TEA),Kir(BaCl2),KV(4-AP)had no obvious effect.Conclusions Midazolam produces remarkable vasodilatation on KCl pre-contracted porcine isolated coronary artery rings.Its relaxtion effect is via concentration-dependent and endothelium-dependent mechanisms and relevant to the production of NO.Na+/Ca2+ exchanger is not involved midazolam′s vasodilatation on KCl pre-contracted porcine coronary artery rings.The relaxant mechanism of midazolam may be concerned with KATP.The Kir,BKCa and KV may be not involved.

Objective To study the effects of ganoderma applanatum polysaccharides(GAPS) on cell morphology, proliferation and PDGFR -?expression in cell lines MGC - 803 , and to explore its potential mechanisms of anti - tumor of GAPS. Methods Cell morphology was observed by inverting microscope. MTT assay was used to investigate the inhibitory effect of GAPS on MGC -803.Expressions of PDGFR -?was analyzed by flow cytometry (FCW). The fluorescence intensity of expressions of PDGFR -?was observed by fluorescence microscope. Results Cells of GAPS group were irregular shaped and grew poorly. GAPS inhibited the proliferation of MGC -803 cells in dose - dependent and time - dependent manners . After MGC - 803 cells were treated with GAPS for 72h, GAPS could down - regulate expression of PDGFR -?observed by fluorescence microscope which was consistented with the results of FCW with statistic significance difference as compared with control group (P

Objective To investigate the clinical significance of piatelet activity in patients with chronic cor Dulmonale.Methods PAC-1 and CD62p was measured with flow cytometry in whole blood samples from 40 patients and 30 normal controls.The pulmonary arterial pressure was detected through Doppler echocardiography.The arterial partial pressure of oxygen and the carbon dioxide were also analyzed.Results PAC-1 and CD62p increased significantiy (P<0.01).Conclusion Platelet activity is positively related to pulmonary artrial systolic pressure,CO2 partial pressure,and negatively related to O2 partial pressure.
9.Research and development of genetic diagnostic method of Staphyloccocus aureus based on loop-mediated isothermal amplification

Objective To develop a method of loop-mediated isothermal amplification(LAMP) to Staphyloccocus aureus rapidly, specifically, sensitively and simply suited for the primary health agency. Methods According to conserved nucleotide of Staphyloccocus aureus and principle of LAMP, we designed a set of LAMP primers and set up an LAMP reaction system. We evaluated the specificity, sensitivity and re-peatability of the method. In addition,we evaluated the linearity between initial template copies 1g value and reaction time (the time when the fluorescent value is 1×10~4). Results The optimal assay showed that it was no cross-reaction with other closely related members of pathogens, and was 10 times more sensitive than PCR. The coefficient of variance between tests was less than 5% ,and the kinetics curves showed a good line-arity between initial template copies lg value and reaction time(r~2=0. 9501). The detection activity could be finished within 1 h with the sensitivity of LAMP was 100% and the specificity was 94.4%, and the accuracy was 96.6%. Conclusion These findings demonstrated that the LAMP had the potential clinical application for detection and differentiation of Staphyloccocus aureus in the public health agency for its sensitive, specific and simple feature.
10.Prognostic Value of a Change in Troponin-Ⅰ Levels in Patients with Sepsis-associated Myocardial Dysfunction

Objective To investigate the expression of cardiac troponin-Ⅰ (cTnⅠ) levels in patients diagnosed with sepsis-associated myocardial dysfunction and explore the relationship between cTn Ⅰ and cardiac systolic and diastolic function.Additionally,we evaluated the prognostic value of cTn Ⅰ as a valuable biomarker.Methods We admitted 65 patients with sepsis.Using echocardiography,the ratio of early diastolic mitral inflow velocity to early diastolic mitral annulus velocity (E/e') and the left ventricular ejection fraction (LVEF) were measured as an evaluation index of left ventricular diastolic and systolic function,respectively.Patients were divided into a cardiac dysfunction and a normal cardiac function group.The cTn Ⅰ level was measured and compared between the two groups,and we determined the correlation between cTn Ⅰ levels and cardiac diastolic and systolic function.Based on assessment of 28-day mortality,cases were divided into a survivor and a death group.A receiver operating characteristic curve was constructed to predict the prognostic value of cTn Ⅰ.Results The cTn Ⅰ level in the cardiac dysfunction group was significantly higher than that observed in the normal cardiac function group (P < 0.05) and showed a positive correlation with E/e'(r =0.421,P =0.008).However,there was no correlation noted between the cTn Ⅰ level and LVEF (P > 0.05).Compared to the survivor group,the level of cTn]Ⅰ was significantly higher in the death group (P < 0.05).The prognostic value of cTn Ⅰ area under the curve was 0.892,with a cut-off value of 0.82 ng/mL (sensitivity =88.0% and specificity =82.5%).Conclusion The cTn Ⅰ level is noted to be significantly elevated in patients with sepsis-associated cardiac dysfunction and shows a positive correlation with left ventricular diastolic function.A cTn Ⅰ level ≥ 0.82 ng/mL can be used as a valuable predictor of mortality in patients with sepsis.
