Objective:To study the prophylactic effect of combined supine mechanical ventilation supporting the management of esophageal atresia in postoperative complications. Methods:80 patients were selected from January 2015 to June 2016 in the hospital for treatment of congenital esophageal atresia surgery, and divided into control group and observation group with 40 cases each according to the postoperative management method. The control group was treated by holding pillow supine posture and implement routine care, and the observation group was maintained 30o supine mechanical ventilation support management, in order to compare incidence of postoperative complications between them. Results:The incidence of postoperative complications in the observation group was lower than that in the control group (x2=6.03, P<0.05). There was no significant difference between the two groups of children with mechanical ventilation time, HR and MAP levels (t=0.39, t=1.73, t=2.04;P>0.05). The children SaO2 levels in the observation group were significantly higher than that in the control group (t=19.24, P<0.05). The patient care satisfaction rate of observation group was 95.0%, significantly higher than 77.5%in the control group (x2=6.38, P<0.05). Conclusion:It works well for patients with esophageal atresia surgery after using 30o supine intervention and mechanical ventilation support management. It can reduce the incidence of postoperative complications, and should be widely applied.

Objective: To observe the clinically curative effect of Jiangtong Xifeng liquid (JTXFL,降通 熄风液)combined with conventional therapy of western medicine on acute cerebral hemorrhage.Methods:Ninety cases were randomly divided into 2 groups.The treated group (n=60) was treated by JTXFL combined with conventional therapy of western medicine.The control group (n=30) was treated with conventional therapy alone.Before an d after therapy the changes in scores of neural functional defect,state of cons ciousne ss,blood pressure,and CT and the curative effects were observed for both groups.Results:The total effective rate was 88.3% in treated group,but it was 76.7% in contro l group, so that it in treated group was significantly superior to that in contr ol group (P<0.05). Conclusions:The curative effects of JTXFL combined with conventional therapy of western med icine on acutely cerebral hemorrhage is very obvious.

Objective To determine whether altered expression of apoptosis pathway genes is related to different apoptosis susceptibility of prostate cancer cells. Methods Androgen sensitive and insensitive prostate cancer cell lines LNCaP and PC 3 were cultured and treated by etoposide,and apoptosis were determined using Hoechst 33258 staining.The cells were harvested and the total RNA was extracted.cDNA probes were prepared and labeled with biotin 16 dUTP,then hybridized to commercially available cDNA arrays including apoptosis pathway specific genes. Results Apoptosis were induced in both PC 3 and LNCaP cells by etoposide,however,PC 3 was more resistant than LNCaP.Compared with LNCaP cells,the 4 fold down regulated genes in PC 3 cells were Bcl10,CIDE A,GADD45a,RIP2,caspase 4,5 and 6,while the 4 fold up regulated gene in PC 3 cells was TRAF4.Caspase 14 and TNFR2 were most strongly expressed genes in the 2 cell lines. Conclusions The altered expressions of apoptosis pathway specific genes are related to the different apoptosis susceptibility,and this may make an important contribution to androgen insensitive state of prostate cancer cells.

Background and purpose: The metastasis-associated in colon cancer-1 (MACC1) is highly expressed in different cancers and has an effect on the proliferation and apoptosis of tumor cells through the regulation of extracellular signal-regulated kinase (ERK)1/2 pathway. However, the role of MACC1 in ovarian cancer has been rarely studied. The study was aimed to suppress MACC1 gene expression by siRNA and explore the relationship between MACC1 expression and chemosensitivity to cisplatin in ovarian cancer cell line SKOV3/DDP. Methods:Empty plasmid p-super-EGFP-1 (negative control group) and p-super-EGFP-MACC1 shRNA (experimental group) were transfected into ovarian cancer cell SKOV3/DDP respectively. SKOV3/DDP cells without transfection were used as blank group. Then, MACC1 mRNA and protein levels were measured by RT-PCR and Western blot, respectively. Cell proliferation and IC50 of cisplatin was determined by methyl thiazolyl tetrazolium test (MTT). Apoptosis rate was determined by lfow cytometer (FCM). ERK1/2 and p-ERK1/2 protein levels were determined by Western blot. Results:Compared with those in blank and negative control groups, MACC1 mRNA and protein levels deceased in experimental group. The IC50 of cisplatin in experimental group was lower than that in the other groups (26.094 vs 47.501/47.089μmol/L, P<0.05). There was a lower expression of p-ERK1/2 in experimental group (0.3979 vs 00.6712/0.6681, P<0.05). Apoptosis rate was significantly higher in the experimental group before and after treatment of cisplatin (1.32%vs 0.66%/0.48%, P<0.05;36.70%vs 18.53%/16.60%, P=0.000). Conclusion:MACC1 gene may be involved in cisplatin resistance phenomenon in SKOV3/DDP cells through ERK1/2 pathway.

