1.Multicenter evaluation on pharmacoeconomics of Ginkgolide Injection in treatment of ischemic stroke

Objective To study the clinical efficacy,safety,and economic efficiency of ginkgolide injection and conventional therapy of patients with cerebral arterial thrombosis in multi center,and to evaluate the economic value of drugs.Methods A prospective cohort study was conducted in this study,patients with ischemic stroke were collected from August 2013 to December 2014.Patients (354 cases) in treatment group were treated with Ginkgolide Injection and routine treatment,and patients (180 cases) in control group could be treated with other drugs for activating blood circulation to dissipate blood stasis on the basis of routine treatment.The patients were telephone followed-up visited 3,6,and 12 months after discharge for pharmacodynamic indexes:evaluation of activities of daily living (ADL) score,self-care rate,cure rate,recurrence rate,and all-cause mortality;economic indicators:the patient work recovery rate,cost effectiveness ratio (CER),and the incidence,and severity of adverse events,to assess the differences in the long-term benefits of different treatment regimens.Results Follow-up in 3,6,and 12 months showed that ADL score,cure rate,self-care rate,and work recovery rate of the treatment group were better than those of control group,and the difference was statistically significant.Follow-up in 12 months showed that,recurrence rate and mortality rate in the treatment group was better than that in the control group,with statistical difference.Follow-up in 6 and 12 months showed that CER of treatment group was smaller than that of the control group.The incidence of adverse reactions was low in the two groups.Conclusion Long term evaluation showed that patients treated with Ginkgolide Injection had better clinical outcomes and better CER than those without it,which proved the effectiveness and economic efficacy of Ginkgolide Inj ection in the treatment of stroke.
2.The phenomenology research of perioperative therapeutic communication for rectal cancer patients

Objective To explore the clinical practice and patient experience of perioperative therapeutic communication in rectal cancer patients, in order to provide theoretical basis for the further clinical application of therapeutic communication. Methods To adopt phenomenology method of qualitative research, collecting data by the direct participation, observation, half structural formula thorough interview and other methods. To establish coding and refine theme. Results Four themes of the rectal cancer patients experience of perioperative therapeutic communication were generalized: emotional adaptation, actively coping with surgery, intraoperative cooperation, postoperative rehabilitation and self-care. Conclusions Application of therapeutic communication system can effectively interfere with patients cognitive behavior, improve communication ability of nurses and extended the nursing service connotation.
3.Interleukin-24 and its antitumor effects

Interleukin 24(IL 24),also called the melanoma differentiation associated gene 7(MDA 7),is stably expressed in human tissues associated with the immune system such as the spleen,thymus,peripheral blood leukocytes and normal melanocytes. IL 24 binds to IL 20 and IL 22 receptor complexes(IL 22R1/IL 20R2 and IL 20R1/IL 20R2),and induces secretion of high levels of IL 6,TNF alpha,and IFN gamma and low levels of IL 1beta,IL 12,and GM CSF from human PBMC. Adenoviral IL 24(Ad IL 24) induces growth suppression and apoptosis selectively in diverse human cancers and tumor cell lines without producing any apparent harmful effect in normal cells. The effects of Ad IL 24 are associated to the ratio of pro apoptotic(BAX,BAK) to anti apoptotic(Bcl 2) proteins,and the up regulation of p38 MAPK and a family of growth arrest and DNA damage(GADD) inducible genes. These demonstrate the potential therapeutic effects of Ad IL 24 in human cancer.
4.Evaluation of reversal effect of 2-DG on multidrug resistance by detecting uptake of 99mTc-MIBI in HNE-1/DDP cells

