1.Effect of chuanxiong and danggui on Na+ and K+ currents of rat hippocampal neurons with hypoxia-reoxygenation

BACKGROUND: The active ingredients of Chinese herbs, such as hongjingtian (rhodiola root, Radix Rhodiolae), srenshen (ginseng, Radix Ginseng), and danggui (Chinese angelica root, Radix Angelicae Sinensis),were found to be effective to inhibit the hypoxia injury.OBJECTIVE: To observe the protecting effect of xionggui diwan (Dripping Pills) on the hippocampal neurons from hypoxia by whole-cell recording patch-clamp technique.DESIGN: An auto-control observation on cells.SETTING: The 88 Hospital of Chinese PLA and Academy of Military Medical Science of Chinese PLA.MATERIALS: The experiment was performed during March to August 2002 in the Institute of Toxicology and Pharmacology of Academy of Military Medical Sciences of Chinese PLA. The neonatal Wistar rats, clean degree, were used. The hippocampal neuron cells out of 9 rats were cultured and divided into the control groups of before and after hypoxia-oxygenation,and xionggui diwan 1 × 10-6 mol/L and 10 × 10-6 mol/L groups. (Xionggui diwan was provided by the 88 Hospital of Chinese PLA and its active ingredient was the volatile ingredient of chuanxiong and danggui.)METHODS: While hypoxia-reoxygenation groups were not given any medicine intervention as control, the treatment of xionggui diwan 1×10-6mol/L was subdivided into xionggui diwan 1 × 10-6 mol/L + non-hypoxia and xionggui diwan 1 × 10-6 mol/L + hypoxia groups, and the treatment of xionggui diwan 10 × 10-6 mol/L was subdivided into xionggui diwan 10 × 10-6 mol/L + non-hypoxia and xionggui diwan 10 × 10-6 mol/L + hypoxia groups. The patch-clamp technique was performed to determine the Na+ and K+ currents of rat hippocampal neurons cultured in vitro before and after hypoxia-reoxygenation.MAIN OUTCOME MEASURES: The Na+ and K+ current amplitude of rat hippocampal neurons cultured in vitro.RESULTS:① The Na+ current amplitude of hippocampal neurons: After hypoxia-reoxygenation, it was significantly lower than that before [(-3.8±1.0, -10.3±0.5) nA, t=3.49, P < 0.01]. ② The K+ current amplitude of hippocampal neurons: After hypoxia-reoxygenation, the activation thresholdof K+ current was changed from -40 mV to -20 mV.③The Na+ and K+ currents of hippocampal neurons before and after hypoxia-reoxygenation with xionggui diwan pretreatment: Before hypoxia-reoxygenation, the ampli tudes of Ⅰ Na and J K of hippocampal neurons at the threshold point were very similar, without significant difference. After hypoxia-reoxygenation,those amplitudes of xionggui diwan 10.0 × 10-6 mol/L group were higher than that of xiongguidiwan 1.0 × 10-6 mol/L group [(-58.5±1.9, -6.9±1.4,-3.8±1.0) nA, t=7.51,P < 0.05].CONCLUSION: It was suggested that xionggui diwan propect the function of hippocampal neurons from hypoxia.
2.Quality control circle activities to improve patients’compliance to head-of-bed elevation

Objective To evaluate the effect of implementing quality control circle (QCC)activities on nurses’ knowledge about ventilator-associated infection(VAP),as well as head-of-bed (HOB)elevation and incidence of VAP in patients.Methods 100 patients who used ventilators in an intensive care unit(ICU)of a hospital were ran-domly selected,50 patients were selected before the implementation of QCC(January-June,2013),and 50 were se-lected after the implementation of QCC (July-December,2103).Condition of nurses and patients who used ventila-tors before and after implementation of QCC were investigated.Results After implementing QCC,the scores of nurses’recognition on knowledge about causes and risks of VAP,significance and standard of HOB elevation,and HOB elevation≥ 30°were all higher than before implementing QCC;the total score was higher than before imple-menting QCC ([18.32±3.14]vs [7.98 ±1 .12],P <0.05 );patients’compliance to HOB elevation was higher than before implementing QCC([96.00%,n=48]vs [44.00%,n=22],P <0.05);incidence of VAP was lower than before implementing QCC ([4.00%,n =2]vs [36.00%,n =18],P <0.05).Conclusion Implementation of QCC activities can effectively improve ICU nurses’recognition on knowledge about VAP and enhance patients’ compliance to HOB elevation,as well as reduce the incidence of VAP,it is helpful for improving healthcare-associat-ed infection management quality.
3.Experimental Study on the Antiaging Effect of Powdered Hindu Lotus (Nelumbo nucifera) Seed

Antiaging effects or powdered Nelumbo nucifera seed were studied on wistar rats by comparing thcir viscera/body, T lymphocyte, SOD, LPO, GSH-Px between fed animals and control. Results showed that there was obvious increase of T lymphocyte in thorax cortex between fed and control, whereas distinct disparity of other indexes can not be found. These results implied that powdered N.nucifera seed has some functions of enforcing immunity.
4.Effect of Xionggui Dripping Pills on Sensitivity of Baroreflex in Selective Sinoaortic Denervated Rats

OBJECTIVE:To observe the effect of Xionggui dripping pills on sensitivity of baroreflex in selective sinoaortic denervated rats.METHODS:The dynamic blood pressures were monitored in selective sinoaortic denervated rats in waking stait,and the changes of baroreflex senitivity(BRS)were measured by modified SU Smyth method.RESULTS:Xionggui drip_ ping pill could improve BRS in sinus denervated rats.CONCLUSION:Since Xionggui dripping pill rectifies autonomic nerve function,it can protect target organs from damage in cardiovascular diseases.

