1.Application of risk management in skin care of hospitalized neurosurgery patients

Objective To investigate the application value of risk management in skin care of hospitalized neurosurgery patients. Methods 179 cases of hospitalized neurosurgical patients from January to December 2011 were chosen as research object.All the patients were divided into the observation group (91 cases,the risk management care group)and the control group (88 cases,the routine care group).The two groups accepted different nursing measures.The skin damage caused by medical factors,family factors and environmental factors for the two groups were taken for statistics and comparison. Results The patients with skin damage due to medical factors,family factors and environmental factors in the observation group were significantly lower than the control group,there was significant difference. Conclusions The application of risk management methods in skin nursing of hospitalized neurosurgery patients has good effect.It can reduce the rate of skin damage of patients significantly and is recommended widely used in hospitalized neurosurgery patients.
2.Therapeutic effect of extracorporeal shock wave therapy on Osgood-Schlatter disease

Objective To investigate the therapeutic effect of extracorporeal shock wave therapy on Osgood-Schlatter disease. Methods Sixty Osgood-Schlatter disease patients were evenly randomly divided into therapeutic group 30 cases and control group 30 cases. The therapeutic group was treated with extracorporeal shock wave therapy. The control group was treated with high frequency electro therapy and rest on knee joint Results The clinical symptom and pain with visual analogous score the tibial tubercle of therapeutic group and control group were obviously decreased after treatment compared with before treatment (P< 0.05), they were (2.2 ± 1.3) scores vs (7.9 ± 2.4) scores, (4.8 ± 2.6) scores vs (8.2 ± 2.5) scores,the therapeutic group in pain resume significantly better than the control group (P<0.05 ). The cured rate of the therapeutic group was 83.33% (25/30), the control group was 63.33% (19/30), there was significant difference between the two groups (P <0.05). Conclusion The rehabilitation therapeutic effect of extracorporeal shock wave therapy on the Osgood-Schlatter disease is remarkable.

6.Expression and purification of nine nucleocapsid proteins from coronaviruses

Objective To define laboratory technology for expression and purification of nucleocapsid proteins of human coronaviruses SARS-CoV,HCoV-229E,HCoV-OC43,HCoV-NL63 and HCoV-HKU1,animal coronaviruses bovine coronavirus,murine hepatitis virus,feline infectious peritonitis virus,and infectious bronchitis virus.MethodsThe pET-30a-based recombinant plasmids containing full length nucleocapsid(N) protein of coronaviruses were transformed into E.coil BL21(DE3) competent cells and were induced to express N proteins by IPTG.The expression products were purified by ion exchange chromatography and Ni2+ affinity chromatography,and were verified by SDS-PAGE and Western blot.Results Nine coronavirus nucleocapsid proteins with correct Mr were solubly expressed and highly purified with purity over 90%.Conclusion Successfully expressed nine recombinant nucleocapsid proteins with high purity in E.Coli,which provides materials to study the function of these coronavirus N proteins.
7.Effect of sitagliptin on autopaghy in mesangial cells induced by AGEs

AIM: To investigate the effect of sitagliptin on the autopaghy and the expression of extracellular matrix in mesangial cells induced by advanced glycation end products (AGEs).METHODS: The cells were divided into 5 groups: control group, AGE group, and sitagliptin (5, 10 and 20 μmol/L) groups.After 48 h, the cell viability was measured by MTT assay, and the content of collagen (Col) Ⅳ in the supernatant of the cell culture was detected by ELISA.The protein levels of beclin-1, adenosine monophosphate-activated protein kinase (AMPK), p-AMPK, p70S6K and p-p70S6K were determined by Western blot.RESULTS: Compared with control group, the viability and the expression of Col IV induced by AGEs in the cultured mesangial cells were significantly increased (P<0.01).Sitagliptin decreased the viability and the expression of Col IV induced by AGEs in the mesangial cells in a dose-dependent manner.AGEs significantly inhibited the protein levels of beclin-1 and p-AMPK, but significantly increased the protein level of p-p70S6K.Compared with AGE group, sitagliptin significantly reversed the above results in a dose-dependent manner.CONCLUSION: Autophagy may mediate the protective effect of sitagliptin on mesangial cells induced by AGEs.
8.Expression of ARK5 in hepatocellular carcinoma tissue and its effect on growth of SMMC-7721 cells

