Abstract: Objective To compare the diagnostic efficacy of the upgraded version of the GeneXpert automated fluorescent quantitative PCR system (GeneXpert MTB/RIF Ultra, GeneXpert Ultra) and the original version of the GeneXpert system (GeneXpert MTB/RIF, Xpert), real-time fluorescent quantitative nucleic acid detection (FQ-PCR), real-time fluorescent thermostatic amplification of Mycobacterium tuberculosis RNA (SAT-RNA), real-time fluorescent thermostatic amplification detection of DNA (thermostatic amplification method) and traditional BACTEC MGIT 960 liquid culture (culture method) for special specimens of tuberculosis, in order to analyze its application value in clinical detection. Methods Using prospective research methods, a total of 170 special specimens (including 47 pleural and ascites effusion samples, and 34 24-hour urinary sediment specimens, 49 tissue specimens and 40 fester specimens) were collected i'an Chest Hospital from January to September 2021. GeneXpert Ultra, Xpert, FQ-PCR, SAT-RNA, isothermal amplification, and traditional culture were used for detection. Clinical diagnosis was used as the standard, and sensitivity, specificity, positive predictive value, negative predictive value, coincidence rate, and Kappa value were compared among the methods. Results The sensitivities of GeneXpert Ultra, Xpert, FQ-PCR, SAT-RNA, isothermal amplification, and traditional culture were 65.18% (73/112), 49.11% (55/112), 37.50% (42/112), 19.64% (22/112), 8.04% (9/112), and 22.32% (25/112), respectively. The sensitivity of GeneXpert Ultra was higher than that of the other five methods, and the differences were statistically significant (χ2=66.25, 42.10, 28.89, 13.09, 4.92, 15.18, all P<0.05). GeneXpert Ultra result analysis showed that: 5.48%(4/73) cases had trace, that is, trace Mycobacterium tuberculosis load, 79.45% (58/73) cases were extremely low, 10.96% (8/73) cases were low, 2.74% (2/73) were medium, , and 1.36% (1/73) were high load. In 4 trace samples, the Xpert detection was negative for all. Of the 73 GeneXpert Ultra positive reports, 63 were rifampicin-sensitive, 6 were rifampicin-resistant, and 4 were rifampicin-resistant but of unclear resistance. Of the 55 Xpert positive reports, 45 were rifampicin-sensitive, 2 were rifampicin-resistant, and 8 were rifampicinresistant but of unclear resistance.. Conclusions The new generation of GeneXpert MTB/RIF Ultra has high sensitivity, specificity and drug resistance detection rate, and its advantage is even more apparent in the pathogenic diagnosis of special
specimens of tuberculosis. It can be used as one of the preferred methods in samples with low bacterial load.

OBJECTIVETo investigate the flavonoids in Rhododendron mariae.
METHODThe constituents in R. mariae were determined by UPLC/Q-TOF-MS. The chromatographic separation was performed on a C18 column (2.1 mm x 50 mm, 1.7 microm) with a gradient elufion of methanol-water containing 0.1% formic acid. The mass specfrometer eqaipped with elecfrospay ionizafion source was usedas defecfor and operated in data was collected under the positive and negative ion modes.
RESULTSeven constituents were identified as myricetin-3-O-beta-D-gluconside (1), myricetin-3'-O-beta-D-xylopyraoside (2), hyperoside (3), isoquercitrin (4), avicularin (5), quercitroside (6) and quercetin (7).
CONCLUSIONIn this study, the main flavonoids in R. mariae were separated by UPLC, and identified through the information of positive ion and negative ion and relative molecular mass which were determined by Q-TOF-MS. It is an accurate and effective method which can be applied for the constituent identification of R. mariae.