6.Design of new type thermal container for infant

Objective To develop infant thermal container with forced protection circuit so as to ensure its security. Methods Temperature database was set in thermal container and Cygnal F005 was used as intelligent controller. The state of the sensor was judged by comparison precisely. Double temperature controller was adopted to ensure normal working temperature. When the container went wrong, it could automatically switch to protection circuit to keep the temperature for infant. Results With precise temperature control and steady working station, the new type thermal container for infant could provide a safe and comfortable condition with clean air and fitting temperature and humidity. Conclusion The container is steady, accurate, safe and convenient.
7.Study on correlation between plasmodium titre with body temperature and prognosis in malaria patients

Objective To investigate the relationship between the Plasmodium malaria titre with the body temperature and prog-nosis to provide a theoretical basis for malaria diagnosis and missed diagnosis.Methods Ninety-three malaria test positive speci-mens were collected and simultaneously performed the plasmodium test by using the colloidal gold immune chromatography assay and thick film blood slide.The body temperature of malaria patients was recorded ,and the malaria test was conducted once when the body temperature was increased by 1 ℃ until positive for malaria detection.Results In 93 tested specimens ,the infection rate of Chinese was 56.98% (53/93) ,and which of foreigners was 43.01% (40/93) ,the difference between Chinese and foreigners malaria infection groups had no statistical significance ,showing that malaria infection had no relationship with race.Among 93 specimens , the initial malaria infection was in 48 cases ,including 47 Chinese and 1 foreigner ,and 45 cases had twice or more malaria infection , in which 33 cases had the titers of 1:10 ,with the recurrence rate of 0.09% (3/33) and the re-burning rate was 0.27% (9/33). The titer of 1:20 had 28 cases ,with the recurrence rate 0.04% (1/28) ,and the burning rate 0.21% (6/28).Titer of 1:30 was 19 people ,with the recurrence rate 0.16% (3/19) and the burning rate 0.37% (7/19).The titer of 1:40 was 13 people ,with the re-currence rate 0.15% (2/13) ,and the re-burning rate of 0.38% (5/13) ,there was no statistically significant difference between the number of recurrent and re-burning patients with the titre(χ2 =2.5 ,P>0.05 ;χ2 =1.96 ,P>0.05).Conclusion There is a positive correlation between malaria with the titer and temperature.The titer is elevated ,and the probability of recurrence and re-buring is increased.
8.Inhibitory Effect of Ginkolide B Derivative on Thrombogenesis

Objective To observe the effect of ginkolide B derivative(XQ) on animal thrombosis.Methods SD male rats were randomized into the model group,troxerutin(48 mg?kg-1?d-1) group,ginkolide B(7.8 mg?kg-1?d-1) group,and low-,middle-and high-dose XQ(in the dose of 3.9,7.8 and 15.6 mg?kg-1?d-1 respectively) groups.The anti-thrombotic effect of XQ was observed on rats mixed thrombosis model induced by arterio-venous shunt method and electrical stimulation.Meanwhile,ICR male mice were randomized into the model group,troxerutin(96 mg?kg-1?d-1) group,ginkolide B(15.6 mg?kg-1?d-1) group,and low-,middle-and high-dose XQ(in the dose of 7.8,15.6 and 31.2 mg?kg-1?d-1 respectively) groups.The anti-thrombotic effect of XQ was also observed on mice acute pulmonary embolism model induced by adenosine diphosphate(ADP).Results XQ at the dose of 7.8 and 15.6 mg?kg-1?d-1 decreased the dry and wet weight of the thrombosis,and obviously prolonged the formation time of rats carotid artery thrombosis as compared to the model group(P

10.Influence of iridosides of cornus officinalis on glomerular mesangial cells cultured with AGEs

Aim To investigate the influence of iridosides of cornus officinalis(ICO) on morphology, cell cycle and oxidative stress of glomerular mesangial cells(GMC) cultured with advanced glycation end products(AGEs). Methods GMC were incubated in culture medium containing AGEs in the presence of ICO and aminoguanidin for 48 hours. At the same time, the control and model groups were added. Then the cultured GMC were stained by mixed fluorescence liquid and observed under fluorescence microscope. Cell cycle of GMC were analyzed using flowcytometry. The content of MDA, activity of SOD and GSH-Px in GMC supernatant were measured. The level of ROS was detected using flowcytometry. Result Morphology analysis showed that the morphology and structure of normal GMC were normal. The structure of most cells in AGEs were unclear, cell counts increased largely and they grow intensively. Cell cycle analysis showed that cell percentage of S phase increased and G_0/G_1 reduced. The level of ROS, MDA remarkably increased, and SOD, GSH-Px activity reduced. When the ICO were added, cell morphology tended to be basically normal and cell counts decreased, the percentale of S phase also decreased. The level of ROS, MDA and the SOD, GSH-Px activity restored in comparison with the model groups. Conclusion ICO can prevent the cultured GMC from lesion caused by AGEs. ICO may protect GMC from AGES to retard the progression of diabetic nepropathy by partially inhibiting the occurrence of oxidative stress.
