Objective: To study the relationship of resting energy expenditure(REE) with wound depth in burn patients and offer some guidance for the nutrition treatment of burn.Methods: Twenty-three burn patients with a similar total burn surface were randomly divided in accordance with the Ⅲ? wound area into Goups A,B,C and D,their total burn surface and Ⅲ?wound area were(66?18)% and(12?9)%,(59?18)% and(20?5)%,(62?6)% and(39?5)%,and(65?16)% and(54?4)%,respectively.Their REE was determined on PBD(postburn day) 1,2,3,7,14,21 and 28 with the indirect measuring method.The measured values of 13 normal adults were used for comparison.Results: The mean value of REE of each burn group was higher than the normal control.REE enhanced significantly with the increase of the Ⅲ? wound area.Conclusion: REE obviously increases in burn patients,and,with a similar total burn surface,the larger the Ⅲ? wound area,the higher REE.Therefore,increased calories should be furnished in the nutrition treatment of burn patients with a larger Ⅲ? wound area.
3.To discuss the relationship between part of risk factors and the characters of coronary artery lesion of different gender patients with coronary heart disease

Objective To discuss the relationship between part of risk factors and the characters of coronary artery lesion of different gender patients with coronary heart disease(CHD). Methods Two hundred and eight patients who were diagnosed CHD by coronary angiography (CAG) were selected. All patients were divided into male group (122 cases) and female group (86 cases). The characters of CHD and part of clinical data in different gender CHD patients were compared and analyzed. The independent risk factors of different gender CHD patients were analyzed. Results The age of onset in female group was later than male group:(69.22±10.12) years vs.(62.80±11.34) years, P=0.000. The incidence rate of hypertension and diabetes in female group were significantly higher than those in male group: 83.7%(72/86) vs. 63.1%(77/122), 53.5%(46/86) vs. 32.8%(40/122);and the levels of total cholesterol(TC), high density lipoprotein cholesterol (HDL-C), low density lipoprotein cholesterol (LDL-C) in female group were significantly higher than those in male group:(5.16±1.26) mmol/L vs. (4.60±1.23) mmol/L,(1.16±0.27) mmol/L vs. (1.05±0.27) mmol/L, (3.17±1.16) mmol/L vs. (2.74±1.06) mmol/L;the level of uric acid (UA) in female group was significantly lower than that in male group: (319.83±90.05)μmol/L vs. (357.91±98.51)μmol/L, there were significantly differences(P<0.01). The level of trigalloyl glycerol (TG) in two groups had no significant difference: (1.91 ± 1.23) mmol/L vs. (1.75±0.97) mmol/L, P=0.298. Logistic regression analysis showed that age and diabetes were the risk factors of CHD in different gender (P<0.01 or <0.05). Conclusions There are different risk factors between different gender CHD patients, while the coronary artery lesion is similar. Diabetes is the most important independent risk factor of different gender CHD patients, which is more important for female patients with more risk factors.
4.Effect of SREBP-2 silencing on tunicamycin-induced endoplasmic reticu-lum stress in chondrocytes

[ ABSTRACT] AIM:To explore the effect of sterol regulatory element-binding protein 2 ( SREBP-2) on tunicamy-cin-induced endoplasmic reticulum stress ( ERS) in chondrocytes.METHODS:After isolation of human normal chondro-cytes and osteoarthritis ( OA) chondrocytes, the normal cells were cultured and treated with tunicamycin and SREBP-2 siR-NA.After 24 h treatment, fluorescent quantitative RT-PCR ( RT-qPCR) was applied to quantify microRNA-185 ( miR-185) levels.The cell apoptotic rate was determined by flow cytometry.The expression of SREBP-2 and ERS-related pro-teins, C/EBP homologous protein (CHOP), phosphorylated eukaryotic initiation factor-2α(p-eIF2α) and activating tran-scription factor 4 (ATF4), and the expression of apoptosis-related proteins, Bcl-2, Bax and caspase-3, were determined by Western blot.The caspase-3 activity kit was used to determine the caspase-3 activity.RESULTS: Compared with hu-man normal chondrocytes, both SREBP-2 up-regulation and miR-185 down-regulation were observed in OA chondrocytes (P<0.05).SREBP-2 siRNA transfection enhanced tunicamycin-inhibited miR-185 level (P<0.05).miR-185 overex-pression reduced tunicamycin-induced SREBP-2 expression ( P <0.05 ) .OA control group and tunicamycin treatment group consistently resulted in ERS and cell apoptosis with concomitant enhancement of CHOP, p-eIF2αand ATF4 proteins, increases in Bax and caspase-3 proteins, and reduction of Bcl-2 (P<0.05).However, SREBP-2 silencing significantly re-versed these effects ( P<0.05) .The apoptotic rates were consistent with the expression tendency of apoptosis-related pro-teins (P<0.05).SREBP-2 siRNA transfection markedly down-regulated tunicamycin-induced caspase-3 activity, which was notably blocked by miR-185 inhibition (P<0.05).CONCLUSION:SREBP-2 silencing may inhibit tunicamycin-in-duced ERS and cell apoptosis via up-regulating miR-185 expression.
5.Effect of miR-33 on proliferation and apoptosis of B16F10 cell line