Objective To study the effect of age and sex on physiological and biochemical indexes of SJ5-SPF chickens at the age of different weeks. Methods Blood biochemical indexes of SJ5-SPF chickens at the age of different weeks were measured using an automatic blood analyzer, including 19 items: ALT, ALP, GGT, AST, LDH, TP, GLB, ALB, TB, DB, IBIL, GLU, TG, TCH, BUN, CRE, K+, Na+, and Ca2+. At the same time, the body temperature (T), respiratory frequency (R), heart rate (HR), diastolic pressure (DBP) and systolic pressure (SBP) of the SJ5-SPF chickens were measured using a RM6240C multi-channel physiological signal acquisition and processing system. Results (1) Among the physiological parameters, the body temperature, respiratory rate, heart rate, diastolic blood pressure and systolic blood pressure showed significant differences between the 4-week old chickens and the chickens at ages of 20, 25, and 25 weeks (P < 0. 05), but there were significant differences between the males and females at the same weeks of age except body temperature. The diastolic blood pressure was only significantly different in the 40-week old male and female chickens (P < 0. 01). The heart rates showed significant differences between the male and female chickens at 4, 20 and 25 weeks of age (P < 0. 01). The respiratory rate and systolic blood pressure in the 4-week old male and female chickens were significantly different (P < 0. 05). (2) The 19 blood biochemical indexes showed significant differences between the male and female chickens: the GGT, ALT, AST, TB, DB, IBIL, GLU, BUN and Na+at an week-age (P< 0. 05), the LDH, TP, GLB, ALB and CRE at two different week-ages (P< 0. 05), the ALP, TG and Ca2+at three different week-ages (P< 0. 05), and the TCH and K+at four different week-ages (P< 0. 05). Conclusions The above results can provide a scientific basis for diagnosis of SJ5-SPF chicken disease, animal quarantine and other related research.

Objective:To investigate the association of plasma EBV-DNA copy number, serum cytokines and B symptoms in patients with extranodal natural killer/T-cell lymphoma, nasal type (ENKTL), unravel the mechanism and assess the prognostic value of clinical indicators. Methods:Clinical data of 173 newly-diagnosed ENKTL patients (116 male, 57 female; median age: 43, 4 to 71 years)were retrospectively analyzed. According to Ann Arbor stage, 126 cases were classified as stage I-II and 47 cases of stage Ⅲ-IV. The primary sites of tumors included nasal cavity (n=100), extranasal upper aerodigestive tract (extranasal UADT, n=34), and extra-upper aerodigestive tract (extra-UADT, n=39). Prior to treatment, 91 patients had B symptoms and 82 cases of without B symptoms. According to plasma EBV-DNA copy levels, all patients were divided into the negative group (n=36), low load group (<10 4 copies/ml, n=73) and high load group (≥10 4 copies/ml, n=64). Serum cytokines including IFN-γ, IL-2, IL-4, IL-6, IL-10 and TNF-α were detected. Correlation analysis was performed by Cochran-Armitage trend test and Spearman correlation analysis. Survival analysis was conducted using univariate and multivariate Cox regression hazard analysis and survival curves were derived from Kaplan-Meier survival analysis. Results:The incidence of B symptoms and fever showed a significant upward trend with the increasing plasma EBV-DNA copy levels. In addition, serum levels of IFN-γ, IL-6 and IL-10 cytokines were higher in patients with B symptoms than those without B symptoms (all P<0.05). Serum IFN-γ, IL-6, and IL-10 levels were also positively correlated with plasma EBV-DNA copy number. The occurrence of B symptoms was associated with high-risk clinical features including advanced stage, primary tumor invasion, regional lymph node involvement, and elevated pre-treatment LDH. Survival analysis showed that stage, B symptoms, plasma EBV-DNA, and the above serum cytokines affected the prognosis of overall survival (OS) and progression-free survival (PFS) (all P<0.05). However, multivariate analysis showed that the occurrence of B symptoms was not an independent prognostic factor of ENKTL patients. Conclusion:This exploratory study suggests that the incidence of B symptoms is associated with increasing levels of EBV-DNA copies and cytokines, and these indicators are also important factors influencing the prognosis of ENKTL patients.
