1.Effect of Ambient Air SO_2 on Mortality of Respiratory Diseases in Shenyang

Objective To assess the effect of ambient air SO2 on mortality of respiratory diseases. Methods Based on the ecological theory, after the adjustment of seasons, temperature, humidity and air pressure, the observed individuals were divided into eight groups. The association between SO2 exposure and mortality rate of respiratory diseases was analyzed using Poisson regression and local auto-regression method. Results SO2 concentration increased by 0.05 mg/m3, the mortality of respiratory diseases increased by 5.90% (95%CI: 2.26%-9.68%) in all groups, in children by 10.23% (95%CI: 2.07%-19.04%), in young-middle-male group by 1.10% (95%CI: -9.51%-12.96%), in young female group by 9.58% (95%CI: -2.57%-23.23%), in young group by 10.23% (95%CI: 2.07%-19.04%), in aged male group by 5.26%(95%CI: 0.01%-10.78%), in aged female group by 8.07% (95%CI: 3.45%-12.89%) and in aged group by 4.66% (95%CI: 1.44%-7.99%) respectively. Conclusion SO2 exposure is found to be associated with the increase of mortality of respiratory diseases, especially in children, aged people and elderly females.
2.Effect of preoperative aprotinin on blood coagulation in patients undergoing meningioma resection

Objective To evaluate the effects of different low doses of aprotinin given before operation on blood coagulation in patients undergoing meningioma resection. Methods Sixty ASA Ⅰ or Ⅱ patients aged 18-60 yrs were randomized to receive intravenous aprotinin 0.5 or 1.0 or 2.0?106 KIU (group Apro 1, 2, 3) or normal saline (control group) before operation. 6% hydroxyethyl starch (HES) 1 000 ml was infused at 25 ml?min-1 after induction of general anesthesia. Venous blood samples were taken before aprotinin was given ( T1 , baseline), before operation after aprotinin was given (T2), while the tumor was being resected (T3), at the end of surgery (T4) and 24h after operation (T5) for routine blood and coagulation tests including Hb, Hct, RBC, WBC and PLT counts, activated partial thromboplastin time (APTT), prothrombin time (PT) fibrinogen concentration (FIB) and thrombin time (TT) and tests using Sonoclot coagulation and platelet function analyzer including Sonoclot activated clotting time (Son ACT), rate of clot formation (CR) and platelet function (PF). Results The 4 groups were comparable with respect to demographic data, duration of operation, uine output, amount of fluid infused and blood loss during operation and volume of the tumor. RBC count, Hb, Hct and PLT count were significantly decreased during and after operation (T2-5) while WBC counts were significantly increased at T5 as compared with the baseline values at T1 in the 4 groups. PT was significantly prolonged and FIB and CR significantly decreased in the 4 groups; APTT was significantly prolonged in group Apro 2 and 3; PF was significantly increased in group Apro 3 during operation at T2-4 as compared to the baseline values at T1 but all returned to normal level after operation at T5 .Conclusions Preoperative aprotinin 1.0 and 2.0?106 KIU can prolong APTT. Aprotinin 2.0?106 KIU provides effective protection of platelet function.
3.Study on design and three-dimension structure of a broad-spectrum inhibitory peptide against staphylococcal enterotoxins superantigen

Objective:Peptides were designed on the basis of high conservative regions of amino acid sequences and structures of the SEs,three-dimension structure of P72 was constructed.Methods: Bioinformatics analysis softwares such as Vector NTI 10.3, InsightII 2000,Discovery Studio 1.7 were used to analyse and predict the space structure of P72.Results:three-dimensional domains of the peptide P72 from SEA, SEB and SEC were quite similar, Peptide P72 was far away from TCRVβchain and MHC class II molecule.Conclusion:The inhibitory activity of peptide P72 may not due to binding to MHCⅡ and TCRVβchain.The exact mechanism of inhibitory activity of P72 should be explored.
4.Association Between Total Suspended Particles(TSP)and Cardiovascular Disease Mortality in Shenyang

Objective To estimate the adverse effects of total suspended particles exposure on cardiovascular disease mortality.Methods Cardiovascular disease mortality and TSP monitoring data from1996to2000in Shenyang were collected.The association between TSP concentration and the mortality of cardiovascular diseases was analyzed using Poisson regression model adjusted for season,temperature and humidity variation using a parametric method.Results It was found that each50?g /m 3 increment of TSP was associated with an OR of1.0122(95%CI =1.0036-1.0209)of cardiovascular disease mortality in the general population,an OR of1.0278(95%CI =1.0160-1.0398)in the male group,1.0406(95%CI =1.0294-1.0519)in the female group,1.0430(95%CI =1.0343-1.0519)in the general aged group,1.0577(95%CI =1.0452-1.0703)in the aged male group and1.0427(95%CI =1.0284-1.0572)in the aged female group.Conclusion The results showed that there was an association between TSP concentration and the cardiovascular disease mortality in Shenyang.It was considered that TSP might be one of the risk factors related to the increase of cardiovascular disease mortality.
5.La accumulation and microstructure change of leg bones of rats fed with La(NO_3)_3 in low dosage for a long term