Aim To evaluate the reversal effect of 2-deoxy-D-glucose ( 2-DG ) on multidrug resistance ( MDR) by observing the uptake change of 99m Tc-MIBI in HNE-1/DDP cells, and to explore its mechanism. Methods The uptake of 99m Tc-MIBI in HNE-1/DDP cells under different concentrations of 2-DG was detec-ted by γ-counter, and the clearance rates of 99m Tc-MI-BI in HNE-1 cells and HNE-1/DDP cells after treated with 2-DG (10 mmol·L-1 ) were compared. The con-tent of ATP in HNE-1/DDP cells was detected after treated with 2-DG. P-glycoprotein ( P-gp ) and multi-drug resistance-associated proteins ( MRP ) expression were measured by Western blot. Apoptotic HNE-1/DDP cells treated with DDP alone or combined with 2-DG (10 mmol·L-1 ) were detected by propidium io-dide ( PI ) staining. Results The clearance rate of 99m Tc-MIBI in HNE-1/DDP cells was 54. 8%, which was significantly higher than that ( - 41. 3%) in HNE-1 cells (P<0. 01). The clearance rate of 99mTc-MIBI in HNE-1/DDP cells was -203. 7% after treat-ment with 2-DG ( 10 mmol · L-1 ) , which could be significantly reduced compared with the control group ( P<0. 01 ) . The level of ATP was 55 . 69% compared with the negative control group and the expression of P-gp and MRP protein decreased dramatically in HNE-1/DDP. With the combination of 2-DG and DDP, the ap-optotic rate of HNE-1/DDP cells reached 49 . 4%which was significantly higher than DDP treated group (22. 5%) . Conclusion Multidrug resistance and the reversal effect of 2-DG on multidrug resistance could be evaluated effectively by detecting the uptake change of 99m Tc-MIBI in HNE-1/DDP cells. The mechanism may be related with the inhibition of ATP level and the re-duced expression of P-gp and MRP protein in cancer cells.
5.Construction of eukaryotic expression vector of mouse microRNA miR-21 and identification its expression activity in 293 cells

Objective To construct the eukaryotic expression vector for mouse microRNA miR-21 and identification its expression activ-ity in 293 cells. Methods The genomic sequence containing pre-miR-21 was amplified from mouse genomic DNA by PCR and cloned into the pRC/CMV plasmid. The constructed recombinant plasmid pRC/CMV-mmu-miR-21 was transfected to 293 cells by lipofectamine 2000, and the stably transfected cells were screened with G418,from which total RNA was extracted for detecting the expression of mature miR-21 by northern blot. In the meantime,a luciferase report plasmid examing the activity of miR-21 named pmiR-21-Luc reporter was also construc-ted,and luciferase activity analysis indicated the product of pRC/CMV-mmu-miR-21 indeed had biological activity. Results Both restriction enzyme digestion analysis and sequencing proved the recombinant plasmids were constructed correctly. The miR-21 was highly expressed in the screened clones of 293 cells and it had good biological activity. Conclusion The eukaryotic expression plasmid of mouse miR-21 was successfully constructed,which laid the foundation of further investigation of the role of miR-21 during skin wound healing.
6.A study on the influence of CRT brightness for image reading

Objective To study the influence of CRT brightness on image reading. Methods Both physical testing and compared viewing were used in the study. (1) Setting the CRT brightness at high and low grade (the blank brightness of CRT was 112 cd/m 2 and 82 cd/m 2, respectively). (2) 21 films with the density ranging from 0.27 to 2.71 were digitized by a scanner and displayed on the CRT. The brightness of CRT that varied with film density was measured by a bright meter, then the relation chart between CRT brightness and the film density was drawn. (3) Thirteen films on which there were many small and low contrast signals were viewed on CRT by three doctors. 7.Exploring in management of women with atypical squamous cells of undetermined significance

