1.Amperometric Immunosensor Based on Layer-by-layer Assembly of Thiourea and Nano-gold Particles on Gold Electrode for Determination of Naphthalene

A new type of amperometric immunosensor for the determination of naphthalene based on self-assemble) layer-by-layer technique was reported. The preparation procedure of such a sensor was characterized and the operation conditions, such as the pH value of supporting solution, incubated temperature and time were studied by cyclic voltammetry. Under the optimized working conditions, the current response of the immunosensor) was proportional to the concentration of naphthalene in the range of 0.5-100 μg/L with a detection) limit of 0.08 μg/L(r=0.9986). This method was applied to the naphthalene detection in river) water) samples and the recovery was ranging in 94.3%-107.0%. The reported immunosensor exhibited high sensitivity and long-term stability for naphthalene detection, and was easy to be prepared and regenerated.
2.An algorithm for separating overlapped protein spots based on valley characteristics.

To separate the overlapped protein spots in two-dimensional gel electrophoresis (2-DE) images, we proposed an auto-separating algorithm based on valley characteristics. Firstly, the marker-controlled watershed algorithm was used to detect the initial outlines of the object regions. Secondly, medial axis transform and hierarchical branch pruning method were applied to the main skeletons of the object regions, and each main skeleton was fitted into line segments to describe the overlap directions. Then, the 3-dimensional model of the object region was scanned on the normal planes of the line segments to find the valley locations. And finally, a validation model was adopted to construct separation lines. The experiments on 2 real scanned 2-DE images showed that the true overlap separate (TOSs) were 78.95% and 85.71%, respectively. The results indicated that the proposed algorithm was better than the existing algorithms and could be used in engineering practice.

3.Studies on clinical features and causal analysis of multiple organs dysfunction syndrome caused by an extremely severe bone marrow form of acute radiation sickness

Objective To sum up the therapeutic experiences and to analyze the causes of multiple organ dysfunction syndrome (MODS) occurred in a patient who subjected to an accidental 60 Co exposure. Methods The patient was diagnosed as MODS by clinical manifestation, the auxiliary examinations and pathologic autopsy. In therapeutic aspects, intensive care, ventilator-assisted breathing, anti-infection, nutritional support treatment and protection management of important organs were given. Results After a successful transplantation of HLA-identical allogeneic peripheral blood stem cell, the complaints of MODS emerged repeatedly as fever, lung infection, respiratory failure, cardiac arrhythmia, circulatory failure, hepatic injury, decreased urine output and paralytic ileus. At last, the patient died of multi-organ failure 75 days after exposure to radiation. The results of auxiliary examinations showed enlargement of heart and cardiac arrhythmia. Biochemical examination also indicated the increased hyper-sensitive C-reactive protein. The results of pathologic autopsy indicated the existence of fungous infections in whole body, pulmonary fibrosis, myocardium degeneration and necrosis and radiation enteritis. In addition, congestive hepatopathy, renal hemorrhage and intracerebral hemorrhage were also found in the examination. Conclusion Some special clinical features were found in this case, and the cause of MODS is mainly related to radiation injury of important organs, low level of immunity and infection. So the measures of anti-infection and protective treatment of multiple organs should be taken. The key and difficulty for preventing such a kind of MODS might be to improve the patient's immunity and to help the tissue reparation after radiation injury.

6.CdTe Nanoparticles Labeled with Anti-Fluorethene-Antibody and Fluorescent Immunoassay of Fluoranthene in Water Samples

The labeled compounds, CdTe was combined with anti-fluoranthene antibody, had good dispersion and stability with the fluorescence intensity enhancing. A direct competitive fluorescent immunoassay with CdTe-anti-fluoranthene antibody to detect fluoranthene in water sample in the environment was developed. The result showed that fluoranthene can be determined in the concentration range from 0.1 μg/L to 1000 μg/L with a correlation coefficient of 0.9983, a sensitivity of (IC_(50)) of 12.4 μg/L and a detection limit (IC_(20)) of 13.1 ng/L. Trace environmental pollutant in environmental water samples were successfully determined with a good accuracy and suitability. The recovery was between 95.1% and 111.0%, with relative standard deviation less than 9%.
7.Value of magnetic resonance imaging on local-regional staging of endometrial carcinoma

