Objective To investigate the application value of risk management in skin care of hospitalized neurosurgery patients. Methods 179 cases of hospitalized neurosurgical patients from January to December 2011 were chosen as research object.All the patients were divided into the observation group (91 cases,the risk management care group)and the control group (88 cases,the routine care group).The two groups accepted different nursing measures.The skin damage caused by medical factors,family factors and environmental factors for the two groups were taken for statistics and comparison. Results The patients with skin damage due to medical factors,family factors and environmental factors in the observation group were significantly lower than the control group,there was significant difference. Conclusions The application of risk management methods in skin nursing of hospitalized neurosurgery patients has good effect.It can reduce the rate of skin damage of patients significantly and is recommended widely used in hospitalized neurosurgery patients.

Objective To investigate the therapeutic effect of extracorporeal shock wave therapy on Osgood-Schlatter disease. Methods Sixty Osgood-Schlatter disease patients were evenly randomly divided into therapeutic group 30 cases and control group 30 cases. The therapeutic group was treated with extracorporeal shock wave therapy. The control group was treated with high frequency electro therapy and rest on knee joint Results The clinical symptom and pain with visual analogous score the tibial tubercle of therapeutic group and control group were obviously decreased after treatment compared with before treatment (P< 0.05), they were (2.2 ± 1.3) scores vs (7.9 ± 2.4) scores, (4.8 ± 2.6) scores vs (8.2 ± 2.5) scores,the therapeutic group in pain resume significantly better than the control group (P<0.05 ). The cured rate of the therapeutic group was 83.33% (25/30), the control group was 63.33% (19/30), there was significant difference between the two groups (P <0.05). Conclusion The rehabilitation therapeutic effect of extracorporeal shock wave therapy on the Osgood-Schlatter disease is remarkable.

Objective To define laboratory technology for expression and purification of nucleocapsid proteins of human coronaviruses SARS-CoV,HCoV-229E,HCoV-OC43,HCoV-NL63 and HCoV-HKU1,animal coronaviruses bovine coronavirus,murine hepatitis virus,feline infectious peritonitis virus,and infectious bronchitis virus.MethodsThe pET-30a-based recombinant plasmids containing full length nucleocapsid(N) protein of coronaviruses were transformed into E.coil BL21(DE3) competent cells and were induced to express N proteins by IPTG.The expression products were purified by ion exchange chromatography and Ni2+ affinity chromatography,and were verified by SDS-PAGE and Western blot.Results Nine coronavirus nucleocapsid proteins with correct Mr were solubly expressed and highly purified with purity over 90%.Conclusion Successfully expressed nine recombinant nucleocapsid proteins with high purity in E.Coli,which provides materials to study the function of these coronavirus N proteins.

Seven-year-program medical education is an important model to train high-level medical person.Through the change of the teaching concept in medicine cytobiology experiment,re-form of the teaching content,and the use of a variety of effective teaching methods,students’com-prehensive quality,innovative thinking and practical ability is improved,which can meet the needs of modern medical education.

By participating in the research practice of basic medicine,the innovative consciousness,the scientific thinking and researching ability of seven-year program medical students can be improved. Meanwhile,the generalization of Open Experimental Teaching and further development of self-design experiment also improve the innovation and research ability of medical students,which can serve as a new approach to the quality education of higher medical talents.

By computer research, the literature of clinical and experimental studies on moxibustion therapy for chronic gastritis in the recent fifteen years was collected, sorted and analyzed, in order to understand the general situation about the clinical and experimental studies on moxibustion therapy for chronic gastritis, summarize the regularity of moxibustion therapy for chronic gastritis, so as to guide the treatment of chronic gastritis.

AIM: To investigate the effect of sitagliptin on the autopaghy and the expression of extracellular matrix in mesangial cells induced by advanced glycation end products (AGEs).METHODS: The cells were divided into 5 groups: control group, AGE group, and sitagliptin (5, 10 and 20 μmol/L) groups.After 48 h, the cell viability was measured by MTT assay, and the content of collagen (Col) Ⅳ in the supernatant of the cell culture was detected by ELISA.The protein levels of beclin-1, adenosine monophosphate-activated protein kinase (AMPK), p-AMPK, p70S6K and p-p70S6K were determined by Western blot.RESULTS: Compared with control group, the viability and the expression of Col IV induced by AGEs in the cultured mesangial cells were significantly increased (P<0.01).Sitagliptin decreased the viability and the expression of Col IV induced by AGEs in the mesangial cells in a dose-dependent manner.AGEs significantly inhibited the protein levels of beclin-1 and p-AMPK, but significantly increased the protein level of p-p70S6K.Compared with AGE group, sitagliptin significantly reversed the above results in a dose-dependent manner.CONCLUSION: Autophagy may mediate the protective effect of sitagliptin on mesangial cells induced by AGEs.

