1.Agroinfection of sweet potato by vacuum inifltration of an infectious sweepovirus

Sweepovirus is an important monopartite begomovirus that infects plants of the genus Ipomoea worldwide. Development of artiifcial infection methods for sweepovirus using agroinoculation is a highly efficient means of studying infectivity in sweet potato. Unlike other begomoviruses, it has proven difficult to infect sweet potato plants with sweepoviruses using infectious clones. A novel sweepovirus, called Sweet potato leaf curl virus-Jiangsu (SPLCV-JS), was recently identiifed in China. In addition, the infectivity of the SPLCV-JS clone has been demonstrated in Nicotiana benthamiana. Here we describe the agroinfection of the sweet potato cultivar Xushu 22 with the SPLCV-JS infectious clone using vacuum inifltration. Yellowing symptoms were observed in newly emerged leaves. Molecular analysis confirmed successful inoculation by the detection of viral DNA. A synergistic effect of SPLCV-JS and the heterologous betasatellite DNA-βof Tomato yellow leaf curl China virus isolate Y10 (TYLCCNV-Y10) on enhanced symptom severity and viral DNA accumulation was conifrmed. The development of a routine agroinoculation system in sweet potato with SPLCV-JS using vacuum inifltration should facilitate the molecular study of sweepovirus in this host and permit the evaluation of virus resistance of sweet potato plants in breeding programs.
2.Influence of viable myocardium in myocardial infract region on cardiac function in diabetic patients with acute myocardial infarction after PCI

Objective:To explore influence of viable myocardium in myocardial infract region on left ventricular remodeling and left cardiac function in diabetic patients with acute myocardial infarction (AMI) after percutaneous coronary intervention (PCI). Method: A total of 208 patients with type 2 diabetes mellitus (T2DM) complicated AMI after PCI underwent myocardial metabolic imaging by 18F- deoxyglucose position emission tomography (18F-FDG PET) and myocardial perfusion imaging by 99mTc-methoxy isobutyl isonitrile single photon emission computed tomography (99mTc-MIBI SPECT). According to whether there was viable myocardium in myocardial infarct region, patients were divided into viable myocardium group (n=115,perfusion didn’t match metabolism) and no viable myocardium group (n=93, perfusion matched metabolism). Indexes of echocardiography were measured in two groups before and after PCI. Influence of viable myocardium in myocardial infarct region on left ventricular remodeling and cardiac function was observed. Result: After myocardial infarction 12 months, compared with no viable myocardium group, there was significant increase in left ventricular ejection fraction (LVEF) [(44.1±7.12)% vs. (46.7±6.98)%] and significant decrease in left ventricular end-diastolic dimension (LVEDd) [ (55.46±4.75) mm vs. (53.17±4.77) mm], left atrial diameter [ (39.25±11.31) mm vs. (35.89±12.08) mm] in viable myocardium group, P<0.05 all. There were no significant difference in ratio of mitral diastolic peak flow velocity (E/A) in two groups after 12 months (P>0.05). Conclusion: In patients with type 2 diabetes mellitus complicated acute myocardial infarction, compared with no viable myocardium patients, LVEF significantly improves, LVEDd significantly decreases in patients with viable myocardium within myocardial infract region.

4.Study of cellular endocytic activity during ultrasound combined with microbubbles enhanced gene transduction of adeno-associated virus

Objective To explore the adeno-associated virus (AAV)gene transduction and cellular endocytosis mediated by ultrasound combined with microbubbles in two types of cells.Methods HeLa and NIH/3T3 cells were infected by rAAV2-EGFP at a concentration gradient to get the optimal concentrations for enhancement.At these concentrations,HeLa and NIH/3T3 cells were infected by rAAV2-EGFP mediated by ultrasound combined with microbubbles.The gene transduction efficiency were observed and measured by fluorescence microscopy and flow cytometry at 48 h after treatment.The cell viability was tested by CCK-8.The number and distribution of cellular clathrin-coated endocytic pits were observed by confocal fluorescence microscopy and transmission electron microscopy on 45 min after treatment.Results The optimal concentrations for HeLa and NIH/3T3 cells were 2000 v.g./cell and 10000 v.g./cell.Ultrasound combined with microbubbles significantly enhanced the transduction efficiency of rAAV2-EGFP (P <0.01) without significant cell viability decrease (P > 0.05 ).Confocal fluorescence microscopy and transmission electron microscopy demonstrated that clathrin-coated endocytic pits were more obviously increased in ultrasound combined with microbubbles mediated AAV transduction group than AAV transduction group. Conclusions Ultrasound combined with microbubbles can efficiently enhance the gene transduction of AAV,whose cellular transportation depends on cellular endocytosis,in two types of cells.Stimulating cellular endocytosis might be one of the mechanisms of enhanced cellular transportation of AAV mediated by ultrasound combined with microbubbles.
5.Dynamic contrast-enhanced MRI in serological negative early stage rheumatoid arthritis compared with osteoarthritis in hand