Objective: To study La accumulation and microstructure change of leg bones of rats fed with La(NO 3) 3 in low dosage for a long term. Methods: After the rats were fed by La(NO 3) 3 in dosage of 2 mg?kg -1 ?d -1 for 6 months, the contents of La and Ca,P in the leg bones were determined by ICP MS and spectrophotometry; the microstructure changes of the leg bones were investigated by electron microscopy and X ray powder diffraction. Results: In the leg bones of tested rats, the contents of La and P increased greatly, and those of Ca did not change obviously, so that Ca/P ratio values decreased in comparison with the control group. Conclusion: La was accumulatied in the rat leg bones and the change of bone microstructure induced after the rats were fed with La(NO 3) 3 in low dosage for a long term.
6.The relationship between serum Sirt1 and EGR1 and other inflammatory cytokines in type 2 diabetic patients with different stages of diabetic nephropathy

Objective To investigate changes of serum silent information regulator 1 (Sirt1) and inflammatory cytokines in type 2 diabetes patients with different stages of diabetic nephropathy.To explore the relationship between serum Sirt1 level and inflammatory cytokines in type 2 diabetic patients with different urinary albumin excretion rates.Methods A total of 436 cases with type 2 diabetes were divided into three groups:normoalbuminuric [D1,n =168],microalbuminuric [D2;n =152],and macroalbuminuric [D3,n =116].Serum Sirt1,hypoxia-inducible factor-1α (HIF-1α),early growth response protein 1 (EGR1),insulin-like growth factors-Ⅰ (IGF-Ⅰ),and monocyte chemotactic protein-1 (MCP-1) were determined by enzyme-linked immunosorbent assay (ELISA).Results The levels of serum Sirt1 in type 2 diabetes patients were significantly lower than that in control group,and with the increase of urinary protein excretion rate,the levels of serum Sirt1 in group D1,D2 and D3 were decreased gradually (P < 0.01).Compared to control,serum inflammatory cytokines (HIF-1α,EGR1,IGF-Ⅰ,and MCP-1) levels were significantly increased in type 2 diabetic patients and gradually increased in the patients of D1,D2 and D3 groups (P <0.01).Furthermore,Serum Sirt1 was negatively correlated with the levels of inflammatory cytokines.Age,duration,fasting blood glucose (FBG),fasting insulin (FINS),homeostasis model assessment insulin resistance index (HOMA-IR),glycosylated hemoglobin (HbA1c),low density lipoprotein (LDL),total cholesterol (TC),triglyceride (TG),serum creatinine (Scr),blood urea nitrogen (BUN),uric acid (UA),HIF-1α,EGR1,IGF-Ⅰ,and MCP-1 were positively correlated with Ln Koc of urinary albumin to creatinine ratio [Ln(ACR)] (P < 0.05);and Sirt1 were negatively correlated with Ln(ACR)(P < 0.01).HIF-1α,MCP-1,IGF-Ⅰ,duration,BUN,Sirt1,UA,LDL,and EGR1 were independent factors that significantly influenced Ln (ACR) (P < 0.05).Conclusions Serum Sirt1 might be a new target for the treatment of diabetic nephropathy.Enhancing serum Sirt1 levels might have a role in delaying the progression of diabetic nephropathy.
7.LONG-TERM EFFICACY AND IMMUNOLOGICAL MEMORY OF PLASMA-DERIVED HEPATITIS B VACCINE 11 YEARS AFTER INITIAL INOCULATION

ObjectiveTo assess the efficacy and the immunological memory of plasma-derived hepatitis B vac- cine 11 years after the initial inoculation. Methods A randomized, double-blind and placebo-controlled trial design was used. Results The immunogenicity and protection rate of the vaccine were good 1 1 years after vaccination, how- ever, from 9 to 11 years after vaccination, the perscn year HBV infection rate showed no noticeable difference be- tween the vaccine group and placebo-controls. Furthermore, the immunological memory remained 11 years after in- oculation, but was significantly lower than that observed in the past 10 years. Conclusion Vaccine protection wanes over the years. More information is needed to define the appropriate time for vaccine booster doses.
8.Effects of SO2 on Mortality of Cardiovascular Diseases in Shenyang

