1.The Study of Nimodipine in Treating Pregnancy Induced Hypertension and its Effect on Intraplatelet Free Calcium

Objective To evaluate the effects of nimodipine (NIM) on pregnancy induced hypertension(PIH) and to assesse the effects of nimodipine on intraplatelet free calcium(PF[Ca 2+ ] i). Methods Fourty eight patients with moderate or severe PIH were randomized to receive either NIM or magnesium sulfate. We observed the change of blood pressure, urc protein, PF[Ca 2+ ] i and symptoms of preeclampsia before and after medication. We compared the pregnancy outcome of the two groups. Objective To observe the clinical efficacy ofXing Nao Kai Qiao (brain awakening and orifice opening) Needling plus mountain-burning fire manipulation in treating post-stroke limb numbness.Method Eighty patients with post-stroke limb numbness were randomized into a treatment group and a control group, 40 cases in each group. The treatment group was intervened byXing Nao Kai Qiao needling plus mountain-burning fire manipulation, while the control group was byXing Nao Kai Qiao needling alone. The sensory function and motor function (Fugl-Meyer Assessment, FMA), National Institute of Health Stroke Scale (NIHSS), and Barthel Index (BI) were observed before and after intervention.Result The component scores of FMA (motor function, sensory function, range of motion, and joint pain intensity), NIHSS scores, and BI scores were significantly changed respectively after 1 and 2 treatment courses in both groups (P<0.05). The component scores of FMA, NIHSS scores, and BI scores after 2 treatment course were significantly different from that after 1 treatment course in both groups (P<0.05). After 2 treatment courses, the sensory function score, joint pain intensity in the treatment group were significantly different from that in the control group (P<0.05).ConclusionXing Nao Kai Qiaoneedling plus mountain-burning fire manipulation is an effective method in treating post-stroke limb numbness.
3.Establishment of rabbit model for acute retinal necrosis and experimental study of infectious pathway in opposite eye

Objective To establish the rabbit model for acute retinal necrosis(ARN) and investigate the possible route of the virus migrates from one eye to the other. Design Experimental study. Participants 41 chinchilla rabbits. Methods HSV-1 COS strain was inoculated into subretina in 41 rabbits to establish ARN animal model. Ganciclovir was used to treat by intraocular and/or intravenous injection 1 day or 3 days after virus inoculation. PCR test of blood samples was performed for HSV detection. Histopathological examinations were also conducted in some eyeballs, optic nerves and brains of the rabbits. Main Outcome Measures Results of blood PCR and histopathology. Results 10 rabbits appeared bilateral retina necrosis (10 to 18 days after virus inoculation) and 15 rabbits appeared central nervous system damages. PCR samples of 5 rabbits obtained before and after the contralateral retina damages appeared (9 to 19 days after virus inoculation) had all negative results. Histopathological damages were found in bilateral eyeballs, optic nerves and brains. Conclusion The ARN model can be established by injection of HSV-1 into subretina of rabbits. Blood samples PCR don't support the hypothesis of virus transmission through circulation system in bilateral ARN cases. Trans-optic chiasm transmission may be a possible route.
4.Therapeutic effects of ganciclovir with different injection methods on experimental retinal necrosis

Objective To observe the therapeutic effects of ganciclovir(GCV)with differentInjection methods on experimental acute retinal necrosis(ARN).Methods The right eyes of 41pigmented rabbits were infected by herpes simplex virus(HSV-1)(COS strain)to establish ARN animalmodel.After 24 and 72 hours,GCV was given by intravitreal injection(10 eyes),intravenous injection(11 eyes)and the intravitreal+intravenous injection(10 eyes);intravitreal injection of GCV anddexamethasone(6 eyes)was also included.Four eyes were not treated as the control.The dosage of GCVin intravitreal and intravenous injection was 800 μg and 5 mg/kg weight,respectively.Retina necrosis wasobserved and the grade was recorded 1-21 days after injection according to the grade standard ofretinopathy.The maximum grades of retinal necrosis in different groups were compared.Results Thegrade of retinal necosis was 3.8 in the control group,and 0.2,0.4,0.8,and 2.2 in intravitreal iniection,intravitrcal+intravenous injection,intravitreal injection with GCV and dexamethasone,and intravenousrejection,respectively,24 hours after the model was set up.The effects of the first 3 groups wereobviously better than the Iast group(P=0.003,0.011,0.045);while the difference among the first 3groups were not significant(P=0.881,0.054,0.107).Seventy-two hours after the mode J was set up,thegrades of retinal necrosis were above 1.4 in 4 groups,and the differences among the 4 groups were notapparent(P=0.214).Conclusions In the animal model of ARN,intravitreal injection with GCV caneffectively decrease the grade of retinal necrosis.The difference among intravitrealinjection,intravitreal+intravenous injection,intravitreal injection with GCV and dexamethasone,and intravenous injection is notsignificant.
6.Effects of Silencing G6PD Expression on The Growth and Apoptosis in Human Skin Melanoma