Objective To assess the value of magnetic resonance (MR) imaging (MRI) in depicting the depth of myometrial infiltration, cervical invasion and lymph nodes metastasis in patients with endometrial carcinoma compared with surgicopathologic findings. Methods Thirty-eight patients with endometrial carcinoma diagnosed by pathology were inspected by MRI in this prospective study. MR images were analyzed by two radiologists to report the depth of myometrial infiltration, infiltration of the uterine cervix and lymph nodes metastasis. MRI and surgicopathologic staging of endometrial carcinoma were based on FICO classification. MRI findings were compared with surgicopathologic findings. Results The overall accuracy rate of MRI in staging endometrial carcinoma was 77.1 %. The results of Kappa statistics showed that there was strong consistency between MRI and surgicopathology for staging of endometrial carcinoma (k= 0.677). Respective diagnostic accuracy, sensitivity, specificity, negative and positive predictive values in assessing myometrial infiltration(>l/2 depth) were 91.4 %, 83.3 %, 95.6 %, 91.7 % and 90.9 %; those in cervical infiltration were 94.2 %, 80.0 %, 96.7 %, 96.7 %, and 80.0 %; and those in lymph node assessment were 96.2 %, 66.7 %, 100 %, 95.8 % and 100 %. Conclusion MRI is highly accurate in depicting the depth of myometrial infiltration, cervical invasion, lymph nodes metastasis and local-regional staging of endometrial carcinoma.
8.Effect of flurbiprofen pretreatment on permeability of blood-brain barrier in a rat model of global cerbral ischemia-reperfusion injury

Objective To investigate the effects of flurbiprofen pretreatment on the permeability of bloodbrain barrier in a rat model of global cerbral ischemia-reperfusion (I/R) injury. Methods Forty-five male SD rats weighing 300-350 g were randomly divided into 3 groups (n = 15 each): sham operation group (group S); global cerebral I/R group (group I/R); flurbiprofen 10 mg/kg + global cerebral I/R group (group F). Global cerebral ischemia was induced by 20 min occlusion of bilateral common carotid arteries combined with hypotension (MAP maintained at 35-45 mm Hg). In group F, flurbiprofen 10 mg/kg was injected iv at 15 min before ischemia. Evans blue 3 ml/kg was injectcd iv at 24 h of reperfusion, then the rats were sacrificed and their brains were immediately removed for determination of the apoptosis rate, brain water content, Evans blue content, TNF-α, IL-1β and IL-10 content, and microscopic examination. Results The apoptosis rate, brain water content, Evans blue content, and TNF-α, IL-1β and IL-10 content were significantly higher in group I/R and F than in group S (P < 0.05 or 0.01).The apoptosis rate, brain water content, and Evans blue content and TNF-α and IL-1β content were significantly lower, while IL-10 content was higher in group F than in group I/R (P < 0.01). Global cerbral I/R-induced changes were significantly attenuated in group F. Conclusion Pretreatment with flurbiprofen can protect bloodbrain barrier against cerebral I/R injury by inhibition of the inflammatory reaction.
9.THE DOSAGE AND ROUTE OF ADMINISTRATION OF CEFTIZOXIME IN SEVERE PULMONARY INFECTION

12 cases of severe pulmonary infection were treated with ceftizoxime, one of cepha-losporins of the third generation. Clinical observation and pharmacokinetics were carried out. The result showed that intravenous infusion was the best route of drug administration. It had the advantages of high serum concentration and prolonged maintenance of serum level. 1 gram of ceftizoxi-me, two times a day, was enough for most pathogens except Pseudomonas aeruginosa.
10.Stent angioplasty for the treatment of symptomatic stenosis of middle cerebral artery

Objective To investigate the safety and feasibility of endovascular stent angioplasty in treating symptomatic stenosis of middle cerebral artery.Methods Endovascular angioplasty with coronary stents was performed in 27 patients with symptomatic stenosis of middle cerebral artery.The clinical results were reviewed and analyzed.Results Of the total 27 patients,successful placement of the coronary stents Was achieved in 24.Angiography immediately after the procedure showed that the stenotic degree of the diseased artery was markedly decreased from preoperative (80±19)%to postoperative (8±4)%,the improvement was very obvious.Percutaneous transcatheter angioplasty had to be employed in two cases because of the failure of stent placement.A mean follow-up period of 18 months was carried out.During the following up period no transient cerebral ischemia attack occurred in 25 patients and no newly-developed cerebral infarction in region fed by the responsible vessels occurred either.Re=irrigation cerebral hemorrhage was seen in one patient,which occurred three hours after the placement of the stent.In one case the placed stent fell off and immigrated into the siphon of internal carotid artery,and the displaced stent Was took out later with a catching apparatus.In another case re-stenosis occurred six months after the stenting.Conclusion Percutaneous endovaacular stent angioplasty is a safe and effective treatment for symptomatic stenosis of middle cerebral artery,although its long-term results need to be further evaluated.