Objective To further approach the effect of miR-33 to melanoma cells line B16F10 proliferation and apoptosis.Methods Constructing targeted miR-33 over-expression mimics and inhibitor,the same B16F10 cells were divided into five groups,control group,miR-33 mimics group,mimics control group,miR-33 inhibitor group,inhibitor control group,then gene transfer technology was used to transfer corresponding gene into B16F10 cells.The effect of miR-33 on B16F10 cell' s proliferation and apoptosis were analysed.Results The relative miR-33 gene expression of miR-33 mimics group (1773.3±245.83) was higher than that of control group,which had statistical significance (P < 0.01).The gene expression of miR-33 inhibitor group (0.6973±0.1958) was lower than those of control group and inhibitor control group.The cell growth rate of miR-33 mimics group was lower than those of control group and the trend after transfection 48 h (1.1875±0.0502) and 72 h (1.7500±0.0933) was significant (P < 0.05).The cell apoptotic ratio of miR-33 mimics group [(1.8050±0.2050) %] was higher than that of control group [(1.13000±0.1414) %] (P < 0.05).Compared with control group the cell proportion ofG1 period in miR-33 mimics group [(62.7000±1.7321) %]increased,the cell proportion of S period [(23.4000±2.5044) %] decreased,both of them had statistical significance (both P < 0.05).Conclusion miR-33 over-expression can restrain the proliferation of B16F10 cells line,promote B16F10 cells' apoptosis.
6.Experimental study on the healing effect of breviscapine injection on deep partial thickness burn wound

ObjectiveTo observe the healing effect of deep partial thickness burns by the injecting of breviscapine and discuss the possible mechanisms. Methods A rat model of deep partial thickness burns was designed,and was injected with breviscapine.The control group was injected with normal saline.The healing time of burn wound of the two groups was recorded,respectively.Seven days later,the tissues of bum wound of each group were extracted and the contents of hydroxyproline,collagenase-1,nitrogen monoxidum,erythrocuprein,and malonaldehyde that were contained in each extracts were measured.The results of each group were statistically analyzed.ResultsThe healing time of burn wound of the experimental group was [ ( 12 ± 1.428 ) days ],which was significantly shorter than the control group [ ( 14.75 ±1.291 )days] ( P <0.05).The contents of hydroxyproline[ (3.17 ± 1.136) mg/g],collagenase-1 [ ( 1.28± 0.651 ) mg/g ],nitrogen monoxidum [ ( 2.62 ± 0.30 ) μmol/gprot ],and erythrocuprein [ ( 221.25 ±25.94) U/mgprot ] in the experimental group were all higher than the control group [ (7.32 ± 2.173 )mg/g,(5.38 ±0.363) mg/g,(7.28 ± 0.40) μmol/gprot,(296.36 ± 29.29) U/mgprot ] ( P < 0.05 or P <0.01 ).However,the content of malonaldehyde [ (6.36 ± 0.93 ) nmol/mgprot ] was lower than the control group [ ( 1.25 ± 0.59) nmoL/mgprot ] ( P < 0.05 ).ConclusionsThe breviscapine injection can decurtate the healing time of deep partial thickness bums and it may be related to the extension of blood vessel,improvement of microcirculation,elimination of oxygen free radical,and degradation of lipid peroxidation.
7.Benign infantile convulsions with mild gastroenteritis:clinical and prognosis analysis of 248 cases

Objective To explore the clinical characteristics and the prognosis of benin convulsions as-sociated with mild gastroenteritis( BICE) in children. Methods To study clinical features of 248 children with BCIE,they followed up for 12 months,and their prognosis were observe. Results Age between 3 months and 3 years,twice or more seizures were occurred on 124 patients. 64 patients suffered the seizures more than 30 mi-nutes. Cranial CT/MRI showed no abnormality. Laboratory examinations were normal, including plasma glu-cose,serum electrolytes and cerebrospinal fluid. Followed up for 12 months,14 patients (5. 6%) suffered from recurrence of BICE,12 patients (4. 8%) suffered from simple febrile seizures,and 8 patients (3. 2%) devel-oped epilepsy. Conclusion The prognosis is good in majority of cases,but some cases may transform febrile seizure or epilepsy. It is more likely to transform epilepsy,when the children suffer from febrile seizure.
8.Results analysis of blood infectious indicators in 331 968 outpatients and inpatients