Objective To investigate the value of dynamic contrast-enhanced magnetic resonance imaging ( DCE-MRI) in differentiating serological negative early stage rheumatoid arthritis (RA) from osteoarthritis (OA)in hand. Methods Plain MRI, enhanced MRI of whole hand and DCE-MRI of single slice were performed in 53 patients suspected of RA or OA and 18 healthy volunteers. Twenty-three of them were diagnosed as RA,including 18 cases of serological negative RA and 18 of them were diagnosed as OA after 3 to 6 months follow-up. The contrast rate,slope, thickening of synovial membrane were measured on DCE-MRI in three groups and the MRI findings were also were detected in both RA and OA groups. The contrast rate and slope of synovial membrane were compared among three groups using rank sum test. The thickening of synovial membrane were compared among three groups using variance analysis. MRI signs of RA and OA group were evaluated with rank sum test Results The dynamic contrast rate of synovial membrane in RA group,OA group and control group was(100.78±61.96)%,(40.44±15.43)% and (23.56±9.14)%,respectively.Individually,RA group to OA group,u=3.101,P=0.002;RA group to control group,u=4.669,P=0.000;OA group to control group,u=3.482,P=0.000.The slope of contrast curve of synodal membrane of RA group,OA group and control group was 72.50°±13.34°,45.39°±9.94°,14.56°±5.75°,respectively.Individually,RA group to OA group,u=8.002,P=0.000;RA group to control group,u=17.102,P=0.000;OA group to control group,u=9.100,P=0.000.The synovial membrane thickening of RA group,OA group and control group was(3.3±0.5),(2.8±0.7)and (1.4±0.6)mm,respectively.Individually,RA group to OA group,q=2.622,P=0.011;RA group to control group,q=9.583,P=0.000;OA group to control group,q=6.961,P=0.000.Conclusion The quantitative index of DCE-MRI,including contrast rate and synovial membrane thickening may provide useful information for differentiating OA from semlogical negative early stage RA.The contrast rate and thickening of synovial membrane in RA group are higher than those in OA group.Many signs of MRI Can help differentiate OA from RA.
6.Effects of hyperbaric oxygen on brain derived neurotrophic factor and the structure of neurons after the cerebral ischemia-reperfusion in mice

Objective To explore the effects of hyperbaric oxygen(HBO) on brain derived neurotrophic factor(BDNF) and the changes of neuron structure after cerebral ischemia-reperfusion(CIR) in mice. Methods Twenty-seven mice were randomly and evenly divided into three groups : a HBO group, a control group and a sham-operation group (SO group). CIR models were established by clamping both carotid arteries in mice for 30 min,and then, untying clamps. 0.20 MPa HBO was applied once a day for 10 days after establishment of models in HBO group,but not in the control and SO groups. Following sacrifice after treatment and acquirement of cortex and hippocamal tissues, HE stain and immunohistochemistry technique were used to observe the changes of neuron structure and expression of BDNF. Results The neural degeneration and necrosis in cortex and hippocampus (issues) were observed under light microscope after CIR, and the amount of abnormal cells in the control group was more than that in HBO group (P
7.Sustaining Quality Improvement Used in Preventing Perineum Wound Infection