Glucose-6-phosphate dehydrogenase (G6PD) derives from the expression of the house-keeping gene G6PD. Recent studies have indicated that G6PD is related to tumor genesis, growth, clinical phenotype, therapy, and prognosis. To elucidate the relationship between G6PD and cancer, three siRNA sequences and one negative control sequence were designed based on the 3' noncoding region of the human G6PD gene. Two complementary single-strand DNA (sense and antisense) were designed and synthesized based on siRNA sequences. The DNA fragments were annealed and ligated to the GFP expression vector pRNAT-U6.2/Lenti. One siRNA with higher interference efficiency than the other two was found after siRNA plasmid transfecting human skin A375 melanoma cells. After lentivirus particle packaging and virus production, the A375 cells were infected, and the single cell clone was acquired and cultured to establish the stable cell strain. Western blotting showed that the endogenous G6PD in the stable A375 cell strain was 0.257 ? 0.074, which was 11.17% of G6PD expression (2.301 ? 0.286) in wild type A375 cells. The final siRNA interference efficiency in this stable cell strain was 88.83%. The G6PD activity of A375-G6PD?驻 was 21.53% of A375-WT. Further study showed that A375-G6PD△ doubling generation time prolonged, and its proliferation was greatly inhibited and the cloning efficiency lowered 25%(P
7.Analysis of Key Factors Affecting Maintenance Cost of Large-Scale Medical Equipment

Objective To explore the key factors which affect maintenance cost of large-scale medical equipment. Methods The basic factors which affect maintenance cost of medical imagining equipment were summarized. The relationships of them are analyzed and then an interpretative structure mode of each factor was set. Additionally, the multiple relationship and controllability between each factor were also analyzed and the judging standard of key factors was set. Results The maintenance policy and mode were the key factors which affect maintenance cost of medical imagining equipment. Conclusion High quality maintenance for equipment condition and scientific optimization of the maintenance policy are necessary to fundamentally reduce the maintenance cost of large-scale medical equipment.
8.Gender dependence of metabolomic phenotypes for human saliva using ultra-high performance liquid chromatography with quadrupole time-of-flight mass spectrometry

Objective · To investigate the gender-related metabolomic differences in human saliva. Methods · The saliva metabolomic profiles of 5 male and 5 female healthy volunteers with matched age, body mass index (BMI), living and tooth-brushing condition were acquired using ultra-high performance liquid chromatography with quadrupole time-of-flight mass spectrometry. Metabolites were identified using publicly accessible databases and further confirmed individually with standard compounds. Both multivariant and univariate statistics were conducted to find inter-gender differences.Results · Forty-eight metabolites in human saliva were identified including 13 amino acids, 6 choline metabolites, 15 carnitines, 4 sphinganine metabolites,7 lysophosphatidylcholine and 3 organic acids. Levels of phenylalanine, acetylcarnitine, propionylcarnitine, butyrylcarnitine, isobutyrylcarnitine,isovalerylcarnitine and sphinganine were higher in the saliva of females than that of males. Conclusion · Rich metabolic information present in human saliva with significant gender diffference which ought to be taken into consideration in study on the metabonomics of saliva.
9.Function and clinical applications of bone morphogenetic proteins in bone repair

Bone morphogenetic proteins (BMPs) are members of the transforming growth factor-β (TGF-β) superfamily, acting as important regulators in embryogenesis and bone and cartilage formation and repair.The complexity of BMPs action has early been unveiled by generous cell and molecular biology approaches and transgenic animal studies.Genetic engineering allows for the mass production of BMPs for clinical use of bone repair, but the desirable scaffold material is also needed to ensure controlled release and maximum biological activity of BMPs at the surgical site.Currently, BMP-2 and BMP-7 have been approved by United States Food and Drug Administration for clinical therapy in long bone fracture, spinal fusion and oral and maxillofacial surgery.The results are satisfactory, but the side effects have been also widely concerned.The functions and clinical applications of BMPs in bon repair are reviewed.
10.Role of adhesion molecule CD44 in respiratory syncytial virus infection induced bronchiolitis

Objective To investigate the role ot adhesion molecule CD44 in respiratory syncytial virus (RSV) infection induced bronchiolitis.Methods Thirty-six children with RSV infection induced bronchiolitis (infection A group),34 children with only RSV infection (infection B group) and 40 healthy children (control group) were selected.The proportion of peripheral blood CD44+ cell was determined by flow cytometry.The levels of peripheral blood interferon (IFN)-γ,interleukin (IL)-4,IL-10 and IL-12 were determined by enzyme-linked immunosorbent assay.Results The expression of CD44 and the level of IL-4 in peripheral blood in infection A group were significantly higher than those in infection B group and control group,there were statistical differences (P < 0.05).The level of IFN-γ in peripheral blood in infection A group was significantly lower than that in infection B group and control group,there were statistical differences (P <0.05).The levels of IL-12 in peripheral blood in infection A group and infection B group were significantly lower than that in control group,there were statistical differences (P < 0.05).There were no statistical differences in the level of IL-10 in peripheral blood among the 3 groups (P > 0.05).In the children with RSV infection,the expression of CD44 in peripheral blood was positively correlated with the level of IL-4 (r =0.798,P < 0.05),the level of IL-12 was negatively correlated with the level of IL-4 (r =-0.186,P <0.05).Conclusion The expression of adhesion molecule CD44 may play a role in the RSV infection induced bronchiolitis,and the high expression of CD44 in children with RSV infection may increase the susceptibility to bronchiolitis.