Objective To assess the advese effects of SO2 on mortality of cardiovascular disease quantitatively. MeThod Based on bionomical theory,after the adjustment of time distribution,temperature,humidity and air pressure,the observed individuals were grouped into total population group,female group,male group,aged group,aged female group and aged male group.The association between SO2 and mortality rate of cardiovascular diseases was analyzed by Poisson regression and local auto-regression method.Ressults A 50?g/m3 increase of SO2 concentration caused 5.33% (95%CI=4.17%~6.49%)increase of mortality rate of cardiovascular disease in total people group,6.96% (95%CI=5.34%~8.60%)in female group,6.43%(95%CI=4.86%~8.03%)in male group,8 14%(95%CI=6 86%~9 46%)in aged group,6 40%(95CI=4 49%~8 34%)in aged female group,9 55%(95%CI=7 74%~11 39%)in aged male gruop.COnclusion SO2 could cause the increase of mortality rate of cardiovascular disease especially ofr aged people with cardiovascular disease.
9.Study on clinical effect of glargine insulin combined with glimepiride in newly diagnosed T2DM patients

Objective To study the efficacy and safety of glargine insulin combined with glimepiride therapy in type 2 diabetes mellitus(T2DM).Methods According to the random number table,100 T2DM patients with unsatisfactory efficacy of oral hypoglycemic agents were randomly divided into treament group and control group, 50 cases in each group.Then,the treatment group was dealed with glargine insulin and glimepiride,the control group were treated with insulin glargine for our treatment protocol.12 weeks after treament,the fasting blood glucose(FPG), 2 hours blood glucose(2hPG),body mass index(BMI),glycosylated hemoglobin(HbA1c),islet β-cell function index(HOMA -β),insulin resistance index(Homa -IR),insulin dosage(INS),time requirds,weight gain and so on were observed.Meanwhile,The occurrence condition of hypoglycemia was also observed.Results In control groups, the post -treatment indexes of FPG,2hPG,HbAlc,HOMA -βand HOMA -IR[(5.7 ±0.8)mmol/L,(8.6 ± 2.5)mmol/L,(6.5 ±0.7)%,(40.35 ±3.12),(2.68 ±1.41 )]were much better than those in pre -treatment [(11.7 ±1.6)mmol/L,(15.1 ±6.1 )mmol/L,(9.2 ±1.1 )%,(19.01 ±2.79),(3.42 ±1.47)](t =23.717, 6.971,14.642,36.052,2.568,P <0.01 or 0.05 for both).In the treatment group,the post -treatment indexes of FPG,2hPG,HbAlc,HOMA -β,HOMA -IR[(5.6 ±0.7)mmol/L,(7.3 ±2.3)mmol/L,(6.3 ±1.0)%,(54.18 ± 3.23),(2.12 ±1.26 )]improved obviously than those in pre -treatment [(11.8 ±1.7 )mmol/L,(15.8 ± 5.2)mmol/L,(9.3 ±1.2)%,(18.71 ±3.12),(3.36 ±1.56)](t =23.846,10.570,13.580,55.849,4.372,P <0.01 for both).After treatment,the indexes of 2hPG,HOMA -β,HOMA -IR,insulin dose,time requirds and ampli-tude of weight gain[(7.3 ±2.3)mmol/L,(54.18 ±3.23),(2.12 ±1.26),(18.06 ±1.43)U /d,(10.1 ±2.4)d, (1.1 ±0.4kg)]etc in the treatment group were significantly ameliorated than those in the control group[(8.6 ± 2.5)mmol/L,(40.35 ±3.12),(2.68 ±1.41 ),(22.46 ±1.77)U /d,(13.2 ±2.6)d,(2.1 ±0.6)kg](t =23.717,6.971,14.642,36.052,2.568,P <0.01 or 0.05 for both).Hypoglycemia events of treatment group were 4 cases(8.0%)and the same in the control group were 13 cases(26.0%),there were statistical significance between the two groups(χ2 =5.535,P <0.05).Conclusion Treatment of glargine insulin combined with glimepiride can improve blood glucose level of T2DM patients,recover their islet β-cell function,relieve the IR,shorten the time requirds,as well as reduce the insulin dosage,range of weight gain and rate of hypoglycemia events.Hence,treatment protocol of glargine insulin combined with glimepiride is an effective clinical treatment for newly diagnosed T2DMpatients.
10.Expression and purification of nine nucleocapsid proteins from coronaviruses

Objective To define laboratory technology for expression and purification of nucleocapsid proteins of human coronaviruses SARS-CoV,HCoV-229E,HCoV-OC43,HCoV-NL63 and HCoV-HKU1,animal coronaviruses bovine coronavirus,murine hepatitis virus,feline infectious peritonitis virus,and infectious bronchitis virus.MethodsThe pET-30a-based recombinant plasmids containing full length nucleocapsid(N) protein of coronaviruses were transformed into E.coil BL21(DE3) competent cells and were induced to express N proteins by IPTG.The expression products were purified by ion exchange chromatography and Ni2+ affinity chromatography,and were verified by SDS-PAGE and Western blot.Results Nine coronavirus nucleocapsid proteins with correct Mr were solubly expressed and highly purified with purity over 90%.Conclusion Successfully expressed nine recombinant nucleocapsid proteins with high purity in E.Coli,which provides materials to study the function of these coronavirus N proteins.