OBJECTIVE To discuss the function of sustaining quality improvement in preventing perineum wound infection.METHODS The perineum wound infection situation in 2003 was retrospectively analyzed,and since then the measures for sustaining quality improvement were implemented.RESULTS In 2004 and in 2005 the perineum wound infection percentage and the stitch inflammation rate were obviously reduced than in 2003.The difference was statisticaly significant.CONCLUSIONS Sustaining quality improvement is an unceasing,gradually advancing and long-term process.The prevention of perineum wound infection is a long time work for obstetrics workers.To carry out strictly the aseptic technical operation,grasp skilled suture technology,choose easily absorbable suture,maintain suitable cleanliness and nursing and use the antibiotic could be effectively to prevent the perineum wound infection and the stitch inflammation occurrence.
8.Using hyperbaric oxygen with mesenchymal stem cells to treat traumatic brain injury

Objective To investigate any therapeutic effect of combining hyperbaric oxygen with mesenchymal stem cells (MSCs) in treating traumatic brain injury (TBI).Methods Eighty healthy adult rats were randomly divided into a control group,a hyperbaric oxygen group,a stem cell group and a combination group,each of 20.TBI was introduced into the rats of all 4 groups.Twenty-four hours after the modelling,the hyperbaric oxygen group received hyperbaric oxygen therapy,the stem cell group received MSCs transplantation,the combination group was given the hyperbaric oxygen therapy an hour after the MSCs transplantation,while the control group was not given any treatment.All of the rats were evaluated using neurological severity scores (NSSs) after the modeling and again after the treatment.They were then sacrificed for HE staining and the expression of nuclear factor kappa B (NF-kB) and brain-derived neurotrophic factor (BDNF) were observed.Results On the 3rd,5th,10th and 20th day after the modeling,the average NSS of the combination group was significantly lower than those of the other three groups.However,the average NSS of the combination group on the 20th day was significantly superior to that on the 3rd and the 5th days.Compared with the control group,the edema of brain cells was less severe in the other 3 groups.The average expression of NF-kB and BDNF in the combination group was significantly higher than in the other 3 groups on the 3rd,5th,10th and 20th day after the modeling.Conclusion Hyperbaric oxygen,especially long term treatment combined with stem cell transplantation,can significantly improve nerve function in the brain after trauma,relieve inflammation and edema in and around the damaged area,and promote the expression of the NF-kB and BDNF.
9.Oxygen treatment on elderly patients with acute ischemic stroke

Objective To study the value of oxygen treatment on elderly patients with acute ischemic stroke.Methods From April 2010 to March 2012,a total of 140 cases of elderly patients with acute ischemic stroke for oxygen therapy combined with drug and rehabilitation were divided into two groups,outside high-flow oxygen group of 71 cases,hyperbaric oxygen group of 69 cases.The neurological functional defects scale was used to evaluate the effect.Two groups of patients were given conventional drug therapy including improving the microcirculation,vegetative neural drugs,and rehabilitation treatment.Results The total effectiveness respectively was 98.6% (70/71),91.3% (63/69),and there was significant difference (P < 0.05).After treatment,the nerve function damage grades of two groups respectively was (10.4 ± 1.7),(15.6 ± 1.8) scores,and there was significant difference compared with that before treatment [(28.2 ± 2.5),(28.7 ± 2.6) scores] (P < 0.05).There was significant difference in treatment adherence between two groups (P < 0.05).Conclusion For patients of elderly patients with acute ischemic stroke,oxygen therapy is necessary,while outside high-flow oxygen therapy is better than hyperbaric oxygen therapy.
10.Effect of CT scanning parameters on CT number

Objective To study the effects on tissue CT number caused by scan protocols.Methods The phantom was repeatedly scanned in different protocols by changing only one of parameters,such as X-ray tube voltage,mAs and recon kernel,while other parameters were ketp unchanged.The CT number of different materials in phantom were measured and analyzed.Results The CT numbers of tissues changed remarkably with the tube voltage and had different relativity for different tissues.The CT numbers had positive correlation with kV for such maierials as polyethyle,lexan,perspex,but for teflon the correlation was negative.The mAs and recon kernel had no effects on CT number.Conclusions The CT number of tissue changes with scanning X-ray tube voltage,so the setting of scan parameters should be taken into account in image diagnosis and radiotherapy.