Objective To detect the expression of ARK5 in hepatocellular carcinoma (HCC)tissue and hepatoma SMMC-7721 cells,and to investigate its effect on the growth of hepatoma cells.Methods The expression levels of ARK5 mRNA and protein were determined by RT-PCR and Western blotting in 30 cases of HCC tissue, paracarcinoma tissue,SMMC-7721 cells,and hepatic cells LO2.The SiRNA of ARK5 and negative control (NC) siRNA were constructed and transfected into the SMMC-7721 cells,and used as experimental group and negative control group;at the same time blank control group was set up. The proliferation activity and apoptotic rate of transfected cells were detected by MTT assay and flow cytometry (FCM).Results The PCR and Western blotting results showed that the expression levels of ARK5 mRNA and protein in HCC tissue and SMMC-7721 cells were significantly higher than those in paracarcinoma tissue and LO2 cells (P<0.05 ). The MTT assay results demonstrated that the inhibitory rates of growth of transfected cells in experimental group at 24,48 and 72 h were (19.39±5.42)%, (23.19±0.53)%,and (20.74±1.23)%;there were significant differences compared with blank control group and negative control group (P<0.01).The FCM results indicated that the apoptotic rate of the transfected cells in experimental group was (15.017±0.945)%,there were significant differences compared with blank control group (8.770%± 0.656 )% and negative control group (8.763%± 1.201%) (P<0.05 ). Conclusion The ARK5 expression level is significantly increased in HCC tissue and hepatoma SMMC-7721 cells;the inhibition of ARK5 expression could suppress the growth of hepatoma cells and induce apoptosis. So ARK5 maybe act as a cancer-promoting gene and induce hepatocellular carcinogenesis.
9.Current advance in the treatment of viral infections by cytotoxic T-lymphocytes after allogeneic hematopoiefic stem cell transplantation

Viral infections remain the common cause of morbidity and mortality in immunocompromised patients receiving allogeneic hematopoietic stem cell transplantation (allo-HSCT).Virus antigens can be recognized by cytotoxic T-lymphocytes (CTL) in the context of HLA class Ⅰ molecules.T-cell-based immunotherapies are now being included in the clinical practice of transplant recipients to prevent and treat viral infections and complications associated with CMV,AdV and EBV.Here,the current status and future feasibility of CTL immunotherapy for virus infections after allo-HSCT are discussed.
10.Genotyping of human papillomavirus among human immunodeficiency virus-positive populations

Objective To compare the prevalence and genotype distribution of human papillomavirus (HPV) between human immunodeficiency virus (HIV)-positive and-negative populations.Methods Patients with condyloma acuminatum or persons who recently had sexual contact with patients with condyloma acuminatum were enrolled into this study,and classified into HIV-positive group (n =62) and HIV-negative group (n =2 716).GeneChip analysis was performed to detect HPV and determine HPV genotypes in mucocutaneous samples collected from the external genitalia of these subjects.Statistical analysis was carried out by chi-square test using the SPSS software version 19.0.Results The prevalence rates of HPV infection,high-risk HPV types and low-risk HPV types were significantly higher in HIV-positive persons than in HIV-negative persons (74.19% (46/62) vs.42.30% (1 149/2 716),67.74% (42/62) vs.29.57% (803/2 716),58.06% (36/62) vs.24.71% (671/2 716),respectively,all P< 0.01).The detection rate of HPV was also increased in HIV-positive men compared with HIV-negative men (92.11% (35/38) vs.37.38% (382/1 022),x2 =45.98,P < 0.01).Although the top three genotypes of low-risk HPV were types 6,43 and 11 in both HIV-positive and-negative groups,the prevalence rate of HPV 6 was 37.10% (23/62) in HIV-positive group,but only 11.12% (302/2 716) in HIV-negative group.The top five genotypes of high-risk HPV were types 16 (22.58%,14/62),52,66,58 and 18 in HIV-positive group,and types 16 (7.77%,211/2716),58,56,66 and 52 in HIV-negative group.Coinfections with multiple HPV subtypes were common in both groups,and the number of concurrent HPV genotypes was as high as 8 in HIV-positive group,and 9 in HIV-negative group.The prevalence rate of coinfections with three or more HPV genotypes in HIV-positive group was significantly higher than that in HIV-negative group (65.21% (30/46) vs.16.71% (192/1 149)).Conclusions Compared with HIV-negative populations,HIV-positive populations show elevated prevalence of HPV infection,high-risk HPV genotypes and low-risk HPV genotypes.Moreover,the prevalence of HPV is higher in HIV-positive men than in HIV-negative men.These findings are worthy of further attention in clinic.