Objective To compare the efficacy and safety of novorapid and novolin R in the treatment of patients with gestational diabetes.Methods From September 2014 to October 2015,the clinical data of 58 patients with gestational diabetes in our hospital were retrospectively analyzed.According to the drug used,the patients were divided into two groups.The observation group was given novorapid,and the control group was given novolin R.Before and after treatment,the total effective rate,fasting blood glucose (FBG),2h postprandial blood glucose (2hPBG),average blood glucose control time,body mass index (BMI),alanine transaminase (ALT),aspartate aminotransferase (AST),serum creatinine (SCr),urea nitrogen (BUN) and the frequency of hypoglycaemia,insulin dosage,the pregnancy outcome were compared between the two groups.Results After treatment,the FBG [(5.14±0.39)mmol/L],2hPBG[(7.06±1.25)mmol/L] of the observation group were significantly lower than those of the control group (t=11.72,5.67,all P＜0.05).ALT[(25.27±8.71)U/L],AST[(27.98±8.53)U/L],SCr[(69.92±11.39)μmol/L],BUN[(5.23±5.75)mmol/L] of the observation group had no obvious differences compared with the control group (t=0.14,0.07,0.04,0.30,all P＞0.05).The total effective rate (93.10％),average blood glucose control time (3.06±0.43)d,insulin dosage (36.15±4.72)U/d of the observation group were significantly better than those of the control group (χ2=4.35,t=14.30,4.92,all P＜0.05).The incidence rates of pregnancy-induced hypertension (3.45％),amniotic fluid (6.90％),cesarean delivery (17.24％),macrosomia(3.45％),high blood bilirubin(3.45％) of the observation group were significantly lower than those of the control group(χ2=5.22,5.50,4.08,9.09,7.73,all P＜0.05).Body weight [(70.6±3.09)g],BMI[(23.9±0.28)kg/m2] of the observation group were better than those of the control group(t=2.20,12.19,all P＜0.05).The incidence rate of hypoglycemic events in observation group(17.24％) was significantly lower than that of the control group(χ2=5.47,P＜0.05).Conclusion Compare with novolin R,the treatment effect of novorapid for gestational diabetes is remarkable and has high security,it is worthy of clinical implementation.

Objective To produce human papillomavirus type 6(HPV-6)virus-like particles with Escherichia coli expression system and study its immunogenicity.Methods HPV-6 L1 gene was inserted into pmkaryotic expression vector pTO-T7 and then expressed in Escherichia coli ER2566.The HPV-6 L1 protein was purified by ammonium sulfate precipitation,ion-exchange chromatography,and hydrophobic interaction chromatography.Then the purified HPV-6 L1 self-assembled into virus-like particle after removing 1,4dithiothreitol(DTr).The morphology of the virus-like particles was investigated with dynamic light scatter and transmission electron microscopy,and the immunogenicity was determined with in vitro pseudownons neutralization as8ay.Results HPV-6 L1 was expressed in soluble form in Escherichia coli.Following the removal of DTT,purified HPV-6 L1 protein could assemble into virus-like particles as 25 am in the radius.And the animal immunization test showed HPV-6 virus-like particles can elite hish titer neutralizing antibodies.Conclusion The bacterially expressed HPV-6 L1 VLP is highly immunogenieity and easy to produce.And it can be good candidate of HPV-6 vaccine.

Objective: To investigate the association between gene polymorphism of vesicular monoamine transporter type 2(VMAT2) and schizophrenia in Han Chinese population. Methods: 430 patients with schizophrenia and 470 age-sex matched controls were recruited from four mental health centers. All patients were diagnosed by two psychiatrists based on the Structured Clinical Interview for DSM Disorders (SCID). The ligase detection reactions (LDR) method was used to assess the polymorphism of the two SNPs (rs363371 and rs363324) of VMAT2. Results: No associations of two SNPs with schizophrenia was found. When we stratified males and females for the analysis, we found that that in the recessive model of rs363371, there was an obvious significant association between rs363371 and schizophrenia in males (OR=0.564, 95% CI=0.357–0.892, p=0.014) but not females. For the association between rs363324 and schizophrenia, no association was found in either males or females. No association was found when stratifying early-onset schizophrenia and late-onset schizophrenia. Conclusion Our findings indicate that both rs363371 and rs363324 were not associated with schizophrenia, while it seemed that the AA genotype of rs363371 plays a protective effect in male Chinese in developing schizophrenia.

Thimerosal has been widely used as a preservative in drug and vaccine products for decades.Due to the strong propensity to modify thiols in proteins,conformational changes could occur due to covalent bond formation between ethylmercury(a degradant of thimerosal)and thiols.Such a conformational change could lead to partial or even complete loss of desirable protein function.This study aims to investigate the effects of thimerosal on the capsid stability and antigenicity of recombinant human papillomavirus(HPV)18 virus-like particles(VLPs).Dramatic destabilization of the recombinant viral capsid upon thimerosal treatment was observed.Such a negative effect on the thermal stability of VLPs preserved with thimerosal was shown to be dependent on the thimerosal concentration.Two highly neutralizing antibodies,13H12 and 3C3,were found to be the most sensitive to thimerosal treatment.The kinetics of antigenicity loss,when monitored with 13H12 or 3C3 as probes,yielded two distinctly different sets of kinetic parameters,while the data from both monoclonal antibodies(mAbs)followed a biphasic expo-nential decay model.The potential effect of thimerosal on protein function,particularly for thiol-containing proteinaceous active components,needs to be comprehensively characterized during formulation development when a preservative is necessary.