Objective Exploring in processing method to triage of women with a cytologic smear showing atypical squamous cells of undetermined significance. Methods All 130 cases were diagnosed with ASCUS received high-risk HPV (hr-HPV)test, colposcopic inspections, reformeded Reid's colposcopic index score and cervical biopsies for pathological examination. Resluts The pathology of 130 ASCUS patients included 31cases of normal and inflammation (23.90%), 29 cases of CINⅠ(22.30%), 14 cases of CINⅡ(10.80%), 55 cases of CINⅢ or cervical carcinoma in situ (42.30%), and 1 case of invasive cervical carcinoma (0.80%). The reformed Reid's colposcopic index score of 130 ASCUS patients included 32 cases of 0 score (24.60%), 36 cases of 1~2 scores (27.70%), 47 cases of 3~4 scores(36.10%), 15 cases of 5~6 scores(11.60%). 2. The colposcopy used to detect the CIN of ASCUS, its sensitivity was 98.99%(98/99), specificity was 51.61%(16/31). The hr-HPV used to detect the CIN of ASCUS, its sensitivity was 96.97%(96/99), specificity was 32.26%(10/31). Conclusion Both the reformeded Reid′s colposcopic index score and hr-HPV testing as the clinical triage methods of ASCUS. The former has good sensitivity and specificity ,which is superior to hr-HPV testing.
8.Design of new type thermal container for infant

Objective To develop infant thermal container with forced protection circuit so as to ensure its security. Methods Temperature database was set in thermal container and Cygnal F005 was used as intelligent controller. The state of the sensor was judged by comparison precisely. Double temperature controller was adopted to ensure normal working temperature. When the container went wrong, it could automatically switch to protection circuit to keep the temperature for infant. Results With precise temperature control and steady working station, the new type thermal container for infant could provide a safe and comfortable condition with clean air and fitting temperature and humidity. Conclusion The container is steady, accurate, safe and convenient.
9.Analysis of Drug Resistance Situation and Influential Factors of Sputum Culture Positive Tuberculosis Pa-tients in A Specialized Subject Hospital

OBJECTIVE:To discuss the drug resistance situation and influential factors of sputum culture positive tuberculosis patients in a specialized subject hospital. METHODS:The information of sputum culture positive tuberculosis inpatients were col-lected from Heze infectious disease hospital during Jan. 1st 2014 to Apr. 30th 2015;the results of drug sensitive test were analyzed retrospectively by using absolute concentration method. RESULTS:Among 259 patients,137 cases were drug-resistant(52.90%), among including 69(26.64%) mono-drug resistant cases,52(20.08%) poly-drug resistant cases,15(5.79%)multi-drug resistant cases and 1(0.39%)extensively drug resistant case;streptomycin (31.66%) and amikacin (15.44%) were drugs with the highest drug-resistant rate;there was no statistical significance in resistant rate between different genders and ages(P>0.05),the rate of drug resistance in first-treatment patients was 47.21%,and that of the re-treatment patients was 70.97%,the difference was statisti-cal significant(P<0.05). In addition to ethambutol and kanamycin,other all had mono-drug resistance,there were 27 combinations of poly-drug resistance and 9 combinations of multi-drug resistance. Logistic regression analysis showed that treatment history was a risk factor for drug-resistance,the risk of re-treatment patient was 2.734 times of first-treatment patients. CONCLUSIONS:Sputum culture positive tuberculosis inpatients patients have high drug resistance and complex spectrum in the hospital;we should pay great attention to the risk of drug resistance in senile patients and re-treatment patients,and rational use of streptomycin,amikacin,isoni-azid and capreomycin.
10.Expression of methionine adenosyltransferase and T lymphocyte activation

Objective: To study the expression and activity changes of methionine adenosyltransferase (MAT) in human peripheral T lymphocytes. Methods: The expression of MAT mRNA was detected by RT-PCR and the activity of MAT was measured. Results: After stimulated by IL-2, PHA and anti-CD3 antibody, MAT-Ⅱ gene expression increased by (8.9? 2.1), (7.7?1.9) and (8.0?1.8) times, respectively, and the expression peak was at 8, 4 and 8 h,respectively; MAT activity continuously increased in 48 h. S-adenosylmethioinie (SAM) moderately induced IL-2 and IFN secretion by human T cells. SAM(0.1 mg/ml) downregulated the expression and activity of MAT-Ⅱ and the secretion of IL-2 and IFN induced by PHA or anti-CD3 antibody in human T cells. Conclusion:MAT is involved in the activation of T lymphocytes, and high dose of SAM may also inhibit its activation through PHA and anti-CD3 antibody.