267 newly-diagnosed patients with type 2 diabetes were divided into normoalbuminuria group [group N-UAlb, urinary albumin to creatinine ratio (UACR)<30 mg/g, n=103], microalbuminuria group(group M-UAlb, UACR 30~300 mg/g, n=88 ) , and macroalbuminuria group ( group L-UAlb, UACR>300 mg/g, n=76). The control group(group NC) consisted of 114 healthy individuals. Serum betatrophin, adiponectin(APN), and interleukin-1β( IL-1β) levels were determined with ELISA methods and the parameters of body mass index (BMI), estimated glomerular filtration rate(eGFR), HbA1C, fasting plasma glucose(FPG), OGTT 2h plasma glucose(2hPG), fasting insulin(FINS), OGTT 2h postprandial insulin(2hPINS), fasting C-peptide(FCP), homeostasis model assessment insulin resistant index(HOMA-IR), and blood lipid were collected. Compared with group NC, the serum betatrophin levels in patients with type 2 diabetes were obviously increased. In patients with type 2 diabetes, betatrophin levels increased along with the increase of UACR and there were significant differences in betatrophin among the three groups(P<0. 01). Betatrophin positively correlated with UACR, HbA1C, FPG, 2hPG, FINS, 2hPINS, HOMA-IR, TC, LDL-C, and TG( r were 0. 785, 0. 225, 0. 136, 0. 241, 0. 386, 0. 223, 0. 411, 0.216,0.193,and0.298,allP<0.05),and betatrophin were also positively correlated with APN and IL-1β(rwere 0. 643 and 0. 710, both P<0. 01). Stepwise multiple regression analysis showed that UACR, HbA1C, FINS, and TG were independent relevant factors affecting betatrophin levels.

ObjcetiveTo study the chimerism status ot bone marrow mesenchymal stem cells (BMSC) following double-unit cord blood stem cell transplantation (DCBT).MethodsBone marrow mononuclear cells were isolated by ficoll density gradient centrifugation from the patient following DCBT and healthy donors.After cultured for about 20 days,BMSC surface makers,the ability to differentiate into adipocytes and osteoblast and the expression of hematopoietic and immune molecules were detected,moreover,chimerism status of BMSC were also evaluated by STR-PCR.ResultsMSC derived from the patient were similar to normal adult BMSC in morphology,surface and differentiation ability.Besides,MSC from the patient can express a variety of hematopoietic and immune molecules normally.The results of STR-PCR showed that the patient demonstrated complete donor chimerism,hematopoiesis in BM and PB were from a single unit cord blood (donor chimerism was 96.4％ and 95.7％,respectively).But the predominant cord blood chimerism of BMSC was 5.4％,the remaining part was from the patient himself.ConclusionSustained hematopoiesis was derived from a single unit cord blood following DCBT.The majority of BMSC were origin from host tissue,mixed chimerism donor part of MSC stemed from the engrafted unit of cord blood.

Objective Sphingolipidoses are a group of rare genetic disorders caused by catabolism defect of sphingolipids by lysosomal hydrolases with diverse presentations,and represent an important health problem to almost all ethnic populations. To date,there is no epidemiologic study on the prevalence of sphinglipidoses,individually,or as a group,in China. We set up a series of enzymatic assays that could make definite diagnoses with the aim to collect data for an epidemiologic investigation of sphingolipidoses and also pave the way to prenatal diagnosis to decrease the rate of inborn error of metabolism. Methods Patients with suspected sphingolipidosis were recruited from pediatric endocrinology and inherited metabolism outpatient clinics of Xinhua Hospital. Leukocytes were isolated with dextran from peripheral bloods. Activities of leukocyte acid β-glucosidase,acid sphingomyelinase,arylsulphatase A,galacto-cerebrosidase,beta-galactosidase were measured using their specific artificial fluorescent substrates,while arylsulfatase A was determined by a colorimetric assay with dipotassium 2-hydroxy-5-nitrophenyl sulfate as the substrate. Results In one year,we identified 17 patients with 5 different kinds of sphingolipidoses,including 3 patients with Gaucher disease,9 patients with Niemann-Pick type A/B,2 patients with metachromatic leukodystrophy,2 patients with Krabbe disease,and 1 patient with GM1 gangliosidosis. We made brief descriptions of disease characters of each different kind disease and compared our results with findings of other ethnic groups. Conclusions Sphinglipidoses was markedly under-diagnosed in China and general pediatricians should be alerted to sphinglipidoses.
