5.THE CLINICAL SIGNIFICANCE OF IgA ANTI-HBc IN VIRAL HEPATITIS B
Medical Journal of Chinese People's Liberation Army 1983;0(05):-
We have developed an antibody-captured sandwich-type enzyme linked immunosorbant assay(ELISA) with monoclonal anti-IgA/a (human) and monoclonal anti-HBc, and determined immunogloblin A against hepatitis B core antigendgA anti-HBc) in sera of 179 cases of various types of HBV infection. IgAanti-HBc in the patients with acute hepatitis B (14 cases) and severe hepatitis (7 cases) was all positive, while in the patients with chronic active hepatitis, cirrhosis, chronic persistent hepatitis, and symptomatic carriers the positive rates were 94% (47/50 cases), 83.3% (10/12 cases), 56.5%(26/46 cases) and 4%(2/50 cases), respectively. Our observations suggested that IgA anti-HBc may reflect hepatic damage in the patients with HBV infection. Its detection may be valuable in the diagnosis and prognosis HBV infection. In addition, the activity of IgA anti-HBc in chronic hepatitis B is related to HBeAg.
6.Effect of molecular chaperone on prokaryotic expression level and activity of tetanus toxin fragment C
Chinese Journal of Biologicals 2025;38(03):272-278
Objective To investigate the effect of molecular chaperone on the prokaryotic expression level and activity of tetanus toxin fragment C(TTC) in order to provide new ideas for the development of new tetanus vaccines. Methods The gene sequence of TTC was optimized for E.coli adaptive codons, and the recombinant plasmid pET30a-HC was constructed, which was identified by double enzyme digestion. The recombinant plasmid pET30a-HC and molecular chaperone pGro7were co-transformed into competent E.coli BL21(DE3) and induced by IPTG to express TTC.The obtained TTC was identified by 12% SDS-PAGE and Western blot, and purified by Ni FF metal chelate affinity chromatography column.Female Kunming mice were immunized with TTC,TTC+Al,TTC+CpG,PBS,and tetanus toxoid(TT) by subcutaneous injection,10 mice in each group. After immunization, blood samples were collected by tail cutting or eyeball picking, and the serum was separated. The antibody titer, antibody typing,IL-4 and IFNγ levels were detected by ELISA.The lung and liver of mice were harvested aseptically, and the safety of TTC was evaluated by histopathological method. Results The recombinant plasmid pET30a-HC was constructed correctly as identified by double enzyme digestion. After adding molecular chaperone pGro7,the soluble expression of the target protein accounted for 40% of the total supernatant expression. After 42 days of the initial immunization, the specific IgG antibody titer in serum of TTC+CpG group was significantly higher than that of PBS,TTC,and TT groups(t=4.753-8.624,each P <0.01),which was also higher than that of TTC+Al group, but there was no significant difference(t=0.966,P> 0.05).There was no significant difference in serum IgG1,IgG2a,IgG2b,IgG3,and IgGM antibody levels among the groups(t=0.163-0.203,P> 0.05),IgG1/IgG2a> 1,indicating that the immune pathway was biased towards Th2 type.Compared with TTC group, the serum IL-4 and IFNγ levels in TTC+Al group had no significant difference(t=2.666 and 2.470,respectively,each P> 0.05),while significantly increased in TTC+CpG group(t=18.080 and8.440,P <0.001 and <0.05,respectively).Compared with TT group, the serum IL-4 level in TTC+CpG group increased significantly(t=8.561,P <0.01),but there was no significant difference in IFNγ level(t=0.472,P> 0.05).No obvious pathological changes were found in lung and liver of mice in each group. Conclusion Molecular chaperone can improve the soluble expression level of TTC with good safety, and combined with CpG adjuvant,it can induce strong immune response.
7.Ethical reflections on Fu biao's twice liver-transplantations:the distributive justice of scarce health-care resouces
Chinese Medical Ethics 1995;0(03):-
Fu biao who got an advanced liver cancer had undergone liver-transplantation twice and died at last, which raises great ethical concern. With respect to distribution, there are four substantial distributive principles, including the principle of equality, the principle of need, the principle of contribution, the principle of effort. When allocating scarce health-care resouses, medical criteirion should be considered firstly the equality of everyone's life.Furthermore, the principle of utility and equality should be embodied for ensuring distributive justice.
8.Effects of ziprasidone on serum cholinesterase concentration in schizophrenia patients
Journal of Pharmaceutical Practice 2020;38(1):88-90
Objective To explore the effects of ziprasidone and clozapine on serum cholinesterase (CHE) concentration in schizophrenia patients. Methods 126 schizophrenia patients were randomly divided into the observation group and the control group. The observation group was treated with ziprasidone. The control group received clozapine. The serum CHE concentration was assayed at 7, 14 and 28 days after treatment. Results ① The PANSS scores in both groups were significantly reduced after treatment, while the control group was lower than the observation group (PANSS score for positive symptoms: 15.3±4.7 vs 22.4±4.8, P<0.01; negative symptoms: 14.6±4.5 vs 21.8±5.2, P<0.01; general psychiatric symptoms: 13.3±3.4 vs 19.2±3.9, P<0.01). ② CHE levels were decreased in both groups after treatment. The control group exhibited greater decrease than the observation group. ③ The observation group had lower adverse reaction rate (34.9%) compared to control group (71.4%). Conclusion ziprasidone has weaker inhibitory effect on CHE activity compared to clozapine. This may explain that ziprasidone has lower therapeutic efficacy and better safety profile.
9.Effect of Acupuncture at Points Taiyang and Fengchi on Heat Shock Protein 70 and Apoptosis during Cerebral Ischemia and Reperfusion in Rabbits
Shanghai Journal of Acupuncture and Moxibustion 2015;(11):1122-1125
Objective To investigate the effect of acupuncture at points Taiyang and Fengchi on the expression of heat shock protein 70 (HSP70) and neural cell apoptosis during local cerebral ischemia and reperfusion in rabbits and explore its protective action on nerve cells and the mechanism. Methods Rabbits were randomized into sham operation, model and acupuncture groups. Each group was randomly allocated to three time points: one, three and seven days. A rabbit model of local cerebral ischemia and reperfusion was made by middle cerebral artery occlusion (MCAO)using an intraluminal thread. Reperfusion was performed at two hours after cerebral ischemia. In the acupuncture groups, points Taiyang and Fengchi were electro-acupunctured at six hours after cerebral ischemia. Treatment was given once daily. The rabbits were sacrificed to take materials at one, three and seven days after reperfusion. Pathological changes in brain tissue were observed by HE staining. Cerebral HSP70 expression was determined by immunohistochemical method. Neural cell apoptosis was examined by TdT-mediated dUTP nick end labeling (TUNEL).Results Pathological staining showed that cerebral infarction area decreased significantly, the number of nerve cells increased and inflammatory infiltration reduced at every time point in the acupuncture group of rabbits compared with the model group. The results of histochemical examination showed that HSP70 expression decreased in the sham operation group and increased in the model group. It increased significantly at every corresponding time point in the acupuncture group compared with the model group (P<0.01). The number of apoptotic cells increased gradually with reperfusion prolongation and reached the peak at three hours in the model group. The number of apoptotic cells decreased significantly at every time point in the acupuncture group and there was a statistically significant difference by comparison with the model group (P<0.01).Conclusions Electroacupuncture at points Taiyang and Fengchi can increase cerebral HSP70 expression and decrease neural cell apoptosis to produce a nerve-protecting effect in rabbits with cerebral ischemia-reperfusion injury.