Objective To analyze the results of detection of infectious indicators of patients from the first affiliated hospital of Nanjing Medical University in recent 5 years ,and to provide a scientific basis for the control and prevention of infectious diseases . Methods The patients with clinical data from January 2010 to April 2014 were retrospectively analyzed ,then the infectious indica‐tors of all the subjects were detected and analyzed .Results HIV positive rate was between 0 .043% to 0 .061% ,positive rate of HCV was between 1 .07 to 1 .41% ,positive rate of TP was between 2 .01% to 2 .17% .HBsAg positive rate in 2010 was 8 .36% ,the positive rate was 7 .81% in 2014 .HBsAb positive rate in 2010 was 35 .36% ,positive rate was 50 .96% in 2014 .Conclusion Effec‐tively cut off the route of transmission could prevent the further spread of infectious disease .
9.Construction of the eukaryotic expression plasmid containing human epidermal growth factor gene with signal peptide

Objective To construct the eukaryotic expression plasmid containing human epidermal growth factor(hEGF) gene with signal peptide(SP).Methods After two pairs of primers were designed and synthesized,the cDNA fragment of hEGF and SP genes were amplified from total RNAs. The amplified cDNA fragments were cloned into pGEM-T vector.The expression plasmids were verified by double endonuclease digestion and DNA sequence analysis. Results With RT-PCR using two pairs of primers,two bands(about 90bp and 180bp) were obtained and confirmed as signal peptide and EGF cDNA fragment with electrophoresis analysis and DNA sequencing after cloned into pGEM-T vector.The SP and EGF cDNA fragments were inserted into plasmid pcDNA3.1(+).The bands of 240bp and 5.4kb were obtained and identified as the full length of SP-EGF cDNA fragment by DNA sequence analysis.Conclusion The eukaryotic expression plasmids containing hEGF gene is successfully constructed.
10.Value of a microRNA risk score model in predicting the prognosis of hepatocellular carcinoma

ObjectiveTo screen out the microRNAs (miRNAs) associated with the prognosis of hepatocellular carcinoma (HCC) through data mining of miRNA transcriptome data of HCC downloaded from The Cancer Genome Atlas (TCGA) database, to establish a miRNA risk score model, and to investigate its value in predicting the prognosis of HCC. MethodsThe miRNA expression data and clinical data of HCC samples were downloaded from TCGA database and R language was used to screen out differentially expressed miRNAs between HCC tissue and adjacent tissue, which were randomly divided into training set and testing set after being integrated into clinical data. Univariate Cox regression analysis and least absolute shrinkage and selection operator (LASSO) Cox regression analysis were performed for the training set to screen out the miRNAs associated with the prognosis of HCC, and then a miRNA risk score model was established. The Kaplan-Meier method was used to evaluate the robustness of the model and whether it could predict the prognosis of patients in the same clinical stage. Finally, the receiver operating characteristic (ROC) curve was plotted and the area under the ROC curve (AUC) was calculated to compare the predictive accuracy of the model versus TNM staging in the training set, the testing set, and the entire set. ResultsA total of 300 differentially expressed miRNAs were screened out and the LASSO Cox regression analysis revealed that hsa-miR-139-5p, hsa-miR-1180-3p, hsa-miR-1269b, hsa-miR-3680-3p, hsa-miR-509-3-5p, and hsa-miR-31-5p were associated with the prognosis of HCC. The risk score was calculated for each sample according to the established miRNA risk score model, and the samples were divided into high-risk group and low-risk group according to the median risk score. The Kaplan-Meier curve showed that in both training and testing sets, the high-risk group had a significantly lower survival rate than the low-risk group (P<0.05). The ROC curve was used to evaluate the prediction efficiency of this model, and the results showed that in the training set, the testing set, and the entire set, the miRNA model had an AUC of 0.817, 0.808, and 0.814, respectively, while TNM staging had an AUC of 0.667, 0.665, and 0.663, respectively. The results of independent prognostic analysis also showed that this miRNA score model could be used as an independent prognostic factor for HCC (P<0.05). ConclusionHsa-miR-139-5p, hsa-miR-1180-3p, hsa-miR-1269b, hsa-miR-3680-3p, hsa-miR-509-3-5p, and hsa-miR-31-5p are associated with the prognosis of HCC, and the miRNA risk score model has a better prediction accuracy than TNM staging in the training set, the testing set, and the entire set. The stratified analysis also shows that the model can predict the prognosis of patients within the same TNM stage, and therefore, it has a certain reference value in clinical practice and can be used as an independent model for predicting the prognosis of HCC patients.
